Biochemical Characterization Of Human Thy 1, Lynda F. Bonewald

The present studies were undertaken in order to a) verify the presence of a human Thy 1 antigen, b) demonstrate its similarity to murine Thy 1, and c) biochemically characterize the molecule. Previously, a 25,000 molecular weight molecule had been isolated from a human T lymphoblastoid cell line, Molt 3, using isolation procedures most frequently used for the isolation of rodent Thy 1. The human Thy 1 (p25) was demonstrated to be biochemically similar to murine Thy 1.2 by molecular weight and ability to bind Lens Culinaris lentil lectin. The Utilization Of Response Surface Techniques In Survival Experiments, Leonard Edward Buchner

Linear regression procedures are described which give an unbiased estimate of the percent functional viability of rat polymorphonuclear leukocytes (PMN's). These statistical techniques allow discrimination between variables which result in increased PMN survival after freezing. A review of current response surface techniques was made as they relate to optimizing the functional viability of PMN's after freezing. Experimental designs based on the steepest ascent and simplex methodologies were considered. Since the hill climbing efficiency of the steepest ascent and simplex methods are comparable, the steepest ascent procedure was selected as it is more readily adaptable to a batch mode analysis.

Some aspects of anuran metabolism are examined, with special emphasis on possible limitations to aerobic metabolism and the effect of chronic hypoxia acclimation on maximal rates of aerobic metabolism and the metabolic fate of lactic acid accumulated after anaerobic metabolism.

Effects Of The Serotonin Depletor Parachlorophenylalanine Upon Shock-Induced Aggression And Pressing Responses In Rats, Robert G. Sewell

The literature concerning the effects of d, l-parachlorophenylalanine (PCPA) upon shock-induced aggression (SIA) is reviewed and found to be inconsistent. PCPA, a known serotonin depletor, has behavioral effects in a variety of other procedures which collectively suggest that PCPA should produce SIA enhancement. The present study was designed to analyze PCPA (300 mg/kg) effects upon SIA in rats restrained spatially close to an inanimate target and panel operandum. The results showed marked increases in both aggressive biting and panel pressing for several days following each PCPA treatment for each subject tested. Strange (unfamiliar) males affect survival of progeny of reproductive female rodents in a variety of ways. In mice, strange males block pregnancy before implantation of embryos (Bruce, 1959) but only in females that are not lactating (Bruce and Parkes, 1961). In voles these males can block pregnancy both before and after implantation (Stehn and Richmond, 1975; Marks and Schadler, 1979; Schadler, 1981). Strange males have also been shown to kill nursing pups in lemmings (Mallory and Brooks, 1978) and mice (Labov, 1980 and vom Saal and Howard, 1982).

Antibody raised against the purified human erythrocyte glucose transporter specifically precipitated four proteins from normal and Rous sarcoma virus-transformed chicken embryo cells: a major protein of Mr 41,000 and minor proteins of Mr 68,000, 73,000, and 82,000. The Mr 41,000 and 82,000 proteins were found only in a membrane fraction, not in the soluble fraction, and displayed a heterogeneous mobility on NaDodSO4/polyacrylamide gel electrophoresis, suggesting glycosylation. The Mr 41,000 and 82,000 proteins were increased in amount after malignant transformation in direct proportion to the increase in hexose transport rate, and the increase was dependent on the expression of the src …

Fifty-seven women over the age of 16, seeking care at a rural health center, completed questionnaires concerning demographic factors and health beliefs and attitudes. Correlation coefficients and Z scores were computed for the variables. There was significant positive correlation in both demographic and health belief and attitude areas. Discussed also are factors possibly contributing to the test results, as well as alternative research models.

A fortunate coincidence, discovered recently, has provided annual ryegrass toxicity researchers with valuable knowledge which may help them understand how the ARGT toxins affect animals, and further their investigations into vaccine development. A toxic antibiotic, identified years ago, has proved to be closely related to the ARGT toxins. Antibiotics are generally regarded as effective agents for the control of many bacterial and fungal infections. However, some of them are toxic to both the bacteria and the host, whether man or animal.

T Cell Growth Factor Receptors. Quantitation, Specificity, And Biological Relevance, Richard J. Robb, Allen Munck, Kendall A. Smith

To examine directly the hypothesis that T cell growth factor (TCGF) interacts with target cells in a fashion similar to polypeptide hormones, the binding of radiolabeled TCGF to various cell populations was investigated. The results indicate that TCGF interacts with activated T cells via a receptor through which it initiates the T cell proliferative response. Internally radiolabeled TCGF, prepared from a human T leukemia cell line and purified by gel filtration and isoelectric focusing, retained biological activity and was uniform with respect to size and charge. A Field Investigation Of Meiofaunal Dispersal: Tidal Resuspension And Implications, Susan S. Bell, K. M. Sherman

Presence of meiofauna in resuspended sediments was investigated in a tidal creek system employing a specially designed core sampler and quick freezing techniques. Forty core samples were taken during 4 successive ebb tides to determine the distribution of meiofauna in (a) overlying water; (b) resuspended sediment; and (c) undisturbed sediment fractions of each sample. Our experiments demonstrate that 13-44 % of the meiobenthic copepod subcommunity was present in overlying and resuspended sediment fractions, while only a small percentage of nematodes was found simultaneously in these layers. Transformation of T lymphocytes, induced by treatment with periodate or with neuraminidase plus galactose oxidase, requires the participation of accessory cells. Procedures were developed for the fractionation of rat lymph node cells, by which most of the lymphocytes can be recovered as a major population of cells that do not respond to mitogenic stimulation unless accessory cells from a separated minor population are added. Further purification led to a 1000-fold overall increase in accessory activity per cell, with a 50-70% yield. The purest preparations were virtually free of macrophages and contained more than 90% typical dendritic cells. Maximum responses occurred …

These studies explore the distribution of blood flow among small samples of placental tissue. Labeled microspheres (14 ± 1 pm) were Injected Into the left ventricle of six un-anesthetized ewes and Into the jugular vein of the fetus In utero. A total of 3,576 samples weighing 31. +11. (S.D.) mg were taken from seventeen cotyledons and counted for flow labels. Maternal and fetal flow within the cotyledons were both normally distributed with standard deviations of 44%. The maternal/ fetal flow ratio was less than 0.5 In 9% of the cotyledon, between 0.5 and 1.5 In 70%, and greater than 1.5 …

Consecutive medical records of 10,622 births were viewed, January, 1962, through December, 1972, from McAllen General Hospital, McAllen, Texas. The overall incidence of birth defects was consistent with rates for the United States. Birth defects detected in Spanish surnamed were significantly higher than for non-Spanish surnamed. Spanish surnamed bore eight times more defects of the central nervous system than non Spanish surnamed. Males had significantly more defects than females, and had almost seven times more defects of the urogenital system. The most distinctive feature of this survey was the high incidence of atelectasis. It has been previously demonstrated that 7,12-dimethylbenz(a) anthracene (DMBA) is an initiator of ovarian granulosa cell tumors. It is also known that DMBA leads to destruction of primordial oocytes, and that tumor induction may involve the interaction of pituitary gonadotropins. However, the effects of DMBA on ovarian steroidogenesis and uterine growth in relation to the tumorigenic process have not been investigated. Therefore, the present study was conducted to examine these parameters using adult C57Bl/6N mice treated in proestrus and diestrus II with 0.1 ml corn oil with or without 1.5 mg DMBA and killed 44 hours later. Plasma samples were obtained from seven hypertriglyceridemic (>150 mg/dl) chronic renal failure patients before and after regular hemodialysis. The effect of the plasma on the lipolytic activity of bovine milk lipoprotein lipase was investigated. The lipoprotein lipase activity, as measured by release of C¹⁴-labeled free fatty acid from triglyceride, was decreased 8-67% when incubated with uremic plasma as compared to the control plasma (44 mg/dl triglyceride). The inhibition noted prior to hemodialysis did not appear to be alleviated by the dialysis but subsequent treatment with an isolated ultrafiltration technique did appear to remove an inhibitory substance as …

A study was undertaken to determine the concentration at which a sealer reaches a point of nontoxicity. An in vitro model utilizing HeLa cells and human fibroblasts was used to observe the effect of various concentrations of eight freshly mixed and eight set root canal sealers. Evaluation of toxicity was determined by cell survival which was measured by incorporation of ³H-thymidine into the nucleic acid components of each cell type used. Each dilution tested was allowed direct contact with the cells for 24 hours before toxicity evaluation. The results indicate that all sealers at concentrations presently used are cytotoxic …

Fifty-six semen samples from males under examination for suspected subfertility were examined to determine if alterations in specific semen parameters could be associated with the presence or absence or Ureaplasma urealvticum. Twenty-seven (48.2%) of the specimens cultured positive for U· urealyticum in U9B urease broth and A-7 differential agar. Both positive and negative specimens have a normal average motility rating. Overall sperm counts (millions per ml) were slightly lower and ejaculate volumes higher in U. urealvticum positive samples. Morphological abnormalicies were not found more frequently in U. urealyticum positive samples. No statistically significant difference between seminal parameters of U. …

A systematic study of the degradation of physiological concentrations of ¹²⁵I-labelled insulin was performed in intact fat-pads, isolated adipocytes and subcellular fractions of isolated adipocytes. The findings indicate that insulin is rapidly degraded to low-molecular-weight peptides and/or amino acids by the intact tissue and isolated cells. Of the total insulin-degradation products present after incubation with an intact fat-pad, 94% is recovered in the medium, indicating that these products are not retained by the cells or tissue. The plasma membranes do not degrade insulin significantly in the absence of reduced glutathione, and over 99% of the cellular degradative capacity is …

The erythropoietin (Ep) dose response of erythroid colony-forming units (CFU-e) and erythroid burst-forming units (BFU-e) from rabbit bone were similar to murine erythroid precursor Ep requirements. Bone marrow and peripheral blood contained a continuum of erythroid precursors at differing stages of maturation. The earliest BFU-e were assayed on day 10. CFU-e were observed in bone marrow, but not in peripheral blood. The most mature erythroid precursors observed in peripheral blood were intermediate BFU-e, assayed on day 5. Speculation: In the fetal lamb during prolonged intrauterine hypoxia, total and regional cerebral blood flows increase to the same extent without evidence of preferential shunting to critical brainstem or subcortical areas. Neuropathologic studies have indicated relative sparing of these areas during similar animal experimental or human neonatal conditions. This suggests that the pattern of hypoxic ischemic insult to the neonatal central nervous system associated with asphyxia may differ from that produced by hypoxia alone. In addition, during asphyxia these pathologic changes may result primarily from hypotension and decreased regional cerebral blood flow, or from regional metabolic derangements …
