A previous report indicated that prototype chlorovirus PBCV-1 replicated in two Chlorella variabilis algal strains, NC64A and Syngen 2-3, that are ex-endosymbionts isolated from the protozoan Paramecium bursaria. Surprisingly, plaque-forming viruses on Syngen 2-3 lawns were often higher than on NC64A lawns from indigenous water samples. These differences led to the discovery of viruses that exclusively replicate in Syngen 2-3 cells, named Only Syngen (OSy) viruses. OSy-NE5, the prototype virus for the proposed new species, had a linear dsDNA genome of 327 kb with 44-nucleotide-long, incompletely base-paired, covalently closed hairpin ends. Each hairpin structure was followed by an identical …

Doxorubicin-Conjugated Dexamethasone Induced Mcf-7 Apoptosis Without Entering The Nucleus And Able To Overcome Mdr-1-Induced Resistance, Kamontip Chaikomon
Chulalongkorn University Theses and Dissertations (Chula ETD)
Doxorubicin is a common chemotherapeutic drug that generally uses in many solid and hematologic malignancies. The main mechanism of doxorubicin is inhibition of topoisomerase II that occurs in the nucleus. Currently, doxorubicin resistant cancer cells which have decreased accumulation of intracellular doxorubicin resulting from an efflux pump, P-glycoprotein encoded by multidrug resistant gene. Several studies reported many different ways to overcome doxorubicin resistance. In this study, we conjugated 3'amino group of doxorubicin to dexamethasone molecule. Despite of lower cytotoxic activity in MCF-7 cells, the conjugated product, DexDOX, exerted its actions in the different fashion to doxorubicin. DexDOX rapidly induced MCF-7 …

Effects Of Estrogens On Astrocytes Exposed To Stressors, Bridget N. Smith
Theses and Dissertations
Hormone replacement therapy (HRT), used by many women to alleviate menopausal symptoms such as hot flashes and mood swings, is often a combination of horomones such as estrogens, progesterone, and conjugated equine estrogens (CEE), extracted from the urine of pregnant mares. Previous studies have found positive correlations between estradiol and cellular protection, but recent research has concluded CEE provide less protective mechanisms as compared to endogenous hormones. This research sought to compare the effects of estrogen treatments (single and combined estrogen) on viability when astrocytes were induced with stressors (epinephrine, cortisol, and low oxygen concentration). Cultured human astrocytes were treated …

Is Gene Therapy A Viable Option For Cancer Treatment?, Aliza Applebaum
The Science Journal of the Lander College of Arts and Sciences
The use of gene therapy as a medical treatment option was first introduced to the world in 1990, when a four-year-old girl became its first patient. Since then gene therapy has met great success but also severe drawback. Incidences with severely negative outcomes on patients gave gene therapy a bad name and many began skeptical towards its use, but the constant work and progress on the safety and effectiveness of gene therapy is making it a more viable route of treatment. This paper focuses on gene therapy as a form of cancer treatment. Viral insertion of the modified genetic material …

Theses and Dissertations--Toxicology and Cancer Biology
Lung cancer is a particularly devastating disease, accounting for the most deaths among all cancer types in the United States. Despite a reduction in the country’s smoking rates, cigarette smoking remains the number one risk factor for lung cancer. Additionally arsenic exposure, which occurs primarily through contaminated drinking water in the U.S., is associated with increased lung cancer incidence. The nucleotide excision repair (NER) pathway is critical for maintenance of genomic fidelity, removing DNA lesions that could otherwise promote DNA mutations and drive carcinogenesis. Tobacco smoking introduces significant amounts of DNA damage and produces characteristic DNA mutations found in lung …

Turkish Journal of Medical Sciences
Background/aim: Psoriasis is a chronic skin disease. To reduce side effects associated with current treatment modalities, new treatment methods are required. Some clinicians have begun to use microphototherapy to treat psoriatic patients with a limited number of lesions (lesions affecting <10% of the body surface area). Materials and methods: Microphototherapy is indicated in patients whose lesions persist despite systemic and topical treatment or when such treatment is contraindicated. Our study aimed to evaluate the efficacy of microphototherapy in psoriasis patients with localized lesions. Patients in this unblinded and prospective study were treated three times a week for 50 sessions using a MedLight CupCUBE Grimed microphototherapy device that emits an ultraviolet B (UVB) spectrum. Results: Forty-five lesions in 23 psoriatic patients were treated. Treatment outcome was evaluated based on the Psoriasis Severity Index (PSI). PSI scores decreased significantly during the treatment. Conclusion: The MedLight CupCUBE Grimed microphototherapy device was effective for the treatment of psoriasis patients with localized lesions. The device can administer UVB radiation to involved regions while protecting uninvolved regions and minimizing radiation dose. However, having to be administered by experienced technicians and the long time required for each treatment session are disadvantages of the technique.

Turkish Journal of Medical Sciences
Background/aim: In this study, we evaluated Thymus revolutus Célak essential oil and its two main constituents, γ-terpinene and p-cymene, as potential oxidative agents against lung cancer and epidermoid carcinoma cells. Materials and methods: Cell viability assessment was made by CellTiter-Blue® cell viability and 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) assay after treatment with 5?600 ?g/mL concentrations of essential oil, γ-terpinene, and p-cymene. Malondialdehyde and 8-hydroxy-2'-deoxyguanosine levels in parental H1299, epirubicin-resistant (drug-resistant) H1299, A549, and A431 cells were also assayed after essential oil, γ-terpinene, and p-cymene had been administered for 24 h. Glutathione S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GRx), and glucose …
