The Role Of Outer Inflammatory Protein A (Oipa) In Vaccination Of Thec57bl/6 Mouse Model Infected By Helicobacter Pylori, Mohaddese Mahboubi, Tahereh Falsafi, Majid Sadeghizadeh, Fatemeh Mahjoub
Turkish Journal of Medical Sciences
Background/aim: Outer inflammatory protein A (OipA) is an important adhesin of Helicobacter pylori. Our goal was to assess the role of OipA in protection of C57BL/6 mice against H. pylori. Materials and methods: C57BL/6 mice were mucosally immunized with recombinant OipA protein, OipA + propolis, propolis, and phosphate-buffered saline. After vaccination, anti-OipA IgA was measured. Mice were challenged three times with 5 x 107 CFU of the H. pylori B19 strain. Two weeks later, bacterial colonization and inflammation in the stomach was analyzed using standard methods. Results: The CFU number in the OipA group was significantly (P < 0.05) lower than that of the control. The CFU number in the OipA + propolis group was higher than those of the OipA and propolis groups. IgA titers were significantly higher (P < 0.0001) in the OipA group compared to the control and OipA + propolis groups. Propolis did not play an adjuvant effect but it interfered with the efficient vaccine effect of OipA. Conclusion: Results show the effect of vaccination by OipA in protection of the mouse model and the importance of OipA in H. pylori pathogenesis. OipA may be proposed as a suitable oral vaccine candidate against H. pylori infection; however, further study is required to determine adjuvant or adverse effects of propolis toward OipA.

Background/aim: The canonical Wingless-type (WNT) pathway is involved in normal hematopoietic cell development and deregulated WNT signaling is implicated in the development of hematological malignancies. Dickkopf 1 (DKK1) acts as a modulator of the β-catenin regulated canonical pathway. Activation of DKK1 leads to apoptosis and growth suppression, whereas silencing by promoter hypermethylation results in abnormal WNT activation. The secreted inhibitor Dickkopf can antagonize WNT signaling by competitively binding to low density lipoprotein receptors (LRPs) 5 and 6. Materials and methods: We studied DKK1 gene promoter methylation and investigated DKK1, β-catenin, LRP5, and LRP6 mRNA levels in B-cell acute lymphoblastic leukemia …

Background/aim: An aneurysm is a pathological enlargement of an artery characterized by the thinning of the elastic fiber layer in the tunica media. Because the aneuritic artery wall is weakened, these vessels can rupture and cause serious bleeding. Surgical methods are often used for the treatment of aneurysms. However, cell-based therapies are less invasive and potentially safer alternatives. In this study, the therapeutic efficacy of rat adipose tissue-derived mesenchymal stem cells (MSCs) was investigated in a new carotid artery aneurysm model.Materials and methods: Arteries were pretreated with elastase to create aneurysms. Gelatin matrices containing MSCs were applied to the outer …

Background/aim: Erythropoietin (EPO) has been proven recently to be a critical mediator in retinal neovascularization (RNV). Previous studies have indicated that the use of recombinant human EPO (rEPO) is a high risk factor in the development of retinopathy of prematurity. In this study, we aimed to investigate the effect of rEPO administration on RNV and its underlying mechanism in a mouse model of oxygen-induced retinopathy (OIR). Materials and methods: A murine model of OIR was used to generate RNV. After daily intraperitoneal injection of rEPO from postnatal day 12 (P12), mice were euthanized at P17. Whole-mount retina staining was used …

Background/aim: The pathogenesis of Raynaud's phenomenon (RP) has not yet been fully elucidated. RP is characterized by exaggerated cold-induced vasoconstriction. Urotensin II (UII) is a potent vasoconstrictor. The aim of the present study was to evaluate plasma UII levels in both primary RP and secondary RP associated with systemic sclerosis (SSc).Materials and methods: Fifteen patients with primary RP, 30 patients with RP secondary to SSc, and 30 healthy controls (HC) were included in the study. Raynaud condition scores (RCS) were determined in the primary RP and SSc groups. Modified Rodnan skin score (MRSS) was determined for the SSc patients. Plasma …

Background/aim: Nasal polyposis is a chronic inflammatory disease affecting the paranasal sinuses and nasal mucosae. It is thought that genetic and molecular mechanisms in inflammatory and apoptotic pathways are the main factors in the etiopathogenesis of nasal polyposis. The aim of this study was to investigate the expression patterns of CD11b, galectin-1, beclin-1, and caspase-3 in nasal polyps.Materials and methods: The mRNA expression levels of CD11b, galectin-1, beclin-1, and caspase-3 protein and western blot analysis of caspase-3 protein were evaluated in inferior turbinate mucosae and nasal polyp tissues. Results: CD11b expression was markedly higher in nasal polyp tissues when compared to …

Background/aim: The purpose of this study was to evaluate the concordance of immunohistochemical (IHC) parameters of breast lesions between the core needle biopsy (CNB) and the surgical resection specimen. Materials and methods: CNB and resection specimens of female patients were retrospectively analyzed. ER, PR, HER-2, and Ki-67 parameters were compared for each patient. A total of 284 cases were assessed. Forty-one and 48 cases were excluded from the HER-2 and Ki-67 examinations, respectively, because the CNBs did not allow for IHC. Results: Concordance rates were 93.3% for ER, 89.4% for PR, 90.1% for HER-2, and 80.9% for Ki-67.Conclusion: CNB is …
