The Psychological, Social, And Bioinformatic Considerations For The Return Of Continental Genetic Ancestry Estimates And Other Secondary Sequencing Results, Jaimie Leighan Richards
The Psychological, Social, And Bioinformatic Considerations For The Return Of Continental Genetic Ancestry Estimates And Other Secondary Sequencing Results, Jaimie Leighan Richards
All ETDs from UAB
With recent advances in sequencing technologies, large scale genome wide sequencing has advanced our understanding of disease risk and etiology. These tests, however, often provide secondary data beyond the scope of the original sequencing objectives, and our understanding of how and if participants want to receive these secondary results is important as clinical and research-based sequencing becomes commonplace. To address this, we completed a systematic review of quantitative research that measures attitudes and preferences on the return of genomic sequencing results. Based on that review, we studied the return of genetic ancestry since these results, popularized in direct-to-consumer sequencing companies …

Characterization Of Cucurbitacin-Inspired Estrone Analogues As Novel Inhibitors Of Human Atp- Binding Cassette Proteins (Abcb1 And Abcc1), Jennifer Kyeremateng
Electronic Theses and Dissertations
ATP-binding cassette (ABC) transporters are a large class of integral membrane proteins that contribute to key physiological functions in all organisms by utilizing ATP binding and hydrolysis to transport diverse substrates across membrane barriers. P-glycoprotein (P-gp/ ABCB1) and Multidrug Resistance protein 1 (MRP1/ABCC1) are widely reported ABC transporters associated with multidrug resistance in cancer. Multidrug resistance (MDR) mediated by P-gp and MRP1 is responsible for treatment failures of many metastatic cancers as a result of reduced accumulation, bioavailability and diminished potency of anticancer drugs. Currently, known P-gp and MRP1 inhibitors are limited due to toxicity, lack of selectivity and low …

Integrative Genome Assembly Of Staphylococcus Epidermidis And Staphylococcus Hominis Strains, Cana L. Brown
Integrative Genome Assembly Of Staphylococcus Epidermidis And Staphylococcus Hominis Strains, Cana L. Brown
Honors Theses
Due to rapid advances in sequencing technology, it is becoming increasingly easier to assemble unknown genomes from millions of short sequencing reads of nucleotides taken from the full genomic sequence (Hernandez, 2008). In this study, we used various computational programs to align reads from unsequenced strains of the bacteria Staphylococcus epidermidis and Staphylococcus hominis into a hitherto undefined, single contiguous genomic sequence. We used SPAdes to assist with template free assembly (Bankevich, 2012), BLAST to identify a suitable reference genome from closely related species (Madden, 2013), Bowtie2 to align our reads to the reference genome, SAMtools to sort and organize …

A Novel Role For Neurotensin In Regulation Of Stem Cell Function In The Small Intestine, Stephanie Rock
Theses and Dissertations--Toxicology and Cancer Biology
Neurotensin (NT) is a nutrient-regulated gut hormone that plays important roles in lipid absorption, obesity, metabolic disorders, and normal and neoplastic growth in the intestine. In this study, we 1) elucidate the mechanisms regulating NT release from endocrine cells, 2) examine the role of NT on proliferation and stem cell function in the small intestine and 3) define the effects of NT on colorectal cancer stem cells. We report that NT release from endocrine cells is enhanced by the MAPK scaffold protein Kinase Suppressor of Ras 1 (KSR1) and the exocyst complex component 70 (Exo70). Moreover, free fatty acid stimulated …

Background: Food insecurity is a global problem that requires a One Health approach. As many households in low- and middle-income nations rely on crops and livestock that they produce to meet their household’s needs, food security and nutrition are closely linked to the health of animals and the environment. Resources controlled by women are more often allocated to uses that benefit the entire household, such as food, health, and educating children, than men’s resources. However, studies of gender control of resources among pastoralist societies are scant. We examined the effect of female and male control of livestock resources on food …

Factors Contributing To Diminished Long-Term Outcomes Of Secondary Infections, Alia Tayara
Honors Theses
It is widely known that patients suffer diminished long-term outcomes in the years following secondary infections, infectious diseases that occur during or after treatment of a primary infection such as influenza or COVID-19. One common secondary infection is pneumonia, which has become the most fatal hospital-acquired infection. The bacterium responsible for pneumonia is known as Pseudomonas aeruginosa. The cytotoxic supernatant in these studies was generated from a PA103 strain of Psuedomonas aeruginosa with a type III secretion system that maintains the ability to transfer exoenzymes ExoU and ExoT into target cells during infection. The focus of the following studies was …
Assessment Of Hypothermia As A Correlate Of Pde4 Inhibitor-Induced Nausea In Mice, Marianna Susan Oditt
Assessment Of Hypothermia As A Correlate Of Pde4 Inhibitor-Induced Nausea In Mice, Marianna Susan Oditt
Honors Theses
Type-4-cAMP-phosphodiesterases (PDE4s) are a group of isoenzymes that hydrolyze the second messenger cyclic adenosine monophosphate (cAMP). PDE4 inhibitors have many therapeutic uses including reducing inflammation, however, they also have side effects like nausea and emesis, which have diminished the clinical utility of PDE4 inhibitors. When administered to mice, PDE4 inhibitors have been shown to cause hypothermia. Given that nausea and emesis in humans are also associated hypothermia, this project served to determine whether PDE4 inhibitor-induced hypothermia may represent a correlate of nausea and/or emesis in mice. To this end, the current literature was screened for articles that explore nausea, emesis …

Mimicking The Effect Of Prolactin On Stat3/Stat5 Activity In Breast Cancer, Jennifer N. Barbuto, Sarah R. Walker
Honors Theses and Capstones
Signal transducers and activators of transcription (STAT) 3 and 5 are commonly constitutively activated in breast cancer. STAT5 can outcompete STAT3 and reduce cell proliferation and metastasis. STAT5 activation is stimulated by prolactin, a natural hormone that can be harmful at high levels. The aim of this study is to identify some possible previously developed drugs that mimic the effect of prolactin and STAT5 without the added risk in MDA-MB231 breast cancer cells. Using the CLUE database query app and STAT5 up- and downregulation signatures, three drugs (X, K, and M) were chosen based on their similarity in signatures to …
Viability Of Physiologically Timed Relaxation Interventions In Children With Asd, Nicholas Mendez
Viability Of Physiologically Timed Relaxation Interventions In Children With Asd, Nicholas Mendez
CMC Senior Theses
Autism Spectrum Disorder (ASD) is an increasingly common developmental disorder that changes how people experience the world and affects individuals’ social interactions and often leads to many adverse behaviors. Current literature dictates that a primary contributor to these adverse behaviors is that those with ASD have difficulty determining their own emotional states and determining the physiological signals that their body sends them. A study by Dr. Sarabadani et al. determined that it was possible to monitor the physiology of an individual with ASD and correlate certain signals to emotions, such as stress. These findings indicate the feasibility of establishing a …

Downregulation Of Mir-34a Via Calorie Restriction Regulates Lipid Metabolism In The Liver Of Long-Living Ames Dwarf Mice, Sarah Ashiqueali
Electronic Theses and Dissertations, 2020-2023
Ames dwarf mice (df/df) display delayed aging relative to their normal siblings, living approximately 40-60% longer. As such, investigating the mechanisms that enable these organisms to have superior life expectancies may prove useful in developing therapies to slow aging and deter age-related disease. Nonalcoholic fatty liver disease (NAFLD) is a condition that is characterized by the accumulation of excess adipose tissue in the liver. Interestingly, its prevalence tends to increase with age with an incidence rate of greater than 40% in individuals over the age of 60. Previous studies highlight the potential of calorie restriction (CR) in promoting longevity, but …

Turkish Journal of Medical Sciences
Background/aim: Treatment of hepatic hemangiomas is a controversial topic, and traditional treatment is surgical excision. Transcatheter arterial embolisation (TAE) and transcatheter arterial chemoembolisation (TACE) have been reported as minimally invasive treatment methods. To the best of our knowledge, there are no studies comparing use of TACE and TAE for hepatic hemangioma treatment. The aim of the study is to compare symptom resolution, size reducing effects, and complications of TACE and TAE for the treatment of giant hepatic hemangiomas. Materials and methods: A total of 104 patients underwent TACE using bleomycin, and 108 patients underwent TAE. The patients were followed-up for …

Background/aim: To evaluate benign and malignant cutaneous-subcutaneous lesions using real-time strain elastography (RTSE) and to compare the findings with histopathologic results. Materials and methods: Over a period of 10 months, 72 patients (38 with benign and 34 with malignant cutaneous and subcutaneous lesions) were prospectively included in this study. Elasticity patterns and strain ratios were examined for each lesion. Lesions were evaluated in 4 groups as yellow-red (soft; pattern-1), green-yellow (moderate; pattern-2), blue-green (hard; pattern-3) and blue (hardest; pattern-4). The stiffness of the lesions was displayed with strain ratios by comparing of a nearby reference tissue. The recorded images were …

Background/aim: This study aimed to evaluate the regression pattern with the distal intramural spread (DIS) of rectum cancer after preoperative chemoradiation. Materials and methods: Specimens from 56 patients who underwent radical resection after preoperative chemoradiation for rectal cancer were examined. The regression pattern (total, fragmented) of the tumor was recorded. DIS status was evaluated by creating sections 0.2 to 0.3 cm thick. Results: A single macroscopic residual area was detected in all specimens. In 10 patients (17.8 %), pathologically complete responses were identified, and DIS was detected in 33 patients (58.9%). The average DIS distance was 0.56 ± 0.3 cm …

Background/aim: To investigate the correlation between depressive-anxiety symptoms, mixed features, disease activity, and functional status in patients with rheumatoid arthritis (RA) in the light of the shared underlying etiology in both disorders. Materials and methods: The study included 556 patients with RA. RA disease activity was measured using the Disease Activity Score 28-joint count C reactive protein (DAS28-CRP), and the patients were evaluated by a Health Assessment Questionnaire (HAQ). The Hospital Anxiety and Depression Scale (HADS), Mood Disorder Questionnaire (MDQ), and Modified Hypomania Checklist (mHCL) were used to evaluate the mixed depression and bipolarity status of the patients. Results: Of …
