Unsuccessful cadaveric kidney transplantation still remains as an important problem in organ transplantation. Cold storage solutions are not very efficient for kidney preservation. Vitamin E is an important dietary antioxidant, which may play a vital role in preventing free radical perfusion injury. We investigated the effect of vitamin E on kidney preservation with Euro-Collins (EC) solution using the isolated perfused dog kidney model. The sulcus angle of the femoral trochlea is particularly important for evaluating the patellofemoral joint. Our experimental study aimed to assess the osseous femoral sulcus angle radiologically in the right and left knees and osteologically in the right and left femurs in males and females. The osseous sulcus angle of 28 male right, 22 male left, 21 female right and 29 female left knees was measured on magnetic resonance images (MRIs) and by a goniometer; the angle was measured in 40 right and 40 left femurs. Recovery Of 1-Chloro-2,4-Dinitrobenzene Detoxification By N-Acetyl- L-Cysteine In Glutathione Predepleted Human Erythrocytes, Deni̇z Yildiz, C.Işil Kuran

Turkish Journal of Medical Sciences

Glutathione is an important thiol-containing compound involved in the detoxification process in erythrocytes. Its thiol group reacts with a variety of xenobiotics in a glutathione S-transferase catalyzed reaction to form conjugates that are effluxed from the erythrocytes by an ATP dependent transport mechanism. A well studied experimental system is the transport of the conjugate of glutathione and 1-chloro-2,4-dinitrobenzene. We investigated whether N-acetyl-L-cysteine protects the free-SH content and restores 1-chloro-2,4-dinitrobenzene detoxification in erythrocytes or replaces glutathione in detoxification process in glutathione predepleted erythrocytes. This study was conducted to determine brucellosis seropositivity in patients suspected of having brucellosis who sought treatment at the Central Laboratory of Dicle University Medical Faculty Hospital in southeast Turkey (Diyarbakır). 20,663 serum samples were collected during the study (1 August 2001-31 December 2002), and 14,480 sera were tested in a 12-month period on a seasonal basis by Rose-Bengal slide agglutination (RBSA), and positive sera were titrated by standard tube agglutination (STA). Titration values of 1/160 and above were considered positive. Of the 20663 sera, 463 (2.2%) tested positive on RBSA. Of these 463 sera, 267 (57.6%) tested positive on …

Norfloxacin is a poorly water soluble drug, and to improve its solubility it was complexed with b-cyclodextrin (BCD). Several ocular patches/inserts of norfloxacin-b-cyclodextrin were prepared in hydroxypropyl methyl cellulose (HPMC) matrix. The influence of rate controlling membranes made of ethyl cellulose (EC) alone and in combination with polyvinyl pyrrolidone K30 (PVP K30) in different proportions on drug release kinetics was studied. The aim of this study was to examine the effects of weight loss induced by pharmacotherapy combined with a hypocaloric diet for a period of 8-weeks on maximal work (Wmax) production capacity and aerobic fitness in obese patients. Twelve obese females were put on an integrated energy restricted diet with orlistat supplement 3 x 120 mg/day. Genetic biomonitoring of human populations exposed to potential mutagens/carcinogens can be performed using different genetic markers. Sister chromatid exchange (SCE) is one of the most extensively used markers of the early biological effects of DNA damaging agents. In order to assess the genotoxicity associated with exposure to cotton dust, we determined SCE frequency in peripheral blood lymphocytes cultured from 20 cotton gin workers and 20 controls. Student’s-t test indicated an increased frequency of SCE in lymphocytes of the workers (14.66 ± 4.18) compared to the controls (10.40 ± 1.37) (P < 0.0001). The Mann-Whitney U test indicated that the duration of exposure to cotton dust (10 years) and the work area were not related to the frequency of SCE (P > 0.05). In this study, the prevalence of Listeria spp. was investigated in the ice cream samples collected from the downtown stores of Kahramanmaras (28 samples) and Adana (30 samples). A total of 58 ice cream samples were analysed by the Food and Drug Administration (FDA) two-stage enrichment techniques. Growth of Listeria spp. was determined in 14 (24.1%) of the samples collected from Kahramanmaras and 10 (17.2%) samples from Adana. The results of the biochemical tests revealed that the bacterial growth in the samples from Kahramanmaras was Listeria grayi whereas 8 (13.8%) samples of Adana were Listeria grayi, the other two samples …

This work was carried out to study the effect of asthma education on asthma knowledge, behavior and morbidity in asthmatic patients. A randomized double blind controlled study was conducted with 54 adults with asthma followed-up at the outpatient clinic of Chest Diseases and Tuberculosis at the Faculty of Medicine of Suleyman Demirel University. We aimed to assess chromosomal aberration frequency by means of FISH chromosome painting and conventional Giemsa staining in peripheral blood lymphocytes of workers at a nuclear waste repository. The results obtained from the exposed group were compared with those from a control group from the administrative staff of the company. Data on health and social status were collected using a questionnaire. Conventional chromosomal analysis after Giemsa staining did not indicate any significant difference between radiation exposed and control workers. LP-BM5 retrovirus-infected C57BL/6 mice develop splenomegaly, lymphadenopathy, hypergammaglobulinemia, and immunodeficiency; thus, this disease has been named mouse AIDS. In this syndrome, CD154/CD40 interactions are required for but do not mediate disease by upregulation of CD80 or CD86. We report here that there is nonetheless a necessity for CD40 signaling competence, specifically an intact tumor necrosis factor receptor-associated factor 6 (TRAF 6) binding site.

Quorum sensing triggers virulence factor expression in medically important bacterial pathogens in response to a density-dependent increase in one or more autoinducing pheromones. Here, we show that phagocyte-derived oxidants target these autoinducers for inactivation as an innate defense mechanism of the host. In a skin infection model, expression of phagocyte NADPH oxidase, myeloperoxidase, or inducible nitric oxide synthase was critical for defense against a quorum-sensing pathogen, Staphylococcus aureus, but not for defense against a quorum sensing-deficient mutant. A virulence-inducing peptide of S. aureus was inactivated in vitro and in vivo by reactive oxygen and nitrogen intermediates, including HOCl and ONOO(-). …

The high levels of preexisting immunity against Adenovirus type 5 (Ad5) have deemed Ad5 unusable for translation as a human vaccine vector. Low seroprevalent alternative viral vectors may be less impacted by preexisting immunity, but they may also have significantly different phenotypes from that of Ad5. In this study we compare species D Ads (26, 28, and 48) to the species C Ad5. In vitro transduction studies show striking differences between the species C and D viruses. Rigorous phylogenetic analyses were used to compare the nucleotide sequences of feline immunodeficiency virus strains isolated from Texas and throughout the world. The envelope V3-V4 sequences and capsid gene of the Texas isolates formed a cluster between subtypes B and E. Statistical comparisons with other published sequences confirmed that the Texas group is a unique cluster, possibly a new subtype, arising from subtype B.

Under conditions that may risk the success of flap surgery, free skin grafting may be an option for the reconstruction of the irradiated area. Clinical observations reveal that skin grafts applied to irradiated area undergo ulceration, resulting in chronic open wounds. The role of hyperbaric oxygen therapy (HBO) in problem wounds is to increase the tissue oxygen tension to optimize fibroblast proliferation and collagen synthesis, to enhance the oxidative killing capacity of the white blood cells and to stimulate angiogenesis.
