Enterohemorrhagic Escherichia coli (EHEC) is the most common cause of hemorrhagic colitis and hemolytic uremic syndrome in human patients, with brain damage and dysfunction the main cause of acute death. We evaluated the efficacy of urtoxazumab (TMA-15, Teijin Pharma Limited), a humanized monoclonal antibody against Shiga toxin (Stx) 2 for the prevention of brain damage, dysfunction, and death in a piglet EHEC infection model. Forty-five neonatal gnotobiotic piglets were inoculated orally with 3 x 109 colony-forming units of EHEC O157:H7 strain EDL933 (Stx1+, Stx2+) when 22–24 h old. At 24 h post-inoculation, piglets were intraperitoneally …

Myocarditis/dilated cardiomyopathy (DCM) patients can develop autoantibodies to various cardiac antigens and one major antigen is β1-adrenergic receptor (β1AR). Previous reports indicate that animals immunized with a β1AR fragment encompassing, 197–222 amino acids for a prolonged period can develop DCM by producing autoantibodies, but existence of T cell epitopes, if any, were unknown. Using A/J mice that are highly susceptible to lymphocytic myocarditis, we have identified β1AR 171–190, β1AR 181–200, and β1AR 211–230 as the major T cell epitopes that bind major histocompatibility complex class II/IAk or IEk …

The Perceived Utility Of Personalized Genomic Medicine In Individuals With A Family History Of Heart Disease: A Pilot Study, Dana Margaret Mittag Jan 2017

The Perceived Utility Of Personalized Genomic Medicine In Individuals With A Family History Of Heart Disease: A Pilot Study, Dana Margaret Mittag

Theses and Dissertations

According to the World Health Organization (2005), cardiovascular disease (CVD) is the number one cause of death in most countries. Assessing a patient’s risk for heart disease may include incorporating factors such as their gender, age, weight, tobacco history, cholesterol, blood pressure, family history, and more recently, genetics. Genome-wide association studies (GWAS) have made it possible to identify risk loci for many of the common, complex disorders, including coronary artery disease (CAD). As the medical genetics community undergoes a shift from a genetics focus to a genomics oriented focus, genomic medicine is becoming more accessible. Research has begun to examine …

Comparison Of Atypical Bacteriophages That Infect Caulobacter Crescentus, Doreen Nguyen Jan 2017

Comparison Of Atypical Bacteriophages That Infect Caulobacter Crescentus, Doreen Nguyen

Theses and Dissertations

Several Caulobacter bacterial genomes have been recently sequenced, and all of the genomes contained one or more clusters of genes with phage origins. This observation indicates that bacteriophages contribute to the Caulobacter gene pool, so in order to understand bacteria genomes we will need to understand phage genomes as well. As part of understanding the phage genomes, we want to isolate novel bacteriophages and study their genomics. This study resulted in the isolation of 12 new phages, including four that differ from the well-studied CbKlike phages. Two of these novel phages are Podoviruses with icosahedral heads and small tails, and …

Previous literature indicates that acute pain can affect executive function, which may be due to a limited amount of shared neural resources of the brain.

Objective: The purpose of this study was to determine if acute pain affects executive function in recreationally active individuals who sustain a musculoskeletal injury.

Methodology: Twenty-four participants who presented with acute pain due to a musculoskeletal injury underwent a neuropsychological battery within 72 hours of injury and within two weeks from the initial testing session. …

Radioactive Contamination Of Natural Biological Systems: Oxidative Balance And Genetic Stability, Daniel Einor Jan 2017

Radioactive Contamination Of Natural Biological Systems: Oxidative Balance And Genetic Stability, Daniel Einor

Theses and Dissertations

The global demand for nuclear energy grows, and so do the risks of an accidental release of nuclear products into the environment. Therefore, understanding the health and ecological effects of contamination by radionuclides should be of great interest for biomedical specialists and the public. Empirical studies unequivocally show that high doses of ionizing radiation (IR) shift the redox-balance of many biological systems and increase the rates of genetic errors. However, the effects of chronic low doses of radiation vary significantly between tissues and between species. The present dissertation comprises the results of experimental and meta-analytical research of a variety of …

Role Of Translation Initiation In Regulation Of Epithelial Junctions And Cell Motility, Fahda Fawaz Alsharief Jan 2017

Role Of Translation Initiation In Regulation Of Epithelial Junctions And Cell Motility, Fahda Fawaz Alsharief

Theses and Dissertations

The integrity and barrier properties of intestinal epithelium are determined by specialized adhesive structures known as intercellular junctions; composed of adherens junctions (AJs), tight junctions (TJs) and focal adhesions that mediate cell-cell and cell matrix interactions, respectively. These two types of epithelial cell adhesions regulate each other during disruption and restitution of the epithelial barrier. Inflammatory cytokines such as interferon gamma (IFNγ) and tumor necrosis factor alpha (TNFα) are elevated during intestinal inflammation. The most notable effects of IFNγ and TNFα on intestinal epithelial homeostasis involve disruption of apical junctions and attenuation of cell migration. Although molecular mechanisms underlying these …

Thyroid Hormone As A Method Of Reducing Damage To Donor Hearts After Circulatory Arrest, William P. Adams Jan 2017

Thyroid Hormone As A Method Of Reducing Damage To Donor Hearts After Circulatory Arrest, William P. Adams

Theses and Dissertations

There is a chronic lack of donor hearts to meet the need for heart transplant both in the US and worldwide. Further, the use of available hearts is limited by the short period between collection and implantation during which the heart can be safely preserved ex vivo. Using mid-thermic Langendorff machine perfusion, we have been able to preserve the metabolic function of a healthy heart for up to 8 hours, twice the limit for current static cold storage. We have also been able to preserve the metabolic function of a damaged DCD Heart collected 30 minutes after cardiac arrest …

Tissue Factor Expression, Regulation, And Signaling In Human Airway Cells, Michael D. Davis Jan 2017

Tissue Factor Expression, Regulation, And Signaling In Human Airway Cells, Michael D. Davis

Theses and Dissertations

Rationale: Tissue Factor (TF) is a transmembrane glycoprotein that canonically functions as the initiator of the coagulation cascade. Increased levels of TF have been associated with inflammatory airway diseases. Since lipopolysaccharide (LPS) is known to elicit and inflammatory response in airway epithelium, we hypothesized that airway epithelial cells release TF when exposed to LPS. Since TF aids in local wound healing, we also hypothesized that inhibition of TF would decrease NHBE growth. The specific aim of this work was to evaluate the effects of LPS exposure on TF production and release from airway epithelia and determine the signaling pathways involved. …

Implication Of Alpha-Synuclein Transcriptional Regulation And Mutagenesis In The Pathogenesis Of Sporadic Parkinson's Disease, Sambuddha Basu

Electronic Theses and Dissertations

Parkinson's disease (PD) is an age-related neurodegenerative disorder characterized by selective loss of dopaminergic neurons (DA neurons) from the substantia nigra (SN) of the mid-brain. PD is classically associated with cytoplasmic inclusion of aggregated proteins called Lewy bodies. alpha-synuclein (α-SYN) coded by the gene SNCA, is one of the major components of Lewy body and neurite along with several other proteins like ubiquitin, neurofilament to name a few. PD is broadly categorized into two groups based on their incidence of occurrence. First is the familial form that occurs due to known genetic aberrations like mutation, gene duplication/triplication in important PD …

Analysis Of The Secondary Neurodegenerative Consequences Of Primary Oligodendrocyte Stress Through The Use Of The Novel Obiden Mouse Model, Daniel Zdzislaw Radecki

Wayne State University Dissertations

The work of this project was to develop, test and characterize a potential novel mouse model of the neurodegenerative disease Multiple Sclerosis (MS). Historically, MS has been identified as a primary autoimmune disease of the central nervous system (CNS). However, treatments based on this view have met with limited success, and in most cases, fail to prevent progression of MS from mild to moderate and severe forms. Original observations regarding axonal and neuronal pathology in the white and gray matter of the CNS were rediscovered in the 1990s. These observations indicated that even in the absence of the immune system, …

Involvement Of Notch Signaling In Interleukin-4-Stimulated Human Macrophages, Naunpun Sangphech

Chulalongkorn University Theses and Dissertations (Chula ETD)

Macrophages respond to various stimuli, resulting in distinct effector phenotypes that range from LPS-stimulated proinflammatory to IL-4-activated pro-healing phenotype. The regulatory mechanism of this polarization is not fully understood. Notch signaling pathway is a conserved signaling pathway involved in polarization of macrophages, especially proinflammatory macrophages. However, the role of Notch signaling in IL-4 stimulated macrophages (M(IL-4)) is not well defined. PPARgamma is a signature nuclear receptor in M(IL-4) that function mainly in regulating lipid metabolism. There are many reports that revealed the interaction of Notch signaling and PPARgamma in various cells but not in macrophages. In this study, the interaction …

Generation And Characterization Of Hla-Universal, Ipsc-Derived Platelets, Phatchara Norbnop

Chulalongkorn University Theses and Dissertations (Chula ETD)

Platelet demand has been increased around the world resulting from many factors including the rise of aging population, hematological malignancy incidence, and hematopoietic stem cell transplantation. In addition, inadequacy of donors or matched donors, time consuming, cost, the risk of transfusion-transmitted infections and transfusion-associated reactions, and platelet transfusion refractoriness are limits of this approach. To solve these problems, here we propose to generate platelets in vitro which do not induce alloimmunity to HLA class I, a major cause of immune factors in platelet transfusion refractoriness. In this study, we have knocked out β2-microglobulin gene (β2m) in iPSCs using paired CRISPR/Cas9 …

Recently, much attention has been paid to natural plant-derived compounds as an alternative strategy for developing anti-obesity agents with minimal detrimental effects. Among various natural compounds, caffeine (1, 3, 7-trimethyl xanthine) and alpha lipoic acid (1, 2-dithiolane-3-pentanoic acid; LA) are the natural agents which show a promising result in decreasing fat deposit. Moreover, we have improved efficiencies of LA by CaCO3 NPs because LA had low bioavailability. In the present study, we investigated the effect of combination between caffeine and LA-CaCO3 NPs in lipid accumulation reduction and lipolysis stimulation in human subcutaneous adipocytes and human visceral adipocytes. Our results showed …

L-arginine (L-arg) is an amino acid that has been used to induce acute pancreatitis (AP) in mice. Curcumin is an active ingredient constituent of rhizomes of the plant Curcuma longa Linn. It has been used for the treatment of inflammatory conditions and shown to exhibit a variety of biological activities such as anti-inflammatory and anti-oxidant. The present study aims to examine the effects of curcumin on oxidative stress, inflammation and apoptosis in AP. Male ICR mice were randomly divided into 4 groups. 1) Control group, mice will be received i.p. of 1% DMSO as a vehicle. 2) AP group, mice …

Metabolic acidosis exerts several cellular and tissue dysfunctions, such as increases mineral bone and protein degradation, induces inflammation, oxidative stress and cell death. These complications lead to multiple organ failure, including chronic kidney disease. Sodium bicarbonate and Shohl's solution are commonly used to treat metabolic acidosis. However, these agents cause frequent gastrointestinal adverse effects. Hence, we invented the new calcium carbonate nanoparticles to use as a drug delivery system for carrying citrate to renal cell which we hypothesize to reduce adverse effects of metabolic acidosis. We studied the physical characteristics and pharmacological effects of calcium citrate nanoparticle (Cacit) and calcium …
