Radioimmunotherapy Of Mice Bearing Breast Tumors With ^{177}Lu-Labeled Trastuzumab, Samira Rasaneh, Hossein Rajabi, Shahram Akhlaghpoor, Shahab Sheybani

In the present study, we investigated the therapeutic efficacy of trastuzumab labeled with lutetium-177 (^{177}Lu) in mice bearing breast tumors. Materials and methods: Trastuzumab was labeled with ^{177}Lu via DOTA as a chelator. In vitro and in vivo quality control tests of incorporated activity, immunoreactivity, stability of ^{177}Lu-trastuzumab, biodistribution, and imaging studies were performed. Therapeutic efficacy and dosimetry studies were carried out in mice with breast tumors. Results: The good in vitro and in vivo quality control results suggested that ^{177}Lu-trastuzumab could be used as a radioimmunotherapy agent. Our aim was to show the effect of one-lung ventilation (OLV) on cerebral blood flow (CBF) by measuring carotid blood flow. This technique has been the subject of experimental and clinical studies. Materials and methods: Carotid doppler flows were measured at 4 different times. Peak systolic velocity (PSV) and end diastolic velocity (EDV) were measured and pulsatility index (PI) and resistive index (RI) calculated. To investigate the protective effects of testosterone propionate (TP) on hypoxic-ischemic brain damage (HIBD) neonatal rat brain by observing oligodendrocyte myelin glycoprotein (OMgp) expression in the cortex and hippocampus after HIBD. Materials and methods: Rats were randomly divided into 3 groups: TP pretreatment, HIBD control, and a sham treatment group. Understanding the evolution of the human immunodeficiency virus type 1 (HIV-1) envelope during disease progression can provide tremendous insights for vaccine development, and simian-human immunodeficiency virus (SHIV) infection of nonhuman primate provides an ideal platform for such studies. A newly developed clade C SHIV, SHIV-1157ipd3N4, which was able to infect rhesus macaques, closely resembled primary HIV-1 in transmission and pathogenesis, was used to infect several pig-tailed macaques. One of the infected animals subsequently progressed to AIDS, whereas one remained a nonprogressor. Enteroviruses like coxsackievirus B3 (CVB3) are common suspects in myocarditis/dilated cardiomyopathy patients. Autoimmunity has been proposed as an underlying mechanism, but direct evidence of its role is lacking. To delineate autoimmune response in CVB3 myocarditis, we used IAk dextramers for cardiac myosin heavy chain (Myhc)-α 334–352. We have demonstrated that myocarditis-susceptible A/J mice infected with CVB3 generate Myhc-α-reactive CD4 T cells and such a repertoire was absent in naïve mice as measured by proliferative response to Myhc-α 334–352 and IAk dextramer staining. Dopamine is one of the key ingredients in the glue that cements social bonds in vertebrates. The D2 dopamine receptor has been implicated in the regulation of monogamous pair bonding in the prairie vole. While dopamine affects courtship behaviors in the male zebra finch, the behavioral role of dopamine acting at D2 receptors in both males and females deserves further attention. Cubilin is an endocytic receptor capable of mediating the endocytosis of multiple ligands including High Density Lipoprotein apolipoprotein A-I (apoA-1) and albumin. It is highly expressed on the epithelium of intestinal villi and renal proximal tubules. In the kidney, endocytosis mediated by cubilin and its co-receptor megalin is the principal mechanism for proximal tubule reabsorption of filtered proteins, such as albumin and apoA-1. Yet, the consequences of cubilin deficiency on the blood levels of albumin and apoA-1 have not been studied. Furthermore, cubilin is downregulated in chronic diseases such as diabetes. DNA mismatch repair (MMR) is a critical genome-maintenance system. It ensures genome stability by correcting mismatches generated during DNA replication, suppressing homologous recombination, and inducing apoptosis in response to severe DNA damage. As a result, defects in MMR lead to genome-wide mutations and susceptibility to both hereditary and sporadic cancer syndromes. The hallmark of cancer cells defective in MMR is their ability to display frequent instability in simple repetitive DNA sequences, a phenomenon called microsatellite instability (MSI). ATP-binding cassette (ABC) transporters are transmembrane proteins that transport a wide variety of substrates across intra and extra-cellular membranes. A few examples of endo and xenobiotic substrates are metabolic products, lipids, sterols, and drugs. An important function of ABC transporters involved in export is to prevent intracellular the buildup of toxic products. Several ABC transporters have also been associated with drug resistance upon treatment with chemotherapeutic agents. P-glycoprotein (P-GP) and the multidrug resistant (MRP) transporters of the ABC C family are examples of transporters that confer chemo-resistance.

To determine the effect of omega-3 (n-3) fatty acid supplementation on plasma orexin A, plasma fatty acids, and anthropometric measurements in patients with narcolepsy. To study the effect of undifferentiated bone marrow stromal cells (BMSCs) on peripheral nerve regeneration using a rat sciatic nerve regeneration model. In recent years, cell transplantation has become the focus of attention, and reliable outcomes have been achieved in regeneration of the sciatic nerve. Materials and methods: A 10-mm sciatic nerve defect was bridged using an inside-out vein graft (IOVG) filled with undifferentiated BMSCs (2 × 107 cells/mL). To investigate the postoperative analgesic effects of epidural tramadol infiltration. Tramadol is a weak opioid that has local anesthetic and antiinflammatory properties. Materials and methods: Sixty patients of American Society of Anesthesiologists class I or II undergoing lumbar microdiscectomy with general anesthesia were included in the study. Tokat Province is an epicenter for Crimean-Congo hemorrhagic fever virus (CCHFV) in Turkey. The aim of this study was to investigate the seroprevalence of Rickettsia conorii and to clarify the epidemiological similarities between CCHFV and R. conorii in Tokat Province. Materials and methods: The prevalence of antibodies reactive with R. conorii was examined by ELISA in 364 sera, 151 of which were seropositive for CCHFV. Results: The overall prevalence of antibodies reactive with R. conorii was 36.81%. To investigate the role of the oxidative stress status of drivers involved in traffic accidents as a risk factor. Materials and methods: Twenty-six drivers and 28 passengers involved in traffic accidents and 31 healthy control drivers and 30 healthy control pedestrians were enrolled in the present study. The total antioxidant status (TAS) and total oxidant status (TOS) were measured in blood samples. Results: The mean value of the TOS level in drivers involved in traffic accidents was significantly higher than that of the control pedestrians (P =< 0.05). The mean value of the TAS level in passengers involved in traffic accidents was significantly lower than that of the control drivers and pedestrians (P =< 0.001 and P =< 0.01, respectively). Conclusion: This is the first study assessing the oxidative markers of individuals involved in a traffic accident compared with a control group. The results reveal that involvement in a traffic accident has an effect on oxidative markers. However, further studies are required in order to use the oxidative stress status of drivers involved in traffic accidents as a marker for estimating the accident risk.

Aim: To investigate the effects of intraabdominal hypertension (IAH) on adrenal glands. Materials and methods: Forty male Wistar albino rats were divided into 4 groups: the control group, sham control group, and 2 study groups. In the study groups, the animals were subjected to IAH of up to 20-25 mmHg for 3 h. At the end of the study, blood samples were collected for cortisol analysis and adrenal gland samples were extracted via laparatomy for histopathological examination. Results: Cortisol levels were decreased compared to the control group (P < 0.05). There were no statistically significant differences demonstrated among the other groups (P > 0.05). Aim: To evaluate the possible roles of phospholipase C, protein kinase C, and endothelin-converting enzyme on contractions induced by ethanol (164 mM) in isolated gastric fundal strips of mice. Materials and methods: After the mice were killed, the stomach of each was removed and longitudinal muscle strips were prepared from the gastric fundus. The strips were mounted in organ baths and their responses were recorded isometrically. Ethanol (164 mM) was added to the organ baths and a steady-state contraction was obtained. The Effect Of N-Acetylcysteine On Growth And Biofilm Formation In Staphylococcus Epidermidis Strains, Sahra Kirmusaoğlu, Seyhun Yurdugül, Mücahi̇de Esra Koçoğlu

Aim: To examine the possible preventive effect of N-acetylcysteine (NAC) on biofilm formation in Staphylococcus epidermidis strains, which cause very important problems such as implant-associated infections in hospitalized and immunocompromised patients worldwide. Materials and methods: The effects of varying concentrations of NAC on the formation of biofilm by 28 strains of Staphylococcus epidermidis was investigated spectrophotometrically by micro-ELISA assay. Results: Significant differences in biofilm formation in the presence of various NAC concentrations were found in 15 methicillin-resistant Staphylococcus epidermidis and 13 methicillin-sensitive Staphylococcus epidermidis strains (P < 0.05). Conclusion: It was found that slime formation was decreased due to the increased concentrations of the NAC treatments. Based on these findings, it is possible to use NAC as an antibiofilm agent on medical devices or in catheter lock solutions to prevent colonization and implant-associated infections.

Aim: To assess the efficiency of hyperbaric oxygen (HBO) therapy on L. tropica, which is the major causative agent of cutaneous leishmaniasis in Turkey. Materials and methods: The effects of HBO on Leishmania tropica promastigote and amastigote forms were studied at a pressure of 2.5 atmosphere absolute (ata). To evaluate the effects of diabetes on nitric oxide synthase (NOS) activity and on gene expressions of B-cell lymphoma protein 2 (Bcl-2) and caspase-3 in the renal tissues of streptozotocin (STZ)-induced rats. Materials and methods: Twenty rats were analyzed in 2 equally distributed groups of healthy controls and STZ-induced diabetic rats. Rats with blood glucose levels of >=250 mg/dL were considered diabetic. Renal tissue sections were analyzed histomorphologically and immunohistochemically. Gene expressions of caspase-3 and Bcl-2 were evaluated by real-time quantitative polymerase chain reaction. The aim of the present study was to investigate the relationship between liver damage induced by carbon tetrachloride (CCl_4) and adenosine deaminase (ADA), 5'-nucleotidase (5'NT), xanthine oxidase (XO) enzyme activities, and malondialdehyde (MDA) levels. The effect of stobadine on purine metabolism was also evaluated. Materials and methods: This study measured the effect of CCl_4 on ADA, 5'NT, XO enzyme activities, and MDA levels, and measured the histopathologic changes in liver cells. We aim ed to study the effect of a glutathione precursor, L-2-oxothiazolidine-4-carboxylic acid (OTCA), on the lung antioxidant defense system in an animal asthma model. Materials and methods: The study was carried out on 24 female C57BL/6 mice.
