Glioblastoma (GBM) is a highly aggressive brain tumor characterized by recurrence and resistance to therapy, which are driven by interactions between glioblastoma stem cells (GSCs), tumor-associated macrophages (TAMs), and the tumor microenvironment. In this study, single-cell RNA sequencing data were analyzed using QIAGEN CLC Genomics Workbench to identify genes associated with tumor–immune interactions. Following clustering, differential expression, and a multi-step prioritization strategy, we identified APOE as a top candidate gene connecting tumor and immune processes, contributing to therapeutic resistance in glioblastoma.

Chronic neuropathic pain severely diminishes quality of life, yet the supraspinal neural circuits that sustain pain perception and mediate analgesic relief remain incompletely characterized. While peripheral and spinal pain pathways are well established, relatively few studies have mapped pain- or opioid-related neuronal activation across the entire brain. This dissertation employed activity-dependent genetic labeling (TRAP2/Ai9), whole-brain tissue clearing, light sheet microscopy, and graph-theoretic network analysis to characterize brain-wide activation patterns across chronic pain and analgesic conditions in mice.

Plants deploy sophisticated adaptive mechanisms to mitigate the detrimental effects of abiotic stresses (drought, salinity, temperature extremes, and heavy metals) and biotic stresses (pathogens and senescence) on growth and productivity. Central to these responses are transcription factors (TFs) that orchestrate stress-responsive gene networks. Among TF families, MADS-box proteins, characterized by their evolutionarily conserved DNA-binding domain, function as pivotal regulators of developmental plasticity and stress adaptation. While recent advances have clarified their roles in abiotic stress tolerance, a systematic integration of their functions in biotic stress responses has yet to be achieved. Combining satellite telemetry and stable isotope analysis offers a framework for inferring sea turtle foraging habitats when spatial isotopic gradients exist. We evaluated this combined approach using carbon (δ13C) and nitrogen (δ1⁵N) isotope values from critically endangered hawksbill turtles (Eretmochelys imbricata) nesting at Buck Island Reef National Monument, U.S. Virgin Islands (N=31), and Long Island (Jumby Bay), Antigua (N = 31). Nesting females were satellite-tracked to identify foraging areas and sampled for skin tissue, and Empirical Bayesian Kriging was used to generate basin-scale δ13C and δ1⁵N isoscapes. Adoption of automatic milking systems (AMS) in the US are expected to increase rapidly in the coming years driven primarily by a desire to improve labor efficiency and flexibility and decrease reliance on hired labor. As such, feeding and milking strategies to best optimize milk production and economic gains tailored to US dairy production systems are needed. Initially introduced as a method to encourage the cow to visit the AMS more frequently, offered feeds through the AMS provide an opportunity to individualize nutrient needs. established and maintained axenic

cyanobacterial cultures to support future

genomic and ecological investigations. By

ensuring low contamination levels and stable

growth conditions, this work provides a

foundation for downstream analyses of bloom-

forming cyanobacteria. It contributes to a better

understanding of freshwater ecosystem health

The large mosasaur Tylosaurus is known to have been a predator of large marine animals. Performing beam theory analysis, a computational technique that uses linear dimensions of a structure to calculate its resistance to bending, on the mandibles of six Tylosaurus specimens from the Smoky Hill Chalk of Kansas indicates dorsoventral and mediolateral bending strength both increase posteriorly along the mandible. Comparisons with Varanus komodoensis indicate relatively greater mediolateral bending strength mid-dentary in Tylosaurus, suggesting that resisting torsional forces of struggling prey was more important at this landmark in Tylosaurus and that more of its jaw length was used …

Modern lifestyles reduce sunlight exposure and increase artificial light at night, potentially promoting vitamin D deficiency (VDD) and melatonin (MEL) suppression. The two hormones regulate immunity, erythropoiesis, and platelet (PLT) indices, but their combined hematological effects are not well characterized. Although traditional management primarily focuses on physical symptom reduction, gaps remain in addressing long-term participation and quality of life, particularly among underserved populations. Bacteria survive hostile conditions by shutting down protein synthesis, but how they restart growth remains poorly understood. Here, we use an E. coli ΔrimM strain, which exhibits a prolonged growth arrest, as a model to investigate how bacteria recover from this state and restore protein synthesis. RimM is a conserved ribosome maturation factor for the 3'-major (head) domain of the 16S rRNA within the bacterial 30S subunit. The loss of RimM causes a longer delay in recovery than other 30S maturation factors, including RbfA. Birdsong is a communication signal shaped by ecological, morphological, and cultural processes. While many songbird species produce relatively simple vocal repertoires, others, such as the Bachman’s Sparrow (Peucaea aestivalis), produce moderately large repertoires with multiple distinct and context-dependent song categories. Dietary nitrate supplementation can improve blood pressure and contractile function of type II (fast twitch) muscle fiber contraction and force production. However, limited data are available for  women and dosing guidelines for women. The purpose of this study is to investigate if nitrate ingestion, provided as nitrate-rich beetroot juice, improves blood pressure and countermovement jumps and cycling sprint performance compared to nitrate-depleted beetroot juice placebo in women. Harmful algal blooms produced by the diatom Pseudo-nitzschia regularly expose marine mammals along the California coast to domoic acid (DA), a neurotoxin that bioaccumulates in marine food webs. In California sea lions (Zalophus californianus), domoic acid exposure is associated with seizures, hippocampal injury, and maternal-fetal transfer of the toxin that can result in abortion. Bloom events have led to large numbers of stranded sea lions along the California coast. Enoyl-ACP reductase (FabI), an enzyme which is inhibited by catechin, is essential for lipid biosynthesis in bacteria (E. coli). Another FabI inhibitor, triclosan, has strong antibacterial effects,1 but overuse can be detrimental to the environment. Flavonoids, a class of dietary compounds, are known to have potent inhibitory effects on FabI and other kinases.2,3,4 Adducts form as a result of the biochemical reaction between flavonoids and cysteine, a modification which can be verified by m/z shifts on the mass spectra.5 Samples of E. coli protein with overexpressed FabI are utilized: the wild type with ordinary primary structure, and mutant type lacking …

The app launched with five activities for exploration: slopefields, phase planes, oscillations, solutions to systems, and numerical methods for approximation. Bifurcations is a new sixth activity that we designed to investigate changes in the long term behavior of solutions to autonomous differential equations. Coral reefs thrive in primarily oligotrophic environments, depending on factors such as temperature and salinity for their stability. While moderate levels of nutrients sustain coral growth, excessive nutrient inputs promote algal blooms which can outcompete corals for space. Over the past century, Maunalua Bay (O'ahu, HI) has experienced dramatic development and increase in population, which has affected the water quality in the nearshore coral reefs of the bay. This study examined the influence of the invasive ice plant Carpobrotus edulis on species richness and biodiversity in Bluffs Park. Because previous research suggests that C. edulis suppresses nearby native flora, our team used quadrat and transect sampling to measure plant diversity at seven distances from the invasion edge (1 m, 3m, 5m, 10m, 15m, 20m, 30 m). Flora were identified using iNaturalist and recorded. Shannon diversity showed no significant change with distance (ANOVA: F₆,₄₂ = 1.34, p = 0.18), and assumptions of normality and equal variance were met. Commercially-available laser-based timing gates are widely implemented for assessing neuromotor function, speed and agility; however, there is limited available data for the reliability and validity of this equipment. The purpose of this study is to establish the reliability and validity of timing gates from DASHR vs. Sport React compared to a manual stopwatch in a battery of neuromotor, performance and agility tests.
