DNA polymerase θ (Pol θ)-mediated end-joining (TMEJ), one of several pathways for repairing DNA double-strand breaks, is traditionally thought to initiate via anchoring at short, consecutive, and perfectly matched microhomologies (MHs). Emerging evidence indicates that Pol θ can utilize MHs containing mismatches both in vitro and in vivo. This revised definition of MH provides a mechanistic explanation for a broader spectrum of Pol θ-dependent repair outcomes. Here, we summarize recent findings on the revised definition of MHs utilized by Pol θ, assess the applicability of this concept across species, and compare TMEJ with other (micro)hom(e)ology-mediated repair pathways. Background: The number and type of genetic alterations required to initiate breast and ovarian cancer remain unclear. While germline BRCA1/2 carriers show markedly elevated cancer risk, it is uncertain whether point mutations or copy number alterations constitute the rate-limiting events of tumourigenesis.
Methods: We developed a statistical framework extending prior incidence-mutation models to estimate the minimal number and type of driver events required for cancer initiation. Somatic mutation and copy-number data from >3000 breast and ovarian cancers in TCGA and METABRIC were compared between germline BRCA1/2 carriers and non-carriers matched on subtypes. Breast surgery for aesthetic purposes, such as breast augmentation or breast reduction, and breast reconstruction after cancer treatment require an accurate structural (anatomical) and mechanical (functional) understanding of the breast components, including the fascial-ligamentous support system of the breast, to achieve optimal results. This paper aims to provide a comprehensive description of the mechanical behavior of the ligamentous and fascial connective tissues of the human female breast. The dromedary camel possesses unique ocular adaptations enabling survival in harsh desert environments. This study aimed to characterize the posterior segment of the dromedary camel’s eye using spectral-domain optical coherence tomography (SD-OCT) and fundus photography. Fourteen healthy adult camels were sedated and imaged. High-resolution OCT images enabled clear visualization and measurement of the retina, choroid, and sclera. The retina exhibited well-defined layers, including the retinal pigment epithelium, photoreceptor layer, plexiform layers, and nerve fiber layer. Water salinity and scarcity constitute major limitations to crop production in arid and semi-arid regions. Introduction of nutritious and stress-tolerant underutilized crops is a promising approach for dietary enrichment, cropping system diversification, remediation of marginal and degraded lands, and building climate resilience. Background: Dual mitogen-activated protein kinase (MAPK) pathway and autophagy inhibition shows synergistic antitumor activity in preclinical models of RAS-mutant cancers. We hypothesized that autophagy blockade with hydroxychloroquine (HCQ) could overcome resistance to MEK inhibition with binimetinib (BINI) and provide clinical benefit in previously treated, KRAS-mutated, metastatic pancreatic ductal adenocarcinoma (PDAC).

The bone marrow endosteum provides a robust source of bone-making osteoblasts by housing various types of osteoblast precursor cells, including endosteal stem cells expressing fibroblast growth factor receptor 3 (Fgfr3) and their downstream progenitor cell populations. In fetal bone development, Dlx5 marks early perichondrial cells that populate the diaphyseal bone and marrow stroma in the postnatal stages. The rare Central Asian psammophilic scorpion genus, Liobuthus Birula, 1898 (Scorpiones: Buthidae), is revised. Five species are delineated according to results from three species delimitation approaches using mitochondrial DNA. Results are supported by diagnostic morphology, including an assessment of the distinctive neobothriotaxy. The type species, L. kessleri Birula, 1898, is confirmed for Iran (northeast), Turkmenistan and Uzbekistan (south). Four new species are described: L. atamuradovi sp. n. In this study, the effects of Aronia melanocarpa and Sambucus nigra L. extracts added at different concentrations (0.5%, 1%,and 1.5%) on the physicochemical, sensory, microbiological, antioxidant capacity (DPPH• and ABTS•+ radical scavenging activity, TPC,TFC, and TAC) antidiabetic (α-amylase and α-glucosidase inhibition), and anti-Alzheimer properties (AChE inhibitory activity) of icecream were investigated. According to the statistical analysis results, extract addition did not have an effect on the total dry matter, fatand ash ratios of the ice cream samples (p > 0.05), but it had an effect on the protein ratio, viscosity, and first drip time at the level ofp < 0.05. The addition of extract positively affected the DPPH• radical scavenging activity IC50 and ABTS•+ radical scavenging activityIC50 values of the ice cream samples, and also increased the TPC, TFC, and TAC values. Indeed, a decrease in the IC50 values of DPPH•and ABTS•+ radical scavenging activities indicates an increase in antioxidant activity. The highest TPC, TFC, and TAC values weredetected in the A-1.5 sample (sample containing 1.5% A. melanocarpa extract) as 749.33 mg GA/kg, 434.70 mg QE/kg, and 290.97 mg/kg, respectively. Similarly, the highest α-amylase inhibitory activity (23.08%), the highest α-glucosidase inhibitory activity (10.41%) andthe highest AChE inhibitory activity (14.70%) were observed in the A-1.5 sample. Extract addition had no effect on microbiologicalproperties and sensory properties except texture. The results of this study showed that A. melanocarpa, S. nigra L. extracts can be usedas a natural additive with antioxidant, antidiabetic and anti-Alzheimer activities in ice cream production.

Mayflies are among the most abundant benthic invertebrates inhabiting natural lotic freshwater systems. Their assemblages vary with environmental, geographical, and hydrological conditions along altitudinal gradients and under different types of watershed disturbance. The main objective of this study was to analyze the composition and β-diversity of mayflies across three river watersheds and to assess how these assemblages differ among them. Rising environmental stressors, including climate change, nutrient loading, pollution, and anthropogenic impacts, have made the continuous monitoring of lake water quality increasingly essential. While in situ methods offer high precision, satellite-based remote sensing is essential for capturing large-scale, long-term changes and accessing remote or extreme environments. To complement these approaches, we developed a cost-effective in situ optical measurement system specifically designed for optically complex inland waters. Pregnancy is considered one of the most physiologically stressful periods in mammals. In the present study, serum and hair cortisol concentrations were measured in nine female Akkeçi goats, including five primiparous (1.5 years old) and four multiparous (3.5 years old), to evaluate chronic stress during pregnancy. The study aimed to determine the effects of zinc-reinforced clinoptilolite supplementation to the diet on selected hematological parameters, serum immunoglobulin G (IgG) concentrations, and enzyme histochemical characteristics in fattening lambs. Electric pressure cookers (EPCs) offer convenience, versatility, and efficiency in everyday cooking. EPCs offer multiple cooking functions beyond pressure cooking, including sautéing, slow cooking, steaming, cooking rice, and preparing yogurt. They are not suitable for pressure canning, as confirmed by multiple Extension research studies and national guidelines. When used properly, EPCs provide a safe and effective way to prepare meals quickly while saving counter and storage space.

Mechanosensitive Piezo1 channels participate in regulating pain sensitivity, insulin secretion, and vascular tension; however, their expression in the autonomic paraventricular nucleus (PVN) and role in modulating sympathetic outflow and cardiovascular function remain unstudied. In this study, unilateral PVN microinjection of the Piezo1 channel blocker Dooku1 (0.1, 1, 10, 100, and 200 pmol) administered to anesthetized male rats increased renal sympathetic nerve activity (RSNA) and mean artery pressure (MAP) in a dose-dependent manner, with maximum increases of 93 ± 30% (p < 0.0001) and 21 ± 5 mmHg (p < 0.0001), respectively, elicited by Dooku1 at 100 pmol. Similarly, PVN microinjection of the peptide Piezo1 channel blocker GsMTx4 (1 nmol) significantly increased RSNA (p < 0.001) and MAP (p < 0.0001). Conversely, PVN-microinjected Piezo1 channel activators Yoda1 (5 nmol) and Jedi2 (5 nmol) did not significantly alter RSNA or MAP. Western blot and qRT-PCR analyses of the hypothalamic PVN showed abundant Piezo1 mRNA and protein expression. Immunofluorescence detection showed that Piezo1 was expressed in pre-sympathetic PVN neurons with axons projecting to the rostral ventrolateral medulla. We conclude that Piezo1 channels expressed in the autonomic PVN neurons play an important role in regulating sympathetic outflow and cardiovascular function.

This research develops an integrated model that links the social benefit requirements of Urban Green Infrastructure (UGI) with the non-physical attributes of Place Identity (PI) to enhance the Sense of Place (SOP) in contemporary urban environments. The need for this integration arises from limitations in current planning frameworks, which often prioritize ecological performance while overlooking cultural, perceptual, and behavioral dimensions that shape user attachment and belonging. The Bermuda fireworm, Odontosyllis enopla Verrill, 1900, is a marine polychaete that displays a unique bioluminescent mating ritual. Despite the first sighting of O. enopla more than 534 years ago, molecular data have been limited. Several syllid mitogenomes are currently available; however, there are only three published genes for O. enopla: two partial mitochondrial genes (16S [508 bp] and cox1 [653 bp]; 1,161 bp total) and one partial nuclear gene (18S [1,339 bp]). Prisoner-on-prisoner sexual abuse is widespread in US male prisons, with inmates “turning each other out” by sexually assaulting each other. While the Prison Rape Elimination Act (PREA) received unanimous support in Congress in 2003, motivated by the Human Rights Watch Report “No Escape: Male Rape in Prisons,” scholars argue that the PREA fails to protect prisoners from prison rape: for example, its standards may be ineffective or criminalize consensual sex between prisoners. Food insecurity is a devastating force in the United States, as shown by its destructive impact on vulnerable groups like children, racial minorities, and those in poverty. Non-white urban communities of color especially have significantly higher rates of household food insecurity due to systemic racism in American food systems. Despite the prevalence of these factors, they are consistently overlooked by public and private forces. Primary membranoproliferative glomerulonephritis (MPGN) is an ultrarare disease characterized by immunofluorescence microscopy as either immune-complex mediated (IC-MPGN) or C3 glomerulopathy (C3), the latter subdivided by electron microscopy to C3 glomerulonephritis (C3GN) and dense deposit disease (DDD). Both IC-MPGN and C3G typically have obvious C3 staining differentiating them from other causes of MPGN histology. Secondary causes must be excluded, including infections, autoimmune disease, and neoplasia. Clinical presentations are variable, including urinary sediment abnormalities, nephrotic syndrome, or a rapidly progressive course. The prognosis is unfavorable with about 50% reaching kidney failure by 10 years. Recurrence following transplantation is frequent, and allograft survival …

Antimicrobial resistance (AMR) poses a critical challenge to global health, with food animal production systems recognized as significant reservoirs of antimicrobial-resistant bacteria. This study evaluated the prevalence and distribution of antimicrobial resistance genes (ARGs) and virulence factors (VFs) across small-scale poultry and cattle farms. A total of 468 samples (soil, feces, water, and natural land soil) were collected from four farms and analyzed using shotgun metagenomics. Proteobacteria (34.91%) were the dominant phylum across environments, followed by Cyanobacteria (15.67%), Actinobacteria (14.95%), Firmicutes (10.57%), and Bacteroidetes (8.69%). Soil salinity is a major constraint on global crop productivity, driving the need for salt-tolerant varieties. While CRISPR-Cas genome editing offers targeted solutions for trait improvement, significant biological and technical bottlenecks limit its application in conferring salt stress resilience. This systematic summarizes findings from 83 peer-reviewed studies (2015–2024) employing CRISPR/Cas technologies to improve salt tolerance in five major crops (rice, wheat, maize, sorghum, barley). Our systematic review reveals that early single-gene edits achieved modest gains (30–50% Na⁺ exclusion) but often showed limited yield gains in field settings, potentially due to compensatory regulation and environmental variation. Mycobacterium tuberculosis (Mtb) remodels host cell functions to support its intracellular persistence. While the effects of Mtb infection on infected macrophages are well characterized, responses of uninfected bystander cells in the same microenvironment remain poorly understood. Here, we show that Mtb infection triggers widespread epigenetic reprogramming not only in infected macrophages but also in neighbouring bystander cells. These bystander macrophages acquire active chromatin marks, exhibit distinct transcriptional changes, and display enhanced immune responsiveness, thereby increasing resistance to both homologous and heterologous infections. This article offers an ecological analysis of Malika Mokeddem's novel N'Zid (2001), moving beyond postcolonial and anthropocentric interpretations. It centers on the protagonist Nora's individuation, arguing this process is intrinsically linked to her evolving relationship with the aquatic environment from which she emerges. The article aims to situate N'Zid within an ecological perspective, particularly relevant in the Anthropocene, an era compelling a reassessment of humanity's place. Snapping shrimp damp the shock waves they produce and use as weapons with a helmet-like structure termed the orbital hood. Here, we ask how structural and material properties contribute to shock wave damping by orbital hoods in Alpheus heterochaelis. Using tensile mechanical testing, we find orbital hoods are approximately half as stiff as carapace and have twice the capacity for viscous energy dissipation. Microstructural features probably contribute to tissue-specific mechanical properties: the endocuticles of orbital hoods have almost twice as many lamellae as those of carapaces despite being half as thick, suggesting a mechanism for enhanced material mobility underlying …
