Background/Objectives: Chronic inflammation and Western-style diets elevate colorectal cancer (CRC) risk, particularly in individuals with colitis, a feature of inflammatory bowel disease (IBD). Diets rich in polyphenol-containing functional foods, such as cocoa, may reduce gut inflammation and modulate the gut microbiome. This study investigated the impact of cocoa polyphenol (CP) supplementation on inflammation and microbiome composition in mice with colitis, fed either a healthy or Western diet, before, during, and after the onset of disease. We hypothesized that CPs would attenuate inflammation and promote distinct shifts in the microbiome, especially in the context of a Western diet. Methods: …

Biomedical Engineering Faculty Publications and Presentations

Pancreatic cancer (PC) is aggressive and deadly; most PC cells overexpress an epidermal growth factor receptor (EGFR). EGFR-targeting peptide (GE11) was conjugated to gold nanoparticles (AuNPs) for delivering rapamycin (Rap), an mTOR inhibitor that reduces proliferation and induces apoptosis. Biophysical techniques confirmed the conjugation. Rap loading efficiency was 41.5%, with sustained release at a lower pH. Conjugates showed higher uptake in PANC-1 cells than unmodified AuNPs. It also inhibited proliferation and induced apoptosis via pathways involving reactive oxygen species, mitochondrial damage, and caspase activation. This approach represents a promising nanoparticle-based therapeutic strategy targeting PC cells, utilizing GE11-EGFR interactions to minimize …

The Nature Of Anthropogenically Driven River Drying: Spatiotemporal Causes And Consequences, Eliza Inez Gilbert Jul 2025

The Nature Of Anthropogenically Driven River Drying: Spatiotemporal Causes And Consequences, Eliza Inez Gilbert

Biology ETDs

Streambed drying naturally occurs in over 60% of rivers and streams worldwide. Climate change and human regulation of surface and groundwater have increased drying in naturally intermittent systems and caused perennial systems to transition to intermittency, impacting water security, water quality, and biodiversity. To understand human-induced drying dynamics, we used 12 years of daily drying data along a 154-km regulated reach of the Rio Grande. We conceptualized river drying as a regime analogous to the natural flow regime paradigm and quantified drying magnitude, rate of change, and duration. Although linear models predicting drying magnitude and rate of change were uninterpretable, …

Nitrogen Addition Additively Boosts Climate-Driven Salsola Tragus (Russian Thistle) Invasion Into Southwestern Usa Grasslands, Melissa A. Bacigalupa

Biology ETDs

Effective management of invasive, non-native plants requires understanding key drivers of invasion, including nitrogen enrichment from atmospheric deposition and agriculture. Long-term studies are essential for capturing complex invasion dynamics yet remain uncommon. We used 22 years of observations and 14 years of nitrogen experiments to examine invasion by Salsola tragus (Russian thistle) into three semi-arid grasslands of central New Mexico. Invasion patterns varied, with rapid expansion in Chihuahuan Desert grassland and the Desert-Plains ecotone but stability in Plains grassland. Under ambient nitrogen, precipitation most strongly correlated with Russian thistle biomass and high native grass biomass was associated with Russian thistle …

Effects Of Perfluorooctanoic Acid (Pfoa) And Elevated Salinity On Growth, Reproduction, And Movement Of Physa Acuta, Juana Baudrix, Andrea Rizzo, Samuel Richardson, Jason Doll, Daniel Elias Jul 2025

Effects Of Perfluorooctanoic Acid (Pfoa) And Elevated Salinity On Growth, Reproduction, And Movement Of Physa Acuta, Juana Baudrix, Andrea Rizzo, Samuel Richardson, Jason Doll, Daniel Elias

Journal of the South Carolina Academy of Science

Freshwater ecosystems face increasing threats from pollutants such as perfluorooctanoic acid (PFOA) and elevated salinity. This study examined the effects of PFOA (1.69 µg/L) and elevated salinity (2.5 g/L) on Physa acuta growth, reproduction, and movement over a 21-day exposure. Movement increased across treatments, and reproductive output also increased with salinity and salinity + PFOA, with increased egg sac production in the combined treatment (p < 0.05), while PFOA alone reduced total egg production. Growth was higher with PFOA but decreased in the salinity and salinity + PFOA treatments. Our results suggest that P. acuta increases reproductive output when exposed to combined stressors, prioritizing egg sac production at the cost of movement and growth. These trade-offs may influence P. acuta population dynamics and freshwater ecosystem processes. While this study provides insights into the effects of PFOA and salinity, further research is necessary to understand the long-term ecological consequences in field conditions.

High Spatial Resolution Crop Type And Land Use Land Cover Classification Without Labels: A Framework Using Multi-Temporal Planetscope Images And Variational Bayesian Gaussian Mixture Model, Minh Tri Le

Mathematics, Physics, and Computer Science Faculty Articles and Research

Previous studies often combined high spatial resolution data (e.g., PlanetScope) with wider spectral range data (e.g., Sentinel-2) and relied on supervised classification methods to produce land use and land cover (LULC) maps. This study proposed a new unsupervised framework to generate crop type and LULC maps at high spatial resolution (< 5 m) using available PlanetScope data solely without requiring ground truths. We used PlanetScope surface reflectance images and their derived spectral indices during growing seasons to create multi-temporal input features, which were fed into an unsupervised Variational Bayesian Gaussian Mixture Model (VBGMM). The VBGMM, unlike the traditional unsupervised classification methods, (1) first estimated optimal parameters that are most suitable based on the input features and then (2) assigned pixels to the cluster with maximum posteriori probability of a mixture of several Gaussian distributions. The crop type and LULC maps were then generated by labeling the derived clusters using the best possible assignment method, referring to the existing crop type or LULC products. We evaluated the produced PlanetScope-based crop type and LULC maps using true labels, corresponding reference maps, and other unsupervised classification methods. The results demonstrated the robustness and effectiveness of the proposed framework in mapping crop types and LULC at 3–5 m pixels across various ecosystems, climate zones, and human-managed landscapes. The spatial patterns of PlanetScope-based maps were (1) highly comparable with all the reference datasets at 10–30 m spatial resolution and (2) better than the traditional GMM and K-means clustering methods. The VBGMM produced classification maps with high confidence, yielding class probabilities above 0.9 for over 90 % of all study areas. The area percentage for all crop type and LULC classes agreed well with their reference maps, with R2 of 0.95 and RMSE of 1.04 %. The confusion matrices using true labels indicated that PlanetScope-based maps achieved a higher overall accuracy of 84 % than the supervised referenced maps of 81 %. Besides, the entropy comparison showed that our framework-based maps were better at capturing fine-scale features such as developed areas within cities that commonly mix with open space and vegetation, deforestation and cropland conversion in South America, smallholder croplands in Africa and Asia, and generating homogeneous crop fields in North America. This study further highlighted the potential for future research to implement our proposed framework to generate timely and extensive annotated datasets, which can be used for operationally training machine learning models to map crop types and LULC, track deforestation, detect wildfires, and delineate flooded areas at larger scales using medium/coarse Earth observations.

Characterization Of The Role Of Rsd-6, A Tudor Domain Protein, In Antiviral Rnai Pathway In Caenorhabditis Elegans, Krishna Prasad Dahal

LSU Doctoral Dissertations

RNA interference (RNAi) triggered by virus produced dsRNA intermediate offers robust innate immunity against invading viruses in plants, insects and animals. Caenorhabditis elegans has been extensively used as a model organism to study host virus interaction. Recently, a forward genetic screen identified RNAi spreading deficient-6 (RSD-6), a Tudor domain protein, as a key component in antiviral RNAi pathway against Orsay virus in C. elegans but its contribution in the pathway remained unclear. Here, we generated a rsd-6 null allele using CRISPR-Cas9 genome editing, crossed it with other known RNAi mutants and analyzed Orsay virus replication in them and in combination …

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

Blanding’s Turtle (Emydoidea blandingii) is a species of concern in North America as many populations have declined across its distribution. Environmental factors associated with alteration of natural ecosystems have played a role in declines leading to many isolated populations. Most populations exist in the Great Lakes region with a number of outlying populations, such as Nebraska. Understanding the current distribution of the species will facilitate regional management strategies to protect this species into the future. In Nebraska, Blanding’s Turtles were known from 23 counties prior to this study. Herein, we report on eight new county records, five (Blaine, …

Simulated Walleye Responses To Various Harvest Regulations In A Nebraska Reservoir, Mikael Ranta, Melissa Wuellner, Keith D. Koupal

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

Recreational fishing provides sociological and economic benefits, and fish populations are managed by state agencies to meet angler expectations. One of the most popular species in Midwest reservoirs are walleye (Sander vitreus), and most of these fisheries have established restrictions on the number and sizes of fish harvested. Regulations are set based on population structure and dynamics and management goals, but often agencies have incomplete information needed to establish regulations. Predictive models can provide insight on population responses and inform managers of the tradeoffs between angler restrictions and population sustainability. Lake McConaughy, Nebraska, supports a popular walleye fishery. …

Metabolic functioning in nearly all eukaryotes relies on molecular machinery dual-encoded by mitochondrial DNA (mtDNA) and nuclear DNA (nDNA) genomes. The two genomes have sustained an extraordinary degree of cooperation across evolutionary time, preserving the capacity for indispensable processes including oxidative phosphorylation and ATP production, which in turn influence many fitness-related traits. How this cooperation is maintained when one member of the pair is debilitated by deleterious mutation is poorly understood, as is the influence of mutation location (mtDNA or nDNA), mating system, or the potentially compensatory effects of mtDNA copy number changes on the process. We asked whether and …

A Novel Microdosimetry-Based Formalism For Cell Survival Modelling Applicable To Hypofractionated Radiotherapy, Oleg N Vassiliev, Radhe Mohan

Faculty, Staff and Student Publications

Objectives

Hypofractionated radiotherapy requires reliable cell survival models for doses much higher than the standard 2 Gy, for which the linear-quadratic (LQ) model is not applicable. We developed an alternative approach applicable to both low doses and high doses used in hypofractionated treatments and radiobiological experiments.

Approach.

We combined a standard microdosimetric technique with a recently introduced non-LQ cell survival model. Our formulation accounts for cell damage by multi-track events involving any number of particles. This is necessary for modelling cell survival at therapeutic doses. We characterise each cell type by the size  of the sensitive volume (SV) and …

The differentiation and suppressive functions of regulatory CD4 T cells (Tregs) are supported by a broad array of metabolic changes, providing potential therapeutic targets for immune modulation. In this study, we focused on the regulatory role of glycolytic enzymes in Tregs and identified phosphoglycerate mutase (PGAM) as being differentially overexpressed in Tregs and associated with a highly suppressive phenotype. Pharmacologic or genetic inhibition of PGAM reduced Treg differentiation and suppressive function while reciprocally inducing markers of a pro-inflammatory, T helper 17 (Th17)-like state. The regulatory role of PGAM was dependent on the contribution of 3-phosphoglycerate (3 PG), the PGAM substrate, …

Unresolved tissue damage is a common feature of Inflammatory Bowel Disease (IBD) that facilitates disease progression. Here, we showed that high animal fat diets (HFD), an environmental risk factor associated with IBD pathogenesis, suppress intestinal macrophage production of critical tissue repair responses after damage. This includes reduced IL-23 production, which drives downstream production of IL-22, which is needed for barrier repair. Indicating that dietary lipids interfere with responses to microbial molecules needed to induce barrier protective functions, we found oleic acid could directly suppress macrophage Il23a induction after lipopolysaccharide (LPS) treatment. Deleting the lipid transporter CD36 on macrophages restored the …

Pharmacy Faculty Articles and Research

Colorectal liver metastasis (CRLM) poses a significant challenge in oncology due to its high incidence and poor prognosis in unresectable cases. Current treatments, including surgical resection, systemic chemotherapy, and liver-directed therapies, often fail to effectively target hypoxic tumor regions, which are inherently more resistant to these interventions. This review examines the potential of a novel therapeutic strategy combining irreversible electroporation (IRE) ablation and Clostridium novyi-nontoxic (C. novyi-NT) bacterial therapy. IRE is a non-thermal tumor ablation technique that uses high-voltage electric pulses to create permanent nanopores in cell membranes, leading to cell death while preserving surrounding structures, and …

Reframing Sleep Diagnostics: A Structured Clinical Guide To Polysomnography Evaluation, Elvie Zulka Kautzia Rachmawati, Mikhael Yosia

AUIQ Complementary Biological System

Polysomnography (PSG) remains the gold standard for evaluating sleep-disordered breathing (SDB) and related conditions, yet its clinical potential is often limited by overreliance on the apnea-hypopnea index (AHI) and similar summary metrics. This narrative review offers a structured, physiology-informed approach to PSG interpretation that integrates sleep architecture, arousal burden, respiratory event morphology, oxygenation patterns, and CO2 trends. Recognizing phenotypic patterns-such as REM-related and positional OSA-through sleep stage and positional stratification is essential for directing targeted therapy. We outline common interpretive pitfalls, including automated scoring errors, overlooked signal artifacts, and the first night effect, emphasizing the need for careful manual …

Dual Action Dynamics: Assessing Hypopharyngeal Cancer Cell Survival Using Combination Chemotherapy, Luke Nieman

Biology Summer Fellows

Targeting glucose metabolism is an attractive strategy for cancer treatment, as cancer cells rely more heavily on glycolysis than oxidative phosphorylation, a phenomenon known as the Warburg Effect (Wang et al 2015). Reversing this metabolic reprogramming can impair cancer cell proliferation. NG52, a small-molecule inhibitor of PGK1, has been shown to effectively reverse the Warburg Effect and reduce cancer cell viability (Wycliffe et al. 2007). Paclitaxel, commonly Taxol, is a chemotherapeutic agent that disrupts mitotic spindle function, causing mitotic arrest and apoptosis (Markman et al 2005). In this study, we aimed to evaluate the individual and combined effects of NG52 …

Synopsis

Across teleosts, feeding by biting substrate-attached prey has evolved multiple times and is associated with convergent morphologies that include a deep body and an elongated, tapered head. However, the functional role of these morphologies in substrate-biting fish is not established. Here, we tested the hypothesis that these morphologies function as control surfaces that affect feeding kinematics during biting. To test this hypothesis, we used simplified physical models of substrate-biting reef fish and examined the role of head, body, and fin morphology in determining feeding kinematics that facilitate the removal of substrate-attached prey. Models simulated the swift lateral movement of …
