Characterization Of A Fatty Acid Synthetase From Corynebacterium Diphtheriae, Herman W. Knoche, Kirston E. Koths

A fatty acid synthetase from Corynebacterium diphtheriae has been purified to a specific activity of 450 nmoles of malonyl coenzyme A incorporated per min per mg. The enzyme is optimally active in 0.5 M phosphate buffer. C. diphtheriae appears to be the most primitive organism having a multienzyme complex for fatty acid synthesis.

The effects of acyl-CoA derivatives (C8 to C20) on the activity

of the fatty acid synthetases from yeast and Corynebacterium

diphtheriae have been examined. Both enzyme

systems are inhibited by the longer chain acyl thioesters

(C16 to C20) and protected against this inhibition by bovine

serum albumin (BSA). Identical relief from acyl-CoA inhibition

is provided by the 6-0-methylglucose-containing

lipopolysaccharide (MGLP), from Mycobacterium phlei. It

is shown that MGLP forms a stable complex with palmitylCoA.

This interaction accounts for the BSA-like effects of

the polysaccharide. BSA and MGLP have two further effects

Tests indicate that the oral administration of diethyl 2-chlorovinyl phosphate (SD1836) and 2,2 dichlorovinyl dimethyl phosphate (DDVP) plastic-incorporated pellets significantly reduces the intestinal nematode parasite Sterliadochona pedispicula in wild rainbow trout. No evidence of fish toxicity was noted with the treatments and dosages tested.

Acanthobothrium urolophi sp. n. is described from a common stingaree, Urolophus testaceus, from South Australia. It differs from all other species in being apolytic and acraspedote, in having hooks 105-115 μ- long, one accessory sucker 80-90 wide on each bothridinm, and 40-72 testes in two longitudinal rows.

Plagioporus hypentelii sp. n. (Trematoda: Opecoelidae) is described from the intestine of the hogsucker, Hypentelium nigricans (LeSueur), in streams of Adams County, Pennsylvania. It most closely resembles P. serotinus Stafford, 1904, but differs from it chiefly in its less elongate shape, smaller size, smaller testes, ovary, and eggs, straight cirrus sac, in hosts and geographic location of the hosts.

In a stimulating analysis, Lack (1967, 1968a) reviewed waterfowl clutch size and egg size data, and concluded that interspecific variations in average clutch size are generally inversely related to those of relative egg size. Thus, he suggested that the average clutch size of each waterfowl species has evolved in relation to the average availability of food to the female around the time of nesting. He hypothesized that in waterfowl, relatively large eggs have probably evolved to provide the newly-hatched young with a large food supply (an idea he subsequently (1968b) questioned) or with an adequate insulating layer of fat. A yellow film of milkstone developed on a laboratory system that simulated unclean stainless steel surfaces. Cursory washing or presoiling with phospholipids and subsequent growth of Pseudomonas sp. in a milk film produced the yellow color in 1 to 2 days. Sequence of treatments, microenvironmental conditions, and microbial growth contributing to the formation of the yellow film were determined, Pseudomonas sp. was isolated from laboratory developed yellow films and from milk soil deposits on farm equipment. This program was a status study of the interaction between Las Vegas Wash, an enriched stream, and Las Vegas Bay, a wedge shaped arm of one of the world's deeper reservoirs. The program centered primarily on identification and counting of planktonic algae from several points in Las Vegas Bay. Additional work on nutrient enrichment of water samples was conducted to aid in interpretation of algal distribution related to nutrient input. In the present study, characteristics of egg masses and egg capsules of the four most abundant California littorinaceans (Lacuna marmorata, L. unifasciata; Littorina planaxis, and L. scutulata) are described. Development within the egg capsule at ambient water temperature is followed to hatching and early weligers are described to facilitate their separation from the plankton.

In the present study, information concerning morphology, ecology, fertilization and larval development of three sympatric species of Eusonus is presented. Morphological similarities are reviewed in part I. These similarities correspond with significant lack of variation found in larval development (part IV). Despite these similarities, as well as the ability to cross fertilize their species, the ecological and reproductive isolating mechanisms presented in part II and II provide a positive indication of the existence of three biologically distinct species. A comparative discussion reviews the efficacy of morphological criteria in differentiating the three closely related Euzonus species.

The importance of showing that Drosophila are capable of learning an instrumental response would, with the information already known from genetic studies, enable researchers to possibly isolate the genetic basis of a simple learning process and thus gain greater insight into this phenomenon. If a reliable technique can be found for earning in Drosophila, a new field of research would be opened which could greatly add to our knowledge of the evolution of behavior.

The following two studies attempted to demonstrate that Drosophila can learn and also investigated which system, vision or olfaction, serves best in this capacity.

The purpose of the study was to determine through electromyography whether a non-pathological fault, such as lateral asymmetry at the pelvic level, would hinder, in significant manner, efficient functioning of the body.

On January 1, 1973, 10 soil samples were collected from areas in Coles County, Illinois, in an effort to isolate soil amoebae. All 10 soil samples were cultured on bacto-nutrient agar and colonies of amoebae genera isolated were Acanthamoeba and Hartmannella. Amoebabacteria cultures prepared from soil isolates were injected into each of 10 adult mice which were sacrificed at the end of the nine days and examined for evidence of infection and pathogenicity. The herbicide glyphosate (MON-0139), an isopropylamine salt of N- (phosphonemethyl)glycine, was tested to determine its effects on the growth of the fresh water green alga Selenastrum capricornutum. Experiments were carried out to determine algicidal, algistatic, and inhibitory concentrations of the compound.

Concentrations from 5 ppm to 1000 ppm were inhibitory to the growth of the alga, but the effects were algistatic, rather than algicidal. At concentrations from 0.1 ppm to 1.0 ppm the growth of the alga was stimulated by the presence of the herbicide, while at concentrations from 0.001 ppm to 0.05 ppm the effects of the herbicide were …

Eighty gizzard shad (Dorosoma cepedianum) from Lake Charleston, Charleston, Illinois were examined for parasites. Eight fish were parasitized. Three fish contained the acanthocephalan Tanaorhamphus longirostris, three fish harbored cestodes of the genus Glaridacris and two fish had the nematode Camallanus oxycephalus. The latter is here reported for the first time from gizzard shad. Twenty fish were infected with the fungus Saprolegnia sp. The numbers of helminth parasites harbored by shad are less than reported for other forage fish. However, the incident of parasitism reported in this study is greater than that reported by previous workers.

Holothurin, a biotoxic principle from the Cuvierian glands of the Bahamian sea-cucumber, Actinopyga agassizi, was studied as an anti-tumor agent capable of retarding tumor growth and prolonging the life of tumor-bearing mice. White Swiss mice injected with Sarcoma 180 had a mean survival time of 18.1 days with an average weight gain representing tumor and ascites fluid accumulation amounting to 25.1 grams in 15 days. Of five white Swiss mice with Sarcoma 180 receiving 0.15 mg Holothurin every other day, one was alive at 57 days and the average weight gain of the group was 10.4 grams in 15 …

Estuarine grass shrimp (Palaemonetes) are common along the Gulf and Atlantic coasts (Faxon 1879, Gunter 1950, Williams 1965, Fleming 1967). These small decapods are found ranging through freshwater areas, brackish estuaries and into inshore saline waters where they are occasionally the predominant animal (Gunter 1950).

A study on seasonal changes in the nutritional components of standing plankton biomass was made from 20 April 1965 to 6 September 1966. Plankton were separated into net plankton and nannoplankton fractions. Nannoplankton3 standing biomass exceeded by 72 times that of net plankton.

Although protein and carbohydrate levels were higher in net plankton on a unit weight basis, total nutrients available from nannoplankton were substantially higher. Some seasonal trends were apparent as changes in the standing biomass in net plankton. Nannoplankton exhibited no seasonal trends. Local burn therapy usually includes the application of medicated ointments and dressings in order to minimize and eradicate infection. As this approach to local therapy may present problems regarding application and removal techniques, it was the purpose of this study to formulate a topical dosage form that may be applied to denuded areas of the skin without inunction. Using the concept of topical films, a dosage form was formulated that is highly water soluble, flexible , and adherent to both denuded and intact areas of the skin.

Numerous investigators have noted that individuals vary widely in their response to the anticoagulant, heparin. Animal model3 have suggested that patients with depressed renal function would be expected to clear heparin from the blood at a slower rate than patients with normal renal function.

Utilizing the Activated Coagulation Time clotting test, a method was developed to determine the anticoagulant half-life of heparin. The study calculated the half-life of heparin in a group of individuals with normal renal function and a population of patients in chronic renal failure. Taxonomic studies involving only adult forms of organisms may not necessarily provide complete information about differentiation of species or about the evolutionary relationships between species grouped in higher taxa. The taxonomic importance of embryonic or larval morphology has been limited only by the lack of detailed comparative morphological work on development. Fretter (1967) has shown that larval shell characteristics are reliable for taxonomic identification of some British prosobranchs. Camptotheca acuminata Decaisne (Nyssaceae) is endemic to China. The plant was brought to the United States only on a few occasions and thus can be considered rare in this country. A search of the plant kingdom for species that produced anticancer substances revealed that ethanolic extracts of Camptotheca acuminata had such a property. The active constituent was identified eventually. Further testing and possible marketing of the active principle, and alkaloid, (camptothecin) depended on a combination of factors: obtaining the plant from China, organization of a mass planting program in the United States or chemical synthesis of the drug.

The present study has attempted to investigate some of the more basic questions concerning the toxicity and accumulation of mercury in the purple shore crab, Hemigrapsus nudus. This rocky intertidal denizen is found in abundance along the entire Pacific coast of the United States from Sitka, Alaska, to the Gulf of California where it dominates the mid-tide pool region. The molecular nature and replicative behavior of R factor 222 was examined in Proteus mirabilis . In deoxyribonucleic acid (DNA} from R+ P. mirabilis , R factor 222 was identified by CsCl density gradient centrifugation as 2 satellite DNA bands at densities corresponding to 50 and 58 moles percent guanine plus cytosine (% GC) . Studies were conducted in DUB/ICR mice with minute virus of mice (MVM), a single stranded DNA virus, and Colorado tick fever (CTF) virus, a double stranded RNA Virus, to determine if CTF virus had teratogenic potential and to characterize further MVM and its teratogenic effects. MVM is a known teratogen, whereas CTF virus had never been investigated previously for teratogenic potential.
