The purpose of this investigation was to study ultraviolet radiation effects on rotifers. The rotifer studied was Philodina acuticornis odiosa Milne. Isolation cultures were used to grow the rotifers individually in separate U-plate depressions. The source of ultraviolet radiation consisted of four mercury germicidal lamps used in combination to deliver a uniform flux of 315 uW/cm2 over the sample. Five groups of rotifers, one control and four experimental groups of 96 each, were used. The rotifers used were 2-10 hours of age at the beginning of the experiment. The experimental groups were exposed to uv radiation for 3 minutes, …

Partial Purification & Kinetic Properties Of Nicotinamide Adenine Dinucleotide Phosphate-Dependent Alcohol Dehydrogenase Of Phycomyces Blakesleeanus, Thomas Hartz
Masters Theses & Specialist Projects
NADP+-dependent alcohol dehydrogenase (alcohol:NADP+:oxidoreductase, EC 1.1.1.2.) was partially purified from a cell-free extract of Phycomyces blakesleeanus (+ mating type, Vanderbilt strain). The fungus was grown in shake cultures containing glucose-asparagine broth, and crude cull-free enzyme extracts were prepared. The crude enzyme extracts were partially purified by a combination of ammonium sulfate precipitations and heat precipitation. Purification resulted in a 40;5 recovery of the enzyme with an increase in specific activity from 0.64 units per milligram of protein to 0.25 units per milligram. The partially-purified enzyme extracts were used to determine the kinetic properties and substrate specificity …

A Chronological Comparison Of The Vegetation Of Vigo County, Indiana Presettlement (1814) Versus Contemporary (1974), Ellen Oliver Donselman
All-Inclusive List of Electronic Theses and Dissertations
A study of the vegetation of Vigo County was undertaken to categorize and map presettlement and contemporary vegetation patterns, and to relate these patterns to climate, topography, soil types and land-use changes. Vigo County was divided into four major topographic units according to geography and soil types. Ttese units served as the basis for this study. The General Land Office Survey of 1814 provided data for the presettlement forest distribution. Sixteen contemporary stands were sampled to obtain data for characterization of present-day vegetation patterns. Standattributes tables and size-class tables were constructed for four contemporary and four presettlement forest units. A …

Studies Of N⁵, N¹⁰-Methylene Tetrahydrofolate Reductase From Porcine Kidney And Mouse L1210-Induced Tumor Tissues, Purification And Interaction With Antifolates, David W. Jayme
Theses and Dissertations
Methylene tetrahydrofolate reductase has been purified 1000-fold from porcine kidney and 400-fold from mouse L1210-induced tumor tissue by classical methods. The enzyme preparations have been demonstrated to be essentially free of contaminating methionine synthetase and serine transhydroxymethylase activity. Studies of the kinetic properties of the kidney and tumor enzymes, with respect to the reverse reaction using N5-methyl tetrahydrofolate as the variable substrate, have indicated Km values of 2.0 and 2.4 x 10-4 M, respectively. Inhibition of this key branch point enzyme in folate metabolism by a number of antimetabolites indicates that several of these antifolate compounds exhibit enzyme inhibition superior …
Lipid Metabolism During Bacterial Growth, Sporulation, And Germination: Differential Synthesis Of Individual Branched And Normal-Chain Fatty Acids During Spore Germination And Outgrowth Of Bacillus Thuringiensis, Kenneth W. Nickerson, Lee A. Bulla Jr., Timothy L. Mounts
Mounts
Papers in Microbiology
The biosynthesis of individual branched- and normal-chain fatty acids during Bacillus thuringiensis spore germination and outgrowth was studied by comparing pulsed and continuous labeling of these fatty acids with [U- 14C ]acetate. The relative specific activity of each fatty acid varies with time as the cell progresses through outgrowth. However, fatty acid synthesis does occur in two distinct phases. Upon germination, acetate is incorporated only into the iso-isomers i-C13, i-C14, and i-C16; no normal or anteiso synthesis occurs. Subsequent to T30, the full complement of branched- and normal-chain homologues is formed …

Biology ETDs
Asymmetric RNA was synthesized in vitro from SV4O form I DNA using Escherichia coli DNA-dependent RNA polymerase. Using this RNA, sheared 32P-SV4O DNA was separated into (+) and (-) DNA strands. (+) and (-) DNA was shown to be complementary strands of SV4O DNA. The separated strands were hybridized to nuclear, informosome, and polysome RNAs extracted at different times in the lytic infection, and to similar RNAs from transformed cells. Further, the size of the RNAs hybridizing to (+) and (-) DNAs was determined by sucrose gradient sedimentation. The results showed:
(1) Nuclear, informosome and polysome RNA from transformed …

"Natural" or unaltered foods have become a fad in recent years. This includes the drinking of unpasteurized cow's milk as well as that designated "Certified." This report is made to call attention to a high prevalence of antibodies to toxoplasma in dairy as compared with range cattle. Previous to this study, few papers have considered the breed or conditions of management of cattle sampled for toxoplasma antibodies (McIlwain, 1969, Arch Environ Health 19: 885-886; Vanderwagen et aI., 1974, J Am Vet Med Assoc 164: 1,034-1,037).

Application Of Satellite Data And Lars' Data Processing Techniques To Mapping Vegetation Of The Dismal Swamp, Jeffrey Allan Messmore
Biological Sciences Theses & Dissertations
This study concerned the feasibility of using digital satellite imagery and automatic data processing (ADP) techniques as a means of mapping swamp forest vegetation. Multispectral scanner data acquired by the Earth Resources Technology Satellite (ERTS-1; renamed LANDSAT-1) was analyzed using ADP techniques developed by Purdue University's Laboratory for Applications of Remote Sensing (LARS). The site for this investigation was the Dismal Swamp, a 210,000 acre swamp forest located south of Suffolk, Va. on the Virginia-North Carolina border. Two basic classification strategies were employed in determining the vegetation mapping capability of ERTS-1 data. The initial classification utilized unsupervised techniques which produced …
