Microbial Characteristics Of An Activated Sludge And Three Commercial Preparations After Exposure To Phenolics, Laurie Gneiding
Microbial Characteristics Of An Activated Sludge And Three Commercial Preparations After Exposure To Phenolics, Laurie Gneiding
Theses
The biological characteristics of four microbial mixtures were examined after successive exposure to phenol and 2-chlorophenol at room temperature. One of the microbial mixtures was an activated sludge from the Livingston, NJ municipal wastewater treatment plant. The other three mixtures were commercial preparations sold by Polybac Corp.(Hydrobac), Sybron Corp. (BI-CHEM DC 1006/1007), and General Environmental Sciences(LLMO).
The characteristics examined were relative counts and species of gram negative/gram positive bacteria, yeast and fungi, protozoa, and higher organisms. For the gram negative bacteria and yeasts, culturing techniques and diagnostic tubes were used to determine the predominant species. Biochemical tests were done on …

Parameters Of Growth In The Embryonic And Neonatal Chick Basilar Papilla, Douglas A. Cotanche, Kathleen K. Sulik
Scanning Electron Microscopy
The growth of the basilar papilla in the chick cochlear duct was studied utilizing light, scanning, and transmission electron microscopy. The ages of the cochleae investigated ranged from embryonic day 6 to post-hatching day 7. The changes in the length and width of the basilar papilla as well as the establishment of its spatula-like shape were correlated with the maturation of the hair cells' apical surfaces and the changes in the cellular organization of the sensory epithelium. The histological reorganization of the distal hair cell nuclei was concomitant with the broadening of the distal region of the basilar papilla and …

Energy Dispersive X-Ray Analysis Of Ocean Ferromanganese Crusts Using Conventional Zaf Corrections, R. F. Commeau, J. A. Commeau, F. W. Brown, F. T. Manheim
Scanning Electron Microscopy
Ocean ferromanganese crusts are composed of interlayered phases of manganese oxide, iron oxide and oxyhydroxide, calcium carbonate/phosphate, silica, and alumino-silicates. These interlayers are so thin and fine-grained that each phase could not be isolated under the beam of a scanning electron microscope for quantitative x-ray microanalysis. A test was conducted to determine if the grain size of the phases was small enough to allow conventional ZAF techniques to be used without serious errors in the results. A "synthetic" ferromanganese crust was prepared by pelletization of a 1:1 mix of two fine-grained (<5 >micrometer) components. The mean of the energy dispersive …5>

An Ultrastructural Analysis Of The Physical Organization Of Collagenous (Type I) Matrices: One Determinant Of Urothelium Maintenance In Vitro, G. M. Hodges, C. Rowlatt, A. R. Howlett, L. K. Trejdosiewicz
Scanning Electron Microscopy
Collagenous matrices, used as cell culture substrata, can be prepared from different collagen types in a variety of forms using a range of polymerization procedures. Type I collagen has been most frequently used either as dried collagen films or hydrated collagen gels. Sheets of isolated bladder urothelium, when plated onto such matrices prepared from type I collagen by different polymerization methods (eg. air-drying; NaOH; NaCl; NH3; or NH3 followed by glutaraldehyde crosslinking) demonstrate the capability of urothelial cells to attach to a variety of differently prepared matrices irrespective of polymerization procedure. In contrast, both cell proliferation and …

The Influence Of Different Cryopreparations On The Distribution Of Ions In Bullfrog Myocard Cells, R. Meyer, M. Schmitz, K. Zierold
Scanning Electron Microscopy
Bullfrog heart muscle trabecula are shock-frozen in liquid propane cooled by liquid nitrogen and then processed for X-ray microanalysis in two different ways: 1. Freeze-drying followed by vacuum embedding. 2. Cryoultramicrotomy and freeze-drying.
Stained sections of freeze-dried, embedded tissue exhibit detailed ultrastructure, but are useless for X-ray microanalysis. Unstained, dry cut plastic-sections are suitable for X-ray microanalysis, but the ultrastructure appears faint. Higher electron optical contrast and peak-to-background ratio of X-ray spectra are generally obtained in freeze-dried cryosections. Both preparation methods show that the X-ray spectra are influenced by the quality of cryofixation. The phosphorus/potassium ratio in nuclei increases with …

Advances In Electron-Beam-Induced-Current Analysis Of Integrated Circuits, J. D. Schick
Scanning Electron Microscopy
Electron-beam-induced-current (EBIC) analysis of integrated circuits and of individual semiconductor devices has become an essential technique for semiconductor device characterization as well as for failure analysis and diagnostics. This tutorial is an update of the paper presented in SEM/1981/I on the same topic and discusses advances and new trends in the applications of EBIC in recent months. Examples of EBIC measurements on high density chips, ion-implanted junctions, and shallow diffusions in thin epitaxial layers are given. Time resolved EBIC (TREBIC) is also explained and new digital readout techniques, including computer-aided data analysis, are described as they refer to detailed analyses …
Computer-Assisted Methods For Design Optimization Of Cardiac Bioprosthetic Valves, Mohamed S. Hamid, Hani N. Sabbah, Paul D. Stein
Computer-Assisted Methods For Design Optimization Of Cardiac Bioprosthetic Valves, Mohamed S. Hamid, Hani N. Sabbah, Paul D. Stein
Henry Ford Hospital Medical Journal
Excessive mechanical stresses on valvular leaflets are considered an important factor in the degeneration of cardiac porcine bioprosthetic valves. This study describes a computer-assisted numerical model of a bioprosthetic valve which estimates the magnitude and distribution of leaflet stresses. The study also describes our initial attempts to determine the influence of stent height upon leaflet stresses. Whereas lowering stent heights has been suggested to improve hemodynamics in the bioprosthetic valve region, results indicate that an increase in leaflet stresses can accompany reductions of stent height.

Computer Applications In The Vascular Registry Of Henry Ford Hospital: Use Of A Simplified Access Method, Joseph P. Elliott Jr., Kunwar Kapoor, John G. Hageman, D. Emerick Szilagyi, Roger F. Smith, Michalene Mcpharlin
Henry Ford Hospital Medical Journal
The Vascular Registry of Henry Ford Hospital was initiated by one of the authors in 1957 to provide a consistent and accurate method of documenting and correlating the clinical results of the treatment of occlusive and aneurysmal arterial diseases. Data collection for each patient includes demographic information, treatment of clinical problems, and followup observations. Initially, the system consisted of keypunch cards and eventually magnetic tape processing. By 1980, it was necessary to further update the system because of the increase in patient volume, the need to minimize manual manipulations, and the demand for research data. The present system uses institutional …

Endometrium Cell Surface Abnormalities In The Syrian Hamster As A Result Of In Utero Exposure To Diethylstilbestrol, J. Gilloteaux, A. W. Steggles
Scanning Electron Microscopy
Scanning electron microscopy (SEM) was used to observe changes in the hamster endometrium cell surface following in utero pre- and/or postnatal exposure to diethylstilbestrol (DES). Some of the changes in cell surfaces are associated with alterations in cell sizes and shapes (from columnar to cuboidal and/or squamous) and in microvilli and mucous secretion. In all cases, DES treated uteri show mucosal cell surface pleomorphism, apocrine secretion and cystic accumulation of secretory material. Microvillous pleomorphism and peculiar linkages attaching one microvillus to others were investigated. Although the function and nature of such linkages is unclear, their presence seems to be more …
The Role Of The Vessels In The Growth Plate: Morphological Examination, K. Draenert, Y. Draenert
The Role Of The Vessels In The Growth Plate: Morphological Examination, K. Draenert, Y. Draenert
Scanning Electron Microscopy
Combined methods of light microscopy to present ossification zones by means of fluorochrome dyes make it possible to explain the contradicting presentations of the vascular system of the growth plate in the literature. The vascular system was casted with methacrylates which can be presented in the scanning electron microscope 3-dimensionally together with the trabeculae as a result of their resistance in the electron beam.
The 3-dimensional presentation in the electron microscope allows a clear distinction between the various vascular sections in the arterial flow system. In the microcorrosion casts the vessels of the epiphyseal side of the growth plate can …

Effects Of Dehydration On Hemoglobin Oxygen Affinity And Blood Cell Volume In Two Anurans, Andrew Christopher Zygmunt
Effects Of Dehydration On Hemoglobin Oxygen Affinity And Blood Cell Volume In Two Anurans, Andrew Christopher Zygmunt
Dissertations and Theses
Two aspects of possible adaptation in cardiovascular performance caused by increased plasma electrolytes were examined. Cells in anisotonic plasma may either act as osmometers or volume regulate. Blood flow rate is dependent upon cell viscosity, which in turn is a consequence of cell volume and membrane deform-ability. Cell volume changes which increase membrane deform-ability will thus potentially extend the limits of dehydration tolerance. It was found in R. catesbeiana and B. marinus that red blood cell is maintain constant volume during dehydration. Cells in vitro initially lose water, but then sodium, potassium and water move into the cell. Cell viscosity …
In Vitro Glucose Utilization In Ob/Ob Mouse Erythocytes & Adipose Tissue, Peggy Fudge
In Vitro Glucose Utilization In Ob/Ob Mouse Erythocytes & Adipose Tissue, Peggy Fudge
Masters Theses & Specialist Projects
An in vitro study of ob/ob mouse and control mouse erythrocytes and adipose tissue showed a difference in the rate of glucose utilization as determined by the production of 3H20. In both ob/ob and control erythrocytes, glucose utilization increased as glucose concentration of the medium was increased, although the ob/ob rate was consistently lower than the control. The similar trend except for the adipose tissues showed a ob/ob adipose tissue at 200 mg/d1 glucose concentration which exhibited a rate less than the rate at 100 mg/d1.
The decreased glucose utilization by adipose tissue in the absence of …

Studies On The Binding Of Human Chorionic Gonadotropin To Polystyrene & A Possible Application, Patrick Mowers
Studies On The Binding Of Human Chorionic Gonadotropin To Polystyrene & A Possible Application, Patrick Mowers
Masters Theses & Specialist Projects
Human chorionic gonadotropin (hCG), labeled with 125I, was used to study the binding properties of hCG to polystyrene beads. The binding capacity of the 0.25-in diameter beads was determined to be approximately 300 mIU/bead. The affinity constant, K, was determined to be essentially O, suggesting that binding is strictly passive. The binding of hCG to polystyrene was proportional to the hCG concentration and temperature. There were highly significant (p<0.001) differences among amounts of hCG bound at 3 C, 25 C and 37 C. There was a highly significant (p<0.001) relationship between the amount of hCG bound and the initial hCG concentration. There did not appear to be any correlation between the percentage of hCG bound and the initial hCG concentration. Attempts to use Staphylococcal Protein A-peroxidase conjugate to detect murine monoclonal antibodies to beta-hCG were not successful.

Toxicological Studies On Aquatic Contaminants Originating From Coal Production And Utilization: The Induction Of Tolerance To Silver In Laboratory Populations Of Fish And The Chronic Toxicity Of Nickel To Fish Early Life Stages, Wesley J. Birge, Jeffrey A. Black, James F. Hobson, Albert G. Westerman
KWRRI Research Reports
Aquatic toxicity studies were performed on two important coal-derived contaminants, silver and nickel. Silver was investigated with regard to metal-induced tolerance in laboratory populations of the fathead minnow (Pimephales promelas). Fish were exposed to acute silver concentrations following acclimation to sublethal exposures of this metal. Based on median lethal times (LT50), animals which had received 14 days prior exposure to 1.5 and 15 μg Ag/L were three to four times more resistant to silver than were previously unexposed organisms. This metal-induced resistance was not a sustained response. After organisms which had been acclimated to 15 μg/L had been transferred to …
An Evaluation And Comparison Of Various Diversity Indices As Illustrated By Portage Creek, Karl Josef Siebert
An Evaluation And Comparison Of Various Diversity Indices As Illustrated By Portage Creek, Karl Josef Siebert
Masters Theses
The diversity index is an established method of measuring water quality. In this study various diversity indices are evaluated for their use in a rapid, accurate, and cost effective biological monitoring program. The study investigated five diversity indices: Approximate, Brillouin's, Shannon's, Simpson's and the Sequential Comparison Index. The diversity indices were tested for accuracy and sensitivity on actual on actual macroinvertebrate samples and simulated data.
The Simposon's Index was found to provide the most accuracy and sensitivity for the indices tested. The importance of considering the evenness element of diversity indices was also supported by the study.

A Restriction Analysis Of Klebsiella Pneumoniae Phage Sbs, Heidi A. Walters-Khan
Loma Linda University Electronic Theses, Dissertations & Projects
A Klebsiella phage was isolated from the San Bernardino sewage and its physiochemical properties analyzed. The present study found the density of the phage to be 1.44 g/ml, the burst size to be 10, and the phage had a latent period of 20-25 minutes. The DNA was found to be double stranded and linear. Its molecular weight was 41.66 + 0.72 kilobase pairs. The DNA was digested with a battery of endoncleases and partial restriction maps determined. The potential of the phage as a vector for Klebsiella is discussed in the text.
