An Investigation Of Salinity Fluctuations In Soils Of A Northern Utah Marshland, Gary Roy Newman
An Investigation Of Salinity Fluctuations In Soils Of A Northern Utah Marshland, Gary Roy Newman
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Ogden Bay Waterfowl Management Area is one of many marshlands bordering the Great Salt Lake that are extremely important as breeding habitats for North American waterfowl. Salinity, both of surface and interstitial waters, is an important factor in maintaining a suitable habitat within the marsh. For this reason, a study was undertaken to determine the factors affecting the salinity of surface and interstitial waters. A small (approx. 100 acre) subunit was studied extensively to determine; 1) variations in surface water salinity with flowrate and water level; and 2) variation in interstitial water salinity with depth and location within the subunit. …

Motility Of The Microtubular Axostyle In Pyrsonympha, George M. Langford, Shinya Inoué
Biology - All Scholarship
The rhythmic movement of the microtubular axostyle in the termite flagellate, Pyrsonympha vertens, was analyzed with polarization and electron microscopy. The protozoan axostyle is birefringent as a result of the semi-crystalline alignment of approximately 2,000 microtubules. The birefringence of the organelle permits analysis of the beat pattern in vivo. Modifications of the beat pattern were achieved with visible and UV microbeam irradiation. The beating axostyle is helically twisted and has two principal movements, one resembling ciliary and the other flagellar beating. The anterior portion of the beating axostyle has effective and recovery phases with each beat thereby simulating the flexural …

In Vitro Experiments On Totipotency Of Dacus Carota, Karl L. Ruser
In Vitro Experiments On Totipotency Of Dacus Carota, Karl L. Ruser
Journal of the Minnesota Academy of Science
The intent of this project was to induce the totipotency of carrot cells in tissue culture. The information gathered would be applied to work with woody plants. Differentiated cells removed from the root were cultured in vitro using a modified White's medium. Callus was initiated and successfully sub cultured. Roots and the production of chlorophyll have been initiated in the cultures. Because some specialized equipment and facilities for tissue culture were not available at the beginning of the project, it took considerable time to gather and improvise them. A roller tube culture mixer was constructed, using the mechanism from a …
Blue-Green Algal (Cyanophyta) Toxins, Charles P. Mason
Blue-Green Algal (Cyanophyta) Toxins, Charles P. Mason
Journal of the Minnesota Academy of Science
In the past toxin production by blue-green algae has been demonstrated by feeding large quantities of algae to vertebrates and observing symptoms leading to animal death. It was the purpose of this investigation to develop a more convenient way to test for the presence of an alga toxin using instead of vertebrates, other algae and those organisms with which it lives and competes in fresh water habitats. Using axenic algae cultures and three different agar media MBL, DAA and NA, the latter to check for bacterial contamination, algae were streaked on the agar surface in petri dishes to form a …

Publications
Tolerance to high moisture conditions in the soil was compared for eighteen crop species, and factors causing differences among species were studied: 1) When the moisture content of a soil was kept high, the soil air contained 10.5 - 17.5% of O2 and 5.0 - 9.8% of CO2, and the soil solution was not very high in Fe, Mn or NO2-N. 2) Cruciferae, Solanaceae, sugar beet, field bean, pea, cucumber and Chrysanthemum coronarium were very susceptible to this condition, but maize, soybean, and onion were tolerant. Rice grew better under this condition than under ordinary upland conditions. 3) Tolerance to …

Probing Mitotic Chromosome Structure And Arrangement Using Micromanipulation, Bruce R. Korf
Probing Mitotic Chromosome Structure And Arrangement Using Micromanipulation, Bruce R. Korf
Student Theses and Dissertations
The Indian deer, (Muntiacus muntjak), has the lowest known diploid chromosome number (seven in the male) for a mammal. Mitotic chromosomes can be removed from cells either by extraction with microneedles or by inducing chromosome expulsion with a drop of silicone oil applied to the cell. The entire diploid complement is removed by virtue of interconnecting fibers among the chromosomes. The chromosomes are brought to the surface of the cover slip for analysis, where each can be identified morphologically with phase contrast optics. The chromosomes are sticky and extensible, and parts of chromosomes can be dissected by cutting with microneedles. …
Studies On Chloroplast Protein Synthesis In Vivo And In Vitro, Karl Stanley Matlin
Studies On Chloroplast Protein Synthesis In Vivo And In Vitro, Karl Stanley Matlin
Student Theses and Dissertations
Protein synthesis in the Chlamydomonas reinhardtii chloroplast was investigated by in vivo and in vitro approaches. The site of synthesis of chloroplast proteins was examined to determine how many proteins were synthesized inside the chloroplast. Cells were labeled in vivo with [35S]-sulfate in the presence of either. chloroplast or cytoplasmic protein synthesis inhibitors, and fractionated into soluble proteins, thylakoid membranes, and chloroplast ribosomal subunits. The use of high specific activity sulfate and improved gel electrophoretic techniques were key features of this study because they permitted minor chloroplast products to be detected for the first time. It was determined …

Fire-Decay: Interactive Roles Regulating Wood Accumulation And Soil Development In The Northern Rocky Mountains, A. E. Harvey, M. J. Larsen, M. F. Jurgensen
Aspen Bibliography
Decay and fire play interactive roles in recycling wood and other organic materials in forest ecosystems, and contribute to the development of high quality soils in the Northern Rocky Mountains. Decayed wood, charcoal, and other decomposed organic matter are the principal media for ectomycorrhizal and nonsymbiotic nitrogen fixing microbes. The activities of these microbes are critical to the growth of forest trees. The balance between decay and fire, as it affects the amount, distribution, and type of organic matter, controls the ability of forest soils to support the growth of trees.
A Comparison Of Cheddar Cheese Yields And Composition Using Conventional Bulk Set Cultures And Superstart Concentrated Cultures, William J. Kipp
A Comparison Of Cheddar Cheese Yields And Composition Using Conventional Bulk Set Cultures And Superstart Concentrated Cultures, William J. Kipp
Electronic Theses and Dissertations
The cheese industry has become by far the largest user of milk for manufacturing. In 1978, cheese production utilized a fourth of the total market supply of milk, up from less than an eighth in 1960 (4). Rising consumer incomes and changing lifestyles and eating habits have increased cheese consumption and sales by 60% since 1960 (21). In the last ten years, annual per capita consumption of cured cheese has increased from 11.6 lb to 17.1 lb (45). Economists state that the cheese market will continue to expand through the 1980's (7, 21, 68). Manufacturing cultured dairy products can be …
Electrophoretic Analysis Of Blood Serum Proteins In Three Species Of Water Snakes (Genus Nerodia), Phyllis J. Garnett
Electrophoretic Analysis Of Blood Serum Proteins In Three Species Of Water Snakes (Genus Nerodia), Phyllis J. Garnett
Journal of the Arkansas Academy of Science
Serum from three species of water snakes (Nerodia rhombifera, N. erythrogaster and N. fasciata) from one geographic region was analyzed electrophoretically on cellulose acetate, and anodic mobility and relative concentration of the fractions were determined by a recording densitometer with an automatic integrator. Classification of fractions was based on mobility (Rf, values), and for identification purposes, bands were labeled in order of decreasing mobility (albumin and alpha₁, alpha₂, alpha₃, beta₁, beta₂, gamma₁, and gamma₂ globulins). Seven fractions were identified in each species with alpha₃ being absent from N. rhombifera and N. erythrogaster, and only one gamma fraction was observed in …

Studies Of The Lysogenic Response As A Function Of Multiplicity Of Infection Of Lambdoid Bacteriophages, Mark Floyd Fedyk
Masters Theses
The lambdoid bacteriophages λimmλ, λimm434 and λcI are ideal subjects for the study of gene product interactions at the molecular level due to their closely related genetic makeup. Previous work in this laboratory has indicated the possibility that phage λimm is sensitive to phage λimm434cI gene product and that phage λimm434 is sensitive to phage λimmλcI gene product. The purpose of this research was to determine if the frequency of lysogeny for phage λimmλ is affected by the multiplicity of coinfection with the λimm434 phage. By comparing the graph of …
The Biodegradation Of Crude Oil By Bacteria Isolated From Brine Pits, Larry Williams
The Biodegradation Of Crude Oil By Bacteria Isolated From Brine Pits, Larry Williams
Masters Theses
The biodegradation of crude oil by bacteria was studied. Hydrocarbon-utilizing bacteria were isolated from sixteen brine pits from an oil producing region of Clark County, Illinois. The bacteria were capable of growth from the vapors arising from crude oil as a sole carbon source. Some strains of bacteria were capable of emulsifying crude oil, apparently through the production of an emulsifying agent(s) that was found to be heat stable and cell bound but which can be partially stripped from the cell by autoclaving. The bacteria appear to belong to the genus Arthrobacter. The chemical environment of the pits was …
The Effect Of Sodium Hexametaphosphate On Cottage Cheese Yields, Stephen T. Dybing
The Effect Of Sodium Hexametaphosphate On Cottage Cheese Yields, Stephen T. Dybing
Electronic Theses and Dissertations
A major concern in the cottage cheese industry is the inefficient use of the available milk proteins. Currently, the methods of cottage cheese manufacture convert an average of only 74.9% of the milk proteins into cottage cheese, while the remainder are removed in the by-product whey (43, 56). As protein is the main determinant of both the yield and nutritional value of cottage cheese, if 25% of the available protein is lost to the whey, then potential cottage cheese yield and nutrition have been lost as well. The proteins lost in the whey are mainly -lactalbumin and β-lactoglobulin, which remain …

The Effects Of Ph On Chloride Flux In Fundulus Heteroclitus, Timothy Alton Washburn
The Effects Of Ph On Chloride Flux In Fundulus Heteroclitus, Timothy Alton Washburn
Master's Theses
The effects of increasing hydrogen ion concentration on rate of chloride influx and efflux were studied in the killifish, Fundulus heteroclitus, adapted to fresh water. Because increasing hydrogen ion concentrations have been shown to inhibit sodium influx and because evidence exists to support coupling of chloride transport with sodium transport, the present study was undertaken to determine what effect pH would have on rate of chloride flux. Chloride efflux rates were studied after the addition of sodium bicarbonate to the medium. No significant pH-dependent changes in efflux rate were observed, except at very low pH. Influx rates were significantly changed, …

Staphylococcal protein A and several different immunoglobulins have been radio-labeled to high specific activities (> 106 cpm/pg) by reductive methylation with tritiated (3H) sodium borohydride. The proteins retain excellent functional and antigenic properties. The utility of these reagents in a variety of assays for cell surface antigens is illustrated. The results indicate that this radiolabeling procedure may become the method of choice for many cell surface and solution immunoassays.
