Seasonal Variation In Population Location Of The Galapagos Tortoise, Geochelone Elephantopus Vandenburghi, On Volcan Alcedo, Isabela Island, Galapagos Archipelago, Kent R. Beaman

Loma Linda University Electronic Theses, Dissertations & Projects

Twenty tortoises, 10 adult males and 10 adult females, were tagged with radio transmitters. Numbers were painted on each carapace to aid in visual sighting. The tortoises were released, and their movement was monitored over a one year period.

The percent recovery of tagged tortoises along with the numbers of non-tagged tortoises counted in sections of the study area, were used to determine tortoise population movement and location.

Weather data were collected and observations were made concerning the vegetation. These observations were compared with those from previous studies to see what impact seasonal climatic conditions and food availability had on …

Isolation And Characterization Of Temperature-Sensitive Protein Synthesis Mutants Of Escherichia Coli By Directed Mutagenesis Of The Defective Bacteriophage Lambda Fus2, Kenton L. Lohman Dec 1985

Isolation And Characterization Of Temperature-Sensitive Protein Synthesis Mutants Of Escherichia Coli By Directed Mutagenesis Of The Defective Bacteriophage Lambda Fus2, Kenton L. Lohman

Electronic Theses and Dissertations

Mutagenesis of the defective transducing bacteriophage lambda fus2 was used to isolate a collection of temperature-sensitive mutants of E. coli in the major ribosomal protein gene cluster. Four mutants were examined in detail. Two of the mutants were resistant to the ribosomal antibiotics neamine and spectinomycin. Another mutant was defective in 50S ribosomal subunit assembly at 42(DEGREES)C. The 30S subunit proteins S17 and S19 were changed in two different mutants. Each protein migrated as a more basic species in two-dimensional gels of ribosomal proteins. Ribosomes from each of the four mutants examined showed a temperature-dependent reduction in translational activity in …

Microbial Characteristics Of An Industrial Mixed Liquor After Exposure To Phenolic Compounds, Patricia L. Boyle Sep 1985

Microbial Characteristics Of An Industrial Mixed Liquor After Exposure To Phenolic Compounds, Patricia L. Boyle

Theses

The microbial characteristics of a mixed population from the Passaic Valley Sewerage Commissioners (PVSC) Wastewater Treatment Plant in Newark were examined. This plant handles about 250 million gallons per day of mixed sewerage, with a large industrial component. The microbial species were characterized using standard plating techniques, morphology, microscopic observation, and biochemical tests. These characteristics were determined for the fresh liquor, and after successive batch exposure to phenol (100 ppm) and 2-chlorophenol (20 ppm). Predator/prey, gram positive to gram negative bacteria ratios, and the fungi and protozoan populations were also determined. These results were compared with those previously obtained using …

Dispersal Of A Heron-Egret Rookery, Douglas I. Hall Sep 1985

Dispersal Of A Heron-Egret Rookery, Douglas I. Hall

Wildlife Damage Management Conference

A rookery composed of an estimated 10,000 herons and egrets (family: Ardeidae) located in Van Buren, Crawford County, Arkansas was successfully dispersed in the Spring of 1983. A diversified scaring program was planned and initiated prior to the onset of courtship display and nest building. The roost relocation was subsequently followed by habitat alteration procedures to make the 5-acre stand of primarily Eastern redcedar (Juniperus virginiana) unattractive as a future roosting site. Although no nesting occurred at the site in 1983, the lack of an early scaring program in the Spring of 1984 resulted in the uncleared …

Isolation And Characterization Of A Novel Motility Mutant In Salmonella Typhimurium, David Matthew Chacko Sep 1985

Isolation And Characterization Of A Novel Motility Mutant In Salmonella Typhimurium, David Matthew Chacko

Loma Linda University Electronic Theses, Dissertations & Projects

A new mutant, BT40, isolated from mutagenized cultures of S. typhimurium was deficient in chemotaxis (Che- phenotype) in tryptone semisoft agar, but had random motility with a tumbling frequency (37%) similar to that of the parental strain, ST23 (39%). In spatial assays accumulation of BT40 cells in capillary tubes containing attractants was decreased by 33%-59% compared to ST23, but BT40 had a normal response to attractants in temporal assays. The methylation level of the chemotactic proteins was decreased in BT40. At room temperature, the mean swimming speed was 11 μm/s for BT40 and 20 μm/s for ST23; more BT40 …

Assay For Rifampicin Resistant M13 Rf Synthesis, Carol Bauer Wheeler

Loma Linda University Electronic Theses, Dissertations & Projects

We have developed a rapid assay which can detect the function of the origin of 0X174 complementary strand DNA in Ml3 by virtue of its ability to initiate the synthesis of M13 complementary strands in the presence of the antibiotic rifampicin. A segment of DNA carrying the sequence recognized by the replication protein n' has been isolated from bacteriophage ØX174 and cloned into an Ml3 bacteriophage vector. Protein n' is a component of the bacterial primosome, a multi-protein assembly which initiates the synthesis of primers during DNA replication in the bacterium Escherichia coli. The recognition site for n' on the …

Factors Influencing Cellulolytic Activity Of The Soil Fungus, Aspergillus Candidus, Jacobo Ortega, Ernest J. Baca Sep 1985

Factors Influencing Cellulolytic Activity Of The Soil Fungus, Aspergillus Candidus, Jacobo Ortega, Ernest J. Baca

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

Effects of pH, incubation time, nitrogen source and carbohydrate source upon production of carboxymethyl cellulase by Aspergillus candidus grown in liquid medium were studied. Maximal cellulase activity per unit of protein occurred in culture fluids with a pH of 5.5, after 10 days of cultivation of the fungus. Ammonium nitrate supported the highest production of the enzyme. However, in the nitrogen-source test, highest accumulation of extracellular protein was found in fluids from cultures with urea as the nitrogen source. Cotton fibers produced the greatest gain in cellulase activity when the carbohydrate source was incremented from 0.5 to 1.0 percent. The …

Scanning Electron Microscopy Studies Of Staphylococcal Adherence To Heart Valve Endothelial Cells In Organ Culture: An In Vitro Model Of Acute Endocarditis, Morris D. Cooper, Christine Jeffery, Debra L. Gall, Andrew S. Anderson

Scanning Electron Microscopy

Organ cultures of human heart valves were used as a model to study the initial pathobiology of acute infective bacterial endocarditis. We used Staphylococcus aureus isolated from a case of infective endocarditis to infect the in vitro culture of the heart valves. Using scanning electron microscopy, we assessed the initial damage, attachment to and invasion of the endothelial cell layer by staphylococci. Our results indicate there is initial damage to the endothelium prior to observation of staphylococci attaching to the endothelial cell. By 12 h post infection, there is significant attachment and damage. At 24 h after infection, destruction of …

Atp Defendent Na+ Transport And The Characterization Of Na+ / Nh4+ Exchange In Basolateral Vesicles Of The Blue Crab, Callinectes Sapidus, Gill, Linda Ann Fuhrman

Master's Theses

ATP dependent Na+ transport was denonstrated in purified ba.solateral vesicles of the blue crab (Callinectes sapidus) gill. Active Na+ uptake was dependent upon the intravesicular presence of either K+ or NH; as a counterion. Final membrane preparations consist of inside-out basolateral vesicles, evidenced by intravesicular ouabain (1 rrM) and extravesicular vanadate (1 rnM) 22Na + uptake inhibition. Na+/NH4+exchange rates were investigated by detennining the NH; concentration in the recovered extravesicular nedium and by using the kinetic capabilities of a DU7 spectrophotareter. These purified basolateral membranes, rich in Na+,K+ …

Scanning Acoustic Microscopy In Materials Characterization, H. R. Vetters, P. Mayr, S. Boseck, Th. Luebben, R. Matthaei, A. Schulz

Scanning Electron Microscopy

The scanning acoustic microscopy is a powerful tool for subsurface imaging and therefore fault detection in coated parts. In this paper several methods are established to reveal the imaging of hidden structures. First efforts were made to find out the information depth due to the various distances between lens and surface of the object. By means of a specially developed test specimen it was possible to estimate the penetration depth for monitoring structural details. The indepth analysis of layered composites is considered by the determination of the V(z)-characteristics. Furthermore the gain of image processing by means of Fourier transformed patterns …

Amphibian Lampbrush Chromosome Loops: Correlative Light Microscopy, Transmission Electron Microscopy, And Scanning Electron Microscopy Observations, M. L. Bonnanfant-Jais, E. N'Da, M. Penrad-Mobayed, N. Angelier

Scanning Electron Microscopy

The RNP matrix of landmark loops in Pleurodeles waltlii lampbrush chromosomes have been examined by light microscopy, transmission and scanning electron microscopy. This study shows that in normal loops as well as granular, globular and dense loops, the RNP matrices are composed of one basic structure: an RNP particle with a diameter of 30 nm. The scanning electron microscope study also clarified the spatial arrangement of the various types of RNP matrices. The specific morphology of the RNP matrices is due to i) the progressive packaging of transcription products and ii) a concomitant and gradual coiling of the loop axis.

Preliminary Experience With Digital Subtraction Angiography In Cardiac Evaluation, Matthew W. Burke, P. C. Shetty, Rajinder Sharma, James F. Brymer, Fareed Khaja

Henry Ford Hospital Medical Journal

iVe report our initial experience using digital subtraction angiography (DSA) techniques for cardiac evaluation. DSA of the heart may be performed with intravenous or right atrial injection of contrast medium (IVDSA) and with left ventricular or aortic root contrast injection (lADSA). The right ventricle and the atria are best demonstrated by IVDSA, while the left ventricle and coronary arteries are best demonstrated by lADSA. The advantages and disadvantages of DSA ofthe bean are discussed. Present equipment limitations restrict the routine use of cardiac DSA, but these should be overcome with advances in technology.

Nuclear Magnetic Resonance: Current And Future Clinical Applications, Jerry W. Froelich, David O. Hearshen, Robert D. Halpert, Suresh Patel

Henry Ford Hospital Medical Journal

Nuclear magnetic resonance has evolved from a laboratory analytical tool to become a rapidly developing discipline in clinical medicine. We present a brief historical overview, an introduction to the basic principles of the phenomenon, and a statement of the current status of clinical imaging. We have elected to use the traditional terminology nuclear magnetic resonance to refer to the imaging component of the field rather than the American College of Radiology (ACR) modification magnetic resonance. We do this out of respect for the founders of the field.

Nodule Initiation Elicited By Noninfective Mutants Of Rhizobium Phaseoli, Kathryn A. Vandenbosch, K. Dale Noel, Yasuko Kaneko, Eldon H. Newcomb

Biological Sciences Faculty Research and Publications

Rhizobium phaseoli CE106, CE110, and CE115, originally derived by transposon mutagenesis (Noel et al., J. Bacteriol. 158:149-155, 1984), induced the formation of uninfected root nodule-like swellings on bean (Phaseolus vulgaris). Bacteria densely colonized the root surface, and root hair curling and initiation of root cortical-cell divisions occurred normally in mutant-inoculated seedlings, although no infection threads formed. The nodules were ineffective, lacked leghemoglobin, and were anatomically distinct from normal nodules. Ultrastructural specialization for ureide synthesis, characteristic of legumes that form determinate nodules, was absent. Colony morphology of the mutant strains on agar plates was less mucoid than that of …

Analysis Of Backscattered Electron Signals For X-Ray Mask Inspection, Michael G. Rosenfield

Scanning Electron Microscopy

A rapid and automated inspection system is a necessity for the detection of defects in x-ray and optical lithography masks. The design of an electron-beam mask inspection system requires a complete understanding of the backscattered electron signal from the various defects which will be encountered. A Monte Carlo simulation program has been used to study the effects of electron-beam size, detector placement, defect type, electron-beam voltage, and absorber thickness on the back-scattered electron signal.

Structure And Surface Exposure Of Protein-Iis Of Neisseria-Gonorrhoeae Js3, Ralph C. Judd May 1985

Structure And Surface Exposure Of Protein-Iis Of Neisseria-Gonorrhoeae Js3, Ralph C. Judd

Biological Sciences Faculty Publications

Colonies of Neisseria gonorrhoeae JS3, each bearing a predominate protein II (PII) type, were derived from a progenitor transparent colony. Five distinct PIIs were identified and isolated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The PII bands were excised from gels of unlabeled whole cells and from gels containing lysates of surface-radioiodinated bacteria. These were subjected to alpha-chymotrypsin digestion and two-dimensional peptide mapping, which allowed for a comparison of both the primary structures of the PIIs and the identification of surface-exposed regions of the molecules. The results demonstrated that PIIs are unrelated to either Protein I or Protein III in structure …
