Observations On The Microstructure And Rheology Of Ovalbumin Gels, I. Heertje, F. S.M. Kleef
Food Structure
Understanding the gelation behaviour of proteins is of importance in order to be able to influence the properties of many food systems and it may lead to proposals for product or process improvement . In this context the formation of heat-set ovdl bumin gels. in different media , has been studied by microstrucmral. rheological and conformational observations. An oval bumin/ water geL prepared at pH 5, shows a granular. inhomogeneous microstructure. At this pH the re are both many interand intramolecu lar interactions. and network formation occurs via aggregates of folded, globular protein chains. On the other hand , gels …
Changes In Feeding And Crawling Rates Of Hydrobia Truncata (Prosobranchia: Hydrobiidae) In Response To Sedimentary Chlorophyll-A And Recently Egested Sediment, Valery E. Forbes, Glenn R. Lopez
Changes In Feeding And Crawling Rates Of Hydrobia Truncata (Prosobranchia: Hydrobiidae) In Response To Sedimentary Chlorophyll-A And Recently Egested Sediment, Valery E. Forbes, Glenn R. Lopez
Valery Forbes Publications
Experiments combining fluorescent particle tracer techniques with time-lapse video recording demonstrated that the gastropod Hydrobia truncata decreases crawling rate on patches rich in chlorophyll-a, while maintaining a constant feeding rate as sedimentary chlorophyll-a content varies. In contrast, H. truncata decreases feeding rate but does not change crawling rate upon encounter with patches of recently egested sediment. Depression of feeding rate is less pronounced if the egested material is enriched in chlorophyll-a. For deposit feeders inhabiting highly reworked sediments, the majority of available material may consist of sediment that has been recently egested. For an animal that responds negatively to recently …

The Skinned Fiber Technique As A Potential Method For Study Of Muscle As A Food, R. G. Cassens, T. J. Eddinger, R. L. Moss
Food Structure
Skeletal, smooth and cardiac muscle cells can be skinned by physical means or a variety of chemical techniques. The skinned fibers have been used to study the molecular mechanisms of contraction and the regulation of contraction by ca++. Skinned fiber preparations are also useful for study of muscle as a food. For example, it is now possible to determine fiber type of skinned fibers following study of their physical properties.
Fixing Conditions In The Freeze Substitution Technique For Light Microscopy Observation Of Frozen Beef Tissue, M. N. Martino, N. E. Zaritzky
Fixing Conditions In The Freeze Substitution Technique For Light Microscopy Observation Of Frozen Beef Tissue, M. N. Martino, N. E. Zaritzky
Food Structure
The freeze substitution histological technique allows the indirect light microscopicobservation and the quantitative evaluation of ice crystal size in frozen tissues. Theuseofchemical fixatives in substitu ting fluids improves morphological and histochemical preservation of the tissue. Fixation conditions become important since this step can introduce modifications in crystal sizes . Effects of temperature on: a) diffusion rate of fixing solution in tissue, b) recrystallization rate of ice in frozen beef and c) variations of the froze~ water fraction were ana lyzed, establishing that 1sothermal freeze fixation constitutes an appropriate method for histological observation. Fixing at temperatures lower than that of the …

Nebraska Center for Virology: Faculty Publications
We investigated the relationship of the presence of antibodies to HTLV-III and immunologic abnormalities in patients with hemophilia. Serum antibodies to HTLV-III were analyzed by ELISA assay, immunoprecipitation of labeled cell extracts, and immunoprecipitation of purified HTLV-III p24. Thirty-four (61%) of the total group (n = 56) had antibody to HTLV-III; 34 (76%) of 45 patients given commercial factor VIII preparations were seropositive, compared with none of 11 patients treated exclusively with cryoprecipitate obtained from volunteer blood donors. Of patients who were seropositive for HTLV-III antibody, 94% had abnormal T4/T8 ratios, and 33% of those whose serum was antibody …

Restriction Mapping And Expression Of Recombinant Plasmids Containing The Arsenic Resistance Genes Of The Plasmid R45, Terry M. Coons
Restriction Mapping And Expression Of Recombinant Plasmids Containing The Arsenic Resistance Genes Of The Plasmid R45, Terry M. Coons
Dissertations and Theses
The trivalent (arsenite) and pentavalent (arsenate) forms of arsenic are introduced into the environment through the use of arsenic in herbicides, pesticides, fertilizers, and the smelting of arsenic-bearing ores. Bacteria resistant to arsenic are readily isolated from surface waters, sewage, and clinical infections. Although some bacterial resistance is provided by inducible phosphate transport systems that discriminate against arsenate, marked resistance is carried on bacterial plasmids.
A 6.9 kilobase fragment previously derived from one such plasmid, R45, and containing the genes for inducible resistance to arsenite and arsenate was ligated into the cloning vectors puce and pUC9 in opposite orientations and …

The Effect Of Soil Storage Methods And Soil Microbes On Carbofuran And Terbufos Degradation, Muriel Monteleone Runholt
Electronic Theses and Dissertations
The purpose of this research was to determine optimal soil storage methods for soils used in carbofuran and terbufos degradation studies. The effect of soil microbes on degradation was also observed. An LD50 study, using 22 day old crickets (Acheta domestica), was undertaken to establish the appropriate LC dose for carbofuran in the soil storage studies. A carbofuran use history failure soil and a noninsecticide use history soil of the same type were collected from the SDSU experimental farm in Centerville, South Dakota. After initial degradation and microbial analysis, the soils were stored air-dried and with a moisture level …
Modeling Of Soil Water Flow And Root Uptake, V. Rasiah
Modeling Of Soil Water Flow And Root Uptake, V. Rasiah
Electronic Theses and Dissertations
The soil water pressure head, h, versus soil water content, 9, and the hydraulic conductivity, K, versus θ, relationships appear as functional coefficients in the non-linear partial differential water flow model. Before the flow equation can be used to simulate the flow of water through soils, the h versus θ and K versus θ relationships must be established. This requires the estimation of the parameters that describe the h versus θ and K versus θ functional relationships. In-situ water retention and hydraulic conductivity measurements were determined from the knowledge of initial and boundary conditions and water content profiles during drainage. …

Technology And Economics Of Commercial Scale Ethanol Production From Fodder Beets, William Ray Gibbons
Technology And Economics Of Commercial Scale Ethanol Production From Fodder Beets, William Ray Gibbons
Electronic Theses and Dissertations
Fodder beets yield two to three times more fuel ethanol per hectare than corn. This increased productivity should reduce feedstock costs and, consequently, ethanol production costs. The major factor that has limited use of this crop is the lack of a proven technology to process fodder beets -- economically and energy efficiently -- to fuel ethanol on a commercial scale. Research reported herein rectifies this situation by examining the technical and economic feasibility of community scale fuel ethanol production from fodder beets using two novel, continuous fermentation systems solid-phase fermentation and diffusion fermentation. Laboratory scale, batch fermentation trials were first …

Kinematic Analysis Of The Chemotactic Locomotion Of Human Blood Neutrophils: The Effects Of Storage At Room Temperature, Jarrett Lee Burton
Kinematic Analysis Of The Chemotactic Locomotion Of Human Blood Neutrophils: The Effects Of Storage At Room Temperature, Jarrett Lee Burton
MUSC Theses and Dissertations
When human blood neutrophils are isolated for the circulation, they begin to lose chemotactic responsiveness. To characterize this loss of function which occurs during storage, an enhanced videomicroscope system was developed and computer-assisted image analysis was utilized to perform kinematic analysis on neutrophils migrating in an agarose assay system. The locomotor behavior of neutrophils was measured in response to a gradient of chemoattractant (chemotaxis) and in response to an isotropic distribution of chemoattractant (chemokinesis). During chemotaxis, neutrophils migrate at ~20 um/min, make few turns and orient toward a chemoattractant with high accuracy and precision. During chemokinesis, neutrophils migrate at a …

Genetics Of The Screwworm Fly, David B. Taylor, Estela Martinez Cuevas
Department of Entomology: Faculty Publications
Three spontaneous eye mutants of Cochliomyia hominivorax, the screwworm fly, are described and characterized. The mutants, yellow eye (ye), orange eye (or), and white eye (wh), segregate independently and are recessive. Yellow eye is recessively epistatlc to orange eye and dominantly epistatic to white eye. All three mutants have complete penetrance, are autosomal, and have little effect on fly viabillty under laboratory conditions.

Recovery Of Parasite Remains From Coprolites And Latrines: Aspects Of Paleoparasitological Technique, Karl J. Reinhard, Ulisses E. Confalonieri, Bernd Herrmann, Luiz F. Ferreira, Adauto J. G. De Araujo
Department of Anthropology: Faculty Publications
Standard techniques for the analysis of prehistoric soils have not been devised. It is unlikely that any single technique is applicable to all types of fecal remains. This is due to various environmental conditions which effect the preservation of helminth ova. In general, gravitational sedimentation is a useful technique for isolating helminth eggs and larvae from coprolites. Latrine soils pose greater problems for helminthological examination. Although various clinical techniques have been successfully utilized in soil study, it is important to remember that some latrine soils have not yielded helminth eggs to any clinical technique. Consequently the paleoparasitologist must be ready …

Isolation Of Nuclei From Physarum Flavicomum: Demonstration Of Nuclear Cyclic Acid Amp Phosphodiesterase, Judith A. Bean, Maurice G. Kleve, Thomas J. Lynch
Journal of the Arkansas Academy of Science
Cyclic AMP phosphodiesterase activity in the nucleus of the myxomycete Physarum flavicomum was demonstrated by cytochemical staining utilizing electron microscopy and by enzymatic assays with tritiated cyclic AMP as the substrate. Cytochemical staining showed Physarum's plasmodial phosphodiesterase activity to be located in the nucleus, along the plasma membrane, in vesicles, and free in the cytoplasm. Nuclear phosphodiesterase, which may be cell cycle dependent, was primarily located in the nucleolus. Nuclei from three to five day old microplasmodial cultures were isolated by the method of Henney and Yee. Whole cells were collected through centrifugation and washed. Pellets were homogenized in a …

Investigation Of Nickle As A Matrix Modifier In The Atomic Absorption Analysis Of Tin, Charles William Saunders
Chemistry & Biochemistry Theses & Dissertations
Nickel has been widely used as a matrix modifier in the flameless atomic absorption analysis of arsenic and selenium. Its utility as a modifier for tin has been investigated using Polarized Zeeman effect Graphite Furnace Atomic Absorption Spectroscopy. Peak area, peak shape, and appearance times were compared in modified and nonmodified standards using the system computer's internal integration and a detector/readout system with 18 msec resolution.
The addition of nickel at 600 mg/L increased the tin absorbance signal five-fold. The improved signal to noise ratio resulted in a two-fold increase in the sensitivity and a lower limit of detection. The …

Transposon Mutagenesis And Excision Of R' Plasmids By Conjugative, Chimeric Plasmid Puw942 In Extra-Slow-Growing Rhizobium Japonicum Strains, Alahari Arunakumari, Anne K. Vidaver
Department of Plant Pathology: Faculty Publications
Transposons Tn501 (specifying mercury resistance) and Tn7 (specifying resistance to trimethoprim and streptomycin) were introduced into extra-slow-growing Rhizobium japonicum by conjugal transfer of the 82 kilobase chimeric plasmid pUW942. Mercury-resistant transconjugants were obtained at a frequency of 10-7 to 10-9. The transfer frequency of streptomycin resistance was lower than that of mercury resistance, and Tn7 was relatively unstable. pUW942 was not maintained as an autonomously replicating plasmid in R. japonicum strains. However, some of the Hgr transconjugants from the RJ19FY, RJ17W, and RJ12S strains acquired antibiotic markers of the vector plasmid pUW942. Southern hybridization of plasmid …

Characterization Of A Staphylococcal Trimethoprim Resistance Gene, Jerald Preston Coughter
Theses and Dissertations
Trimethoprim resistance(Tp r) is encoded by conjugative plasmids in clinically significant staphylococcal isolates. Two genetically and physically similar plasmids from S. aureus, pG01 and pGOS, have Tp r genes that map in different locations on these plasmids. In order to study the relatedness of the Tp r genes and their products to other known Tp r genes, a 1.2 kb fragment of pG01 and a 4.2 kb fragment of pGOS were cloned in E. coli and used as probes for in situ filter hybridization experiments.
A 500 base pair subclone of the original 1.2 kb fragment containing only the staphylococcal …
The Effect Of A Bacteriophage On The Competition Of Rhizobium Japonicum Strains For Nodulation Of Glycine Max (L.) Merr, Lori Kelley
Masters Theses
The symbiotic relationship between Rhizobium japonicum (Kir.) Buch. and Glycine max (L.) Merr. was the concern of this thesis. This relationship occurs naturally in soybean fields. In the field there are many different strains of Rhizobium. Some of these strains fix atmospheric nitrogen more efficiently than others resulting in a higher crop yield. The problem is the more efficient nitrogen fixers are out-competed by less efficient nitrogen fixers. In order to solve this problem the more competitive strains have to be controlled. There are two main ways to do this. One is to increase the number of the desired …

Characterization Of Small Metallic Clusters By Electron Energy Loss Spectroscopy, M. Acheche, C. Colliex, P. Trebbia
Scanning Electron Microscopy
Small spherical tin and gallium clusters (diameters in the range 2 to 80 nm), prepared with a liquid metal ion source, were analysed in a scanning transmission electron microscope (STEM) by electron energy loss spectroscopy (EELS). Both volume and surface plasmon excitations were investigated and their dependence on the cluster size interpreted by classical and quantum mechanical models. A blue shift of the volume plasmon energy with decreasing radius R of the cluster was clearly detected. The full width at half maximum (FWHM) of the peak, related to the damping of the volume plasmon excitation, showed a dependence in 1/R. …

Intraocular Lens Implant Image Quality: The Optico-Retinal Interface, David D. Bogorad
Henry Ford Hospital Medical Journal
Improvements in intraocular lens implants during the past ten years have revolutionized cataract surgery. Both anterior and posterior chamber lenses are currently in wide use, but the optical superiority of the posterior chamber location has yet to be demonstrated. To do so, a more detailed analysis is required of visual function than is clinically routine. Also required is appreciation that the retina is an integral part of the eye's optical system because of its directional characteristics, which are known as the Stiles-Crawford effect. The optical quality of the pseudophakic eye is affected by the optics of the implant, its centration, …
Microencapsulation As An Alternative To Emulsification For Preparing As A For Use In Papermaking, William J. Corriveau
Microencapsulation As An Alternative To Emulsification For Preparing As A For Use In Papermaking, William J. Corriveau
Masters Theses
The sizing chemical ASA is emulsified to make i t compatible with the papermaking process. Unfortunately, emulsified ASA is chemically reactive with water at the elevated temperatures found in papermaking systems. As an alternative, encapsulated ASA was made and investigated for its stability towards water at elevated temperatures.
A handsheet procedure was developed to analyze emulsified and encapsulated ASA. It was concluded that the procedure gave statistically reproducible results. Investigations with the procedure revealed important variables concerning the laboratory and m ill application of ASA. Evidence indicated that retention of ASA was critical to sizing development.
Encapsulated ASA made for …

Hereditary Sclerocornea, David Barsky, Steven P. Dunn
Henry Ford Hospital Medical Journal
A rare case of total hereditary sclerocornea is presented along with clinical history, histopathology, and a review of the possible pathogenesis and current literature on the subject. It is quite possible that imperfect cleavage of the anterior chamber angle may be the predisposing factor in scleralization of the cornea due to a failure of neural crest cell differentiation. If surgical intervention to correct for the corneal opacification is indicated, corneal transplantation should be performed as soon as possible after birth, similar to the indications for removal of dense congenital cataracts to avoid deprivation amblyopia.
