Assembly And Alignment Of Fibronectin-Coated Gold Beads Into Fibrils By Human Skin Fibroblasts, Donna M. Pesciotta Peters, Deane F. Mosher

Scanning Microscopy

The assembly of fibronectin into fibrils was monitored by high voltage electron microscopy using 18 nm colloidal gold beads bound to fibronectin (Au18-fibronectin) or an amino terminal 70 kd fragment of fibronectin (Au18-70 kd) that blocks the incorporation of fibronectin into disulfide bonded fibrils. Subconfluent cultures of human skin fibroblasts were incubated with the colloidal gold complexes for 0.25, 0.5, 1.5 and 5 h. In fibroblast cultures incubated with Au18-fibronectin and Au18-70 kd fragments for 0.25 and 0.5 h, the complexes of Au18-fibronectin and Au18-70 kd fragment were …

Expression Of Major Histocompatibility Complex Antigens On Macrophages: Correlative Study Using Flow Cytometry, Radioimmunoassay, And Colloidal Gold Immunolabeling, Lynne E. Guagliardi, Donna M. Paulnock, Ralph M. Albrecht Feb 1987

Expression Of Major Histocompatibility Complex Antigens On Macrophages: Correlative Study Using Flow Cytometry, Radioimmunoassay, And Colloidal Gold Immunolabeling, Lynne E. Guagliardi, Donna M. Paulnock, Ralph M. Albrecht

Scanning Microscopy

Correlative scanning electron microscopy (SEM), radioimmunoassay (RIA), and flow cytometric analysis were used to characterize levels of class I and class II major histocompatibility complex-encoded (MHC) antigen expression on peritoneal exudate cells of mice chronically infected with Chlamydia psittaci. Analysis of peritoneal macrophages by all three techniques revealed a marked induction of H-2 K,D (class I) and I-A, l-E (class II) antigens on cells from infected C3H mice when compared to uninfected controls.

Scanning electron micrographs further document that the increases in class I and II MHC antigens are due to an increase in la/H-2 bearing cells as well …

Vascular Changes In Popliteal Lymph Nodes Due To Antigen Challenge In Normal And Lethally Irradiated Mice, Douglas A. Steeber, Chris M. Erickson, Kees C. Hodde, Ralph M. Albrecht Feb 1987

Vascular Changes In Popliteal Lymph Nodes Due To Antigen Challenge In Normal And Lethally Irradiated Mice, Douglas A. Steeber, Chris M. Erickson, Kees C. Hodde, Ralph M. Albrecht

Scanning Microscopy

The microvascular system of the murine popliteal lymph node was investigated using scanning electron microscopy of microcorrosion casts. Time-dependent changes in the microvasculature following regional antigen challenge in normal and lymphocyte-depleted mice were studied. Normal lymph node microvasculature exhibited a significant increase in both the vascular bed and post-capillary venules containing high-endothelium in response to antigen challenge. Lymph nodes of lymphocyte-depleted mice showed no microvascular size increase following antigen challenge and a reduction in the amount of high-endothelium was observed.

The Effect Of Controlled Diffusion Of Ions On The Formation Of Hydrated Calcium Oxalate Crystals, H. Lachance, R. Tawashi Jan 1987

The Effect Of Controlled Diffusion Of Ions On The Formation Of Hydrated Calcium Oxalate Crystals, H. Lachance, R. Tawashi

Scanning Microscopy

Calcium oxalate monohydrate (COM), calcium oxalate dihydrate (COD) and calcium oxalate trihydrate (COT) were grown from solutions under controlled release of the reacting ions. The mass transfer kinetics of ions released from an insoluble polyethylene matrix, from an osmotic pump system and from the hydrolysis of diethyl oxalate were studied under different experimental conditions. It was possible with simple laboratory techniques to grow well-formed crystals of COM, COD and COT, suitable for single crystal work. Results obtained show that the degree of hydration, size, and morphology of the crystals formed, are controlled by interfacial kinetic factors.

Correlative Light And Electron Microscopy Of Platelet Adhesion And Fibrinogen Receptor Expression Using Colloidal-Gold Labeling, Steven L. Goodman, Ralph M. Albrecht Jan 1987

Correlative Light And Electron Microscopy Of Platelet Adhesion And Fibrinogen Receptor Expression Using Colloidal-Gold Labeling, Steven L. Goodman, Ralph M. Albrecht

Scanning Microscopy

Differences in the shape change responses of platelets to various polymers may determine the thrombotic potential of these materials. Substrate-dependent variation in the expression and motility of the platelet fibrinogen receptor may underlie these differences due to this ligand's essential role in platelet aggregation. In this study we examine platelet activation on polyetherurethaneureas (PEUUs) presently being evaulated for vascular prosthetic applications. These polymers are prepared as 50-100nm thin films suitable as substrates for consecutive light microscopy, high voltage electron microscopy (HVEM), and SEM. 18nm colloidal gold coupled to fibrinogen permits visualization of that receptor's motility in living cells by video-enhanced …

Microbial Cell Division And Separation: Effect Of Citrate On The Growth Of Group N Streptococci, S. Ito, T. Kobayashi, K. Ozaki, T. Morichi, M. Saitoh Jan 1987

Microbial Cell Division And Separation: Effect Of Citrate On The Growth Of Group N Streptococci, S. Ito, T. Kobayashi, K. Ozaki, T. Morichi, M. Saitoh

Food Structure

In the presence of citrate, some strains of lactic streptococci grow as long chains of innumerable cells. The results with citrate-negative and citrate-resistant variants suggest that citrate is involved in the cell separation system of the streptococci. Observations of the long chains under scanning and transmission electron microscopes suggest that citrate inhibits a certain step near the final cell separation, or that citrate can stimulate initiation of cell division at multiple sites, thereby leading to the cells in long chains.

Modification Of Low Density Lipoprotein And The Effects Of Modified Low Density Lipoproteins On The Endocytic And Secretory Repertoire Of The Macrophage, Ruth Rebecca Montgomery

Student Theses and Dissertations

This study examines two aspects of the complex interaction between modified low density lipoprotein (LDL) and macrophages in the initiation and development of the atherosclerotic lesion. The first of these concerns the observed biological modification of LDL that leads to its subsequent enhanced uptake by macrophages, and the role of oxidation in that process. The second aspect of this thesis studies the effects of modified LDL on the functions of macrophages. Studies on the oxidation of LDL showed that LDL isolated from human plasma anticoagulated with EDTA (EDTA-LDL) was 4-fold more resistant to oxidation by reagent H2O2 …

Host-Parasite Interrelations: Bacterial Adherence To And Invasion Of Epithelial Cells, Debra Bessen Jan 1987

Host-Parasite Interrelations: Bacterial Adherence To And Invasion Of Epithelial Cells, Debra Bessen

Student Theses and Dissertations

Enteroinvasive Escherichia coli (EIEC) preferentially invaded multinucleated HeLa cells which arose naturally in monolayer cultures at a low frequency. Data suggests that enhancement of the penetration step accounts partly for increased invasion of multinucleated cells. Treatment of HeLa cell monolayers with polyethylene glycol (PEG) led to cell fusion, and invasion of PEG-treated monolayers by EIEC was increased by an average of 7-fold compared to untreated HeLa cells. Treatment of HeLa cells with PEG had no significant effect on the low level of cell-association displayed by noninvasive organisms and thus, PEG-treated HeLa cells appeared to retain their selectivity for EIEC. The …

The Influence Of Ingredients And Processing Variables On The Quality And Microstructure Of Hokkien, Cantonese And Instant Noodles, R. Moss, P. J. Gore, I. C. Murray

Food Structure

Light and scanning electron microscopy have been used as a part of a research program designed to study the influence of flour quality and processing variables on three of the noodle types that are popular in South East Asia . The noodles selected for study were Cantonese, Hokkien and modern instant. They represent the range of cooking and processing variables that are commonly encountered in these products . Flour particle size and the choice of alkaline ingredients influence protein development during the sheeting process . A continuous protein matrix is required in t he raw noodle if it is to …

Effects Of Drying Techniques On Milk Powders Quality And Microstructure: A Review, Marijana Caric, Miloslav Kalab Jan 1987

Effects Of Drying Techniques On Milk Powders Quality And Microstructure: A Review, Marijana Caric, Miloslav Kalab

Food Structure

The quality of milk powders is markedly affected by the composition and properties of the milk, the manufacturing procedures, thermal processing during manufacture and, in particular, the drying technique itself. A variety of physico-chemical analytical methods, including scanning electron microscopy, has been used to obtain information on the effects of the various factors on the microstructure of the milk powders. Roller-drying, which has recently lost its commercial importance, produces a sheet of dried milk that is powdered in a hammer mill. The resulting powder consists of compact particles with sharp edges. Powders obtained by spray-drying are in the form of …

Coliphage As An Indicator Of Fecal Pollution In Marine Waters: Assay, Validation, And Application, Donald S. Mccorquodale Jr. Jan 1987

Coliphage As An Indicator Of Fecal Pollution In Marine Waters: Assay, Validation, And Application, Donald S. Mccorquodale Jr.

HCNSO Student Theses and Dissertations

Escherichia coli, the preferred bacterial indicator for fecal pollution in fresh waters, does not conform to the concept of an indicator microorganism because it is rapidly killed or inactivated by seawater. This series of papers investigated the value of coliphage, a virus which infects E. coli, as an indicator of pollution in saline waters. In order to be an accurate indicator an organism must (1) be ubiquitous in wastewater, (2) survive and be detectable at least as long as the harmful organisms, and (3) be easy to isolate and identify.

A review of the literature determined that coliphage …

Symbiosis Between Social Spiders And Yeast: The Role In Prey Attraction, William James Tietjen, L. Rao Ayyagari, George W. Uetz Jan 1987

Symbiosis Between Social Spiders And Yeast: The Role In Prey Attraction, William James Tietjen, L. Rao Ayyagari, George W. Uetz

Faculty Scholarship

A number of predatory animals use deception, including odors, signals, and bait-like body appendages or objects to lure prey (Alcock, 1984). Several spider species are known to emit chemical odors that mimic the sex attractants of certain noctuid or saturniid moths as a means of luring males to be captured (Tietjen and Rovner, 1982). Here we provide evidence that the social spider species, Mallos gregalis, uses a scented "bait" to attract prey. Our results indicate that this odor is produced by yeasts growing on the carcasses of previously fed-upon flies which these spiders incorporated into their webs.

The Strain Improvement Of Aspergillus Ficuum Nrrl 3135, Eric A. Utt Jan 1987

The Strain Improvement Of Aspergillus Ficuum Nrrl 3135, Eric A. Utt

Retrospective Theses and Dissertations

A strain improvement program was initiated to increase the yield of extracellular phytase (EC 3.1.3.8) in Aspergillus ficuum NRRL 3135. A minimal plating medium containing 2.99 g liter-1 a-ketoglutaric acid was developed to decrease and stabilized conidiogenesis time and facilitate the isolation of single colonies. Exposure of A. ficuum conidia to 0.5 mg ml-1 N-methyl-N’-nitro-N-nitrosoguanidine (NTG) for 15 min at pH 6.5 caused a one log kill and an increase in the mutation rate of 800-fold. Treatment of A. ficuum conidia with 46 ergs mm-2 sec -1 UV radiation for 30 sec resulted in a one …

Variant Forms Of Ataxia Telangiectasia, A. M. R. Taylor, E. Flude, B. Laher, Michael W. Stacey, E. Mckay, J. Watt, S. H. Greens, A. E. Harding

Bioelectrics Publications

Two ataxia telangiectasia patients with unusual clinical and cellular features are described. Cultured fibroblasts and PHA stimulated lymphocytes from these two patients showed a smaller increase of radiosensitivity than cells from other A-T patients, as measured by colony forming ability or induced chromosome damage respectively, after exposure to ionising radiation. The response of DNA synthesis to irradiation of these cells was, however, the same as for other A-T patients. Cells from a third patient with some clinical features of A-T but with a very protracted course also showed low levels of radiation induced chromosome damage, but colony forming ability and …

The Effects Of Frozen Storage And Repeated Freeze-Thawing On Salmonella In Varied Concentrations Of Waste Activated Sludge, Joseph Toman Jr. Jan 1987

The Effects Of Frozen Storage And Repeated Freeze-Thawing On Salmonella In Varied Concentrations Of Waste Activated Sludge, Joseph Toman Jr.

Masters Theses

Commonly, waste activated sludge is applied to farmland as a fertilizer supplement and soil conditioner. The use of waste activated sludge is not only an efficient way of sludge disposal, but also an economical one. Much concern has been raised about the safety of such a practice. Harmful pathogens, like Salmonella, are commonly present in waste material. Treatment at wastewater plants is successful in eliminating these organisms from final effluent, but the solids generated are full of indigenous microorganisms, including Salmonella.

Work of this paper attempts to determine the safety of the sludge product after simulated winter conditions …

Antibiotic Effects On Coliform Enumeration, Keith Leonard Wozniak Jan 1987

Antibiotic Effects On Coliform Enumeration, Keith Leonard Wozniak

Masters Theses

The inhibitory effects of Bacillus polymyxa on Escherichia coli were studied. Significant coliform inhibition was observed. The inhibitory agent (presumably polymyxin) was active against coliforms after a one hour holding time. Different suspending menstrua were analyzed for their effects on coliform inhibition. It was observed that Peptone water provided the highest recovery of coliforms while phosphate buffer provided less protection. Two methods of enumeration were also studied as to their ability to quantify coliforms inhibited by B. polymyxa. Comparable inhibition was shown using both methods. No significant differences exist between the Standard Plate Count and Membrane Filter techniques. Overall, …

Chickpeas As A Protein And Energy Supplement For High Producing Dairy Cows, Darryl Lynn Hadsell Jan 1987

Chickpeas As A Protein And Energy Supplement For High Producing Dairy Cows, Darryl Lynn Hadsell

Electronic Theses and Dissertations

Thirty lactating Holstein cows were randomly assigned to one of three diets to evaluate chickpeas as a dietary supplement from wk 4 to 16 postpartum. Diets contained chickpeas at 0, 50, and 100 % of the concentrate dry matter in place of corn and soybean meal. Total mixed diets, fed individually, contained concentrate corn silage and alfalfa hay at 52, 32, and 16 % of the dry matter. Milk yield (34.5, 35.1, 35.7 kg/d) was higher for cows fed 100 % chickpeas than 0 % chickpeas. Fat (3.06, 3.09, 3.28 %) was highest for cows fed 100 % chickpeas. Milk …

Comparison Of Gelled And Meat Diets For Rearing Screwworm, Cochliomyia Hominivorax (Diptera: Calliphoridae), Larvae, David B. Taylor, Robert L. Mangan

Department of Entomology: Faculty Publications

A liquid diet solidified with an acrylamide-acrylate polymer gelling agent (Water-Lock) was compared with a standard meat diet for rearing screwworm, Cochliomyia hominivorax (Coquerel), larvae. Size and yield of larvae reared on gel diet were equal to, or greater than, those of larvae reared on meat diet. Duration of larval development was increased by ca. 13 h on gel diet; however, variability of pupal weight and larval survival over generations were reduced when gel diet was used. Larval size was also more uniform over the 3-d period when larvae leave the rearing medium to pupate (crawl-off). Gelled diet was more …
