Scanning Electron Microscopy Of Vascular Smooth Muscle Cells From Rat Muscular Arteries, William J. Krizmanich, Robert M. K. W. Lee Jun 1987

Scanning Electron Microscopy Of Vascular Smooth Muscle Cells From Rat Muscular Arteries, William J. Krizmanich, Robert M. K. W. Lee

Scanning Microscopy

The use of scanning electron microscopy (SEM) for the examination of medial smooth muscle cells (SMC) is limited by the presence of connective tissue mass in the adventitia obscuring the detailed structure of the SMC. This study deals with the description of a method which selectively removes the connective tissue, allowing three dimensional observation of SMC using SEM. A number of methods were tried. One method involving post fixation with 2% Os04 used in conjunction with HCl proved most successful in removing the extracellular matrix of large mesenteric arteries from spontaneously hypertensive rats (SHR) and age-matched Wistar-Kyoto normotensive rats …

Preservation Of In Vivo Morphology Of Blood Vessels For Morphometric Studies, Robert M. K. W. Lee Jun 1987

Preservation Of In Vivo Morphology Of Blood Vessels For Morphometric Studies, Robert M. K. W. Lee

Scanning Microscopy

Preservation of in vivo morphology of the blood vessels is important for morphometric studies. With each fixation method (e.g., immersion fixation, perfusion fixation), there are inherent pitfalls which can easily distort vessel wall morphology, thereby creating artifacts. Alteration of the vessel wall cell volume due to tissue preparation is another common source for errors. Selection of proper morphometric protocols for comparative studies is crucial. Lack of attention in these areas is a cause of confusion and contradiction in the literature. In this review, the merits of various commonly used methods for vessel wall preparation and measurements are discussed.

Surface Studies Of Calcium Oxalate Dihydrate Single Crystals During Dissolution In The Presence Of Urine, M. Akbarieh, B. Dubuc, R. Tawashi Jun 1987

Surface Studies Of Calcium Oxalate Dihydrate Single Crystals During Dissolution In The Presence Of Urine, M. Akbarieh, B. Dubuc, R. Tawashi

Scanning Microscopy

Single crystals of Calcium Oxalate Dihydrate (COD) were grown from solution under controlled release of the reacting ions. Dissolution of COD was studied at different pH levels and in different dilutions of urine. The descriptors of the contour were determined during dissolution of COD using a quantitative morphological technique. The shape parameters and surface ruggedness were determined from Fourier and fractal analysis. The results obtained give quantitative information on the dissolution kinetics and the surface geometry of COD crystals in normal and diluted urine. Dissolution inhibition and morphological changes of COD crystals during dissolution were attributed to selective adsorption of …

Multiplane Sectioning And Scanning Electron Microscopy As A Method For Studying The Three-Dimensional Structure Of Mature Dental Enamel, S. Risnes Jun 1987

Multiplane Sectioning And Scanning Electron Microscopy As A Method For Studying The Three-Dimensional Structure Of Mature Dental Enamel, S. Risnes

Scanning Microscopy

A method is described where teeth are sectioned/ground along at least two planes, etched, and viewed at various angles in the SEM in order to study the three-dimensional structure of enamel. This multiplane sectioning-scanning electron microscopy (MPS-SEM) method has been applied to the study of rat and human enamel. The method demonstrates in a direct way the complex three-dimensional structure of rat incisor enamel; the path of prisms and the distribution of interprismatic substance. The different appearance of alternate prism rows as seen in the longitudinal plane is seen to be due to prisms in alternate rows being inclined to …

Endothelialization Of A New Dacron Graft In An Experimental Model: Light Microscopy, Electron Microscopy And Immunocytochemistry, G. Pasquinelli, P. Preda, T. Curti, M. D'Addato, R. Laschi Jun 1987

Endothelialization Of A New Dacron Graft In An Experimental Model: Light Microscopy, Electron Microscopy And Immunocytochemistry, G. Pasquinelli, P. Preda, T. Curti, M. D'Addato, R. Laschi

Scanning Microscopy

Two types of synthetic vascular grafts, Dacron Triaxial and Dacron Gelseal Triaxial, were implanted into both the common carotids of sheep. The animals were sacrificed 1, 2, 8, and 16 weeks after surgery. Multiple specimens, obtained from grafts and anastomoses, were studied by light microscopy, transmission and scanning electron microscopy. A parallel immunocytochemical analysis was performed on some specimens. Dacron Triaxial grafts failed to develop a complete neointimal coverage. Myofibroblasts and fibroblasts were the dominant cells in such synthetic graft. Moreover, focal areas of stripping, platelet deposition, and thrombosis were observed at 8 and 16 weeks.

In contrast, a stable …

Cell Polarity And Water Transport In Thyroid Epithelial Cells In Separated Follicles In Suspension Culture, Lucio Nitsch, Seymour H. Wollman May 1987

Cell Polarity And Water Transport In Thyroid Epithelial Cells In Separated Follicles In Suspension Culture, Lucio Nitsch, Seymour H. Wollman

Scanning Microscopy

Separated thyroid follicles maintained in suspension culture can be used to study the properties of thyroid epithelium in the virtual absence of other cell types and to study the effect of extracellular materials on the follicles. They can be prepared by enzymatic separation of thyroid into single cells followed by reaggregation of the epithelial cells and also by collagenase treatment of thyroids to release follicles and sheets of epithelia that can be separated from other materials by differential filtration. The follicles can exist with normal orientation or inverted (inside out). The follicles are inverted in the presence of high serum …

Ultrastructural Analysis Of Dynamic Cellular Processes: A Survey Of Current Problems, Pitfalls And Perspectives, Helmut Plattner, Gerd Knoll May 1987

Ultrastructural Analysis Of Dynamic Cellular Processes: A Survey Of Current Problems, Pitfalls And Perspectives, Helmut Plattner, Gerd Knoll

Scanning Microscopy

Dynamic phenomena in cells that can be analyzed on the ultrastructural level comprise so different aspects as ion shifts, conformational changes of macromolecules, membrane particle rearrangements, lipid phase transitions, protein--protein interactions (notably ligand-receptor interactions, including their sorting and sequestration), reversible membrane-to-membrane contacts, membrane fusions, transcellular transport phenomena, restructuring of cytoskeletal elements, ciliary and flagellar beat, cell shape changes, etc. Only some of these phenomena can be analyzed under stationary conditions, while others are unidirectional and sometimes very rapid. Therefore, the methodical approaches to be used (primary methods and follow-up procedures) might be widely different. Quite different methods are available, such …

Rise To Power Of Senator Joseph R. Mccarthy: Reflections Of The Cold War Mccarthy Era In American Film, Scott Lainer May 1987

Rise To Power Of Senator Joseph R. Mccarthy: Reflections Of The Cold War Mccarthy Era In American Film, Scott Lainer

Honors Theses

The common bond between much of the film industry and Joseph McCarthy was insecurity and the drive for national approval. If one grasps the specific characteristics of McCarthy the man, and the methods of these politically "inspired" movies, we can to better place the period into context and acknowledge the fact that, if the citizenry is not aware, and is again caught by an ever-building wave of trickle down sentiment, the 1950s might not prove to be an isolated period in American history. Insecurity was not a fifties novelty. It still exists, and could potentially escalate anti-Communist policy and sentiment …

The Use Of Scanning Electron Microscopy In Studying Enamel Caries, R. P. Shellis, A. S. Hallsworth May 1987

The Use Of Scanning Electron Microscopy In Studying Enamel Caries, R. P. Shellis, A. S. Hallsworth

Scanning Microscopy

SEM studies related to carious change in dental enamel are reviewed, and their contribution to understanding the mechanism of formation of the early enamel lesion and of its repair evaluated. SEM has contributed significantly to understanding the mechanism of enamel dissolution at the level of the single crystal. Etching studies have yielded useful information on the effect of enamel structure on the pattern of acid dissolution at the microscopic level and have highlighted the importance of re- precipitation phenomena in modifying the pattern of mineral loss. High-resolution studies have provided interesting quantitative data on changes in crystal size, and also …

Plasmid-Linked Maltose Utilization In Lactobacillus Spp. Isolated From Meat, Mei-Ling Liu May 1987

Plasmid-Linked Maltose Utilization In Lactobacillus Spp. Isolated From Meat, Mei-Ling Liu

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Five strains of Lactobacillus plantarum and four strains of Lactobacillus species isolated from fresh meat were examined for the presence of plasmid DNA. All strains examined contained between one and five plasmids ranging in molecular mass from 1.3 to 51.6 (Mdal). Plasmid-curing experiments suggest that maltose utilization is associated with a 51 Mdal plasmid in Lactobacillus sp. DB29 and 42 Mdal plasmids in Lactobacillus spp. DB27, DB28, DB31. Southern blot DNA-DNA hybridization showed homology between the maltose plasmid from Lactobacillus sp. DB29 and several plasmids from the other Lactobacillus spp.

The purpose of this research was to determine whether or not canning fruit using fluoridated water made a difference in the fluoride contents of the juice and the pulp. Four cities in Utah were chosen. Two cities were fluoridated (Brigham City and Helper) and two cities were not fluoridated (North Ogden and Price). Three homes were chosen within each city, and three jars of canned fruit (one jar each of apricots, cherries, and peaches) were selected from each home. The fruit samples were freeze-dried, and then the fluoride was measured with a fluoride ion electrode. A difference was found in …

Purification And Characterization Of An Inhibitor Of Thymidine Uptake From Culture Supernatants Of Human Tonsil Lymphocytes, Abdolreza Zarnegar May 1987

Purification And Characterization Of An Inhibitor Of Thymidine Uptake From Culture Supernatants Of Human Tonsil Lymphocytes, Abdolreza Zarnegar

Electronic Theses and Dissertations

Lymphocytes from human tonsils were cultured in the absence of serum for 3 days. In the presence of the concentrated culture supernatant the proliferative response of PBL, to con A, as measured by the uptake of ('3)H-tdr, was significantly reduced. The suppressor substance was referred to as SMAL (suppressor of mitogen activated lymphocytes). The estimated molecular weight of SMAL under nondenaturing conditions was 100,000-300,000. SMAL also suppressed the incorporation of ('3)H-tdr by a variety of mouse and human tumor cell lines. The activity of SMAL was sensitive to pronase and heating at 100(DEGREES)C for 30 minutes but insensitive to RNase. …

X-Ray Microanalysis Of Growth Cartilage After Rapid Freezing, Low Temperature Freeze Drying And Embedding In Resin, J. Appleton Apr 1987

X-Ray Microanalysis Of Growth Cartilage After Rapid Freezing, Low Temperature Freeze Drying And Embedding In Resin, J. Appleton

Scanning Microscopy

This paper reviews the work undertaken on the X-ray microanalysis of growth cartilage particularly in relation to matrix calcification. Attention is focused on the methodology available to accurately localize and retain the elements of interest. A method is described which involves rapid freezing, low temperature freeze drying in a custom built device, and embedding in Spurr resin. The results show that little tissue damage occurs and that elements of physiological interest are accurately localized at the sub-cellular level. Where damage occurs to chondrocytes as a result of freezing, however, dense intra mitochondrial granules are seen suggesting a phase transformation takes …

Scanning Electron Microscopy Of Vascular Corrosion Casts In Comparative Studies On Renal Vascular Structure, H. Ditrich, H. Splechtna Apr 1987

Scanning Electron Microscopy Of Vascular Corrosion Casts In Comparative Studies On Renal Vascular Structure, H. Ditrich, H. Splechtna

Scanning Microscopy

In comparative microcorrosion casting studies on renal vascular systems the following demands should be met: The preparation procedure (anaesthesia, operation, flushing of the blood vascular system, ...) should be in accord with the specific physiological properties of the animal under investigation and the casting procedure (injection, curing, maceration, ...) should be kept constant as far as possible. If these points are considered, comparative data, even of quantitative nature, can be obtained from corrosion casts. Examples of results at the organ, single vessel and intercellular level as well as correlation of the results with physiological data are given.

Evaluation Of Two Methods For Estimating Belowground Production In The Great Dismal Swamp, Millie Symbula

Biological Sciences Theses & Dissertations

Two methods were used to estimate belowground production in a maple-gum stand in the seasonally flooded Great Dismal Swamp of Virginia. The first was an auger method which involved sequential core sampling. The second method was an implanted soil mass technique. Nylon mesh bags were filled with peat and buried at the study site. New growth into these implants was taken to be production.

Mean annual lateral root biomass and necromass to 40 cm, determined from soil cores, were 583 g·m-2 and 281 g·m-2, respectively. Production estimates for roots ≤ 5 mm in diameter, using soil cores, …

Is There A Calcification Factor Common To All Calcifying Matrices?, E. Bonucci Mar 1987

Is There A Calcification Factor Common To All Calcifying Matrices?, E. Bonucci

Scanning Microscopy

This paper reviews the principal morphological findings pertinent to the early phases of the calcification process and explores the possibility that there may be a calcification factor common to all calcifying matrices. Three structures have a main role in calcification: collagen fibrils, matrix vesicles, and crystal ghosts. Only crystal ghosts are present in all calcified tissues, so that only they can be taken into consideration as a common calcification factor. They are organic molecules which have the same morphology as that of the inorganic structures present in the calcified matrix, which means that they can be considered as templates for …

Trifluoperazine Inhibition Of Fibrinogen Receptor Redistribution In Surface Activated Platelets: Correlative Video-Enhanced Differential Interference Contrast Light Microscopic, High Voltage Electron Microscopic And Scanning Electron Microscopic Studies, O. Trifluoperazine Inhibition Of Fibrinogen Receptor Redistribution In Surface Activated Platelets: Correlative Video-Enhanced Differential Interference Contrast Light Microscopic, High Voltage Electron Microscopic And Scanning Electron Microscopic Studies, O. E. Olorundare, S. L. Goodman, R. M. Albrecht

Scanning Microscopy

Video-enhanced differential interference contrast (VDIC) light microscopy in conjunction with fibrinogen labelled colloidal gold was employed as a probe to follow the mobility of the fibrinogen receptor on platelets. Correlative studies by both high voltage and scanning electron microscopy confirms localization of labels relative to platelet ultrastructural and surface characteristics, respectively. Treatment of platelets with trifluoperazine prior to and after incubation with fibrinogen-gold labels results in a concentration dependent inhibition of receptor movement. The results obtained from this study suggest that phosphorylation of myosin by the Ca++ -calmodulin dependent enzyme, myosin-light chain kinase, is important in the fibrinogen redistribution …

A Comparative Study On In Vitro Invasion Rates By Melanoma Cells In The Human Amniotic Basement Membrane Model Versus In Vivo Tumor Nodule Formation In C57bl6 Mice, B. Persky, G. W. Chmielewski Feb 1987

A Comparative Study On In Vitro Invasion Rates By Melanoma Cells In The Human Amniotic Basement Membrane Model Versus In Vivo Tumor Nodule Formation In C57bl6 Mice, B. Persky, G. W. Chmielewski

Scanning Microscopy

Invasion by murine B16-F10 melanoma cells was studied using the human amniotic basement membrane (HABM) assay. B16-F10 cells were collected after a single passage through the amnion and grown to near confluency. The cycle of plating, passaging, collecting, and culturing B16-F10 cells was repeated five times. The invasion rate for B16-F10 cells remained relatively unchanged after six passages through the amnion. Injection of first-passage B16-F10 cells into C57BL6 mice resulted in 29 lung tumors per animal whereas sixth-passage cells resulted in 300+ lung tumors. While there exists no correlation of the number of cells penetrating the amnion with colonization number, …
