Lifileucel, a tumor-infiltrating lymphocyte (TIL) cell therapy approved for advanced melanoma, demonstrates promise for treating other solid tumors, including endometrial cancer (EC). The current study evaluates the feasibility of manufacturing TILs from EC tumors using Iovance’s proprietary 22-day Gen2 manufacturing process. Key parameters, including TIL yield, viability, immune phenotype, T-cell receptor clonality, and cytotoxic activity, were assessed. Of the 11 EC tumor samples processed at research scale, 10 (91%) successfully generated >1 × 109 viable TIL cells, with a median yield of 1.1 × 1010 cells and a median viability of 82.8%. Our previous research identified 12 small Cajal body-specific RNAs (scaRNAs) with reduced expression in the right ventricle in infant patients with tetralogy of Fallot. Likewise, we showed that there were significant changes in mRNA processing in the RV in these patients. ScaRNAs play a crucial role in the biochemical maturation of spliceosomal RNAs (pseudouridylation and 2′-O-methylation). We showed that variations in scaRNA1 levels resulted in changes in alternative splicing in human cells. A Wachspress-Type Rational Characteristic Method For Solving First-Order Hyperbolic Equations, Mohamed Al-Lawatia

This paper introduces a characteristic method for solving linear first-order hyperbolic partial differential equations in two space dimensions. The algorithm employs rational Wachspress-type basis functions applied to a hexagonal discretization of the domain, integrated within the Eulerian-Lagrangian Localized Adjoint Method (ELLAM) framework. The proposed scheme preserves the strengths of characteristic methods and delivers accurate numerical results, even with large time steps. Computational tests conducted on standard benchmark problems illustrate the effectiveness of the method.

This paper investigates the onset of thermal convection in a bounded system comprising a horizontal viscous fluid layer of infinite extent overlying an anisotropic horizontal permeable layer saturated with the same fluid. It is assumed that this system is heated from below and cooled from above in the presence of internal heat generation. The present article deals with the effect of angular rotation on the propagation of plane harmonic waves in a homogeneous, isotropic magneto-thermo elastic medium. The governing equations of a generalized magneto-thermo elastic solid are formulated and solved with the method of plane harmonic solution. Three types of waves, namely, plane longitudinal, magneto, and magneto-thermal waves, are significantly affected by the angular rotation of the solid. Introduction: Foreign-born and foreign trained International Medical Graduates (FIMGs) face greater challenges in acculturation to their host countries than IMGs who train abroad and return to practice in their home country. As FIMGs are likely to fulfill a shortage of physicians in High Income Countries in the foreseeable future, we conducted a systematic review of literature to identify acculturation interventions that help FIMGs assimilate better in their host country health systems. This improves their productivity and satisfaction, allows health systems to be more accepting of FIMGs, and most importantly, enhances patient outcomes.

This study provides a comprehensive assessment and synthesis of the literature on short-selling. It performs a lexicometric analysis, providing a quantitative review of 1093 peer-reviewed journal articles to identify and illustrate the main themes in short-selling research. Almost half the published literature on short-selling is thematically clustered around portfolio management techniques. Other key themes involve short-selling as it relates to risk management, strategic management, and market irregularities. Descending hierarchical classification examines the overall structure of the textual corpus of the short-selling literature and the relationships between its key terms. The prevalence of depression among young American adults has risen significantly over the past two decades, but the mechanisms that underlie the population-level changes in depressive symptoms severity remain unclear. We used data from the 2005–2023 National Health and Nutrition Examination Survey (NHANES) and hierarchical age–period–cohort (HAPC) analysis to examine developmental (i.e., age), life stage (i.e., period), and generational (i.e., cohort) distribution of depressive symptoms among American adults aged 20–30 years old over an 18-year period. Our model results indicate that the probability of either mild, moderate, or severe depression increased among young adult females and males between 2005 and …

Breast cancer is the leading cause of cancer-related deaths among women worldwide and the second most common cause of cancer deaths in women in the United States. Genetic variants that increase the risk of breast cancer include mutations in the breast cancer susceptibility genes 1 and 2 (BRCA1 and BRCA2). Apoptosis, or programmed cell death, is a natural process that helps the body remove aged cells. However, in cancer, deregulated apoptotic signaling—especially the activation of anti-apoptotic mechanisms—enables cancer cells to evade this process, leading to uncontrolled proliferation, tumor survival, therapeutic resistance, and cancer recurrence. Pleural Fibrosis (PF) involves excessive extracellular matrix (ECM) deposition, mainly by myofibroblasts, leading to pleural thickening and impaired lung function. Pleural mesothelial cells (PMCs) contribute via mesothelial and mesenchymal transition (MesoMT), triggered by cytokines like TGF-β. Our lab identified Myocardin (MyoCD), a transcriptional co-activator of cardiac and smooth muscle, is a master regulator of pleural fibrosis through interactions with serum response factor (SRF) and Smad2/3 transcription factors which initiate fibrosis signaling. RNA sequencing and qPCR analysis revealed that silencing MyoCD decreased the expression of CTNNB1, MDFI, and Tspan2. MDFI emerged as a potent candidate, as this gene affecting the expression …

Since the late 1990s, the United States has been in the midst of an opioid epidemic that continues to present significant public health challenges. Opioid-related mortality, particularly involving fentanyl, has disproportionately affected vulnerable populations. The purpose of this quantitative study was to examine the associations between educational level, homelessness, and alcohol use with opioid-related overdose deaths involving fentanyl, while controlling for age, gender, and marital status, among Black and White populations in the U.S. state of Georgia. To investigate the experiences of adolescents with chronic pain who participated in an intensive interdisciplinary pain treatment program, this secondary study analyzes the themes that emerged regarding the spontaneous utilization of music in coping strategies for chronic pain. During research interviews focused on coping skills and treatment engagement, participants spontaneously reported using music as an effective coping strategy for managing pain. A deductive thematic analysis revealed key themes related to their usage, including using music as a distractor, motivator and in other ways as coping strategies. We tested if hyperoxic conditions can reduce the proportion of active myofibroblasts, which are assumed to be a major driver of head and neck radiation-induced fibrosis, as measured by expression levels of pro-fibrotic genes. Radiated, non-cancerous soft tissue from the head and neck and skin/soft tissue from non-radiated flap donor site were collected from each patient. Myofibroblast density was quantified using immunofluorescence staining with α-SMA and DAPI and visualisation under confocal microscopy and compared between baseline non-radiated and radiated tissue from the same patient. Pumas (Puma concolor) are among the species identified as having the potential to enhance ecosystem function. Previous research highlights sufficient ecological habitat to support pumas in the eastern United States; however, their reintroduction requires social and institutional support as well. To this end, we conducted research to assess attitudes about puma reintroduction among key constituencies like hunters, rural residents, and young people. We sampled 2756 respondents across seven states (Massachusetts, Maine, New Hampshire, New York, Pennsylvania, Vermont, and West Virginia). The commercial launch of ChatGPT in November 2022 and the fast development of large language models have catapulted the regulation of artificial intelligence to the forefront of policy debates. A vast body of scholarship, white papers, and other policy analyses followed, outlining ideal regulatory regimes for AI. The European Union and other jurisdictions have moved forward by regulating AI and LLMs. One overlooked area is the political economy of these regulatory initiatives—or how countries and companies can behave strategically and use different regulatory levers to protect their interests in the international competition on how to regulate AI.

The emergence of a chronic physical illness is unexpected in emerging adulthood and is accompanied by stress. Additionally, married partners of individuals with physical illnesses are likewise affected by the new diagnosis. Marriage outcomes of couples facing chronic illnesses together vary, being sometimes positive and sometimes negative. The current study examined the relationship between the emergence of chronic physical illness in emerging adults and marital communication of both the patient and partner, with marital satisfaction as a moderator. Introduction: SARS-CoV-2 was declared a pandemic in March 2020. Studies have characterized some of the negative impact of the pandemic on public health workers (PHW), but few have explored the strength and coping strategies used. Our study documents the experiences of PHW in Iowa during the pandemic. Methods: We used an innovative mixed methods data collection tool, SenseMaker® to gather stories from PHW in Iowa between March and July 2022. This study aims to provide an update on infectious scleritis (IS). A comprehensive search was conducted using Google Scholar, Scopus, and PubMed. IS is a rare but critical condition that presents significant diagnostic challenges and often leads to poor visual outcomes. It accounts for approximately 5%-10% of scleritis cases and warrants significant attention due to its delayed diagnosis. Ocular surgery is the primary contributor to IS, accounting for a substantial proportion of cases ranging from 58% to 83%, followed by trauma contributing to approximately 10% of cases.
