Circadian rhythms are critical for maintaining homeostasis and regulating physiological functions, and consequentially impact disease progression; yet, they remain largely overlooked in in vitro models used in preclinical research. One major barrier to rigorously testing the role of circadian rhythms in these models is the lack of accessible tools that seamlessly integrate into standard culture setups and are capable of sustainably delivering time cues to cells and tissues in long term experiments. Here, we present the ThermoClock, a low-cost, Arduino-based automated temperature control system capable of delivering independent temperature programs to multiple cultures simultaneously. ``Set it and forget it'' is a common practice with automatic irrigation systems and can lead to excessive irrigation, unhealthy root systems, and water waste. The Water Dogs program in Nebraska focuses on educating Extension Master Gardener volunteers (EMGVs). The goal was to increase their understanding of turfgrass water needs, efficient irrigation methods, and soil health management practices, and to promote greater engagement of EMGVs with homeowner audiences to share and demonstrate the knowledge gained and practices learned through ``Neighbor-to-Neighbor'' workshops. The overall goal is to conserve water and improve soil and plant health.

The two-spined trap-jaw ant, Odontomachus haematodus (Linnaeus), is native to South America and was introduced to the United States via Mobile, Alabama in the mid-twentieth century. Misidentification as a native congener for several decades has inhibited the study of the ecology of O. haematodus in its introduced range within the Gulf Coastal Plain. We conducted a multimethod survey to: 1) assess the abundance of O. haematodus across urban parks in Hattiesburg, MS, the northernmost published record in this species’ exotic range, 2) establish which sampling methodologies are most effective for conducting urban surveys of O. haematodus, and 3) study …

Dietary strategies and plant-based fermented foods have attracted increasing interest for supporting gastrointestinal health, particularly under conditions of drug-induced mucosal stress. Soybean fermentation broth (SFB), a fermented soybean-derived product, may possess enhanced nutritional and functional properties through fermentation-associated generation of bioactive peptides and antioxidant metabolites. This study evaluated the gastroprotective effects of SFB in an indomethacin-induced gastric injury model in rats and investigated the underlying mechanisms. Indomethacin markedly induced gastric ulcer formation (16.61 ± 1.38 mm2), whereas SFB pretreatment significantly attenuated ulcer development in a dose-dependent manner, reducing ulcer area to 6.66 ± 0.98, 3.89 ± 0.63, and …

Garambullo (Myrtillocactus geometrizans) is a Mexican fruit rich in bioactive phytochemicals. Its fermented extract (FEG), obtained after in vitro gastrointestinal digestion and fermentation, was evaluated in HT-29 and SW480 colorectal cancer cells. FEG reduced cell viability (IC50: 2985 and 3200 μg/mL, respectively) and induced apoptosis and necrosis. In 3D assays, FEG inhibited colony formation, with complete suppression in HT-29 cells. Molecular docking further suggested that butyric acid identified in FEG interacts with Caspase-3 and RIPK1. These findings support the antiproliferative and pro-death effects of FEG in colorectal cancer models.

Circadian rhythms regulate fundamental aspects of physiology and behavior. The disruption of circadian rhythms has been associated with metabolic disorders, cardiovascular disease, obesity, and impaired immune function. Feeding behavior and gut microbiota represent two essential nodes within this regulatory network, as both are highly responsive to environmental cues such as diet composition and sleep-wake cycles. Research has shown that high-fat diets, excessive fructose, and chronic alcohol consumption perturb peripheral circadian rhythms and impair metabolic pathways, contributing to obesity and insulin resistance. Conversely, dietary interventions such as time-restricted feeding and macronutrient-specific entrainment can restore rhythmic gene expression and prevent metabolic pathologies. …

Cannabis sativa L. or hemp is a cosmopolitan, multi-purpose plant and area of its cultivation has been growing in recent years. Its public perception is influenced by potential to produce narcotic substances. However, hemp seed can be a valuable source of nutrients for humans and may be of particular interest in more sustainable food consumption patterns, including vegetarian and flexitarian diets.

With the continuous rise in economic supply and demand, an enormous increase in waste, litter, and pollution has been seen globally, with plastic pollution being the most abundant category. Microplastic (< 5 mm), derived from larger plastics, pollution is intensely widespread across marine ecosystems and can be transferred across trophic levels through predator-prey interactions. Recent studies have shown that ingestion of MPs can damage cells, tissues, and internal organs and increases oxidative stress leading to malformations, reduced feeding and growth, , and slower developmental rates in a wide array of organisms. This study examined the trophic transfer of microplastics (MPs) from the anemone Exaiptasia pallida to its specialist predator Berghia stephanieae, and evaluated the effects of exposure on predator behavior. A dose-response experiment demonstrated that MP retention in E. pallida decreased over time, although higher exposure concentrations resulted in greater initial microplastic concentrations. Trophic transfer to B. stephanieae was confirmed through feeding experiments. Behavioral assays revealed that B. stephanieae did not exhibit a preference between MP-exposed and control prey. Agricultural land conversion (ALC) creates a dual sustainability challenge: the loss of productive farmland to urban expansion and the conversion of forested land into new agricultural areas. Farmland loss may reduce food production capacity, while forest conversion for agriculture may increase environmental degradation. Metastatic breast cancer remains one of the most difficult cancers to treat, with a five-year survival rate that drops from 99% for localized disease to just 33% once distant metastases are present. Benthic infaunal communities are widely-used indicators for assessing estuarine health, especially in systems that experience frequent disturbances and hydrological fluctuations. However, using benthic infauna to predict ecosystem responses to future disturbances remains a challenge. The Indian River Lagoon (IRL), spanning 40% of Florida's east coast, is a prime example of a complex estuary that has experienced decades of hydrological alterations leading to reduced water quality, sediment composition, and biodiversity. Microbial communities regulate essential ecological and metabolic processes across both engineered and host-associated ecosystems. Although these systems differ substantially in composition and function, they are governed by common ecological principles including community succession, resilience, species interactions, and responses to environmental and biological perturbations. This dissertation investigates how perturbations influence microbiome structure, function, and stability through three complementary studies spanning anaerobic digestion and the human gut microbiome. First, the effects of an extremophilic biological pretreatment using Caldicellulosiruptor bescii were evaluated in industrial-scale anaerobic digesters treating dairy manure. Background: Breast cancer remains a major global health burden. Particularly in Indonesia, limitations of conventional chemotherapy, such as severe side effect, toxicity, and high cost, indicating the necessitate the development of alternative therapeutic agents. Peperomia pellucida, a medicinal plant rich in bioactive phytochemicals, has shown promising anticancer potential; however, its cytotoxic activity against MCF-7 breast cancer cells and the influence of solvent polarity remains underexplored. Freshwater mussels (family Unionidae) are among the most endangered aquatic species worldwide, making effective monitoring vital for their conservation. Traditional surveys are laborious, time-consuming, and often miss rare or low-density populations. Environmental DNA (eDNA) is a highly sensitive, non-invasive approach for freshwater mussel detection and monitoring; however, uncertainties in ecological interpretation continue to limit its broader application in freshwater biodiversity monitoring. Micro-and Nanoplastics (MNPs) are ubiquitous biological pollutants that have been identified throughout a variety of biological samples. MNPs have been detected in several human tissues, such as the brain, kidneys, and liver. Lung tissue remains understudied for MNP accumulation despite the large surface area and clear path for exposure. Using Pyrolysis-Gas Chromatography Mass Spectrometry (Py-GC/MS), human lung tissue was evaluated for 12 different polymers, enabling quantification of total plastic per gram of tissue to be identified. Nucleotide Excision Repair (NER) is a fundamental cellular process that removes bulky and helical distorting lesions from the genome through the separate responses of its two subpathways – global genomic (GG-NER) and transcription coupled (TC-NER) DNA repair. The pathways converge with the recruitment of the TFIIH complex, a key step that is mediated by the translocase and helicase activities of its XPD subunit. Early life adversity (ELA) increases lifelong neuropsychiatric vulnerability. Yet how ELA reorganizes brain-wide activity and circuit coordination across later experience remains unclear. This dissertation tests the hypothesis that ELA alters adult brain-wide activity and responses to threat through disrupted coordination among neural systems that regulate emotional experience, including prefrontal-limbic and monoaminergic systems. Longitudinal manganese-enhanced MRI of adult mice exposed to standard or fragmented early care, combined with computational processing and statistical modeling, quantified brain states before, during, and long after innate predator threat. Marine heatwaves are increasing in frequency and duration, with widespread bleaching events and mortality impacts observed on Florida’s Coral Reef. Temperature is a major driver of physiological and developmental processes in marine organisms, and continued climate warming is expected to affect reproduction and recruitment in estuarine species. The fossa, Cryptoprocta ferox, is the largest carnivorous mammal endemic to the island of Madagascar. C. ferox is currently listed as a vulnerable species by the IUCN Red List (2015) due to deforestation. Most fossas are now located in zoos for breeding and repopulation purposes, but research on captive mating has been limited due to the low numbers of successful breeding attempts in captivity. Mechanisms for the maintenance of chronic viruses are poorly understood, particularly for archaeal viruses. Here, we identify the product of Sulfolobus spindle-shaped virus 1 (SSV1) ORF a291 as a putative virally encoded toxin required for growth inhibition but dispensable for viral replication and virion production. Viruses lacking ORF a291 replicated their genomes and formed morphologically normal spindle-shaped particles, yet failed to inhibit the growth of uninfected Saccharolobus solfataricus. Substitution of residues at a predicted N-terminal signal peptide cleavage site abolished growth suppression without affecting replication, suggesting that secretion is essential for toxin function. Background: Venous thromboembolism (VTE) includes deep vein thrombosis (DVT) and pulmonary embolism (PE), the latter of which often originates from DVT and can be fatal.
Objectives: The aim of this study was to identify causal genetic factors that alter risk of PE among those with a presumed DVT.
Methods: Using a case-only design of VTE, we conducted meta-analyses of genome-wide association studies (GWASs) in the International Network Against Venous Thrombosis Consortium. Over time, environmental plastic will degrade into smaller pieces, which sea turtles can mistake for prey. Fasciola hepatica, the common liver fluke, is endemic in the Pacific Northwest and Gulf Coast of the United States. To provide updated information on F. hepatica prevalence, as part of the USDA National Animal Health Monitoring System Sheep 2024 study, sheep operations in the top 30 sheep-producing states were identified, and producers were invited to participate. For each operation, feces were collected from at least eight sheep and pooled to form a single composite sample. [Objective] The variations in inorganic element content and the safety of heavy metals and harmful elements in Ranunculi ternati radix from different origins were investigated. [Methods] The quantitative analysis of 24 elements in the samples was performed by inductively coupled plasma mass spectrometry (ICP-MS). Health risk assessment was conducted using estimated daily intake (EDI), target hazard quotient (THQ), and carcinogenic risk (CR).
