Monoamine Oxidase As A Target Enzyme In Pineal And Testicular Tissue: Adrenal Cortical Steroids And Arginine Vasotocin, Randall James Merrill

The purpose of the research was twofold: (a) To ascertain what effects the adrenal glucocorticoids and catecholamines have on pineal N-acetyltransferase and monoamine oxidase activity, and (b) to observe what effect arginine vasotocin (a pineal polypeptide) has on testicular monoamine oxidase activity.

Rat pineal N-acetyltransferase activity was numerically increased when rats were injected with cortisol. It was increased significantly when the animals were injected with cortisol plus norepinephrine. Adrenal demedullation significantly increased N-acetyltransferase activity in starved rats when compared with intact control animals. Moreover, bovine pineal monoamine oxidase activity was markedly inhibited in a dose-dependent manner when incubated in vitro …

The purpose of this study was to develop a diagnostic test and conduct a survey for the neonatal calf diarrhea virus (NCDV) and human infant reolike diarrhea virus. Two immunologic methods were developed in this investigation.

Immune electron microscopy (IEM) and the fluorescent viral precipitin test (FVPT) are the methods used to detect NCDV and the human virus. Both methods are based upon the principle that viral aggregates form when the virus is reacted with anti-NCDV antibody. There is a paucity of information concerning the occurrence and distribution of the Saprolegniaceae of Illinois. These fungi, commonly called "water molds", are of particular interest as many are potential parasites of fish, crustaceans, and other aquatic plant and animal life. It is of considerable interest to students of the fungi and of practical value to the fisheries biologists and environmentalists to have available a means of identification for these organisms.

Cartilage from newborn mice homozygous for chondrodysplasia contains normal quantities of protein and chondroitin sulfate. Mutant fetal cartilage incorporates normal amounts of sulfate, glucose, and glucosamine. Overnight rinsing with water removes about 25% of the incorporated sulfur and existing uronic acid from mutant ribs and about 55% from mutant limbs. Control ribs and limbs are affected very little by such a rinse. The diffusability of chondroitin sulfate (represented by incorporated sulfur and uronic acid) may be due to a lack of covalent bond formation between chondroitin sulfate and collagen.

Despite the extensive data that have accumulated on the detrimental effect of molds on the growth of farm animals, very little is known as to the beneficial role of fungi in animal feed. Recent studies at South Dakota State University research laboratories revealed, however, that only 164 of 392 strains of Aspergilli were found to be toxic. When the cultures were grown on sterile soybeans or wheat, some of the tested cultures exerted positive effects on growth of chicks and mice. A Taxonomic And Distributional Survey Of The Benthic Blue-Green Algal Flora Of The Skokomish River Estuary, Louis J. Boitano

This paper presents a taxonomic and distributional survey of the benthic blue-green algal flora within an estuary system. Physical parameters were considered in the seasonal growth and distribution of algal species. A majority of the observed species occur in all habitats of the estuary and are present throughout the year. Seasonal growth rates and distribution are primarily dependent upon light intensity, temperature, salinity, and substrate condition. The largest extent of growth and distribution occurs in the late spring and summer months at which time the abiotic factors are most favorable for the development of benthic algal communities.

Phenological and cultural studies of common dandelion (Taraxacum officinale Weber) were conducted from 1969 to 1971 at Bowling Green, Kentucky. The phenological studies were based upon dandelion plants which were found growing in a Kentucky bluegrass lawn or in open lots. At this location the dandelion flowered throughout the year. Maximum flowering occurred during April and a secondary peak occurred in September and October. The number of flowering stems per square meter was not correlated significantly with precipitation, temperature, or day length. Length of stem was positively correlated with temperature and day length. The plants exhibited diurnal variation in flowering …

Light And Electron Microscopy Investigation Of Developing Wheat Caryopses Infected By Tilletia, Melvin Douglas Grove

Caryopses of wheat were studied during infection by Tilletia caries and T. controversa. Initially the pathogens penetrated embryos of the host. After the embryo tissue was consumed by the fungi, endosperm tissue was penetrated and consumed. Initial penetration of endosperm tissue was in localized areas. During infection the fungus mycelium spread intercellularly, but occasionally was seen intracellularly. As the host tissue was consumed teliospores were formed. At maturity a host cell layer 4-6 cells thick remained which retained the mature teliospores. Histochemical studies indicated that carbohydrates and lipids of the host disappeared soon after invasion by the pathogens. Nuclei and …

In an attempt to elucidate the role of nematodes in a grassland ecosystem nematode taxa, number and biomass data was gathered over a three year period at the Cottonwood International Biological Program Grassland Biome site in western South Dakota. The effects of grazing intensity, sampling date and sampling depth were studied. Nematicide treatment of range grasses in field and greenhouse studies was also used to evaluate the role of nematodes. Biomass data was used to estimate the intake of plant feeding nematodes. Quantitative Growth Relationships Of Microcoleus Vaginatus (Gom.) Ex Drouet And Alcaligenes Sp. In A Sucrose-Salts Medium, James Rexford Brownell

The blue-green alga, Microcoleus vaginatus (Gom.) ex Drouet 1968, and a bacterium, Alcaligenes sp., collected from the same habitat, were cultured together and separately in mineral salts media with and without sucrose. Growth of axenic M. vaginatus was stimulated by sucrose. In association with the bacterium in sucrose-containing media algal growth was suppressed, while in inorganic media it was stimulated. Factors involved in this algal-bacterial system probably included competition for the carbon source, changes in pH, and accumulation of cellular waste and lytic products.

Genetic control of decapitation-induced fission in planaria is mediated by stimulator and inhibitor molecules. Resolution of the two factors was accomplished by filtration chromatography of whole planaria extract. The stimulator (M.W. 2500 to 3500) was purified with ion exchange chromatography and increases the multiple fission rate. The inhibitor (M.W. 20,000 to 50,000) is non-dialyzable, heat labile and is inactivated unless kept frozen. Storage at 4°C results in a loss of inhibitory capacity with a concomitant stimulator increase, suggesting a protein-stimulator complex as the inhibitor. The flowering pattern of flax (Linum usitatissimum, L.) is cyclic in nature with periods of blossoming separated by periods of rest. To study the regulation of flowering cycles, four experiments were conducted. The first two dealt with nutritional aspects of cyclic flowering by imposing environmental stresses which conceivably might regulate flowering pattern nutritionally. The third and fourth experiments dealt with regulation from hormonal systems and included in vitro regulation of bud growth by growth regulators as well as characterization of endogenous hormonal systems by fluorimetry and bioassays. In order to obtain proper muscle development in tissue culture, certain conditions must prevail. There is a delicate balance of factors, and elimination of even one of these factors leads to poor and improper development. Muscle cells grown in the absence of fibroblasts usually fail to differentiate. Therefore, proper development depends upon the presence of two cell types: myoblasts and fibroblasts. Investigators have demonstrated that the fibroblasts are necessary in that they produce collagen which is necessary for proper muscle development. Newer diagnostic techniques, including radioisotope liver scans, bacteriologic culture procedures for anaerobes, serologic tests for amebiasis, ultrasonic echograms, and hepatic arteriography, permit more specific identification and localization of hepatic abscesses. Large pyogenic abscesses are of particular Importance to surgeons; are frequently caused by anaerobic organisms; and require early, direct, adequate, and sufficiently prolonged surgical drainage. The clinician should be alert also to recognition and management of the primary lesion responsible for the hepatic abscess, associated abscesses elsewhere, and the possible presence of an underlying liver neoplasm.

A study of the phytoplankton periodicity and its correlation with chemicophysical parameters, diel variation of the plankton, and the cyclomorphotic variation of Ceratium hirundinella was made in Shanty Hollow Lake, Warren County, Kentucky, from October, 1969, to August, 1970.

Phytoplankton samples were collected at zero and six feet with a Kemmerer sampler (31 October through 8 December) and with a ten -liter Birge-Juday plankton trap (23 December through 9 August). Deoxyribonucleic acid (DNA) analyses were used to assess on a molecular level, the relationships among representatives of the genera Streptomyces, Nocardia and Mycobacterium. The methods developed in this study have been used for routine analysis of DNA from a large number of actinomycetes and have given reliable and reproducible data.

DNA isolated from various actinomycetes was characterized by buoyant density determinations in CsCl from which the mole fraction guanine plus cytosine (GC) content was calculated. All the streptomycete DNA preparations studied had buoyant densities in the range of 1.7287 to 1.7312 g cm-3 which corresponded to GC compositions of …

The chemical composition of algae grown in batch culture depends mainly on environmental conditions, nutrient availability, presence of predators, cell age, and species. The effects of nutrient availability and cell age on the composition of three unialgal cultures (algae + bacteria) and one hetergeneous culture (algae + bacteria + microscopic animals) were evaluated. The cultures were grown in batch culture under both nutrient-abundant and nutrient deficient conditions and the changes in compositions were observed. Samples of aqueous humor were obtained from the anterior chamber of patients with uveitis. Bacterial L-forms, or cell wall deficient forms, were recovered from 11 of 19 eyes with unexplained uveitis. These bacterial variants were cultured on newer, highly specialized media and stained with acridine orange. This stain demonstrates fluorescence of nucleic acids under UV microscopy.
