The shooting method consists of guessing unknown initial values, transforming a second-order nonlinear boundary value problem (BVP) to an initial value problem and integrating it to obtain the values at the right end to match the specified boundary condition, which acts as a target equation. In the shooting method, the key issue is accurately solving the target equation to obtain highly precise initial values. Due to the implicit and nonlinear property, we develop a generalized derivative-free Newton method (GDFNM) to solve the target equation, which offers very accurate initial values. A noise abatement method using an eigen function in conjunction with an optimizer (Genetic Algorithm) under a space-constrain situation is proposed in this study concerning a high order wave propagation effect. An eigen function is used to describe an acoustical field and shorten the acoustical model in a four-pole transfer matrix form. The system’s four-pole matrix estimates the Transmission Loss (TL) for the silencers by multiplying individual four-pole matrices.

The aim of this study was to identify the sources of uncertainties of typhoon wave field monitoring using HF radar and to quantitatively assess the bias of the wave parameters, such as significant wave height and mean period, retrieved under various conditions. The strategy was to apply a purely numerical simulation of the Doppler-range spectra and then compare the estimation results to the target. Marine debris is a global problem that has not been resolved. This has encouraged the emergence of marine debris cleaning technologies, one of which is a conveyor ship. However, how effective conveyors are in collecting waste has not been studied. In this paper, experimental research on conveyors as marine debris collectors is investigated. The capability of catamarans with or without solid wing conveyors to collect marine debris is explored. The paper considers the second-order nonlinear boundary value problem (NBVP), which is equipped with nonlinear integral boundary conditions (BCs). Two novel iterative algorithms are developed to overcome the difficulty of NBVP with double nonlinearities involved. In the first iterative algorithm, two nonlocal shape functions incorporating the linear integral terms are derived, and a nonlocal boundary shape function (NBSF) is formulated to assist the solution. The growing threat of antimicrobial resistance is among the largest concerns in the world today. One method under development to combat this issue is the encapsulation of microbes in microfluidic droplets for single-cell testing. This method may be able to circumvent the need for a traditional positive cell culture which consumes the majority of the testing time using current diagnostic methods. This dissertation presents a method by which to deterministically encapsulate microbes using an artificial intelligence object detection algorithm and a Droplet-On-Demand microfluidic device. The parathyroid glands (PTGs) are often damaged during thyroid surgeries due to a lack of methods identifying PTGs and assessing their viability. Damage to PTGs can cause hypocalcemia, a deficiency of calcium in the body. This complication can lead to detrimental consequences with economic burden. Vascular smooth muscle cells (VSMCs), which provides structural integrity and regulates the diameter of vasculature, are of great potential for modeling vascular-associated diseases and tissue engineering. Here, we presented a detailed comparison of differentiating human pluripotent stem cells (hPSCs) into VSMCs (hPSCs-VSMCs) in four different culture methods, including 2-dimensional (2D) culture, 3-dimensional (3D) PNIPAAm-PEG hydrogel culture, 3-dimensional (3D) alginate hydrogel culture, and transferring 3- dimensional alginate hydrogel culture to 2-dimensional (2D) culture. Active food packaging materials enhance the shelf-life of food products while reducing food waste. The current study aims to develop a biodegradable active food packaging material. The food packaging material was developed with the incorporation of clove essential oil, sodium alginate, gelatin, and nanoclay films were prepared. The influences of nanoclay and clove on the surface, optical, mechanical, chemical, barrier, and pH-indicating properties were studied. The lightness and yellowness increased by 1.06 folds and 3.34 folds when compared to clove (control), respectively. Bionanocomposite packaging eco-friendly alternatives with enhanced characteristics. This study aimed to develop a bionanocomposite intelligent packaging. Sodium alginate, gelatin, Curcumin (Cur), glycerol, and Nanoclay (NC) films were prepared. The influences of nanoclay and curcumin on the surface, optical, mechanical, chemical, barrier, and pH-indicating properties were studied. The results showed that the lightness of films was reduced by 1.28 folds compared to NC (control) film, while the yellowness of films increased by 5.82 folds. Imaging of both the positions and orientations of single fluorophores, termed single-molecule orientation-localization microscopy, is a powerful tool for the study of biochemical processes. However, the limited photon budget associated with single-molecule fluorescence makes high-dimensional imaging with isotropic, nanoscale spatial resolution a formidable challenge. Here we realize a radially and azimuthally polarized multi-view reflector (raMVR) microscope for the imaging of the three-dimensional (3D) positions and 3D orientations of single molecules, with precisions of 10.9 nm and 2.0° over a 1.5-μm depth range. This work presents an in-depth numerical investigation into a hypothesized two-layer central pattern generator (CPG) that controls mammalian walking and how different parameter choices might affect the stepping of a simulated neuromechanical model. Particular attention is paid to the functional role of features that have not received a great deal of attention in previous work: the weak cross-excitatory connectivity within the rhythm generator and the synapse strength between the two layers. Fibrin is a natural polymer already used in extreme clinical settings as an adhesive and for shallow wounds when activated by the enzyme thrombin. However, current formations contain irritants, are slow drying, and are not easily washed off. This study used a spray-on bandage composed of thrombin and fibrinogen on a monolayer of 3T3 mouse fibroblast cells to test the enhancement of the body's natural healing without the risks of current formulations. The glass transition of branched polymers is determined by multiple structural parameters that dictate their inter- and intramolecular interactions, and ultimately, their molecular packing in the amorphous phase. Here we examined the impact of side chain length, backbone length, molecular weight composition, and topology on the glass transition behavior of bottlebrush polymers. Through examining precision bottlebrush polymer libraries (PBP, ĐSC = 1.0), we find the infinite molecular weight Tg is reached at a specific brush length after which the effect of the side-chain length dominates. Arctic warming is affecting snow cover and soil hydrology, with consequences for carbon sequestration in tundra ecosystems. The scarcity of observations in the Arctic has limited our understanding of the impact of covarying environmental drivers on the carbon balance of tundra ecosystems. In this study, we address some of these uncertainties through a novel record of 119 site-years of summer data from eddy covariance towers representing dominant tundra vegetation types located on continuous permafrost in the Arctic. Tissue indentation response is an important metric for understanding how different musculoskeletal regions respond to loading and is a function of the tissue’s form. Modem imaging techniques provide information about the internal structures of human tissue. Ultrasound remains one of the most common imaging techniques performed, given its portability and low costs. Prior work and data collection on 100 patients involved the collection of ultrasound images at eight different locations across the musculoskeletal extremities. The impact of secondhand smoking to health of general population, and specifically children, is a well-known phenomenon that researchers have studied for years. In this dissertation, I extend work done within a secondhand smoke intervention to understand and quantity the impact of intervention on flow of smoke air particle concentration from the main room where smoking generates air particle contamination to a room where a child living in the home sleeps.
