Laparoscopy, a minimally invasive surgical technique, has revolutionized the field of surgery by replacing traditional open procedures with smaller incisions and reduced patient trauma. However, the current design of manual laparoscopic devices limits the range of motion for surgeons, restricting their maneuverability to only two degrees of freedom (DOF). This dissertation is a study aimed at addressing this limitation by designing and developing a robotic articulating laparoscopic instrument capable of providing 7-DOF movement.

Multivariate Categorical Data (MCD) plays a significant role in many industries, and the ability to understand the data is critical for insight and decision making. Visualization is a key tool for understanding the data. This dissertation designed and implemented a novel technique for visualizing MCD called Pivoting Parallel Charts (PPC). The design of PPC was informed by studying several existing MCD visualization techniques.
PPC visualizes MCD as a sequence of parallel axes with affixed bar charts. A user-specified axis, called the pivot, acts as the crucial point of consideration for all data relationships. The bar charts are color-coded by the …

Cancer is one of the deadliest diseases worldwide and has been responsible for millions of deaths. However, developing a satisfactory smart multifunctional material combining different strategies to kill cancer cells poses a challenge. This work aims at filling this gap by developing a composite material for cancer treatment through hyperthermia and drug release. With this purpose, magnetic nanoparticles were coated with a polymer matrix consisting of poly (L-co-D,L lactic acid-co-trimethylene carbonate) and a poly(ethylene oxide)–poly(propylene oxide)–poly(ethylene oxide) triblock copolymer. Elucidating physical transport phenologies in large lakes can aid understanding of larval recruitment dynamics. Here, we integrate a series of climate, hydrodynamic, biogeochemical, and Lagrangian particle dispersion models to: (1) simulate hatch and transport of fish larvae throughout an illustrative large lake, (2) evaluate patterns of historic and potential future climate-induced larval transport, and (3) consider consequences for overlap with suitable temperatures and prey. Simulations demonstrate that relative offshore transport increases seasonally, with shifts toward offshore transport occurring earlier during relatively warm historic and future simulations. Dimension reduction methods are used to visualize the output of unsupervised learning models when applied to complex data. These techniques improve interpretability by transforming a high-dimension space to a lower-dimension space (usually 2D or 3D). The results are typically viewed as 2D scatter plots, and class centroids may be added to increase interpretability. Although useful, the relationship of these class centroids to the underlying feature space remains opaque. The innovative aspect of this work is to create a strong link between the dimension-reduced space and the underlying high-dimension feature space by adding selected feature centroids to the 2D scatter plots. …

A novel cylindrical cavity resonator-based measurement method was developed to characterize the dielectric property of the low-loss dielectric liquid in the paper. The proposed method can be used to accurately measure the dielectric constant (Dk) and the loss tangent (Df) of the low-loss coolant for signal integrity analysis. The cylindrical cavity resonator works at tangential magnetic (TM010) mode is used in the paper to demonstrate the proposed method. Microwave displacement sensors have garnered significant research interest in recent years and have found successful applications in industrial automation and aerospace engineering. However, most microwave sensors are limited to measuring in-plane displacement, with the assumption that out-of-plane displacement remains constant during operation. In this work, we propose and experimentally demonstrate a novel concept for 2-D micro displacement sensing that simultaneously measures both in-plane and out-of-plane displacement. In this paper, we propose and demonstrate a novel concept of segmented fiber optic sensors by integrating the fiber Bragg grating (FBG) reflector modality and a hybrid interrogation technique enabled by microwave photonics. As a proof of concept, a radiofrequency Fabry-Perot interferometer (FPI) based on an optical fiber with two FBGs as the two reflectors of the Fabry-Perot (FP) cavity is constructed. By measuring the frequency response of the FPI device followed by a joint-time-frequency-domain analysis, the interferogram of the FPI in the microwave domain and the time-domain signal of the FBGs can be unambiguously reconstructed. Motivation: This study investigates the flexible refinement of AlphaFold2 models against corresponding cryo-electron microscopy (cryo-EM) maps using normal modes derived from elastic network models (ENMs) as basis functions for displacement. AlphaFold2 generally predicts highly accurate structures, but 18 of the 137 models of isolated chains exhibit a TM-score below 0.80. We achieved a significant improvement in four of these deviating structures and used them to systematically optimize the parameters of the ENM motion model.

Exceeding physiological limits of the cell membrane potential compromises structural integrity, enabling the passage of normally impermeant solutes and disrupting cell function. Electropermeabilization has been studied extensively at the cellular scale, but not at the individual membrane lesion level. We employed fast total internal reflection fluorescence (TIRF) imaging of Ca(2+) entry transients to discern individual lesions in a hyperpolarized cell membrane and characterize their focality, thresholds, electrical conductance, and the lifecycle. A diffuse and momentary membrane permeabilization without a distinct pore formation was observed already at a -100 mV threshold. Polarizing down to -200 mV created focal pores with a …
