Intracellular reactive oxygen species (ROS) generation is widely suggested as a trigger for biological consequences of electric field exposures, such as those in electroporation applications. ROS are linked with membrane barrier function degradation, genetic damage, and complex events like immunological cell death. Dihydroethidium (DHE) is commonly used to monitor ROS in cells. DHE is linked to intracellular ROS by a primary oxidation product, Ethidium (Eth+), that shows increased fluorescence upon binding to polynucleotides. We observed changes in DHE-derived fluorescence in Chinese hamster ovary (CHO) cells post 300-ns electric pulse exposures, comparing them to tert-butyl-hydroperoxide (t-BHP) induced oxidative stress. …

The integration of Indigenous and Western science plays an essential role in Tribally led collaborations for land management. This process of woven sciences is rooted in reciprocal relations and partnerships guided by Tribal Nations. Our cohort was invited by Wiyot Tribal Representatives to investigate a culvert located within the wetlands of Mouralherwaqh— a parcel of land reacquired by the Wiyot Tribe in 2022. Nanosecond pulsed atmospheric pressure plasma jets (ns-APPJs) produce reactive plasma species, including charged particles and reactive oxygen and nitrogen species (RONS), which can induce oxidative stress in biological cells. Nanosecond pulsed electric field (nsPEF) has also been found to cause permeabilization of cell membranes and induce apoptosis or cell death. Combining the treatment of ns-APPJ and nsPEF may enhance the effectiveness of cancer cell inactivation with only moderate doses of both treatments. "Underground mine planning engineers face significant challenges when determining what geometry provides the most profitable and safe stope for extraction. Several techniques and optimization algorithms have been developed in recent years, but most fail to find optimal solutions because they are heuristic or LP-based without efficient geometric constraints. This thesis work proposes a two-dimensional binary linear programming (BILP) model for determining the optimal combination of blocks in a stope that maximizes the economic value of the layout of stopes for a sublevel deposit. This parametric study aimed to analyze the effects of increased magnetic field exposure (MFE) on the growth and production of the bioactive compounds of Chlorella (C.) vulgaris. With the intent of studying the effect of an increased MFE, the magnetic field typically experienced by life on Earth was amplified by an order of magnitude. In the increased-MFE environment, six treatments of C. vulgaris with two repetitions for each treatment were exposed to a magnetic field of 5 Gauss (500 µT) about each axis, which was generated in a state-of-the-art Helmholtz cage. This paper introduces a path planning algorithm for a system with squid dynamics in a cluttered environment. We capture the complex interactions of fin, arms, and body patterning by analyzing experimental data collected from observing squid motion. We extract nine motion primitives to build the control sequence for a time-optimal trajectory. This task is formulated as a mixed-integer program, and we generate the minimum-time trajectory using a sample-based approach. In This Research, We Explored the Synthesis and Application of Thiolactone Acrylamide (TLA) Polymers for Neutralizing Reactive Oxygen Species (ROS), Produced as a Result of Traumatic Brain Injury (TBI). We Synthesized Well-Defined TLA Polymers using RAFT (Reversible Addition-Fragmentation Chain Transfer) Polymerization Across a Range of Monomer-To-Chain Transfer Agent ([M]0/[CTA]0) Ratios to Achieve a Range of Molar Masses. Polymerizations Were Carried Out using a Trithiocarbonate CTA (Chain Transfer Agent) with AIBN (Azobisisobutyronitrile) as the Initiator in N'N-Dimethylacetamide (DMAc) at 60 °C. The assessment of greenhouse gases to identify ways to reduce global warming was of enormous importance worldwide. This study assessed the carbon footprint of an organization and the carbon footprint of products within the herbal soap manufacturing industry, following the guidelines of the Thailand Greenhouse Gas Management Organization (TGO). It also evaluated the environmental toxicity of soap production. The study found that the organization’s carbon footprint amounted to 551 tCO2 equivalent, with Scope 3 emissions being the highest at 359 tCO2 equivalent, followed by Category 1 products and services, which accounted for 208 tCO2 equivalent. Surface-enhanced Raman spectroscopy (SERS) is an ultrasensitive spectroscopic technique that generates signal-enhanced fingerprint vibrational spectra of small molecules. However, without rigorous control of SERS substrate active sites, geometry, surface area, or surface functionality, SERS is notoriously irreproducible, complicating the consistent quantitative analysis of small molecules. While evaporatively prepared samples yield significant SERS enhancement resulting in lower detection limits, the distribution of these enhancements along the SERS surface is inherently stochastic. Acquiring spatially resolved SERS spectra of these dried surfaces, we have shown that this enhancement is governed by a power law as a function of analyte concentration. TGF-β1 is a potent regulator of epithelial-mesenchymal transtion (EMT), a cellular
transdifferentation process that promotes cellular motility and matrix remodeling, critical to tumorigenesis and metastasis. TGF-β1-induced EMT promotes the assembly of FN, which synergistically binds latent TGF-β1 to aid in its activation. The purpose of of this work is to further probe the effects of autocrine TGF-β1 signaling during EMT progression by utilizing CRISPR-Cas9 deletion of the TGFB1 gene in in vitro models of triple-negative breast cancer. However, labeled activity recognition datasets are costly to collect and highly varied, which challenges machine learning techniques that rely on large datasets. Electrospun polymeric biodegradable scaffolds are essential in tissue engineering, particularly for Engineered Tissue Vascular Grafts (ETVGs), which promise advancements in treating coronary artery disease, peripheral arterial disease, congenital cardiovascular defects, and renal disease. These scaffolds present a solution to issues with autologous graft availability and durability. While large-diameter grafts in low-pressure environments have seen success, small-diameter grafts in high-flow scenarios remain challenging. Understanding polymeric scaffold degradation and behavior during incubation, especially under dynamic mechanical loading, is vital for clinical translation of small-caliber ETVGs.

The purpose of this thesis is to test biopolymers synthesized from oilseeds or produced from agricultural wastes for large bale net wrapping as a viable alternative to conventional petroleum-based plastics (PBP). The goal is to test and assess novel polymers to guide the synthesis of a benign biodegradable polymer. Biobased net wrapping will significantly reduce ingestion of PBPs in animals such as ruminants to improve digestive health and minimize cattle loss due to plastic disease. Problematic soil like expansive soil, and frost-susceptible soil causes significant damage to civil infrastructure. The use of calcium-based stabilizers in the treatment of sulfate-rich expansive soils is not suggested due to the formation of ettringite. Infrastructure such as pavements and embankments built on expansive soil are often exposed to the damaging impacts of freeze-thaw cycles in areas prone to seasonal freezing making them vulnerable to cracking and spalling. Schwann cells are the principal glia in the peripheral nervous system, supporting normal peripheral nerve function, particularly in the myelination of motor and sensory axons of the peripheral nerves. Disruptions in normal Schwann cell function such as injury and disease can lead to decreased motor function, sensory capability, and neuropathic pain. Efforts to treat this do not always have consistent or favorable outcomes, so there has been an increased interest in developing tissue engineering solutions to address these problems.
