The goal of the Malaria Vaccine Program at the Walter Reed Army Institute of Research (WRAIR) is to develop a licensed multi-antigen, multi-stage vaccine against Plasmodium falciparum able to prevent all symptomatic manifestations of malaria by preventing parasitemia. A secondary goal is to limit disease in vaccinees that do develop malaria. Malaria prevention will be achieved by inducing humoral and cellular immunity against the pre-erythrocytic circumsporozoite protein (CSP) and the liver stage antigen-1 (LSA-1). The strategy to limit disease will target immune responses against one or more blood stage antigens, merozoite surface protein-1 (MSP-1) and apical merozoite antigen-1 (AMA-1). The …

We developed a real-time controller for a 2 degree-of-freedom robotic system using xPC Target. This system was used to investigate how different methods of performance error feedback can lead to faster and more complete motor learning in individuals asked to compensate for a novel visuo-motor transformation (a 30 degree rotation). Four groups of normal human subjects were asked to reach with their unseen arm to visual targets surrounding a central starting location. A cursor tracking hand motion was provided during each reach. The effects of cattle grazing on stream stability have been well documented for the western portion of the U.S., but are lacking for the east. Stream and riparian damage resulting from grazing can include alterations in watershed hydrology, changes to stream morphology, soil compaction and erosion, destruction of vegetation, and water quality impairments. However, few studies have examined the successes of best management practices (BMPs) for mitigating these effects. This Thesis presents an analytical approach for generation of control inputs for a non-holonomic mobile robot in coverage control. The diffusion of single n-butane and n-decane molecules on a model stepped surface, Pt(655), and on a corresponding flat surface, Pt(111), is investigated using molecular-dynamics simulations and anisotropic united atom model. The surface step on Pt(655) causes the alkane molecules to adsorb on the lower terrace in all-trans conformations with their long molecular axes adjacent and parallel to the step edge, and to diffuse anisotropically along the surface step via a constant wiggly motion without rotation or marked deviation from the parallel adsorption configuration. Discrete models are used in describing various microscopic phenomena in many branches of science, ranging from biology through chemistry to physics. Arrays of evanescently coupled, equally spaced, identical waveguides are prime examples of optical structures in which discrete dynamics can be easily observed and investigated. As a result of discretization, these structures exhibit unique diffraction properties with no analogy in continuous systems. Recently nonlinear discrete optics has attracted a growing interest, triggered by the observation of discrete solitons in AlGaAs waveguide arrays reported by Eisenberg et al. in 1998. Enhanced Biological Phosphorous Removal (EBPR) performance was found to be adequate with reduced Return Activated Sludge (RAS) flows (50% of available RAS) to the anaerobic tank and smaller than typical anaerobic zone volume (1.08 hours hydraulic retention time or HRT). Three identical parallel biological nutrient removal (BNR) pilot plants were fed with strong, highly fermented (160 mg/L VFAs), domestic/industrial wastewater from a full scale wastewater treatment facility (WWTF). The pilot plants were operated at 100%, 50%, 40% and 25% RAS (percent of available RAS) flows to the anaerobic tank with the remaining RAS to the anoxic tank. In addition, varying …

We propose a novel method to overcome significant problems of baseline drift and sensitivity degradation in amperometric biosensors based on oxidase enzyme reactions. A novel glucose microsensor with a built-in electrochemical oxygen manipulation microsystem is introduced to demonstrate three novel functionalities; one-point in situ self-calibration (zero-point), broadening of dynamic range and increase in sensitivity. The influence of electrochemically generated oxygen microenvironment on the sensor output within a fluidic structure is investigated.

Backpropagation algorithm is the most commonly used algorithm for training artificial neural networks. While being a straightforward procedure, it suffers from extensive computations, relatively slow convergence speed and possible divergence for certain conditions. The efficiency of this method as the training algorithm of a radial basis function neural network (RBFN) is compared with that of particle swarm optimization, for neural network based identification of a small power system with a static compensator. The comparison of the two methods is based on the convergence speed and robustness of each method.

There is an urgent need for tools to unravel the complex interactions and functionalities of genes. As such, there has been much interest in reverse-engineering genetic regulatory networks from time series gene expression data. We use an artificial neural network to model the dynamics of complicated gene networks and to learn their parameters. The positive and negative regulations of genes are defined by a weight matrix, and different genes are allowed to have different decaying time constants. We demonstrate the effectiveness of the method by recreating the SOS DNA Repair network of Escherichia coli bacterium, previously discovered through experimental data.

Large-scale time series gene expression data generated from DNA microarray experiments provide us a new means to reveal fundamental cellular processes, investigate functions of genes, and understand their relations and interactions. To infer gene regulatory networks from these data with effective computational tools has attracted intensive efforts from artificial intelligence and machine learning. In NiTi, a reversible thermoelastic martensitic transformation can be induced by temperature or stress between a cubic (B2) austenite phase and a monoclinic (B19') martensite phase. Ni-rich binary compositions are cubic at room temperature (requiring stress or cooling to transform to the monoclinic phase), while Ti-rich binary compositions are monoclinic at room temperature (requiring heating to transform to the cubic phase). The stress induced transformation results in the superelastic effect, while the thermally induced transformation is associated with strain recovery that results in the shape memory effect. The primary purpose of this study is to identify the 'universal' rate formulations with scaled-dependent parameters for the biological reduction of hematite. Three possible rate formulations were proposed to describe the bioreduction rate of hematite, and two kinds of simulation were conducted to validate the formulations and parameters with both batch and column experimental data: a reaction-based biogeochemical (batch) modeling with BIOGEOCHEM 1.0 and a reactive biogeochemical transport (column) modeling via HYDROGEOCHEM 4.0.
