In bacterial adaptation to the dynamic environment, metabolic genes are typically thought to be the executors, whereas global transcription regulators are regarded as the decision makers. Although the feedback from metabolic consequence is believed to be important, much less is understood. This work demonstrates that the gluconeogenic genes in Escherichia coli, ppsA, sfcA, and maeB, provide a feedback loop to the global regulator, cAMP receptor protein (CRP), in carbon source transition. Disruption of one of the gluconeogenic pathways has no phenotype in balanced growth, but causes a significant delay in the diauxic transition from glucose to acetate. To investigate the …

The composition of natural bone comprises of collagen (Cg) as organic phase which acts as a matrix and inorganic calcium phosphate, particularly hydroxyapatite (HA), which is used as reinforcement. This study involved the wet-spinning of collagen fibers incorporating HA in mixed and coated form. The weight percent ratio of collagen and HA in the composite fibers was 95 to 5 respectively. HA coating on pure collagen fibers was achieved by immersing them in HA solution formed by mixing HA in water. In this research, the Haptic Master robotic arm and virtual environments are used to induce motor learning in subjects with no known musculoskeletal or neurological disorders. It is found in this research that both perception and performance of the subject are increased through the haptic and visual feedback delivered through the Haptic Master. These system benefits may be extended to enhance therapies for patients with loss of motor skills due to neurological disease or brain injury.

The healthy heartbeat is traditionally thought to be regulated according to the classical principle of homeostasis whereby physiologic systems operate to reduce variability and achieve an equilibrium-like state. However recent studies reveal that under normal conditions, beat-to-beat fluctuation in heart rate display the kind of long-range correlations typically exhibited by the dynamical system far away from equilibrium. Heart rate variability (HRV) has proven to be a useful noninvasive tool to study the neuronal control of the heart. Recently, nonlinear dynamic methods based on chaos theory and fractal dynamics have been developed to uncover the nonlinear fluctuations in heart rate. Approximate Entropy (ApEn), Sample Entropy (SampEn) and Multi-scale Entropy (MSE) are measures based on chaos theory that quantify the regularity in time series. Disclosed are system and method for a characterizing system consisting of a fluid sample on a two-sided stage, utilizing data obtained by applying, from both sides thereof, beams of electromagnetic radiation to a fluid coated surface in a containing cell volume. The beams can have the same or different wavelength content and/or polarization state and can be applied at the same or different magnitude angles-of-incidence, and a third typically unpolarized beam can be applied at a normal angle-of-incidence.

The buildup of phosphorus (P) in the soil is a major factor limiting the operating life of a wastewater land treatment system. In this study, we evaluated changes of chemical properties, P profiles, and adsorption isotherms in the soils of a Muskegon wastewater land treatment system, which has received wastewater for ~30 years. It was found that the pH in the 15-cm topsoil increased from ~5-6 in 1973 to ~7.4-7.8 in 2003; a large amount of salt (e.g., Ca, Mg) in wastewater was adsorbed by the soil; the soil Al content (either exchangeable or oxalate extractable) decreased, while the oxalate-extractable …

Anthrax spores, in weaponized form, are dangerous biological warfare agents. Handheld technology for the rapid detection of anthrax is greatly needed to improve national security. Methods to detect anthrax spores are diverse, with most taking at least an hour for positive identification. A viable option for rapid detection is analytical pyrolysis (AP), which produces chemicals containing taxonomical information (biomarkers). AP methods are reviewed and critically analyzed to show that reproducible detection of anthrax spores in a rapid manner (< 5 min) with a handheld device is not currently possible. A promising alternative to AP is the use of a catalyst to produce biomarkers from anthrax spores with improved selectivity and reproducibility. Catalytic materials having promise for this include platinum, nickel, and superacids. Experiments evaluating several of these materials are described. A biomarker mass spectral library was created, based on information available in the scientific literature, to facilitate analysis and identification of the biomarkers produced experimentally. The RAMFAC algorithm was used to deconvolute chromatographic peaks to produce clean mass spectra and match them against entries in the biomarker library. While the library is not complete, its use with the RAMFAC algorithm enabled detection of many important biomarkers in experiments involving catalytic breakdown of anthrax spores. Experimental results from preliminary tests of several catalysts are presented and discussed. Addition of catalysts in the form of platinum nanoclusters and superacids to bacterial spores in a commercial pyrolyzer effected an increase in the amount of biomarkers produced at mild conditions over traditional pyrolysis methods. Electroformed nickel mesh, on the other hand, demonstrated low catalytic activity for the production of biomarkers, likely due to poor contact of the spores with the mesh. Biomarkers similar to those published in the literature were observed, including dipicolinic acid, picolinic acid, propionamide, acetamide, diketopiperazines, fatty acids, furfuryl alcohol, and DNA bases. A statistically designed factorial study was used to determine the importance of temperature, spore loading, and nanocluster loading on the production of three important biomarkers. The relative importance of these variables differs for each of the three important biomarkers, suggesting they are produced by different reaction mechanisms.

Water restrictions, pollution control, volume balances, and the emergence of stormwater utilities have lead to the development of an automated intelligent system (“I-Water”) for water use and control. With the use of this system, water stored in stormwater ponds or in the surficial aquifer is not discharged to surface waters because it is used to meet water demands, such as, lawn irrigation, environmental protection, agriculture, drinking and industrial uses.

Chitosan [<(1-4)-2 amino-2-deoxy-d-glucose], the natural polyaminosaccharide derived from N-deacetylation of chitin [<(1-4)-2 acetamide-2-deoxy-d-glucose], has been shown to possess attractive biological and cell interactive properties. Recently chitosan and chitosan analogs have also been shown to support the growth and continued function of chondrocytes. In the present study, chitosan substrates are crosslinked with a func-tional diepoxide (1,4 butanediol diglycidyl ether) to alter its mechanical property, and the viability and proliferation of the canine articular chondrocytes seeded on the crosslinked surface are further assayed. Of interest is the impact of substrate stiffness on the growth and proliferation of articular canine chondrocytes. Cross linked scaffolds were also sub-jected to degradation by chitosanase to examine the impact of cross linking on enzyme- assisted degradation. The hydrophilicity and compression modulus of the crosslinked sur-faces were measured via contact-angle measurements and compression tests, respec-tively. Scanning electron microscopy (SEM) and fluorescent staining were used to ob-serve the proliferation and morphology of chondrocyte cells on noncrosslinked and crosslinked surfaces. The crosslinked chitosan was found to be nontoxic to chondrocytes and more hydrophilic. Its compression modulus and stiffness increased, which may im-prove the scaffold resistance to wear and in vivo shrinkage once implanted. The in-creased stiffness also seemed to serve as an additional mechanical stimulus to promote chondrocyte growth and proliferation. The cell morphology on crosslinked scaffolds seen by SEM and fluorescent stain was the typical chondrocytic rounded shape. The method proposed provides a nontoxic way to increase the mechanical strength of the chitosan scaffolds.

A recombinant C-terminus heavy chain fragment from Botulinum neurotoxin serotype E (BoNT/E) is proposed as a vaccine against the serotype E neurotoxin. This fragment, rBoNTE(Hc), was produced intracellular in Pichia pastoris GS115 by a three-step fermentation process, i.e., glycerol batch phase and a glycerol fed-batch phase to achieve high cell densities, followed by a methanol fed-batch induction phase. Assessing the effect of foam layers on transport phenomena is of significant interest in many industries, so a method for predicting foam layer properties has been developed. A model of the propagation of radiation from an amplitude-modulated laser beam through a non-absorbing foam layer has been developed using diffusion theory. Measurements predicted by diffusion theory were compared to results generated using Monte Carlo methods for a variety of foam layer properties in both the time-domain and the frequency-domain. We present in this paper an investigation on a special class of wireless sensor networks for monitoring critical infrastructures that may extend for hundreds of miles in distances. Such networks are fundamentally different from traditional sensor networks in that the sensor nodes in this class of networks are deployed along narrowly elongated geographical areas and form a chain-type topology. Based on careful analysis of existing sensor network architectures, we first demonstrate the needs to develop new architecture and networking protocols to match the unique topology of chain-type sensor networks. Protein C (PC) is an important anticoagulant, antithrombotic, and antiinflammatory in blood plasma. PC deficiency can lead to severe venous thrombotic events, including lung embolism, stroke, and heart attack. In the body, PC is activated only when it is needed, and, therefore, PC does not cause bleeding problems that currently available anticoagulants may do. Purification of PC from plasma is currently done by immunoaffinity chromatography using monoclonal antibodies (Mabs), which is very expensive. Knowledge of high-frequency currents in the chip and chip-package are necessary for EMI analysis and prediction, though measurement of these currents may be difficult to obtain in many cases. One possibility is to estimate currents from near-field scan data. In this paper, measurements were made of the magnetic field over a simple circuit and a chip package. The current flowing in the circuit and the chip lead frame was estimated from the compensated near-field data and compared with measurements made directly on the pins. A training program for producers in manure use planning was evaluated. The potential for runoff nutrient loss to surface water can reduced and the value of manure increased through better planning of manure use. Training was conducted throughout Nebraska in 2002 and 2003. Livestock producers who attended evaluated the event at the end of the last class. The need for and impact of the training was greater for continuing operational and maintenance skills of manure utilization that are regularly needed for strategic planning skills. We recommend that training focus primarily on operational and maintenance skills.

This thesis seeks to extend the PDG methodology by developing a generalized formal method for nesting PDGs in an interconnected system. A procedure for decomposing an individual PDG into reusable modules will be defined and a software architecture will be presented which takes advantage of these reusable modules. This method breaks the PDG structure into discrete elements known as PDG objects, PDG modules and PDG services. Each of these elements forms a distinct unit of reuse and each can be seen as a "little" PDG. Two different industrial implementations of this method are presented. These examples show that it is …

This paper introduces the new ASAE Standard D384.2, Manure Production and Characteristics. This new standard provides an equation-based approach that integrates dry matter and nutrient intake as well as animal performance into the final estimate of total solids, nitrogen, and phosphorus excretion for seven livestock and poultry species. The manure excretion estimates of the new standard are compared with the past ASAE standard as well as other commonly used reference values for three specie groups. A commercial feedlot study was conducted to determine manure nutrient flow in six feedlots representing 6,366 cattle. On average, cattle involved in this summary were yearlings (BW = 353 kg) and gained 183 kg over 123 d. It was calculated that 11.5% of the feed nitrogen and 16.9% of the feed phosphorus was retained by the animal with the remaining nutrients excreted. On average, 25.6 kg of N and 4.1 kg of P (DM basis) were excreted per fed beef animal. The purpose of this project was to quantify the odor and gas emission benefits of a fine ground rubber cover on laboratory scale manure storage units and to evaluate the ability of a fine ground rubber cover applied to a commercial swine manure storage unit to remain intact and functional over an extended period of time. A three-inch cover of fine ground rubber reduced odors by 77 to 99 percent from the manure storage tanks over a six-week period. However, consistent reductions of NH3 emissions were not observed and H2S emissions from all manure storage treatments were …

Bacteria in biofilms adhere well to surfaces and are quite difficult to remove. Oral plaque is one example of a biofilm. Many researchers have studied ways to remove plaque and bacteria from surfaces. It has been found that the passage of a bubble across a surface to which bacteria has adhered can remove the bacteria from the surface. Biofilms of Streptococcus mutans were grown on glass coverslips as a simple model for oral plaque. The coverslips were mounted in a Plexiglas chamber filled with artificial saliva. Visualizing and interacting with engineering analysis results can provide valuable insights into a system's performance and aid in engineering decision-making. Currently, the majority of analysis codes are developed as isolated solutions focusing only on the most prominent physical influence to a system, such as thermal, structural, fluid, etc. Frequently, more than one of these physical influences combined to force engineers to evaluate complex, coupled systems.
