Florida is surrounded by water, and its many internal lakes and rivers have long been recognized for their excellent fishing and boating. This notoriety draws land developers to the lake shores to establish residential and commercial infrastructure. This land development brings with it flood plain alteration, water level stabilization, and increased nutrients which cause adverse impacts to our lakes. The reduction of membrane productivity (i.e. membrane fouling) during operation occurs in virtually all membrane applications. Membrane fouling originates from the method by which membranes operate: contaminants are rejected by the membrane and retained on the feed side of the membrane while treated water passes through the membrane. The accumulation of these contaminants on the feed side of the membrane results in increased operating pressures, increased backwashing frequencies, increased chemical cleaning frequencies, and increased membrane replacement frequencies. Self-assembled lipid tubules are particularly attractive for inorganic synthesis and drug delivery because they have hollow cylindrical shapes and relatively rigid mechanical properties. In this thesis work, we have synthesized lipid tubules of 1,2-bis(tricosa-10,12-dinoyl)-sn-glycero-3-phosphocholine (DC8,9PC) by self-assembly and polymerization in solutions. We demonstrate for the first time that both uniform and modulated molecular tilt orderings exist in the tubule walls, which have been predicted by current theories, and therefore provide valuable supporting evidences for self-assembly mechanisms of chiral molecules. The objective of the study was to compare the trihalomethanes (THMs) produced from ferrate with hypochlorite and to determine how different the THM production would be for a given degree of disinfection (3 log reduction in Heterotrophic Plate Count (HPC)). Different water samples were collected from Lake Claire, Atlantic Ocean, and secondary effluent from an advanced wastewater treatment plant. THM formation was determined using a standard assay over 7 days at room temperature. Nanocrystalline hydroxyapatite (HAp) powder of size 10-20 nm was synthesized applying microwave radiation using calcium nitrate tetrahydrate and sodium phosphate dibasic anhydrous as the starting materials. Microwave power of 600 W and Ca/P ratio of 1.66 in the starting chemicals served as the major factors in the synthesis of nanocrystalline HAp powder. Phase composition and evolution were studied using X-ray diffraction (XRD) technique. Morphology, agglomeration and particle-size of the synthesized powder were studied using Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM) techniques. Orthophosphates and polyphosphates are rarely present at significant levels in raw water source but are purposefully added to the water in various forms to inhibit corrosion, iron oxidation (red water), or calcium carbonate precipitation (scale formation). Orthophosphates serve as building blocks for polyphosphates, which includes structures in linear chain, branched chains (metaphosphate) and "glassy" polyphosphate polymers. This study represents an analysis of comprehensive temperature and moisture content data collected from the anaerobic portion of a bioreactor landfill at the New River Regional Landfill, Florida, USA. The main focus of the study was the analysis of effects of the liquid injection on the temperature inside the landfill. When the leachate or groundwater at lower temperature than the landfilled waste is injected into the landfill, it has an initial cooling effect on the waste until the biological activity, enhanced by the additional moisture, releases heat. Simulation studies are presented that probe the possibility of using high-field (>100kV ∕ cm), short-duration (∼50ns) electrical pulses for nonthermal and reversible cessation of biological electrical signaling pathways. This would have obvious applications in neurophysiology, clinical research, neuromuscular stimulation therapies, and even nonlethal bioweapons development. The concept is based on the creation of a sufficiently high density of pores on the nerve membrane by an electric pulse. This modulates membrane conductance and presents an effective "electrical short" to an incident voltage wave traveling across a nerve. Net blocking of action potential propagation can then result. Anaerobic biodegradation in sewage sludge and aerobic biodegradation in compost of polyhydroxybutyrate (PHB) were investigated to determine degradation kinetics and identify changes in polymer properties. To determine the impact of a natural plasticizer on biodegradation, tributyl citrate (TBC) was blended with some test specimens. For comparison to another bioplastic, polylactic acid (PLA) was biodegraded in the anaerobic study. Evolved gaseous carbon was measured to assess biodegradability according to ASTM standards D5210 and D5338. Mass loss experiments were performed to facilitate determination of degradation kinetics. This thesis presents a novel design for a passive alignment feature for vertically stacking chips in a micro fluidic device. Each chip is the size of a standard microscopic slide (75mm—25mm) and is a highly specialized functional unit designed and fabricated to process fluidic samples. The fluidic samples in the device flows from one chip to another using inter connecting holes 500μm-100μm in diameter. Antisense oligodeoxynucleotides (AS ODNs) are short single-stranded pieces of DNA that are designed to hybridize with messenger RNA and thus offer a powerful technique for controlling gene expression. Phosphorothioate AS ODNs are a well-established modification of the traditional phosphodiester structure that increase the in vitro half-life of AS ODN activity. In this work, we describe a method for controlling the hybridization activity of PS ODNs in cell culture with a photo-reversible mechanism. The genome halving problem, motivated by the whole genome duplication events in molecular evolution, was solved by El-Mabrouk and Sankoff in the pioneering paper [SIAM J. Comput., 32 (2003), pp. 754–792]. The El-Mabrouk–Sankoff algorithm is rather complex, inspiring a quest for a simpler solution. An alternative approach to the genome halving problem based on the notion of the contracted breakpoint graph was recently proposed in [M. A. Alekseyev and P. A. Pevzner, IEEE/ACM Trans. Comput. Biol. Breakpoint graph analysis is a key algorithmic technique in studies of genome rearrangements. However, breakpoint graphs are defined only for genomes without duplicated genes, thus limiting their applications in rearrangement analysis. We discuss a connection between the breakpoint graphs and de Bruijn graphs that leads to a generalization of the notion of breakpoint graph for genomes with duplicated genes. We further use the generalized breakpoint graphs to study the Genome Halving Problem (first introduced and solved by Nadia El-Mabrouk and David Sankoff). Sediment capping remains an effective remediation technique for contaminated sediments due to its capability to contain contaminants and provide a sound habitat for indigenous biota. This research investigated the effect of capping on biodegradation of organic contaminants in Anacostia River sediments using both experimental and modeling techniques. 14C–Phenanthrene mineralization was monitored under various redox conditions in Anacostia River sediments. Mineralization of 14C–phenanthrene was strongly linked to sulfate reduction, which was the more energetic process than any other anaerobic condition in Anacostia sediments.
