Diafiltration by means of the ultrafiltration system of probiotic fermented Mung beans (Phaseolus radiatus L.) concentrate has been performed to reduce or eliminate salts and smaller impurities than the nominal cut-off of the membrane of 20,000 nominal weight cut-off (NWCO). These processes have been conducted as an attempt in order to get a probiotic product with organoleptic acceptability, composition, and the optimal total lactic acid bacteria (LAB) counts because the presence of salts will affect on the viability of LAB and the cell lysis of LAB and limit its utility in food products. This report assesses the economics of microalgae biofuels production through an analysis of five production scenarios. These scenarios, or cases, are based on technologies that currently exist or are expected to become available in the near-term, including raceway ponds for microalgae cultivation, bioflocculation for algae harvesting, and hexane for extraction of algae oil. Process flow diagrams, facility site layouts, and estimates for the capital and operations costs of each case were developed de novo. Surface resistance to nonspecific protein adsorption, cell/bacterial adhesion, and biofilm formation is critical for the development and performance of biomedical and analytical devices. Significant needs and efforts have been made in the development of biocompatible and bioactive materials for antifouling surfaces, but much of the work retains an empirical flavor due to the complexity of experiments and the lack of robust theoretical models. Integrated constraint-based metabolic and regulatory models can accurately predict cellular growth phenotypes arising from genetic and environmental perturbations. Challenges in constructing such models involve the limited availability of information about transcription factor-gene target interactions and computational methods to quickly refine models based on additional datasets. In this study, we developed an algorithm, GeneForce, to identify incorrect regulatory rules and gene-protein-reaction associations in integrated metabolic and regulatory models. We applied the algorithm to refine integrated models of Escherichia coli and Salmonella typhimurium, and experimentally validated some of the algorithm's suggested refinements. This thesis presents the design of a novel piezoelectric device with dual functions of studying soft biological tissues. The device consists of a central circular plate and a set of flexural beams. The flexural beams are partially covered with piezoelectric film for integrated transduction. It can function as a force sensor for measuring the mechanical behavior of a tissue sample and a load applier for mimicking physiological loads experienced by the tissue sample in vivo, while the mechanical behavior can be visually observed under a microscope simultaneously. Liming has been used to mitigate effects of acidic deposition in forest ecosystems. This study was designed to examine the effects of calcium (Ca) supply on the spatial patterns and the relations between soil and soil solution chemistry in a base-poor forest watershed. Watershed 1 at the Hubbard Brook Experimental Forest in New Hampshire, USA was experimentally treated with wollastonite (CaSiO3) in October, 1999. Myocardial infarction (MI) is a condition in which blood supply to the heart is insufficient. MI is associated with two forms of cell death: apoptosis and necrosis. 99mTc-duramycin (99mTc-D) is a novel radiopharmaceutical that detects cell death by recognizing externalized phosphatidylethanolamine. The objective of this study was to develop a compartmental model for 99mTc-D uptake kinetics in normal and infarct myocardium, and utilize this model to compare the uptake kinetics of 99mTc-D in MI with that of another radiopharmaceutical, 99mTc-C2A-GST.

This study was conducted to evaluate the effect of pulsed electric field (PEF) treatment on anthocyanin extraction from red cabbage using water as a solvent. Mashed cabbage was placed in a batch treatment chamber and subjected to PEF (2.5 kV/cm electric field strength; 15 μs pulse width and 50 pulses, specific energy 15.63 J/g). Extracted anthocyanin concentrations (16 to 889 μg/mL) were determined using HPLC. Heat and light stabilities of the control and PEF-treated samples, having approximately the same initial concentrations, were studied. PEF treatments enhanced total anthocyanin extraction in water from red cabbage by 2.15 times with a higher …

Cartilage tissue engineered constructs using mesenchymal stem cells were stimulated with 3 different stimulation algorithms to achieve characteristics mimicking the superficial tangential zone of articular cartilage. The stimulation algorithm of both compression and tension without an offset had the best properties out of all the evaluated groups.

This project examined the use of process mapping as a tool to show the process of developing medical devices from a broad perspective that includes research, innovation, development, regulation, and marketing. The medical device development cycle, on a broad scope, is not well defined. The lack of a universal language with which to describe this process has made it difficult to understand and communicate. In this project, data was collected from peer-reviewed sources, summarized in a literature review, drawn out by hand into a series of lower-level process maps and finally assembled into a single process map. As Cal Poly moves towards an environmentally friendly campus, the Energy Harvesting from Exercise Machines (EHFEM) project looks to generate a new source of renewable energy to help power the Cal Poly Recreation Center. The project focuses on retrofitting exercise machines to generate electricity. As of December 2009, Audrey Nakamura, Justin Arakaki, and Praveen Lawrence developed a design which allows the contribution of electricity generated from multiple exercise machines into the power grid.

Motivated by the goal of building a device capable of detecting when a user takes a bite of food, we examine whether there is a correlation between linear wrist motion and bite detection. Such a device could be used to manage weight loss by tracking eating trends over time and providing real time feedback to the user regarding eating rate or total consumption. Hollow fiber membranes (HFMs) formed through phase inversion methods exhibit specific physicochemical characteristics and generally favorable surface and mechanical properties, supporting their use in diverse applications including ultrafiltration, dialysis, cell culture, bioreactors, and tissue engineering. Characterization of, and modifications to, such membranes are important steps in achieving desired characteristics for specific applications.
HFMs subject to gas, irradiation, and chemical sterilization techniques were characterized based on several analytical techniques. It was revealed that these common sterilization techniques can cause inadvertent changes to HFM properties. A single-step, extractive reaction for extraction of lipids such as biodiesel components, omega-3 fatty acids, or other triglycerides from microbial cells was examined. Conventional methods for lipid extraction use toxic solvents, and require multiple steps and long processing times. When the goal is to produce fatty acid methyl esters or FAMEs, the extracted lipids are subjected to a separate transesterification reaction with simple alcohols in the presence of an acid or base catalyst. An immediate demand exists for new tuberculosis (TB) antibiotics due to the ever-increasing spread of drug-resistant strains. The drug-development process goes through four phases, the first (Phase 0) of which is to demonstrate and investigate drug effectiveness and toxicity. This research investigated the effectiveness of the Hypericum perforatum herb (commonly St. John's wort (SJW)) in its growth inhibition of mycobacteria and its viability effect on human lung cells.
Organic-solvent SJW extracts were effective at inhibiting every nonpathogenic genetically sequenced Mycobacterium isolate currently available (six isolates) in preliminary studies. Quantitative studies of five Mycobacterium isolates showed an order of concentration sensitivity …

Factor analysis was used to develop a more detailed description of the human hand to be used in the creation of glove sizes; currently gloves sizes are small, medium, and large. The created glove sizes provide glove designers with the ability to create a glove design that can provide fit to the majority of hand variations in both the male and female populations. The research used the American National Survey (ANSUR) data that was collected in 1988. The Aviation industry has developed extensively since its establishment by the Wright Brothers in 1903. Over time, the highly male-dominated industry experienced significant changes to incorporate female aviators. Determined women initiated this process through participating in the aircraft engineering sector and pursuing careers as pilots. However, these women faced various challenges, which resulted in setbacks to their effective growth in the industry. In fact, vital issues encountered in the past are still present and often overlooked in today’s aviation industry. Agricultural irrigation and drainage canals in Taiwan are generally constructed using concrete. Concrete is used for the safety of conveyance efficiency. It also reduced loss and is structurally safe. The irrigation ditches and soil revetments in Taiwan fitted with a strong concrete lining. The smooth nonporous concrete surface makes the biology perch difficult and causes a monotonous flow. Crude oil degradation has been carried out using biopile reactor in total petroleum hydrocarbon (TPH) concentration of 5%, 10% and 15%. The thermophilic microorganisms used from isolation result and identification are Aeromonas salmonicida, Bacillus pantothenticus, and Stenotrophomonas maltophilia. The biodegradation of biopile reactor is done by various concentrations of TPH, total plate count (TPC), and volatile suspended solid (VSS) per day for 30 days. The biodegradation of kinetic parameter calculated consists of μ, μm, Y, Yt, Yobs, Kd, Ks from TPH concentration decision, and TPC and VSS in every microorganism with t (observation time) of 0 hour to 168 hours. …

The micro-alga Chlorella vulgaris was cultivated under a variety of environmental conditions in various culture media solutions to assess and optimize growth rate and biomass productivity. Efforts during this work investigated growth parameters at the micro-scale in an air-lift bubble system with the goal of interpreting performance characteristics that can be applied to a larger serpentine tubular Photo-bioreactor. Maximum growth rates and biomass yields were 0.65 d-1 and 2.003 g biomass/L and achieved in seven days using urea in de-ionized water under a 24:0 Photoperiod (Light:Dark). Chlorella vulgaris was cultured in microbioreactors using Bold’s Basal medium at varying nitrogen and phosphorus concentrations to define nitrogen and phosphorus utilization standards. Nutrient concentration was varied between 137 mg/L to 7.33 mg/L NO3-N and between 55.2 mg/L to11.0 mg/L PO4-P in five test scenarios. All were grown under a constant photoperiod at 22±2 °C and a mixture of 4 to 10% carbon dioxide/air. Maximum yield and growth rate occurred with the highest initial nitrogen and phosphorus concentrations. Tampa (US), Guyana (SA), and Bolivia (SA), are geographically, socially, economically, and politically unique which make them ideal sites to study issues of mercury and sustainability. Mercury’s innate ability to bioaccumulate and biomagnify in aquatic and terrestrial ecosystems poses a severe threat to both human and environmental health. The most vulnerable populations affected by mercury consumption include coastal communities, children, women of child-bearing age, the indigenous poor and persons with high environmental/occupational exposure factors. Communities in the regions of Florida, Bolivia, and Guyana whose diets are high in fish and are environmentally/occupationally exposed to mercury may be at a higher …
