A cost-effective and highly sensitive portable diagnostic device is needed to enable much more widespread monitoring of health conditions in disease prevention, detection, and control. Miniaturized and easy-to-operate devices can reduce the inherent costs and inefficiencies associated with healthcare testing in central laboratories. Hence, clinicians are beginning to use point of care (POC) testing and flexible clinical chemistry testing devices which are beneficial for the patient.

The need for a renewable energy source to replace automobile gasoline has become more critical in the past decade. Filtration processes, whether on the microfluidic, clinical treatment systems, or industrial scale (e.g., point-of-care diagnostics, dialysis, and biopharmaceutical manufacturing, respectively), are often inseparable from membrane clogging (fouling). As a consequence, most, if not all, filtration systems require frequent maintenance to maintain functionality and efficiency. The thesis of this project hypothesizes that Dielectrophoresis can be combined with standard filtration to reduce filter fouling, extending membrane life, and enabling continuous operation. This project investigates a method to reduce fouling, add specificity and efficiency, and decrease the cost and challenge of filtration based biofluid separations.

Current recombinant protein production systems require several months to develop. Existing systems fail to provide timely, flexible, and cost-effective therapies to protect against emergency mass-casualty infections or poisonings. As the identity of many new biological threat agents are unlikely to be known in advance, pre-emptive manufacturing and stockpiling of countermeasures cannot be performed. Preparedness for a biological catastrophe requires a radical solution to replace the current slow scale-up and manufacture of lifesaving medical countermeasures. The lungs are considered as one of the fastest portals of entry to the bloodstream and oral inhalation (OI) has long been accepted as the preferred mode of administration of therapeutics to the respiratory tract. Glioblastoma Multiforme is a primary malignancy of the central nervous system and is fatal for patients despite surgical resection and radiotherapy. Oncolytic Herpes Simplex Virus (oHSV) vectors deleted of the γ134.5 neurovirulence gene are potential therapies for treating glioblastoma tumors. Although replication in permissive cells is considered the primary mechanism of oHSV mediated tumor clearance, there is evidence that the immune system is part of the tumor clearance mechanism during treatment with oncolytic viruses. Approximately 2.2 million cattle carcasses require disposal annually in the United States. Land burial is a convenient disposal method that has been widely used in animal production for disposal of both daily mortalities as well as during catastrophic mortality events. To date, greenhouse gas production after mortality burial has not been quantified, and this study represents the first attempt to quantify greenhouse gas emissions from land burial of animal carcasses. In this study, anaerobic decomposition of both homogenized and unhomogenized cattle carcass material was investigated using bench-scale reactors. Methamphetamine (Meth) is a widely abused stimulant and its users are at increased risk for multiple infectious diseases. To determine the impact of meth on the immune system, we utilized a murine model that simulates the process of meth consumption in a typical addict. Our phenotypic analysis of leukocytes from this dose escalation model revealed that meth affected key immune subsets. Meth administration led to a decrease in abundance of natural killer (NK) cells and the remaining NK cells possessed a phenotype suggesting reduced responsiveness. Dendritic cells (DCs) and Gr-1(high) monocytes/macrophages were also decreased in abundance while Gr-1(low) monocytes/macrophages appear …

T. pallidum can survive a primary immune response and continue growing in the host for an extended period of time. T. pallidum is thought to bind serum fibronectin (FN) through Tp0483 on the surface to obscure antigens. A Tp0483 fragment (rTp0483) was adsorbed onto functionalized self-assembled monolayers (SAMs) with FN. FN capture by adsorbed rTp0483 depended greatly on surface chemistry with COO- groups being best for FN binding. Hemocompatibility was determined by analysis of plasma protein adsorption, intrinsic pathway activation, and platelet activation. rTp0483+FN bound an equal or lesser amount of fibrinogen (Fg), human serum albumin (HSA), and factor XII …

PID is a polymicrobial infection of the female reproductive tract, and is associated with pelvic pain, abnormal uterine bleeding, and tubal damage that can lead to ectopic pregnancies and infertility. Proper hand hygiene is known to reduce hospital acquired infections. Currently the most widely accepted method used to measure hand hygiene compliance is through direct observation. This method often provides data that may not accurately represent true hand hygiene compliance due to such factors such as the Hawthorne Effect. Perhaps a better approach is to use counting devices to capture hand hygiene opportunities and activities. This dissertation presents two applications of discrete-event simulation (DES) to represent clinical processes: (1) a model to quantify the risk of the maternal obese and diabetic intrauterine environment influence on progression to adult obesity and diabetes, and (2) a model to evaluate health and economic outcomes of different smoking cessation strategies. The first application considers the public health impact of the diabetic and obese intrauterine environment's effect on the prevalence of diabetes and obesity across subsequent generations. Currently, capturing a sample and sending it to a laboratory can take weeks to get results, which can be much too long. A PDMS microfluidic reactor was made in Cal Poly’s class 1000 clean room for the purpose of synthesizing quantum dots.  The device master mold was made from a silicon substrate and SU-8 features 50μm tall.  The PDMS reactor was cast from that mold.  The flow rates of fluid through the channels, heating of the reactor and pressure in the reactor were measured in order to characterize the potential for synthesizing quantum dots.  Flow rates of 20 mL/hr through 4 mL/hr were tested to characterize the consistency of amount of time the fluid remains in the reactor at a constant flow …

To determine if varying city water qualities from around the United States can support a developing “oxidation corrosion of silver thin films” method of creating aqueous ionic silver solutions for the anti-microbial treatment of healthcare linens, a test was devised holding all variables constant with the exception of water quality parameters. Electroporation (EP) is a simple in vivo method to deliver normally impermeable molecules, such as plasmid DNA, to a variety of tissues. Delivery of plasmid DNA by EP to a large surface area is not practical because the distance between the electrode pairs, and therefore the applied voltage, must be increased to effectively permeabilize the cell membrane.
