The finite state projection algorithm provides modelers a new way of directly solving the chemical master equation. The algorithm utilizes the matrix exponential function, and so the algorithm’s performance suffers when it is applied to large problems. Other work has been done to reduce the size of the exponentiation through mathematical simplifications, but efficiently exponentiating a large matrix has not been explored. This work explores implementing the finite state projection algorithm on several different high-performance computing platforms as a means of efficiently calculating the matrix exponential function for large systems. Current progress in the development of vaccines has decreased the incidence of fatal and non-fatal infections and increased longevity. However, new technologies need to be developed to combat an emerging generation of infectious diseases. DNA vaccination has been demonstrated to have great potential for use with a wide variety of diseases. Alone, this technology does not generate a significant immune response for vaccination, but combined with delivery by electroporation (EP), can enhance plasmid expression and immunity. Point-of-use (POU) drinking water treatment is a common method of providing drinking water in disaster relief situations when critical water infrastructure is damaged. In these cases, POU treatment devices can be used to treat local water until relief organizations set up more permanent water provision methods. One such POU technology is PŪR® Purifier of Water, a combined coagulation/flocculation and disinfection chemical treatment sachet produced by Procter & Gamble. This research investigates the use of personal ventilation (PV) in a typical office space as a means of contaminant removal from ones breathing zone (BZ). For this work, a validated computational model was developed and used for PV assessment under different scenarios. Experimental data of Khalifa et al. (2009), Ito (2007) and Rim et al. (2009) were used to validate a computational model that is capable of simulating indoor chemical reactions with excellent agreement compared with the experiments. We have previously shown that a hydrophobic alkylating compound, 1,5-iodonaphthyl-azide (INA) can efficiently inactivate the virulent strain of Venezuelan equine encephalitis virus (VEEV), V3000 in vitro. In this study, we have evaluated the safety of INA-inactivated V3000 and V3526 and the protective efficacy of INA-inactivated V3000. INA-inactivated V3000 and V3526 did not cause disease in suckling mice. RNA isolated from the INA-inactivated V3000 and V3526 was also not infectious. Immunization of adult mice with INA-inactivated V3000 induced an anti-VEEV antibody response and protected mice from virulent VEEV challenge. The protective efficacy of INA-inactivated V3000 increased with the use of adjuvants. …

The development of a vaccine against malaria is a major research priority given the burden of disease, death and economic loss inflicted upon the tropical world by this parasite. Despite decades of effort, however, a vaccine remains elusive. The best candidate is a subunit vaccine termed RTS,S but this provides only partial protection against clinical disease. This review examines what is known about protective immunity against pre-erythrocytic stage malaria by considering the humoral and T cell-mediated immune responses that are induced by attenuated sporozoites and by the RTS,S vaccine. With improved survival in burn patients, Clostridium difficile infection (CDI) remains a significant potential complication. The incidence of, risk factors for, and outcomes of CDI in severely burned patients are poorly studied and remain unclear. This study involves retrospective case control and cohort studies using electronic medical records from February 1, 2002 to January 31, 2009 at the US Department of Defense’s only burn unit. Dendrimers are ideal materials to be used in the emerging field of nanomedicine. Their nanoscale size and high density of functional groups on their peripheries allow them to be used for various biomedical applications. This work exploits dendrimers as drug delivery vehicles and a versatile platform for capturing biomarkers with improved sensitivity and specificity. Hydroxyl terminated poly(amidoamine) dendrimer (PAMAM-OH) was modified with a linker having amine group at the end and conjugated to two drugs, erythromycin (EM) and allopregnanonolne, respectively and evaluated in vitro. Methicillin-resistant Staphylococcus aureus is estimated to cause more U.S. deaths annually than HIV/AIDS. The emergence of hypervirulent and multidrug-resistant strains has further amplified public health concern and accentuated the need for new classes of antibiotics. RNA degradation is a required cellular process that could be exploited for novel antimicrobial drug development. However, such discovery efforts have been hindered because components of the Gram-positive RNA turnover machinery are incompletely defined. In the current study we found that the essential S. aureus protein, RnpA, catalyzes rRNA and mRNA digestion in vitro. A purification process for the manufacture of a recombinant C-terminus heavy chain fragment from botulinum neurotoxin serotype C [rBoNTC(Hc)], a potential vaccine candidate, has been defined and successfully scaled-up. The rBoNTC(Hc) was produced intracellularly in Pichia pastoris X-33 using a three step fermentation process, i.e., glycerol batch phase, a glycerol fed-batch phase to achieve high cell densities, followed by a methanol induction phase. Hence, the decision support models for mitigation of pandemic outbreaks must be user-friendly and operational, and also incorporate valid estimates of disease transmissibility and severity. Current physiologically-driven operator cognitive state assessment technology relies primarily on electroencephalographic (EEG) signals. Traditionally, gel-based electrodes have been used; however, the application of gel-based electrodes on the scalp requires expertise and a considerable amount of preparation time. Additionally, discomfort can occur from the abrasion of the scalp during preparation, and the electrolyte will also begin to dry out over extended periods of time. Silver nanoparticles (AgNPs) have received tremendous attention for their antimicrobial properties; however, many gaps in knowledge exist. To address these issues, three research objectives were examined. The first objective hypothesized AgNPs can be size selected and concentrated via tangential flow ultrafiltration. The second objective hypothesized a high-throughput method could be developed to screen nanoparticle antiviral-activity and cytotoxicity simultaneously. The third objective hypothesized AgNPs inhibit viruses by preventing viral entry.

Nanoformulations of crystalline indinavir, ritonavir, atazanavir, and efavirenz were manufactured by wet milling, homogenization or sonication with a variety of excipients. The chemical, biological, immune, virological, and toxicological properties of these formulations were compared using an established monocyte-derived macrophage scoring indicator system. Measurements of drug uptake, retention, release, and antiretroviral activity demonstrated differences amongst preparation methods. Interestingly, for drug cell targeting and antiretroviral responses the most significant difference among the particles was the drug itself. We posit that the choice of drug and formulation composition may ultimately affect clinical utility.

Background: In a globalized word, prevention of infectious diseases is a major challenge. Rapid detection of viable virus particles in water and other environmental samples is essential to public health risk assessment, homeland security and environmental protection. Current virus detection methods, especially assessing viral infectivity, are complex and time-consuming, making point-of-care detection a challenge. Faster, more sensitive, highly specific methods are needed to quantify potentially hazardous viral pathogens and to determine if suspected materials contain viable viral particles. Buildings are a significant energy consumer and are responsible for an increasingly large percent of worldwide greenhouse gas emissions, currently between 30 and 40 percent. Energy efficiency presents unique opportunities for building owners to reduce their environmental footprint and add value through cost savings, tax deductions, and increased market value. An analysis of 183 samples of efficiency measures in seven technology categories found that 74% of efficiency investments had a positive net present value. Building automation system and chiller plant improvements had the highest mean energy and carbon dioxide savings per square foot. Additionally, building automation systems had, on average …

This Dissertation characterizes the first asphaltene DSSC (dye-sensitized solar cell), in which light to electricity conversion efficiency has improved from 0 % to 1.8 %. Four natural organic asphaltene factions, namely, un-fractionated asphaltene, Fr. 1 Asphaltene, Fr.2 asphaltene, Fr.3 and Fr.4 were extracted and tested for various solar cell parameters of TiO2 based dye-sensitized solar cell DSSC. DSSCs were fabricated using the four individual asphaltene dyes. The photovoltaic performance of the cells was examined by determining the various parameters such as open-circuit voltage, short-circuit current, fill factor, and series resistance. The objectives of this research were to 1) determine the variability of fecal-indicator bacteria in Gulf of Mexico marine waters, 2) determine the effect of environmental variables on enterococci concentrations, 3) determine if beach sand at Holly Beach, LA was capable of acting as a reservoir for fecal-indicator bacteria, and 4) determine if a model could be developed to reduce the time it takes to issue a beach swim advisory. Fecal-indicator bacteria concentrations varied from year to year in historical data collected from a Gulf Coast beach. Afternoon fecal-indicator bacteria concentrations were consistently lower than morning concentrations, demonstrating daily variability. …

Ovarian cancer is the fifth leading cause of cancer death among women in United States and has an alarming 1.4% (1 in 71) lifetime risk. The lack of overt symptoms and the absence of a reliable screening test to detect ovarian cancer result in over 70% of women being diagnosed after the disease has spread beyond the ovary resulting in a poor prognosis. Non-viral delivery of plasmid DNA has traditionally relied upon physical forces applied directly to target tissues. These physical methods typically involve contact between an applicator and the target tissue and often cause transient patient discomfort. To overcome the contact-dependent limitations of such delivery methodologies, an atmospheric direct current plasma source was developed to deposit ionized gas molecules onto localized treatment sites. The deposition of charged species onto a treatment site can lead to the establishment of an electric field with strengths similar to those used for traditional electroporation.
