A process was developed for production of a candidate vaccine antigen, recombinant C-terminal heavy chain fragment of the botulinum neurotoxin serotype E, rBoNTE(Hc)in Pichia pastoris. P. pastoris strain GS115 was transformed with the rBoNTE(Hc) gene inserted into pHILD4 Escherichia coli—P. pastoris shuttle plasmid. The clone was characterized for genetic stability, copy number, and BoNTE(Hc) sequence. Expression of rBoNTE(Hc) from the Mut+ HIS4 clone was confirmed in the shake-flask, prior to developing a fed-batch fermentation process at 5 and 19 L scale. Our investigations in 2007 focused on continuing collection and analyses of tree rings from living trees and building a long ring-width series from interstadial wood. These interstadial forests were extensive and their preservation, although discontinuous and fragmented, is remarkable. In situ stumps, still rooted in growth position, and logs held within sediments remain from forests that existed at various times between glaciations over approximately the last 10,000 years.
We have analyzed previous and new collections of ring-width tree-ring series, including a multiple species collection from near Excursion Ridge in the southeast portion of the Park. These tree-ring series have a …

Polypeptides, linear macromolecules, are formed from amino acid residues by linkage of peptide bonds. Proteins are polypeptides too, with more complex conformations contributing to specific functionalities. Disulfide bonds are very important to maintain the structure and functions of proteins, which will form between two cysteine (Cys) residues under oxidizing circumstance.

Cys containing polypeptides are designed and synthesized by F-moc (9-Fluorenylmethyloxycarbonyl) chemistry. The number and position of Cys residues can be controlled by amino acid sequences design and following peptide synthesis, which is important to gain insights on the nature of polyelectrolyte multilayer film assembly and stability.

Eastern North America receives elevated atmospheric mercury deposition from a combination of local, regional, and global sources. Anthropogenic emissions originate largely from electric utilities, incinerators, and industrial processes. The mercury species in these emissions have variable atmospheric residence times, which influence their atmospheric transport and deposition patterns. Forested regions with a prevalence of wetlands and of unproductive surface waters promote high concentrations of mercury in freshwater biota and thus are particularly sensitive to mercury deposition. Through fish consumption, humans and wildlife are exposed to methylmercury, which markedly bioaccumulates up the freshwater food chain. Average mercury concentrations in yellow perch fillets …

A model for the prediction of color release from a pilot distribution system was created in 2003 by Imran. This model allows prediction of the release of color from aged cast iron and galvanized steel pipes as a function of water quality and hydraulic residence time. Color was used as a surrogate measurement for iron, which exhibited a strong linear correlation. An anomaly of this model was an absence of a term to account for pH, due to the influent water being well stabilized. The objective of this study was to evaluate the practical limits of EPA's NONROAD 2005 to accurately simulate Central Florida conditions, especially with regard to lawn and garden equipment. In particular we investigated a NONROAD emission inventory using default inputs and then created a locally specific emission inventory. These emission inventories were prepared for Orange, Osceola, and Seminole county and focused only on the VOC and NOx emissions caused by lawn and garden equipment. The high concentration of ground level ozone can cause a variety of health problems including chest pain, coughing, throat irritation, and congestion or it can worsen bronchitis, emphysema, and asthma. In this study, the bacterial growth dynamics of 14 pilot drinking water distribution systems were studied in order to observe water quality changes due to corrosion inhibitor addition. Empirical models were developed to quantity the effect of inhibitor type and dose on bacterial growth (biofilm and bulk water). Water and pipe coupon samples were taken and examined during the experiments. The coupons were exposed to drinking water at approximately 20°C for at least 5 weeks to allow the formation of a measurable quasi- steady-state biofilm. Bulk water samples were taken every week. Research objectives. Psychophysical acceleration threshold is a tool for detecting deficits in dynamic postural control. Our lab has shown differences in the acceleration threshold among young adults, elderly adults, and elderly adults with diabetes. Electromyography, Semmes-Weinstein monofilaments, and hearing tests investigate the underlying physiological mechanisms for the detriments in postural control. Due to peri-sway perturbations, the motion of a person's sway affects the signal to noise ratio for perturbed stance. This dissertation consists of four papers concerning impacts of zinc orthophosphate (ZOP) inhibitor on iron, copper and lead release in a changing water quality environment. The mechanism of zinc orthophosphate corrosion inhibition in drinking water municipal and home distribution systems and the role of zinc were investigated. Fourteen pilot distribution systems (PDSs) which were identical and consisted of increments of PVC, lined cast iron, unlined cast iron and galvanized steel pipes were used in this study. The Mahalanobis Taguchi System (MTS) is a diagnosis and forecasting method for multivariate data. Mahalanobis distance (MD) is a measure based on correlations between the variables and different patterns that can be identified and analyzed with respect to a base or reference group. MTS is of interest because of its reported accuracy in forecasting small, correlated data sets. This is the type of data that is encountered with consumer vehicle ratings. This paper asked professionals in the legal system to evaluate the current state and effectiveness of laws to identify and deter computer crime. Responses were evaluated with a formal structural equation model. The results generally show that legal professionals believe potential jurors have minimal knowledge of computer crime issues. More importantly, they also believe that judges have little knowledge or experience. A similar lack of knowledge by defense attorneys indicates that it could be difficult for a person accused of computer related infractions to find adequate representation. A simple experiment for measuring single-phase permeability of fully saturated porous medium is introduced. The experiment utilizes radial flow of a non-volatile wetting fluid through a porous medium such as ceramic tile, concrete or sand. The radial position of flow front is measured as a function of time and the collected data are analyzed using Darcy's Law to determine the permeability. In addition, the phenomenon of the multiphase flow through the medium with a broad pore size distribution is demonstrated.

In this study, acoustic profiling techniques are used to resolve the two-dimensional spatial structures of the oscillating boundary layer in a phase-averaged sense over a wave orbital amplitude. The principal objective of this work is to examine the spatial variability of turbulent properties over irregular roughness. Field observations are used to investigate the interaction between bottom roughness, flow structure and turbulent properties within the near-bed flow. The overall focus of the research is to relate wave energy dissipation or friction factor estimates to physical roughness elements and understand the dynamics of such processes in the near-bed flow.
