An atomic force microscope (AFM) is used to scan high-resolution images on the nano scale. The lambda DNA used for this project are 48,502 base pairs in length and are double-stranded. This project utilizes the NanoSurf Core AFM in order to characterize and manipulate strands of lambda DNA which have been deposited on a mica surface. The deposition process of the DNA on the mica surface was developed by the National Institute of Standards and Technology and the University of Colorado - Boulder. Using unmanned aerial systems (UAS) for leaf sampling is contributing to a better understanding of the influence of climate change on plant species, and the dynamics of forest ecology by studying hard-to-reach tree canopies. Currently, multiple skilled operators are required for UAS maneuvering and using the leaf sampling tool. This often limits sampling to only the canopy top or periphery. Glioblastoma (GBM) brain tumors are highly aggressive gliomas due to genetic and cellular heterogeneity. Current GBM treatment consists of surgical resection of the tumor combined with radio- or chemo-therapies. While these treatments have increased the life expectancy for GBM patients up to 20 months, they have had little effect on the 5-year survival rate. The complex cellular and genetic composition of the tumor makes current treatments less effective long term. One approach to developing more effective GBM treatments is to customize nanoparticle-based drug delivery systems that can directly target the aberrant gene expression patterns within a particular GBM tumor. Peripheral arterial occlusive disease (PAOD) is characterized by atherosclerosis, which is the buildup of plaque, consisting largely of cholesterol, in the arterial walls. This plaque accumulation eventually blocks blood flow to the limbs, causing symptoms such as intermittent claudication and tissue death in cases of critical limb ischemia. The body compensates for the reduced perfusion by enlarging pre-existing bypass arteries, known as collaterals, in a process called arteriogenesis. However, in many cases, collateral networks constructed through arteriogenesis fail to enlarge sufficiently or function effectively in patients. This thesis comprises two studies involving design and application of soft material systems. The goal of the first study was to design, fabricate, and characterize hydrogel lattice structures with consistent, controllable, anisotropic mechanical properties. Lattices, based on four types of unit cells (cubic, diamond, vintile, and Weaire-Phelan), were printed using stereolithography (SLA) of polyethylene glycol diacrylate (PEGDA). In order to create structural anisotropy in the lattices, unit cell design files were scaled in one direction by a factor of two in each layer and then printed. Disinfection can promote safe drinking water. Chlorine and chloramines are the two most used disinfectants. While these disinfectants are effective in inactivating disease-causing pathogens, they also produce hazardous byproducts when they react with organic compounds (precursors). Nitrogenous disinfection byproducts (N-DBPs) are more toxic than their counterparts. The increased use of nitrogen-based agrochemicals and antibitics has led to their detection in different water sources. Duchenne muscular dystrophy (DMD) is an X-linked muscle wasting disease caused by mutations in the Dmd gene resulting in non-functional dystrophin. Adeno-associated virus (AAV)-mediated delivery of classical CRISPR systems resulting in double-stranded breaks (DSBs) has yielded promise as a curative DMD therapeutic. However, the consequences of AAV-CRISPR require more thorough characterization. PCR-enriched short-read amplicon sequencing at cut sites is the most common method of sequencing CRISPR editing, but PCR bias results in unreliable quantification of on-target edits. In this study, the greenhouse gas emissions from milk production in the United States were calculated using a cradle-to-gate analysis. This paper presents a novel control method for quadcopters that achieves near-optimal tracking control for input-affine nonlinear quadcopter dynamics. The method uses a reinforcement learning algorithm called Single Network Adaptive Critics (SNAC), which approximates a solution to the discrete-time Hamilton-Jacobi-Bellman (DT-HJB) equation using a single neural network trained offline. The control method involves two SNAC controllers, with the outer loop controlling the linear position and velocities (position control) and the inner loop controlling the angular position and velocities (attitude control). Atherosclerosis is the most prevalent pathology of cardiovascular disease with no known cure. Despite the many systemic risk factors for atherosclerosis, plaques do not form randomly in the vasculature. Instead, they form around bifurcations and the inner curvature of highly curving arterial segments that contain so-called disturbed blood flow that is low in magnitude and multidirectional over the cardiac cycle.
