Antiinfective Properties Of Ursolic Acid-Loaded Chitosan Nanoparticles Againststaphylococcus Aureus, Fatemeh Ghasemzadeh, Ghasem Najafpour, Maedeh Mohammadi

The present study aimed to synthesize ursolic acid-loaded chitosan nanoparticles (UA-Ch-NPs) as an antiinfective agent against 21 Staphylococcus aureus isolates. The UA-Ch-NPs were synthesized by a simple method and then characterized by TEM, FTIR, DLS-zeta potential, and XRD analyses. According to the characterization results, highly dispersed spherical nanoparticles with a mean diameter of 258 nm and a zeta potential of + 40.1 mV were developed. The antibacterial properties of UA-Ch-NPs were investigated and their inhibitory effect on biofilm formation was demonstrated by AFM. In this study, the synthesis of new monostyryl (BDPY-2) and distyryl BODIPY dyes (BDPY-4, BDPY-5) containing pyridine groups has been reported for the first time. The acetylcholinesterase from Electrophorus electricus (AChE), butyrylcholinesterase from equine serum (BuChE), α-glucosidase from Saccharomyces cerevisiae and DNA hydrolytic cleavage actions of BDPY-2, BDPY-4, BDPY- 5 were investigated using various techniques. The results indicated that the compounds had varying inhibition properties against AChE, BuChE, and α-glucosidase. BDPY-4 was the most potent compound on AChE with IC50 of 54.78 ± 4.51 μM, and Lineweaver-Burk plots indicated that the compound is bound to a site other than the …

Supramolecular solvents (SUPRASs) have rapidly gained more attention as a potential substitute to organic solvents in the sample preparation and preconcentration process. The essential properties of SUPRAS solvents (e.g., multiple binding sites, different polarity microenvironments, the opportunity to tailor their properties, etc.) these qualities offer numerous opportunities to advance innovative sample preparation and pretreatment platforms compared to the traditional solvents. The vic-dioximes are compounds with various industrial uses and scientific applications. Many coordination compounds have been synthesized based on vic-dioximes. This study presents the synthesis and full characterization of two vic-dioximes based on dichloroglyoxime, p-aminobenzoic acid, and p-aminotoluene. Their structures were proved by IR, 1H, 13C and 15N NMR spectral analysis, and single crystal X-ray diffraction. One of the reported vic-dioximes, bis(di-p-aminotoluene)glyoxime mono-p-aminotoluene trihydrate showed good to moderate antimicrobial activity against both nonpathogenic gram-positive and gram-negative bacteria (Bacillus subtilis and Pseudomonas fluorescens), phytopathogenic (Xanthomonas campestris, Erwinia amylovora, E. carotovora) and the fungi (Candida utilis and Saccharomyces cerevisiae) at MIC - …

A novel four component modified Biginelli reaction for the synthesis of C-2 functionalized dihydropyrimidines has been established. The approach uses assembly of less explored acetyl acetone with aromatic aldehyde, thiourea, and dimethyl sulphate to construct a novel 5-acetyl 2-methylthio dihydropyrimidine system, which works as an efficient well-designed intermediate for generating C-2 modified Biginelli libraries with nitrogen nucleophiles. Phenyl hydrazine, semicarbazide, and aryl semicarbazides are successfully used as N-nucleophiles to generate C-2 functionalized dihydropyrimidine derivatives, which fulfil the demands of active pharmacophore. This study was conducted to investigate the physicochemical, phytochemical, in vitro antidiabetic and anticancer potential of Olea ferruginea R bark. After extraction using Soxhlet, in vitro antidiabetic and cytotoxic activity on human hepatocellular carcinoma (HepG2) cells was assessed by nonenzymatic glycosylation of hemoglobin assay, alpha-amylase inhibition assay, glucose uptake by yeast cells, and 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide assay, respectively, and gene expression via real-time polymerase chain reaction. Chromatographic separation of n-hexane and ethyl acetate fraction of a crude methanol extract obtained from aerial parts of the Scorzonera aucheriana DC collected from Sivas province of Turkey yielded nine natural compounds; two new 3-caffeoyl-quinic acid analogs (1-2), one new taraxasterol oleate (3), and six known triterpenoids taraxasterol (4), taraxasterol acetate (5), ptiloepoxide (6), lupeol (7), lupeol acetate (8), and β-sitosterol (9) were characterized. The structures of the isolated compounds were elucidated on the basis of NMR (1H, 13C, COSY, HMBC, HSQC, and TOCSY), UV, FT-IR and LC-Q-TOF-MS spectrometric data.

CO2 valorization through chemical reactions attracts significant attention due to the mitigation of greenhouse gas effects. This article covers the catalytic hydrogenation of CO2 to methanol and dimethyl ether using Cu-Ho-Ga containing ZSM-5 and γ-Al2O3 at atmospheric pressure and at temperatures of 210 °C and 260 °C using a CO2:H2 feed ratio of 1:3 and 1:9. In addition, the thermodynamic limitations of methanol and DME formation from CO2 was investigated at a temperature range of 100?400 °C. 3-acetyl coumarin derivatives (1a-d) are formed as a result of condensation of salicylaldehyde derivatives and ethyl acetoacetate and were converted into coumarin-selenophene hybrid compounds (2a-d) in the basic medium by modified Gewald reaction in the presence of malononitrile and selenium. Products are characterized by nuclear magnetic resonance (NMR). The prepared compounds are screened for their anticancer activity against DU-145 cell line. In addition, selected target compounds are evaluated for apoptosis and oxidative stress on DU-145 (prostate carcinoma) cell lines.

Three new dibenzimidazolium salts bridged by 2-methylenepropane-1,3-diyl group were synthesized. Their dinuclear N-heterocyclic carbene Ag(I) complexes were prepared by the reactions of these salts with Ag2O. The structures of the synthesized compounds were defined by nuclear magnetic resonance (NMR), Fourier-transform infrared spectroscopy (FT-IR), elemental analysis, and LC-MSMS (for complexes) techniques. Stability of the silver complexes was confirmed by 1H NMR spectroscopy. Catalytic activities of Ag(I) compounds were tested for three-component coupling reaction of some aldehydes, amines, and phenylacetylene.

Two naphthalene pyrazole ligands were synthesized using KOH/DMSO and Cu catalyst and characterized with FT-IR, ESIMS, 1H, and 13C NMR spectroscopies. The crystal structures of 1-(2-methylnaphthalen-1-yl)-1H-pyrazole (MeNap-Pz) ligand have been determined with X-ray crystal structure analysis. Reaction of the ligands with Cu(NO3)2x3.5H2O gave two new complexes and characterized with magnetic susceptibility, molar conductance, FT-IR, LCMS-MS, ICP-OES, NMR, thermogravimetric analysis, and ESR spectra. The spectral data of the ligands are coordinated to the metal ion through the nitrogen atoms of the pyrazole ring. The reaction of 3,4-dichlorophenyl-1,3,4-oxadiazole-2(3H)-thione with piperidine derivatives via Mannich reaction was used to generate eleven novel compounds in moderate to good yields. Synthesized molecules were characterized according to their structure with 1H NMR, 13C NMR and FT-IR spectral foundations, which were compatible with literature informations. Antimicrobial activity and cytotoxicity studies were done by disc diffusion and NCI-60 sulphordamine B assay methods. The antimicrobial test results revealed that synthesized compounds have better activity against gram-positive species than gram-negative ones. In this study, a new series of Mannich bases, 3-(aminomethyl)-6-{3-[4-(trifluoromethyl)phenyl]acryloyl}-2(3H)- benzoxazolones (1a-g), were synthesized by the Mannich reaction. Inhibitory effects of the newly synthesized compounds towards carbonic anhydrases (CAs) and acetylcholinesterase (AChE) enzymes were evaluated to find out new potential drug candidate compounds. According to the inhibitory activity results, Ki values of the compounds 1 and 1a-g were in the range of 12.3 ± 1.2 to 154.0 ± 9.3 nM against hCA I, and they were in the range of 8.6 ± 1.9 to 41.0 ± 5.5 nM against hCA II. Green Fluorescent Protein (GFP) has grown in popularity and new applications are currently being developed. There are certain residues that are highly conserved through all the naturally occurring fluorescent proteins variants, and some of their functionality is yet to be determined. This is the case for three glycines that appear in a GXGXGX motif in the second β-sheet at positions 31, 33, and 35 with conservations of 100%, 87% and 95%, respectively.

Several Bacillus amyloliquefaciens isolates obtained from wheat residue have been studied as biological agents to control wheat diseases. Notable traits of isolates 1BA and 1D3 include growth with high salt (10% NaCl), at temperatures up to 50°C and over a wide pH range. BIOLOG Gen III plates were used to study and further characterize Bacillus amyloliquefaciens 1BA and 1D3. Both isolates showed tolerance to high salt concentration supporting previous studies. They also grew in the presence of lithium chloride, potassium tellurite, and sodium bromate. Both isolates grew at pH 5, with almost identical carbon source utilization fingerprints. This thesis describes a chemical method for alternative oligonucleotide purification that is non-chromatographic and gel-free and allows to routinely synthesize and purify long functional RNA strands. The purification of long RNAs is based on the bio-orthogonal inverse electron demand Diels-Alder (IEDDA) chemistry between trans-cyclooctene (TCO) and tetrazine (Tz). Target oligonucleotide strands are selectively tagged with Tz and can be captured and purified from the failure sequences with immobilized TCO. Organophosphates are a group of insecticides that are commonly used as chemical warfare agents (CWAs), since they can act as nerve agents by inhibiting acetylcholinesterase. Acetylcholinesterase is a vital enzyme of the central nervous system (CNS). This enzyme facilitates the breakdown of acetylthiocholine into thiocholine and acetate. Gram-negative bacterium Neisseria meningitidis, responsible for human infectious disease meningitis, acquires the iron (Fe3+) ion needed for its survival from human transferrin protein (hTf). For this transport, transferrin binding proteins TbpA and TbpB are facilitated by the bacterium. The transfer cannot occur without TbpA, while the absence of TbpB only slows down the transfer. Thus, understanding the TbpA-hTf binding at the atomic level is crucial for the fight against bacterial meningitis infections. Usnic acid (UA) was extracted from Usnea longissima (L.) Ach. in supercritical carbon dioxide (SC-CO2) medium. The selected process parameters were extraction temperature (35-45 °C), amount of co-solvent (0%-5%) and extraction time (5-9 h). These parameters were applied to Box-Behnken design (BBD) belonging to response surface methodology (RSM) to determine optimum process parameters for the highest amount of UA in the extract. g UA/100g lichen, extraction yield % and UA content values were calculated in the range of 0.045-0.317, 2.77-5.4 and 71%-82% in different experimental conditions, respectively. The optimum conditions were predicted as 42 °C, 4.3% (ethanol) and 7.48 h. …
