Molecular chaperones have an essential role for the maintenance of a balanced protein homeostasis. Here, we investigate how protein kinases are recruited and loaded to the Hsp90-Cdc37 complex, the first step during Hsp90-mediated chaperoning that leads to enhanced client kinase stability and activation. We show that conformational dynamics of all partners is a critical feature of the underlying loading mechanism. The kinome co-chaperone Cdc37 exists primarily in a dynamic extended conformation but samples a low-populated, well-defined compact structure. Exchange between these two states is maintained in an assembled Hsp90-Cdc37 complex and is necessary for substrate loading. Breathing motions at the …

Biofilms are the default state of microbial life and their formation on surfaces influences many processes. The coordinated behavior of microorganisms including their growth into biofilms occurs through a process termed quorum sensing during which they excrete specific signaling molecules called autoinducers. Recently it has been observed that Ag+ and Cu2+ ions interact with N-hexanoyl-L-homoserine autoinducers thereby inhibiting quorum sensing. Such inhibitors are called quorum quenchers. In this study, the biosynthesis of Au and Ag nanoparticles (AuNp and AgNp) with a green chemistry approach was performed by using bioextracts of various fruits and vegetables (Lycopersicon esculentum (LE), Cucumis sativus (CS), Malus domestica (MD), Cucurbita pepo (CP), Apiumgraveolens var. rapaceum (AVR), Prunus cerasifera (PC), Oleraceae var. botrytis (OVB)). Synthesis And Reactions Of Di(Thiophen-2-Yl)Alkane Diones: Cyclocondensation, Teki̇n Artunç, Abdullah Menzek

Known 1,6-di(thiophen-2-yl)hexane-1,6-dione (2) and novel 1,7-di(thiophen-2-yl)heptane-1,7-dione (4) were obtained from the reactions of thiophene with the corresponding diacyl chlorides. Furthermore, compounds with furan and pyrrole units in place of thiophene units in compound 2 were obtained in the same way. Bromination of 2 and 4 gave bromides regioselectively. The reaction of each of the compounds 2 and 4 in HOAc medium yielded cyclocondensation products. In total, four known and eleven novel compounds were synthesized.

The novel pyridine bearing schiff base substituted metal-free (9), zinc(II) phthalocyanine (10), and its quaternized derivative (11) were designed and synthesized. These phthalocyanines were fully characterized by spectroscopic methods (FT-IR, UV-Vis, MALDI-TOF, and 1H NMR). The photo-physicochemical properties of these phthalocyanines were investigated in both DMSO and DMF for 10 and in both DMSO and aqueous solution for 11. The addition of pyridine bearing Schiff base groups as peripheral ligands showed an improvement in the photophysical and photochemical properties. The aim of this work is to research the interactions of water-soluble nitrogen-containing copolymers with essential amino acids in aqueous media. For this, poly(N-vinylformamide-co-N-vinylimidazole) and poly(N-vinylcaprolactam-co-N-vinylimidazole) random copolymers were synthesized by free radical polymerization. The products obtained are characterized by GPC, DLS, and FTIR. The copolymers have a narrow molecular weight distribution and low dispersity. The interactions of the obtained copolymers with histidine, proline, arginine, leucine, phenylalanine, and methionine were researched by UV spectroscopy, FTIR, and TEM. The Investigation Of Parameters Affecting Ibrutinib Release From Chitosan/ Tripolyphosphate/Carbon Nanofiber Composite Microspheres, Canan Armutcu

This study described the performance of carbon nanofiber modified chitosan (CNF@CS) composite microspheres for the controlled release of the Ibrutinib (IBR) drug. The surface morphology, particle sizes, and functional group contents of the microspheres were characterized by attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), scanning electron, and optical microscopy measurements. The obtained data demonstrated that the addition of CNF to the microsphere increased the encapsulation efficiency of the IBR while allowing the controlled and gradual release of the drug. Isolation and characterization of anticancer activity guided secondary metabolites of endemic Astragalus leucothrix Freyn& Bornm were aimed. Aerial parts of the plant were extracted by maceration method in the solvent system methanol-chloroform (1 : 1) at room temperature. The obtained crude extract was dissolved in purified water. Then, the extract was partitioned with n-hexane, chloroform, ethyl acetate, and n-butanol, respectively. Adsorption isotherms, kinetic and thermodynamic parameters for Cd(II) and Pb(II) ions in water solutions by using Amberlyst 15 resin were performed and evaluated by utilizing solid phase extraction method with the batch system at 298, 308, and 318 K. Flame atomic absorption spectrometry was utilized for absorbance measurements of Cd and Pb in solutions. The Langmuir, Freundlich, and Dubinin?Radushkevich isotherm models, respectively were implemented to equilibrium results obtained. In this study, a water-soluble metal-free phthalocyanine (SPC) containing sodium 2-mercaptoethanesulfonate substituents at the peripheral positions was used to investigate the algaecidal properties and oxidative effects on the growth of two microalgal species, Arthrospira platensis and Chlorella vulgaris. Although OD at 560 nm and chlorophyll-a content were decreased in Arthrospira platensis during 7 days depending on dose and time, increases in both OD at 750 and chlorophyll-a content at 8 ppb (parts per billion) concentration on the 7th day were observed in Chlorella vulgaris. Molecular imprinted polymers (MIP) have key-lock pattern binding properties specific to the size and shape of target molecules. In this study, we have prepared detection platforms based on a molecularly imprinted surface plasmon resonance (SPR) sensor that can detect bovine serum albumin (BSA) sensitively, selectively, quickly, and in real time. Ubiquitination and other post-translational modifications play a major role in the proliferation of many neurodegenerative diseases, developmental disorders, and cancers, and as such are subjects of recently increased biochemical interest. Expanding upon such research, this study confirmed a robust method of ubiquitin expression and purification, then used 15N-1H HSQC to analyze ubiquitin samples containing varying concentrations of a deep cavitand with affinity for lysine and arginine side chains. In the pure ubiquitin spectrum, arginine side chain chemical shifts were detected in the nitrogen 80-85 ppm range and lysine side chain signals were undetected. Guanine nucleotide-binding proteins, or G-proteins, are crucial to cell signaling by acting as molecular switches through interactions with membrane-bound receptors. G-proteins have become of greater interest due to the possibility of novel drug targets. The G-protein alpha subunit, Gα, binds GTP before localizing to the membrane to interact with other G-protein subunits and G-protein-coupled receptors (GPCRs). Gi lacks a hydrophobic region to promote localization to the membrane, but a co-translational lipidation of the N-terminal residue stabilizes this interaction.

Electrochemical impedance spectroscopy (EIS) is an important non-destructive tool that allows for a deeper understanding of the electrochemical processes and mechanisms occurring in an electrochemical cell. Equivalent electrical circuits (EECs) are used to model the impedance data into electrical components, such as resistors and capacitors, on a circuit. RNA being composed of multiple covalently linked nucleotides is thought to have been a precursor to life circa 4.3-3.8 billion years ago. Non-enzymatically formed adenosine monophosphate (AMP), more specifically, is a vitally important subtopic of the self-assembly of the first RNA sequence. The goal of this study was to synthesize AMP non-enzymatically under benign conditions that are likely to have existed on early Earth. Glycolipids with diacetylene functional groups are fascinating compounds with many practical uses. Among these, diacetylene-containing gelators are especially important because they can form photopolymerizable gels, which are useful stimuli-responsive materials. In this study, novel silver nanoparticles (AgNPs) were successfully synthesized and functionalized with an antibacterial agent, namely chlorothymol (denoted C@AgNPs). The resulting colloid (C@AgNPs) was purified by two comparative methods: ultrafiltration and ultracentrifugation. Ultrafiltration proved to be more efficient in purifying and size selecting (10 kD filter) and concentrating the C@AgNPs than ultracentrifugation. The physicochemical properties of the filtered C@AgNPs were then characterized by UV-Vis absorption spectroscopy, inductively coupled plasma optical emission spectroscopy (ICP-OES), Raman spectroscopy, Cytoviva hyperspectral imaging, and Scanning electron microscopy. This project focuses on the optimization of benzothiazole-based chromophores to utilize them as fluorescent tags functionalized onto stimuli-responsive, or “smart,” polymers as non-viral gene delivery vectors for gene therapy applications. Blue-fluorescent emissive chromophores will allow tracking capabilities for the transfection pathway of the vector to be monitored as it delivers the DNA payload within intercellular space. The most appropriate chromophore must be covalently bound to a free amine within the vector, which is accomplished through NAS chemistry from fluorophenyl-benzothiazoles (F-BTZ-CBz) derivatives to amidated hyperbranched poly(ethyleneimine)(HPEI-IBAm0.61). Benzotriazoles are a class of contaminant of emerging concern which are commonly used as anticorrosive agents in aircraft deicer and anti-icing fluids (ADAFs). The analogues 1H-benzotriazole (BTZ), 4-methyl-1H-benzotrizole (4m-BTZ), and 5-methyl-1H-benzotriazole (5m-BTZ) are commonly found in environmental occurrence together. The two methylated isomers, collectively known as tolytriazole (TTZ), have different toxicity and stability. These contaminants are highly water soluble and resistant to biodegradation, making them persistent through water treatment. Eighteen compounds were obtained from hexane and dichloromethane extracts from the rhizomes of Stemona curtisii Hook f. Their chemical structures were determined by extensive spectroscopic studies including 1D and 2D NMR, MS, UV, IR, and comparison with data from the literature. This study aimed to isolate γ-oryzanol from the crude oil acid fraction (a by-product of rice bran oil refinery) and to evaluate their Angiotensin-Converting Enzyme 2 (ACE2) inhibitory activity. Chromatographic fractionation of the crude oil acid fraction gave 3 isolated fractions of γ-oryzanol containing cycloartenyl ferulate, campesteryl ferulate, sitosteryl ferulate, stigmasteryl ferulate, and cyclobranyl ferulate. The γ-oryzanol containing cycloartenyl ferulate and campesteryl ferulate as major component showed good ACE2 inhibitory activity with the IC50 value 10.57 μg/mL.
