A total integrated electrocolorimetric sensing approach consisting of gel-based electromembrane extraction and colorimetric detection in one-step process was developed. This system was designed using colorimetric reagents preadded to the agarose gel for the determination of the following two model analytes: iodide for negative ions and nickel for positive ions. In this system, when a voltage was applied, the analytes were extracted and transferred from the sample solution (donor phase) to the gel (acceptor phase). In this work, rapid screening and onsite monitoring of Cr(VI) using paper-based has been developed. Layered double hydroxides (LDHs) or hydrotalcite compounds are referred to anionic clays with specific anion adsorption and exchanging properties. These behaviors are similar to ionophores and ionic additives in ion selective electrodes (ISEs). Therefore, in this study LDHs was used to function as tailor-made ionophores for particular anions and ionic additives in ISEs. MgAl LDHs were prepared, and then modified the intercalated anions to be NO3-. The selective polymeric membrane was casted using PVC and MgAl LDHs-NO3-. Novel approaches were developed for the colorimetric detection of norfloxacin and ceftazidime using chemically modified polymer resin and cotton sponge as sensing materials, respectively. For norfloxacin detection, the optimum condition was achieved by using a resin modified with 6.0 µM Congo red at pH 6 to extract norfloxacin in 10 mL of sample at pH 2. The reaction was completed within 15 minutes. Regarding ceftazidime detection, a cotton sponge functionalized with polyethyleneimine (PEI) was fabricated and characterized by FTIR, SEM, and ninhydrin test. In this dissertation, nickel(II) complexes featuring N4-Schiff base macrocycles derived from diphenylamine-2,2'-dicarboxaldehyde (1-Ni and 2-Ni) with a new crystal structure ([2-Ni]Me) were synthesized and investigated for electrochemical CO2 reduction (ECR). Cyclic voltammetry (CV) of two nickel complexes, 1-Ni and 2-Ni, illustrated the catalytic response whereas one, [2-Ni]Me, virtually remained peak-shaped in the presence of CO2, indicating the feasibility of ECR activity for these nickel complexes. To develop heterogeneous ECR catalysts in aqueous media, all nickel complexes were adhered on N-doped graphene (NG) through non-covalent interaction, obtaining Ni@NG hybrid catalysts. The goal of this project is to explore a new method to efficiently remove Pb(II) ions from water by processing Undaria pinnatifida into immobilized beads using sodium alginate and calcium chloride. The resulting biosorbent was characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy (SEM-EDS). Using immobilized U. pinnatifida, we investigated the effect of various factors on Pb(II) ion removal efficiency such as temperature, pH, ionic strength, time, and underlying biosorption mechanisms. Mitochondria are cellular organelles that are becoming more recently studied. One of their main functions is the production of energy through cellular respiration, which is crucial to cell life. However, they are also associated with numerous disease states. This study investigated the protein adsorption interactions of bovine serum albumin (BSA) with three different functionalized 5 nm gold nanoparticles (AuNPs) in order to determine the binding affinity and total amount of protein adsorbed to each AuNP surface chemistry. In the present study, Ag nanoparticles (AgNPs) were synthesized from Salvia officinalis and Thymus vulgaris, known as phytotherapy plants. The obtained silver nanoparticles were characterized using SEM, XRD, FTIR, and UV/Vis spectra. The antioxidant capacities of Salvia officinalis-mediated AgNP (SO-AgNP) and Thymus vulgaris-mediated AgNP (TV-AgNP) were analyzed in vitro using 1,1-diphenyl-2-picrylhydrazyl and iron chelating activity assays. DPPH activities were 83.74% and 57.17% for SO-AgNP and TV-AgNP at concentration 200 mg/L, respectively. Both green synthesized AgNPs exhibited good iron chelating activity. In addition, the DNA cleavage activities of SO-AgNPs and TV-AgNP were investigated with agarose gel electrophoresis technique. …

The aim of this study was the determination of chromatographic profiles of major whey proteins (WP): α-lactalbumin (α-La), β-lactoglobulin A and B, (β-Lg A and B), bovine serum albumin (BSA) in dairy beverages based on Zonar milk serum. The studied WP were separated by high-performance liquid chromatography (HPLC) on Aeris XB-C18 column using gradient elution with 0.1% trifluoroacetic acid (TFA) in water and 0.1% TFA in 80% acetonitrile, at 214 nm detection. Different derivatives of imatinib were synthesized by a 3-step reaction method. The structures of the new compounds were characterized by spectroscopic methods. For quantitative evaluation of the biological activity of the compounds, MTT assays were performed, where four BCR-ABL negative leukemic cell lines (Jurkat, Reh, Nalm-6 and Molt-4), one BCR-ABL positive cell line (K562), and one non-leukemic cell line (Hek293T) were incubated with various concentrations of the derivatives. Although imatinib was specifically designed for the BCR-ABL protein, our results showed that it was also effective on BCR-ABL negative cell lines except for Reh cell line. Hybrid nanostructures decorated with glycopolymers are appropriate for biomedical applications. In this paper, the results are obtained from nanographene (NG) decorated with glycoblock copolymer to increase their potential in use in therapies and in examining lectin interactions. A pyren-1-ylmethyl 4-cyano-4-((phenylcarbonothioyl)thio)pentanoate (CPADB-py) chain transfer agent was used in the synthesis of methyl methacrylate glycoblock copolymers (P2 and P3) by reversible addition-fragmentation chain transfer (RAFT) polymerization to adhere the polymer to the nanographene surface. Microwave irradiation has become a routine technique in homogeneous and effective heating in organic synthesis. However, its application in enzyme-containing reactions is limited since it can cause denaturation of the enzyme. In this study, we have briefly investigated the effect of microwave heating on the conjugation reaction of horseradish peroxidase (HRP) with aldehyde derivative of dextran (D-CHO). Quercetin, a multifunctional therapeutic agent, is used in various types of cancer. However, its therapeutic effect is limited by virtue of poorly aqueous solubility and instability in the physiological medium. To overcome these limitations, we aimed (i) to design quercetin loaded liposomes with unlinked-PEG4000 with regard to not only surface modification but also solubility enhancement, and (ii) to investigate the antineoplastic effects on HeLa cells. PEG4000 increased the quercetin solubility 2.2 fold. In this study, a new copolymer, which is a combination of two chemical structure (9,10-di(furan-2-yl)anthracene and 3,4-ethylenedioxythiophene was synthesized via electrochemical synthesis methods in the electrolyte medium of 0.1 M TBAPF6/ ACN solution. PEDOT homopolymer was also electrosynthesized for comparator experiments of the polymer formations in the same medium. The characterizations of polymers were achieved with general optical and electrochemical characterization techniques. The corresponding electrochromic copolymer shows blue and lilac color in its neutral and oxidized states, respectively. The synthesis of fluorescent carbon quantum dots (CQDs) and their applications have attracted great attention due to their excellent properties. Especially, the unique visible-light absorption and photo-induced electron transfer properties make CQDs available in photocatalytic degradation of organic dye pollutants in water resources. Herein, we synthesized nondoped CQDs and boron-doped CQDs (B-CQDs) by hydrothermal method and compared their photocatalytic degradation activity of methylene blue (MB) and methyl red (MR) dyes under visible light irradiation. It has been reported for many globular proteins that the native heat capacity at 25°C, per gram, is the same. This has been interpreted to indicate that heat capacity is a fundamental property of native proteins that provides important information on molecular structure and stability. Heat capacities for both proteins and DNA has been suggested to be related to universal effects of hydration/solvation on native structures. The in-water ozonolysis of alkenes is an emerging alternative method to the conventional ozonolysis since it eliminates the requirement for a reductive work-up step using expensive and hazardous reductants such as dimethyl sulfide, triphenylphosphine, and zinc in acetic acid. To date, catalase protein from bacterial species is not the most widely used commercial catalase. Catalase, the enzyme that breaks down hydrogen peroxide in most living organisms, is an invaluable enzyme in the food, agriculture, and pharmaceutical industries. In 2019, a novel species of a psychrotolerant bacterium was discovered in Cold Harbor, NY. This species, Psychrobacter koprana, exhibits highly active catalase. Preliminary data from activity assays demonstrates that P. koprana catalase has the same activity as commercially available catalase.
