Synthesis And Biological Activities Of Acyl Derivatives Of Hydroxylated Eudesmanolides, Rawinnipha Wettayanugul

This research focused on the synthesis of two hydroxylated eudesmanolides, reynosin (RNS, 2) and santamarine (STM, 3) from costunolide (1) isolated from Saussurea lappa roots, and the synthesis of their acyl derivatives to study on their biological activities. A total of 20 acyl derivatives of hydroxylated eudesmanolides, including 14 new compounds: 2(a-c), 3b, 3c, 3e, 3f, 3(h-j), and 3(l-o). The compounds were then evaluated for their anti-melanogenic, anti-colorectal cancer, and anti-inflammatory activities. It was observed that STM (3) exhibited anti-melanogenic and anti-colorectal cancer activity with IC50 values of 8.02 and 15.55 µM, respectively, which was greater than RNS (2) (IC50 …

In this thesis, two types of polymeric gels made from naturally derived polymers have been developed for bio-catalysis and wound dressing applications. In the first part, the heterogeneous biocatalyst with tunable geometry was fabricated using a 3D printing technique. The hydrogel-based ink as a supporting material for the immobilization of laccase was prepared from alginate (Alg) combined with Pluronic® F127-dimethacrylate (F127-DMA). In this work, we have developed the mild and efficient desulfurization process in water such as 1) conversion of amine to isothiocyanate and 2) guanylation of thiourea into guanidine. For the first project, isothiocyanate was successfully synthesized from amine and carbon disulfide in the presence of cesium carbonate (Cs2CO3) and diacetoxyiodo benzene (DIB) as desulfurizing agent under 1 wt% TPGS-750-M/H2O. With optimized condition at room temperature in our hand, isothiocyanate derivatives were isolated by column chromatography in low to high yields (37-94%). In this study, we have developed portable and cost-effective electrochemical sensors by utilizing a screen-printed graphene electrode (SPGE). This dissertation focuses on the development of a non-invasive wearable chemical sensor for the detection of sweat biomarkers using electrochemical techniques. This dissertation can be divided into two parts. In the first part, a thread-based electrochemical immunosensor was developed for the detection of cortisol to identify adrenal disorder. The conductive threads were modified with MXene, gold nanoparticles and L-cysteine to increase conductivity and specific surface area for antibody immobilization leading to enhanced sensitivity. Selective binding of organic compounds is the cornerstone of many important industrial and pharmaceutical applications. Here, we achieved highly selective binding of aromatic compounds in aqueous solution and gas phase by oxygen-enriched graphene oxide (GO) nanosheets via a previously unknown mechanism based on size matching and polarity matching. Oxygen-containing functional groups (predominately epoxies and hydroxyls) on the nongraphitized aliphatic carbons of the basal plane of GO formed highly polar regions that encompass graphitic regions slightly larger than the benzene ring. Many wildlife species are live captured, sampled, and released; for polar bears (Ursus maritimus) capture often requires chemical immobilization via helicopter darting. Polar bears reduce their activity for approximately 4 days after capture, likely reflecting stress recovery. To better understand this stress, we quantified polar bear activity (via collar‐mounted accelerometers) and body temperature (via loggers in the body core [Tabd] and periphery [Tper]) during 2–6 months of natural behavior, and during helicopter recapture and immobilization. Massive Australian wildfires lofted smoke directly into the stratosphere in the austral summer of 2019/20. The smoke led to increases in optical extinction throughout the midlatitudes of the southern hemisphere that rivalled substantial volcanic perturbations. Previous studies have assumed that the smoke became coated with sulfuric acid and water and would deplete the ozone layer through heterogeneous chemistry on those surfaces, as is routinely observed following volcanic enhancements of the stratospheric sulfate layer. Here, observations of extinction and reactive nitrogen species from multiple independent satellites that sampled the smoke region are compared to one another and to model calculations. The …

Background

Biochar ozonization was previously shown to dramatically increase its cation exchange capacity, thus improving its nutrient retention capacity. The potential soil application of ozonized biochar warrants the need for a toxicity study that investigates its effects on microorganisms.

Results

In the study presented here, we found that the filtrates collected from ozonized pine 400 biochar and ozonized rogue biochar did not have any inhibitory effects on the soil environmental bacteria Pseudomonas putida, even at high dissolved organic carbon (DOC) concentrations of 300 ppm. ปัจจุบันน้ำเสียกลายเป็นปัญหาใหญ่ที่ผู้คนต้องเผชิญในชีวิตประจำวัน แหล่งน้ำเสียประมาณ 70% มาจากน้ำเสียครัวเรือน ซึ่งมีสารประกอบไนโตรเจน (N) และฟอสฟอรัส (P) ที่มีความเข้มข้นสูง แต่กลับไม่ได้นำมาใช้ให้เกิดประโยชน์ งานวิจัยนี้จึงขอเสนอการพัฒนาตัวกรองชีวภาพที่มีชีวิตแบบจุลินทรีย์ผสมระหว่างจุลสาหร่ายและแบคทีเรียที่ส่งเสริมการเจริญเติบโตของพืชในการบำบัดน้ำเสียสังเคราะห์เพื่อศึกษาประสิทธิภาพในการกำจัดสารประกอบ N P และค่าความต้องการออกซิเจนทางเคมี (COD) จากน้ำเสียสังเคราะห์ระหว่างระบบแขวนลอยและตัวกรองชีวภาพที่มีชีวิต ตัวกรองชีวภาพที่มีชีวิตได้รับการพัฒนาโดยใช้เทคนิคการตรึงเซลล์ด้วยสารยึดเกาะบนพื้นผิวของผ้าฝ้ายที่ใช้เป็นวัสดุรองรับ จากการทดลองพบว่าสารยึดเกาะกัวร์กัม/คาราจีแนน (GG/CG) แสดงเปอร์เซ็นต์ความเสื่อมสภาพน้อยกว่า 5 % ซึ่งต่ำกว่าสารยึดเกาะอื่น ๆ และไม่เป็นพิษต่อเซลล์ หลังจากการบำบัดน้ำเสียสังเคราะห์เป็นเวลา 7 วัน พบว่าระบบตัวกรองชีวภาพที่มีชีวิตมีประสิทธิภาพการกำจัดสารอาหารและ COD สูงกว่าระบบแขวนลอย โดยเฉพาะอย่างยิ่งตัวกรองชีวภาพที่มีชีวิตในอัตราส่วนการผสม 3:1 ของจุลสาหร่าย Chlorella sp. และแบคทีเรีย Bacillus subtilis TISTR1415 ที่ตรึงด้วยสารยึดเกาะ GG/CG มีประสิทธิภาพในการกำจัด NH4+ PO43- และ COD สูงสุดมากถึง 91.81 % 99.44 % และ 83.15 % ในขณะที่ระบบแขวนลอยแสดงประสิทธิภาพในการกำจัดเพียงแค่ 56.75 % 58.71 % และ 69.57 % ตามลำดับ นอกจากนี้ ค่า pH ในระบบตัวกรองชีวภาพที่มีชีวิต (จาก 8.81 เหลือ 7.26) ลดลงมากกว่าระบบแขวนลอย (จาก 8.81 เหลือ 8.13)

In this study, antimicrobial nanofibers were produced with the mixtures of polyvinyl alcohol (PVA) and cyclotide-rich fractions by electrospinning method. After extraction, the first separation was carried out with C18 flash chromatography and then fractioned into five separate parts by reversed-phase high-pressure liquid chromatography (RP-HPLC). The molecular weights of cyclotides in each fraction were determined by quadrupole time-of-flight liquid chromatography-mass spectrometry (Q-TOF LC-MS). Cyclotide-rich fractions were mixed with 10% of PVA solution and nanofibers were produced from this biocomposite mixture by electrospinning method. Over the last 20 years, several novel anti-anxiety therapies have entered the drug development pipeline, but none have made it to market.

Computational modeling is an invaluable tool in the drug discovery process either for small ligand or protein therapeutics. The widespread availability of protein X-Ray Crystal and Cryo-Electron Microscopy (Cryo-EM) structures has allowed for more accurate molecular dynamics (MD) simulations that are not reliant on methods such as homology modeling, which may produce structures that require significant computational time to demonstrate their stability. A new series of Pb(II) cyanoximates were synthesized and characterized by elemental analysis (C, H, N, S), IR spectroscopy, thermal analysis, and X-ray crystallography. The goal was to synthesize compounds with a PbL2 (L= selected known chelating cyanoximes) composition for use as potential new non-linear optical (NLO) materials, specifically second harmonic generators (SHG), which must belong to non-centrosymmetric space groups. However, researchers have shown contradicting effects of CeO2-NPs on barley grains with focus mostly on concentration effects and not sizes of these CeO2-NPs. Therefore, this study aimed at comparing the effects of 0nm (control), 8nm, and 40nm (250 mg CeO2-NPs/kg soil) on the metabolite profile of barley grains. The long-term goal of this thesis is to design novel chemical protective garments that both shield and sense for chemical warfare agents. The overarching hypothesis of this work is that joining textiles with a chemically responsive temporary adhesive will reduce the permeability of and detect hazardous analytes across fabric boundaries. Counterfeit pharmaceuticals pose a threat to society that can include inaccurate amounts of the active pharmaceutical ingredient (API), no API, or containing off-target compounds. For example, there are many recent examples of counterfeit pharmaceuticals containing potentially lethal doses (> 2 mg) of fentanyl (i.e., a synthetic opioid). Current measures to combat illicit pharmaceuticals (e.g., unique packaging and product serialization) have merit, however with evolved technologies, counterfeiters can relatively easily simulate these measures and continue to distribute illicit pharmaceuticals. Drug abuse within the United States is a dynamic and pervasive issue that affects the quality and life expectancy of our society. One of the major drug classes that are constantly utilized is opioids. Opioids are prescribed for pain management in patients suffering from cancer and other medical illnesses. A nanocomposite containing Fe3O4, chitosan (Ch), and hexacyanoferrate (HCF) was synthesized in the form of powder. The physicochemical properties of this nanocomposite are determined using different techniques including Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), scanning electron microscopy (SEM). The existence of Cs (Cs) ions onto the surface of the nanocomposite was verified by dispersive X-ray spectroscopy (EDX). FTIR spectra confirmed that the nanocomposite was well coordinated. Due to the length of time required to develop specific antiviral agents, the World Health Organization adopted the strategy of repurposing existing medications to treat Coronavirus disease 2019 infection. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) main protease is possible biological target for potential antiviral drugs. We selected various compounds from PubChem database based on the structure of main protease inhibitors in Protein Data Bank database. Activated carbon, having high surface area and porosity, is a carbonaceous material that comes from organic main sources. In this work, activated carbon materials were produced from human hair that belongs to people who can be classified into two age groups (0-18 or 18-40 years). Activated carbons were characterized by nitrogen adsorption/desorption isotherms (BET), X-ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Raman spectroscopy, scanning electron microscopy (SEM), and elemental analysis techniques. In this study, biological activities of silver nanoparticles (AgNP) synthesized by green synthesis method using the fruit extract of Gilaburu (Viburnum opulus L.) plant were investigated. The characterization of the synthesized AgNPs was performed by ultraviolet visible region spectroscopy (UV-Vis), Fourier transform - infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and X-ray diffraction (XRD) techniques. Teucrium species that belong to the family Lamiaceae have been traditionally used for their medicinal properties. T. polium is one the most widespread members of the genus for its use in the treatment of several diseases. In this study, the essential oil and phenolic composition of the aerial parts from T. polium were assessed by GC-FID, GC/MS, and LC-MS/MS as well as for its total phenolic content. Sorbitol is one of the platform chemicals and can be produced from various renewable and sustainable sources via different processes. Hydrothermal liquefaction is an effective and promising approach to produce sorbitol, since the subcritical reaction media and appropriate catalysts provide a selective production of platform chemicals. In this study, sorbitol was produced from different renewable sources (cellulose and glucose) in the presence of Ru-based catalysts (Ru/SiO2, Ru/AC, Ru/SBA-15, and Ru/SBA-15-SO3) under subcritical conditions. A fluorescent probe based on bovine serum albumin stabilized copper nanoclusters (BSA-CuNCs) was developed for the selective and sensitive determination of ornidazole (ORN). The nanoclusters were synthesized via a one-pot hydrothermal process in basic media. The synthesized and characterized BSA-CuNCs have less than 3 nm particle size and exhibited blue emission at 405 nm when excited at 325 nm. Synthesized and characterized nanoclusters were successfully applied as a turn-off fluorescent probe for the determination of ORN in pharmaceutical dosage forms.
