Terra-Lsd1 Phase Separation Promotes R-Loop Formation For Telomere Maintenance In Alt Cancer Cells, Nicholas J. Reiter

The telomere repeat-containing RNA (TERRA) forms R-loops to promote homology-directed DNA synthesis in the alternative lengthening of telomere (ALT) pathway. Here we report that TERRA contributes to ALT via interacting with the lysine-specific demethylase 1A (LSD1 or KDM1A). We show that LSD1 localizes to ALT telomeres in a TERRA dependent manner and LSD1 function in ALT is largely independent of its demethylase activity. Instead, LSD1 promotes TERRA recruitment to ALT telomeres via RNA binding. In addition, LSD1 and TERRA undergo phase separation, driven by interactions between the RNA binding properties of LSD1 and the G-quadruplex structure of TERRA. Importantly, the …

Microplastics (plastic pieces less than 5 mm) have emerged as a pervasive form of pollution acknowledged as a universal threat to both environmental and human health. Their small size and widespread distribution pose significant risks to marine life, terrestrial ecosystems, and ultimately human health. Microplastics have been reported in food and drink items designated for human consumption, in cosmetics and consumer goods designed for human use, and within the air we breathe, each providing an entry route for microplastics to enter the human body. Fuel cells are considered a promising technology for achieving cleaner and more sustainable energy production and utilization. However, one of the current major challenges is the economic viability associated with the extensive use of Pt group metal (PGM) catalysts, particularly on the cathode side. Therefore, non-PGM single-atom catalysts (SACs) have received considerable attention as alternatives for PGM catalysts in the oxygen reduction reaction (ORR), which occurs at the cathode of the fuel cell. Among various SAC structures, the non-PGM/nitrogen-doped carbon-based structure (M-N-C) stands out for its excellent ORR performance.

Despite notable advancements in the design and synthesis of novel chemotherapeutic drugs, cancer continues to be a leading cause of death in the United States. Metallodrugs, especially Au complexes, have played a prominent role in the search for the next generation of chemotherapeutic agents. Herein, I report on the synthetic strategies towards the rational design of stable Au (Au)-based agents, study their reactivity in biological environment, and offer insights into the cytotoxic mechanism of action in cancer. Chronic inflammation and dysfunctional inflammatory responses are believed to be contributing factors to the severity of many diseases. Since the emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), it has been recognized that levels of inflammatory markers and the severity of coronavirus disease-2019 (COVID-19) cases are closely related. The work within this dissertation begins with establishing an alternative platform for the diagnosis of COVID-19 and explores ways to enhance the diagnostic utility of this platform. Over the past decades, nucleic acids have emerged as crucial frameworks with significance in biological systems and functional material engineering. Cytosine-rich deoxyribonucleotide sequences can form quadruplex structures a.k.a. i-motifs. Recently, i-motifs have attracted scientific interest in biological and material design areas.
In this work, we systematically investigate an effect of structured and unstructured modifications such as hairpins, stems, loops and overhangs on the kinetic and thermodynamic properties of DNA i-motifs. We demonstrate that the modifications can influence i-motif’s stability and folding/unfolding rates: thus, double stranded stems and hairpins are the best at stabilizing i-motifs and increasing kinetic rates of folding. …

Supramolecular and Molecular chemistry offer an expanding array of applications for the development of smart devices, with calixarene-based molecular baskets playing a pivotal role in host-guest chemistry. A key concept in this field is the design of "smart" molecules with gate-like features, which can selectively transport chemical or biochemical species that are volatile, reactive, or sensitive to typical solution environments. These molecular hosts are engineered with predefined groups that respond to external stimuli, such as changes in concentration, temperature, or pH.

The work outlined in this dissertation describes acid-base catalyzed projects facilitating the construction of key synthetic analogs with vast therapeutic applications. Three major projects have been covered demonstrating various synthetic routes developed by our research group to furnish key derivatives of heterocyclic compounds, including coumarins, carbohydrate moieties, and beta-lactams. By their catalytic nature, acids, and bases are known to enhance the reactivities of compounds. This allows synthetic chemists to design or chart new and better reaction pathways leading to the delivery of desired reaction products. DIAPH1 is diaphanous related formin which polymerizes actin into filaments and is important to cytoskeleton dynamics.[1, 2] The dysregulation and aberrant activation of DIAPH1 is known to cause deafness[3, 4] and clotting disorders,[5] and is recently implicated in the inflammatory responses associated with cardiovascular and diabetic pathology.[6-9] Despite this, activation of the full length structure and its regulation are poorly understood.[10-12] Therefore, the main objective of this dissertation is to explore the structural, autoinhibitory, and regulatory properties of DIAPH1 in the context of disease pathology. Glycogen synthase kinase-3 (GSK-3) is a serine/threonine kinase which plays a center role in the phosphorylation of a wide variety of proteins, generally leading to their inactivation. As such, GSK-3 is viewed as a therapeutic target. An ever-increasing number of small organic molecule inhibitors of GSK-3 have been reported. Phenylmethylene hydantoins are known to exhibit a wide range of inhibitory activities including for GSK-3β. A family of fourteen 2-heterocycle substituted methylene hydantoins (14, 17–29) were prepared and evaluated for the inhibition of GSK-3β at 25 μM. In this study, reproducible label-free detection and quantification of the antiretroviral drug emtricitabine (FTC) down to 78 ng/mL in human plasma by surface enhanced Raman spectroscopy (SERS) is presented. A novel plasma sample pretreatment method using silver nitrate and silver colloidal nanoparticles (Ag CNPs) was used to prepare the plasma samples for analysis. The pretreated plasma samples were evaporated to dryness on an aluminum surface and a computer-controlled Raman scanning system was used to collect spatially resolved SERS spectra of the entire surface. "Pathological processes often involve complex biochemical changes which can be assessed using advanced mass spectrometry. In this present dissertation, two fields were studied: traumatic brain injury (TBI), and water contamination by L. pneumophilia. TBI is a pressing public health concern for which current clinical tools remain inadequate. We present newly developed mass spectrometric methods to access metabolites associated with blast induced TBI. We applied these methods to the biofluids of soldiers conducting explosives training. Significant changes in several metabolites were observed between pre- and post-blast specimens, including changes that increased with repeated exposure. These changes point to the possibility of …

"The development of highly sensitive and efficient analytical methods utilizing advanced instrumentation is necessary to help improve disease diagnosis and therapeutics. A major neuro-consequence of traumatic brain injury (TBI) is oxidative stress from the generation of reactive oxygen species and the depletion of antioxidant defenses. Alteration in concentrations of certain small molecules also occurs with the disease progression and can help understand TBI pathophysiology. Phosphorescent materials are essential in bioimaging, anti-counterfeiting, and other applications due to their prolonged light emission. However, current phosphorescent materials face challenges such as complex synthesis, high costs, and instability. Carbon dots (CDs), with size less than 10 nm, are a promising alternative due to their biocompatibility, tunable optical properties, and simple synthesis. This thesis explores the synthesis of CDs using ethylenediamine (EDA) and phosphoric acid via a microwave-assisted method, followed by silica encapsulation using a modified Stöber process to form CDs@SiO2, aiming to enhance their stability and optical properties. These semiconductors are integral components in electronic devices such as transistors, diodes, organic semiconductors, organic field-effect transistors, and organic light-emitting diodes. Illicit and hazardous substances pose a serious effect on the human body which can create longer-term issues on the mental and physical health which potentially lead to fatalities. Current methods for detecting illicit substances include analytical techniques like mass spectrometry (MS) and gas/liquid chromatography (GC/LC) which are known for accuracy and sensitivity. However, these instruments are bulky, costly, time-consuming, and require highly trained personnel. While the oxidative chemistry of transition metals such as iron and copper is a highly developed area of investigation, the study of similar chemistry with nickel is much younger. However, nickel offers rich coordination chemistry with oxygen and other oxidants and is a promising avenue of research for applications such as sustainable hydrocarbon functionalization. Herein, we summarize the progress made recently in nickel coordination chemistry relevant to hydrocarbon functionalization and offer our perspectives on open questions in the field.

Polyol-induced extraction (PIE) is an extraction technique for separating organic solvents from water, using glycerol and polyol as separating agents. Glycerol binds water releasing the organic solvent as a separate liquid phase. this thesis, investigates how the PIE process may be used to separate caffeine and capsaicin. When acetonitrile and water are used as a 1:1 solvent mixture, two immiscible phases are created upon addition of the polyol, glycerol. แบตเตอรี่ลิเธียมไอออนนับเป็นเทคโนโลยีการกักเก็บพลังงานที่ได้รับความนิยมอย่างแพร่หลายในปัจจุบัน เนื่องจากมีความหนาแน่นพลังงานสูงและประสิทธิภาพในการชาร์จ-คายประจุที่ดี อย่างไรก็ตาม ยังประสบข้อจำกัดหลายประการ อาทิ ความไม่ปลอดภัยจากการใช้สารอิเล็กโทรไลต์อินทรีย์ ต้นทุนการผลิตที่สูง และข้อจำกัดด้านทรัพยากรลิเธียมในธรรมชาติ ส่งผลให้เกิดความสนใจในการพัฒนาเทคโนโลยีแบตเตอรี่ทางเลือกที่มีความยั่งยืนมากยิ่งขึ้น หนึ่งในระบบที่ได้รับความสนใจคือแบตเตอรี่สังกะสีชนิดชาร์จซ้ำได้ซึ่งมีจุดเด่นด้านความปลอดภัยที่สูง ต้นทุนต่ำ ความเป็นมิตรต่อสิ่งแวดล้อม และการเข้าถึงทรัพยากรสังกะสีที่สะดวก อย่างไรก็ดี แอโนดสังกะสีในระบบอิเล็กโทรไลต์น้ำยังเผชิญกับอุปสรรคสำคัญหลายประการ เช่น การเกิดเดนไดรต์ การปลดปล่อยแก๊สไฮโดรเจน การก่อตัวของชั้นซิงก์ออกไซด์ที่ไม่ทำปฏิกิริยาไฟฟ้าเคมี และการกัดกร่อนของแอโนด ซึ่งล้วนเป็นปัจจัยที่ลดทอนเสถียรภาพและประสิทธิภาพของแบตเตอรี่โดยรวม ด้วยเหตุนี้ งานวิจัยฉบับนี้มีวัตถุประสงค์เพื่อพัฒนาอิเล็กโทรไลต์กึ่งแข็ง บนพื้นฐานของพอลิอะคริลาไมด์ ร่วมกับเจลาตินในอัตราส่วนร้อยละ 3, 5 และ 7 โดยน้ำหนักของอะคริลาไมด์ เพื่อยกระดับความเสถียรทางเคมีไฟฟ้าและสมรรถนะของขั้วแอโนดสังกะสี ผลการทดลองพบว่าเซลล์สังกะสีแบบสมมาตรที่ใช้อิเล็กโทรไลต์กึ่งแข็งฐานพอลิอะคริลาไมด์ที่เติมเจลาตินร้อยละ 5 แสดงค่าความต่างศักย์เกิน ระหว่างกระบวนการพอกพูนและละลายของสังกะสีได้อย่างมีเสถียรภาพสูงสุด พื้นผิวของขั้วแอโนดภายหลังการทดสอบยังคงความเรียบเนียน ไม่พบการเกิดเดนไดรต์ และแสดงร่องรอยการกัดกร่อนน้อยที่สุด นอกจากนี้ ยังให้ศักย์ไฟฟ้าสำหรับการเกิดปฏิกิริยาปลดปล่อยแก๊สไฮโดรเจนสูงที่สุด (-0.158 มิลลิโวลต์) และค่ากระแสต่ำที่สุดจากปฏิกิริยาดังกล่าว (-15 มิลลิแอมแปร์) สะท้อนถึงประสิทธิภาพในการยับยั้งปฏิกิริยาดังกล่าวได้อย่างมีประสิทธิภาพ อีกทั้งยังแสดงค่าการนำไฟฟ้าไอออนสูงถึง 0.966 มิลลิซีเมนส์ต่อเซนติเมตร สำหรับระบบแบตเตอรี่สังกะสี-แมงกานีสไดออกไซด์ พบว่าอิเล็กโทรไลต์กึ่งแข็งฐานพอลิอะคริลาไมด์ที่มีเจลาตินร้อยละ 5 ให้ค่ากระแสพีคจากการวัดไซคลิกโวลแทมเมทรีสูงที่สุด และมีพื้นที่ใต้กราฟมากที่สุด บ่งชี้ถึงกระบวนการสอดแทรกและละลายของไอออนสังกะสีที่เกิดขึ้นได้อย่างมีประสิทธิภาพ ส่งผลให้สามารถให้ค่าความจุจำเพาะสูงสุดที่ 242 มิลลิแอมแปร์ชั่วโมงต่อกรัม ที่กระแส 0.025 แอมแปร์ต่อกรัม และแสดงเสถียรภาพต่อการชาร์จ-คายประจุในระยะยาวได้อย่างน่าพึงพอใจ

This research aims to expand the application of DHP in biological systems by employing it as a biosensor for detecting caspases, a family of protease enzymes linked to diseases like cancer and neurodegenerative disorders. In this study, DHP derivatives—BZD, Fmoc-Asp-NH-DHP, and zWEHD-NH-DHP—were synthesized and tested for their potential as protease sensors. Initial screening of BZD and Fmoc-Asp-NH-DHP with common proteases, including papain and bromelain, revealed no enzymatic activity. To facilitate in vitro analysis, caspase-1 enzyme was successfully purified using an E. coli BL21(DE3)pLysS expression system. ปัจจุบัน การบริโภคเชื้อเพลิงฟอสซิลสำหรับเครื่องยนต์สันดาปภายในมีแนวโน้มลดลง เนื่องจากการเข้ามาแทนที่ของยานพาหนะไฟฟ้า ส่งผลให้เกิดอุปทานส่วนเกินของไบโอดีเซลในอนาคต เพื่อรักษาสมดุลของตลาดน้ำมันปาล์ม ไบโอดีเซลที่ผลิตจากน้ำมันปาล์มจึงถูกนำมาใช้เป็นวัตถุดิบในการผลิตเชื้อเพลิงชีวภาพอากาศยาน ตามนโยบายขององค์การการบินพลเรือนระหว่างประเทศที่สนับสนุนการใช้เชื้อเพลิงการบินที่ยั่งยืน ในงานวิจัยนี้จึงมุ่งเน้นไปที่กระบวนการไฮโดรคอนเวอร์ชันของเมทิลเอสเทอร์ของกรดไขมันที่ผลิตจากน้ำมันปาล์มไปเป็นเชื้อเพลิงชีวภาพอากาศยานสำหรับเครื่องบินโดยใช้เครื่องปฏิกรณ์แบบเบดคงที่ เนื่องจากข้อจำกัดของตัวเร่งปฏิกิริยานิกเกิล ซึ่งมีแนวโน้มเกิดการเกาะกลุ่ม การเกิดไฮโดรจิโนไลซิสอย่างรุนแรง และการสะสมโค้กระหว่างกระบวนการ งานวิจัยนี้จึงได้ศึกษาการเติมแกลเลียม ในตัวเร่งปฏิกิริยาโลหะคู่ บนตัวรองรับซีโอไลต์ชนิดเบตาที่ผ่านการปรับปรุงด้วยการชะด้วยกรด (beta-HF) โดยใช้สารละลายกรดไฮโดรฟลูออริก/แอมโมเนียมฟลูออไรด์ นอกจากนี้ ยังได้ศึกษาผลของการเติมซีเรียม เพื่อใช้เป็นตัวสนับสนุนลงในตัวเร่งปฏิกิริยา NiGa/beta-HF เพื่อเพิ่มสมรรถนะของตัวเร่งปฏิกิริยา โดยทำการศึกษาที่อุณหภูมิ 360 องศาเซลเซียส ภายใต้ความดันแก๊สไฮโดรเจน 30 บาร์ ค่า WHSV 2.45 ชั่วโมง-1 และใช้อัตราการไหลของไฮโดรเจน 60 มิลลิลิตร/นาที จากการศึกษาพบว่าตัวเร่งปฏิกิริยา 5Ni5Ga-5Ce/beta-HF ให้สมรรถนะการเร่งปฏิกิริยาดีที่สุด โดยให้การเลือกเกิดเชื้อเพลิงชีวภาพอากาศยานสูงถึง 41.6% มีอัตราส่วนไอโซ-/นอมัล-อัลเคนที่ 0.90 และปริมาณสารประกอบออกซิเจนต่ำสุดที่ 14.3% ส่งผลให้เชื้อเพลิงชีวภาพที่ได้มีค่าความร้อนสูงสุดที่ 37.4 เมกะจูล/กิโลกรัม ซึ่งสามารถนำไปผสมกับน้ำมันอากาศยานเจ็ท A-1 ในสัดส่วน 10/90% โดยปริมาตรได้ โดยไม่ทำให้คุณภาพของน้ำมันอากาศยานเจ็ท A-1 ลดลง งานวิจัยนี้จึงถือเป็นแนวคิดที่มีศักยภาพสำคัญในการผลิตเชื้อเพลิงการบินที่ยั่งยืนเพื่อลดการพึ่งพาน้ำมันอากาศยานเจ็ท A-1

บริษัท สยามมิชลิน จำกัด (โรงงานพระประแดง) จังหวัดสมุทรปราการ มุ่งมั่นในการจัดการทรัพยากรน้ำอย่างมีประสิทธิภาพและยั่งยืนในกระบวนการผลิตยางรถยนต์ การผลิตยางรถยนต์และยางรถจักรยานยนต์มีการใช้น้ำจำนวนมากในกระบวนการผลิต จึงเป็นเหตุให้เกิดน้ำเสียจำนวนมากตามมาเช่นกัน น้ำเสียเหล่านี้จะถูกส่งเข้าระบบบำบัดน้ำเสียแบบเลี้ยงตะกอนเร่งเพื่อบำบัดและมีสมบัติได้ตามมาตรฐานน้ำทิ้ง แนวทางในการลดการปล่อยน้ำเสียหลังการบำบัดแล้วสู่สาธารณะ คือ การนำน้ำเสียที่ผ่านการบำบัดเหล่านี้กลับมาใช้เพื่อสามารถหมุนเวียนทรัพยากรให้กลับมาใช้ใหม่อย่างมีประสิทธิภาพสูงสุด และยังสอดคล้องกับแผนการบริหารด้านความยั่งยืนในการจัดการสิ่งแวดล้อมของบริษัทอีกทางหนึ่งด้วย ในงานวิจัยนี้จึงศึกษาความเป็นไปได้ในการนำน้ำเสียที่ผ่านการบำบัดแล้วกลับมาใช้ใหม่ในกระบวนการอบยาง โดยศึกษาและปรับปรุงประสิทธิภาพอัตราการไฟล (%Recovery) ของระบบรีไซเคิลรีเวอร์สออสโมซิสแบบขั้นตอนเดียวด้วยสารกรองแก้ว (AFM®ng) พบว่ามีประสิทธิภาพเพิ่มขึ้นจากเป็นร้อยละ 64 จากนั้นศึกษาเปรียบเทียบความเป็นไปได้ของโครงการระหว่างระบบรีไซเคิลแบบสองขั้นตอน (Double-pass RO) มีร้อยละผลตอบแทนของโครงการ (%ROI) และระยะเวลาคืนทุน (Payback period) เท่ากับ ร้อยละ 242, 1.22 ปีและระบบรีไซเคิลแบบสองขั้นตอนร่วมกับระบบการกรองระดับอัลตร้า (UF-RO/RO) เท่ากับร้อยละ 189, 1.97 ปีตามลำดับ แสดงให้เห็นว่าระบบรีไซเคิลแบบสองขั้นตอน (Double-pass RO) มีผลตอบแทนด้านการลงทุน (ROI) ที่ดีที่สุด และระยเวลาการคืนทุนที่เร็วกว่า แม้ว่าระบบรีไซเคิลแบบสองขั้นตอนร่วมกับระบบการกรองระดับอัลตร้า (UF-RO/RO) จะมีคุณภาพน้ำดีกว่า แต่เมื่อพิจารณาต้นทุนแล้ว พบว่าไม่แตกต่างกันมากนัก เนื่องจากระบบรีไซเคิลแบบสองขั้นตอน (Double-pass RO)มีร้อยละผลตอบแทนการลงทุนที่ดีที่สุด และระยะเวลาคืนทุนเร็วกว่า บริษัทฯ จึงเลือกใช้ระบบนี้ในการผลิตน้ำป้อนเข้าสู่หม้อต้มไอน้ำ ส่งผลให้สัดส่วนการใช้น้ำประปาในการอบยางลดลงร้อยละ 26 สัดส่วนการใช้น้ำรีไซเคิลเพิ่มขึ้นถึงร้อยละ 42 และลดการปลดปล่อยก๊าซคาร์บอนไดดอกไซด์ได้ 111.63 ตันคาร์บอนไดออกไซด์ต่อปี

Hepatitis B, caused by the hepatitis B virus (HBV), is a significant global health concern, primarily affecting the liver and leading to severe complications, including drug resistance and chronic infection. Numerous studies have linked these conditions to the presence of hepatitis B core antigen (HBcAg) and hepatitis B e antigen (HBeAg). CdS quantum dots (CdS QDs), acting as photosensitizers, or sometimes solo photocatalysts in CO2 reduction, have offered a myriad of advantages including quantum confinement effect, tunable redox potential, and photoreduction selectivity. However, CdS QDs are frequently compromised by thermal and oxidative instability, as well as photocorrosion, which restricts their practical applications. In this research, an effective method endowing CdS QDs by assembling with amphiphilic metallopolymer was developed to enhance their stability and synergistic activity. The catalytic system generated in situ from the cationic Ru–H complex [(C6H6)(PCy3)(CO)RuH]+BF4– (1) with 2,3,4,5-tetrachloro-1,2-benzoquinone (L1) was found to be highly effective for promoting the deaminative coupling reaction of 2-aminoaryl aldehydes with branched amines to form 2-substituted quinoline products. The analogous deaminative coupling reaction of 2-aminoaryl ketones with branched amines led to the regioselective formation of 2,4-disubstituted quinoline products. A number of biologically active quinoline derivatives including graveolinine and a triplex DNA intercalator have been synthesized by using the catalytic method.

Lysine specific demethylase-1 (LSD1) serves as a regulator of transcription and represents a promising epigenetic target for anticancer treatment. LSD1 inhibitors are in clinical trials for the treatment of Ewing’s sarcoma (EWS), acute myeloid leukemia, and small cell lung cancer, and the development of robust inhibitors requires accurate methods for probing demethylation, potency, and selectivity. Here, the inhibition kinetics on the H3K4me2 peptide and nucleosome substrates was examined, comparing the rates of demethylation in the presence of reversible [CC-90011 (PD) and SP-2577 (SD)] and irreversible [ORY-1001 (ID) and tranylcypromine (TCP)] inhibitors.
