INTRODUCTION: Dysfunctional microRNAs and GABAergic interneurons are features of Alzheimer's disease (AD). The role of neuronal microRNA155 (miR155), elevated in both AD and Down syndrome (DS), remains unknown.
METHODS: We utilized in silico analyses of published databases, MIR155-deleted and -overexpressing human induced pluripotent stem cell (hiPSC)-derived cells, cortical organoids, and amyloid beta precursor protein (APP)/PS1-miR155 knockout mouse.
RESULTS: MIR155HG (miR155 host gene) colocalizes with APP in a neuron-specific, topologically associated domain (TAD) in chromosome 21. In human neural stem cells (NSCs), neurons, and cortical organoids, MIR155 deletion enhanced NSC proliferation and GABAergic interneuron generation. Over the past decade, the Model Organism Development and Evaluation for Late-Onset Alzheimer's Disease (MODEL-AD) consortium has transformed preclinical Alzheimer's disease (AD) research by addressing critical limitations in traditional mouse models that failed to translate to human disease. By leveraging human genetic discoveries, MODEL-AD has developed > 70 genetically informed mouse models, standardized phenotyping pipelines, and an open-access data infrastructure aligned with late-onset AD biology. These models incorporate human risk variants, environmental factors, and aging to better capture disease complexity, including emerging recognition of mixed pathologies such as vascular contributions, Lewy body disease, and TDP-43 proteinopathy. Microenvironment remodelling impacts tumour growth and metastasis, but whether remodelling promotes pre-malignant clonal fitness remains unknown. Here, using single-cell RNA-sequencing of the bone-marrow microenvironment in a mouse model of DNMT3A-mutant clonal haematopoiesis (CH), we identify mesenchymal stromal cells (MSCs) in a molecular state of cellular senescence. Elevated bone-marrow MSC senescence is also observed in humans with CH driven by several common somatic mutations. MSC senescence is induced by mutant haematopoietic cells in a contact-independent manner through production of soluble factors including TNF-α and IL-6. Vascular-related factors are now considered major contributors to most forms of dementia, including Alzheimer's disease. However, the degree to which vascular deficits contribute to risk, onset, and progression of cognitive impairment and dementia has only recently been appreciated. Our understanding of the mechanisms by which vascular deficits drive cognitive decline in dementia is still limited. Further, the testing of therapeutic approaches to prevent vascular deficits to treat dementia are few. These factors motivated the establishment of the Vascular Contributions to Cognitive Impairment and Dementia (VCID) Center Without Walls (CWOW) network, comprising institutes across the United States. This review shares the ongoing work of the global Worldwide Innovative Network (WIN) Consortium for Precision Medicine to synthesize emerging cancer treatment data and to define the requirements for a common global cancer database that can truly support precision oncology. We performed a narrative review of emerging cancer treatment data, molecular profiling technologies, and existing clinicogenomic databases, focusing on how tumors are characterized, how subgroups are defined, and how demographic, lifestyle, and environmental factors are captured. INTRODUCTION: Post mortem proteomic analysis of Alzheimer's disease (AD) brain tissue has identified novel target genes and proteins for potential therapeutic development. Moesin (MSN) and CD44 were identified as candidate targets. Using human induced pluripotent stem cells (hiPSCs)-derived neurons, we assayed how reducing gene expression of MSN and CD44 affected amyloid beta secretion, tau phosphorylation, and transcriptional state.
METHODS: Knockdown of MSN and CD44 in hiPSC-derived neurons was performed using short hairpin RNA (shRNA). Amyloid precursor protein processing and intracellular tau phosphorylation was measured using chemiluminescent ELISA assays. Global gene expression was analyzed by bulk RNA sequencing (RNA-seq).

The purpose of this phenomenological study was to explore educators’ lived experiences implementing schoolwide Positive Behavioral Interventions and Supports (PBIS) discipline systems and to understand how those experiences influenced school culture in private PK–12 schools. Historically, PBIS focused on specific outcomes, such as reductions in discipline referrals, suspensions, and behavioral incidents. Though these indicators are critical, they do not consider in what way educators experience disciplinary systems, nor how their lived experiences affect routine interactions with students. The purpose of this qualitative phenomenological study was to research secondary STEM educators’ perceptions of PD needs. This study focused on the lived experiences of educators actively teaching in STEM subject disciplines in grades nine through 12. The essential question for this research study was: How do Tennessee secondary STEM teachers perceive the relevance, applicability, and impact of PD opportunities? The research was rooted in Knowles’ (1984) theory of adult learning, Andragogy, and Learning Forward’s (2022) Standards for Professional Learning.

As plastic use shifts from non-degradable to biodegradable materials, the effects of biodegradable plastic fragments on crop physiology remain unclear. This study investigated how biodegradable plastic fragments influence maize development, transpiration, and defense signaling. Among five plastic types tested, poly(butylene adipate-co-terephthalate) (PBAT), polyhydroxyalkanoates (PHA), polycaprolactone (PCL), polylactic acid (PLA), and thermoplastic starch (TPS). PBAT and TPS showed the most significant effects and were selected for further oxylipin analysis. Results indicate that biodegradable plastic fragments can alter oxylipin signaling pathways in subtle but meaningful ways, suggesting that these materials are not inherently risk-free alternatives to conventional plastics.

Continuum robots are a class of continually deformable robots inspired by snakes and tentacles that are commonly used in medical, search and rescue, and exploration applications. The robots are highly nonlinear and difficult to accurately model and control. Precise control of the pose of a continuum robot is typically reliant upon a system model. The function of a material is defined not only by its chemical composition, but also by the structural organization of its constituents across multiple length scales, from the architecture of individual building blocks to their collective arrangement. Developing strategies to access, engineer, and reconfigure that structure on demand represents a central opportunity in the design of adaptive materials. Here, we present a thermoresponsive polymer-colloid platform that enables high-precision tuning of colloidal structures using only thermal stimuli. Integrated Silicon Photonics leverages light-speed operation with lower power and higher data rates than electronic counterparts. To achieve this, light is sent through a silicon circuit, where it is manipulated to perform various operations. Primarily, the light for these circuits comes from laser sources. Semiconductor lasers are typically used for their small form factor and ability to be heterogeneously integrated into larger, more complex systems. School-based interventions that support emotional regulation and student well-being are increasingly emphasized within Multi-Tiered Systems of Support (MTSS). Yoga has emerged as a potential school-based practice; however, its outcomes, alignment with MTSS frameworks, and implementation characteristics remain unclear. This systematized review examined (a) psychological, emotional, and behavioral outcomes associated with school-based yoga interventions, (b) their alignment with Tier 1 and Tier 2 supports, and (c) implementation characteristics that support feasibility in school settings. A systematic search identified 21 U.S.-based studies, which were analyzed using thematic analysis. Traumatic knee injury leads to posttraumatic osteoarthritis (PTOA) and significant skeletal muscle weakness, resulting in chronic disability. The current standard of care frequently fails to prevent musculoskeletal dysfunction, underscoring the need to identify therapeutic mechanisms of PTOA. Using an established preclinical anterior cruciate ligament (ACL) transection model of PTOA and leveraging an innovative SPiDER-senescence-associated β-galactosidase stain to discern senescent cells, we investigated cellular senescence at single-cell resolution and identified anti-inflammatory macrophages as a predominant contributor to the senescent cell burden in both muscle and knee joint after injury. Three new rare earth borates, Na2.62Ln2.12(BO3)3 (Ln = Pr, Nd, Sm), were synthesized via a high-temperature solution reaction using a BaCO3–H3BO3–NaF flux and structurally characterized by single-crystal X-ray diffraction analysis. Sodium barium lutetium borate NaBaLu(BO3)2 crystals were synthesized using a high-temperature solution method. The crystal structure of this layered orthoborate is described and corresponds to that of an extensive orthoborates family, NaBaR(BO3)2 (R = Sc, Y, Yb, Tb – Lu). NaBaLu(BO3)2 was doped by Eu3+ and Tb3+ to explore its performance as a host material and to investigate the luminescent properties of the Eu3+ and Tb3+ doped materials. Springsnails in the genus Pyrgulopsis in western North America have long been a dilemma for taxonomists. Defining species boundaries has been complicated by the presence of undiscovered or cryptic taxa, range over- or underestimation, species complexes representing an unknown number of species, and named species synonymous with other taxa. To address these issues, we conducted an array of molecular species delimitation analyses using sequences from up to 2 mitochondrial and 2 nuclear genes from over 5000 specimens representing 140 of the 151 currently recognized taxa. This study examined how maternal attachment style, parity, race, and motherhood expectations, individually and in interaction, predicted the quality of the mother-infant bond during the postpartum period. Grounded in a maternal feminist framework, the study used a quantitative, cross-sectional correlational design. Postpartum mothers aged 20–39 (N = 87) completed validated measures of maternal attachment style, motherhood expectations, mother-infant bonding, and depressive symptoms. Multiple regression compared a Primary Model containing the four focal predictors and planned interaction terms with a Secondary Model that added maternal depressive symptoms as a covariate. Melanoma is an aggressive form of skin cancer characterized by rapid progression, metastatic potential, and the frequent development of resistance to targeted therapies. Although the Inducible cAMP Early Repressor (ICER) has been identified as a potential tumor suppressor in melanoma, the molecular mechanisms underlying its role in tumor progression remain poorly understood. Building upon the transcriptomic findings of Wheelan et al. (2025), this study investigates the relationship between ICER and ten candidate genes (gdf6a, pdgfra, col4a4, fgfr2, pdgfrb, cdkn1a, fgf5, akt1, met, and notch3) associated with BMP signaling. One of the most robust amphibians is the American Bullfrog, Lithobates catesbeianus, which is a widespread species that is invasive worldwide, and is broadly tolerant of a range of environmental conditions. Despite this tolerance, survival and performance of adult L. catesbeianus can be negatively affected by increasing temperatures and salinity resulting from anthropogenic impacts, such as climate change and road salts. This study focuses on the effects of acute salinity exposure (mimicking road salt influx into a pond) on L. catesbeianus tadpoles at different larval stages. Improving water retention and other hydraulic properties is key to managing soil water and sustaining crop production under fluctuating and extreme weather events. We assessed how wood biochar (836 g C kg-1) applied at 0, 6.25, 12.5, 25, and 50 Mg ha-1 affected: i) soil hydraulic properties after 5 years and ii) corn and soybean yield, and biomass production of cereal rye cover crop (CC) and CC mix (cereal rye, radish, and winter pea) in sandy, sloping, and semi-arid soils in the central U.S. Great Plains over 6 years. This Improvement Science Dissertation in Practice concentrated on the persistent problem of freshman course failure at a large, diverse, Title I high school serving a military-connected community in Central Texas. Although intervention programs existed, an increasing number of ninth-grade students were consistently failing one or more courses during their first year of high school, which revealed several gaps in the campus’s system of support. The Improvement Science framework and two Plan-Do-Study-Act (PDSA) cycles were used to examine the factors contributing to freshman course failure and to assess the interventions implemented to improve academic success as students transitioned into high school. …

This study’s purpose was to explore the correlations between peak velocity in a rotational throw with a medicine ball, peak force (PF) and rate of force development (RFD) in an isometric mid-thigh pull, time to failure in a plank test, and peak velocity in a sword thrust task, which was also tested for reliability. Twenty-seven recreationally trained male participants between the ages of 18-35 completed the study. This study looks at whether using an end-gun on a center pivot irrigation system is financially worthwhile for Utah alfalfa producers. End-guns are used to water extra acreage beyond the end of a pivot, they can also apply water less evenly, which may reduce production on those outer acres. To examine this tradeoff, the study compares farm returns with and without an end-gun using historical Utah alfalfa price and yield data along with estimates of irrigation cost, added acreage, and possible yield loss. The results suggest that end-guns can improve profitability in many cases, mainly because they bring more acres …

This study examined teachers’, teacher assistants’, and classroom aides’ perceptions, knowledge, and implementation of the Orton–Gillingham approach to reading instruction. A mixed-methods design was employed, incorporating quantitative survey data (N = 37) and qualitative interview data (N = 9). Descriptive statistics indicated generally low levels of perception, knowledge, and implementation among participants. One-way analyses of variance (ANOVA) were conducted to examine differences based on instructional setting, years of experience, and level of Orton-Gillingham training. Results revealed statistically significant differences in knowledge and implementation by training level and years of experience, whereas no significant differences were found in perceptions across groups. …

The primary purpose of this study was to examine the well-being of California school board members. Using an adapted Workplace PERMA-Profiler, a quasi-experimental study was conducted with 144 school board members to investigate the study's research questions. Pearson correlation analyses and hierarchical multiple regression were used to examine the relationships between overall well-being and the Workplace PERMA-Profiler domains. Differences in overall well-being and PERMA domain scores across age groups and years of service were examined using one-way analyses of variance with Tukey HSD post hoc comparisons. Results indicated that each PERMA domain was positively associated with overall well-being, whereas loneliness …
