Xanthine Oxidase Catalyzed Oxidation Of Acetaldehyde Substrate And Carboxylate Inhibition, Patrick A. Goodman Jan 1973

Xanthine Oxidase Catalyzed Oxidation Of Acetaldehyde Substrate And Carboxylate Inhibition, Patrick A. Goodman

All Master's Theses

In recent years, interest in an enzyme which catalyzes the oxidation of a variety of aldehydes and purines has been growing steadily. In fact, this enzyme commonly known as xanthine oxidase, has been implicated by some (1) in the regulation of nucleic acid synthesis and cellular growth - an important consideration in the study of caner. Unfortunately, detailed investigations directly pertaining to biological aspects of this enzyme have as yet been somewhat limited. Nevertheless, the amount of information concerning the nature of the enzyme and its catalytic effects upon different classes of substrates, through far from complete, is indeed significant.

A Crystalline Fragment Of The Double Helix: The Structure Of The Dinucleoside Phosphate Guanylyl-3',5'-Cytidine, Roberta Ogilvie Day, Nadrian C. Seeman, John M. Rosenberg, Alexander Rich

Department of Chemistry: Faculty Publications

The sodium silt of guanylyl-3',5'-cytidine crystallizes in a monoclinic unit cell with one molecule in the asymmetric unit. Each molecule is related to another molecule by a 2-fold rotation axis which results in the formation of an antiparallel; right-handed .double helix with complementary hydrogen bonding between the guanine and cytosine residues. The crystal is heavily hydrated with 36 water molecules in the unit cell. The geometry of this crystalline double helix is. very similar to those which have been derived from studies of fiber x-ray diffraction patterns of double-stranded RNA; even though the latter do not yield data at atomic …

The Atomic Absorption Determination Of Ultra-Trace Tellurium In Rocks Utilizing High Sensitivity Sampling Systems, Richard D. Beaty Jan 1973

The Atomic Absorption Determination Of Ultra-Trace Tellurium In Rocks Utilizing High Sensitivity Sampling Systems, Richard D. Beaty

Doctoral Dissertations

"The sampling boat and the graphite furnace have been shown to possess the required sensitivity to detect tellurium at ultra-trace levels, in a variety of sample types, by atomic absorption. In the sampling boat approach, tellurium in sample solutions is chemically separated and concentrated by extraction into methyl isobutyl ketone before measurement. For samples exhibiting extraction interferences or excessively high background absorption, a preliminary separation of tellurium by coprecipitation with selenium is described. Using this technique, tellurium can be quantitatively detected down to 5 nanograms and linear response is observed to 100 nanograms. Relative standard deviations of better than 7% …

The Effect Of Organic Solvents On The Sensitivity Of Rare Earths Atomic Absorption, Keith A. Geiken Jan 1973

The Effect Of Organic Solvents On The Sensitivity Of Rare Earths Atomic Absorption, Keith A. Geiken

Dissertations and Theses @ UNI

Atomic absorption of the rare earths erbium, europium, and dysprosium was studied in 10% organic solvent-water systems, water miscible solvent-water systems, and in 100% solvent solutions of the metal acetylacetonates. Sensitivity was found to depend on the solvent system used and instrument parameters. All solvent systems used enhanced absorption, with (1) higher number of carbon atoms, (2) tertiary and secondary alcohols, and (3) high boiling point solvents producing the largest increase in absorption. No significant viscosity relationships existed in the 10% solvent-water systems for either miscible or partially soluble solvents; however, viscosity might have been a factor in absorption in …

Acidities In Ethanol/Water Solvents: The Pka Of Hf, Raymond M. Merrill Dec 1972

Acidities In Ethanol/Water Solvents: The Pka Of Hf, Raymond M. Merrill

Chemistry and Chemical Biology ETDs

The acidity of hydrofluoric acid in mixed ethanol/ water solvent systems was studied by determining the pKa of HF in a number of different ethanol concentrations. The effect of changing temperature on the acidity of HF in these media was also examined. The pKa's were determined using a lanthanum fluoride membrane Fluoride Specific Ion Electrode and a glass pH electrode versus the same reference electrode. The method eliminates the need to determine liquid junction potentials. By determining the pKa for HF in each ethanol/water mixture, at a number of different temperatures, thermodynamic parameters, such as free energy, enthalpy, entropy, and …

Synthesis Of Derivatives Of Pyrido [3,4-D]Pyridazines, Diane (D'Ottavio) Martin Jun 1972

Synthesis Of Derivatives Of Pyrido [3,4-D]Pyridazines, Diane (D'Ottavio) Martin

Chemistry and Chemical Biology ETDs

The current interest in the biological activity of certain 1 ,4-disubstituted pyrido[2 ,3-d]pyridazines prompted the investigation of various derivatives of the isomeric pyrido[3,4-d]pyridazine ring system. Thus, 1,4 dichloro[3,4-d]pyridazine was treated with thiourea and the intermediate thiouronium salt hydrolyzed to give pyrido[3, 4-d]pyridazine-1,4(2H,3H)-dithione which was subsequently alkylated to give 1,4-bis(alkylthio)pyrido[3,4-d]pyridazines. On the other hand, 1,4-bis(arylthio)pyrido[3,4-d]pyridazines were prepared by the reaction of 1,4-dichloropyrido[3,4-d]pyridazine with various substituted thiophenols. Furthermore, treatment of 1,4-dichloropyrido[3,4-d]pyridazine with a variety of substituted anilines gave the corresponding 1,4-disubstituted aniline derivatives, whereas reaction with alkylamines afforded only the 1(4)-chloro-4(1)-alkylaminopyrido[3,4-d]pyridazines. When 1-chloropyrido[3,4-d]pyridazine-4(3H)-one or 4-chloropyrido[3,4-d]pyridazine-1(2H)-one was treated with hydrazine, the corresponding …

Enzymatic Synthesis Of Oligoribonucleotides Of Defined Base Sequence, Mark J. Rowe May 1972

Enzymatic Synthesis Of Oligoribonucleotides Of Defined Base Sequence, Mark J. Rowe

Theses and Dissertations

The synthetic reaction of ribonuclease T_1 has been studied as to its potential for the synthesis of oligoribonucleotides of defined base sequence. Conditions have been determined for maximum synthesis with respect to percentage incorporation of both guanosine 2',3'-cyclic phosphate (donor), and cytidine (acceptor). With regard to maximum acceptor incorporation, a donor blocked at the 5'-hydroxyl has been used to drive the reaction. It has also been shown that acceptor incorporation decreases as the chain-length increases up to the hexanucleotide. Synthesis of a 3'-blocked substrate for polynucleotide phosphorylase for use in chain termination studies was attempted using adenylate kinase. In this …

Synthesis And Metal Coordination Properties Of Some Cyclic Polyethers And Polythioethers, Barry Lant Haymore May 1972

Synthesis And Metal Coordination Properties Of Some Cyclic Polyethers And Polythioethers, Barry Lant Haymore

Theses and Dissertations

Thermodynamic values for cation-cyclic polyether interactions for several systems are reported and discussed in light of ligand geometry, cation selectivity, solvent effects, structure, stoichiometry and the nature of the coordinate bond. Cyclic polyethers are shown to be remarkably good ligands even in water and to have marked selectivity for d0, d10 and d10 s2 cations. Syntheses of one new cyclic polyether and several new mixed cyclic polyether-thioethers are reported. Cyclic chelates with smaller rings are shown to prefer smaller cations, but this selectivity is masked by the hydration energy of the cation. Except for thiophillic cations such as Hg2+ and …

Potentiometric and conductometric measurements of additions of acids and bases to purified sulfolane and of various acid-base titrations were made. The series of carboxylic acids included benzoic, orthomethoxybenzoic, and three different monochlorobenzoic acids. The bases investigated included sodium methoxide, pyridine, and diphenylguanidine. Interpretation of …

Continuous Wave Co2 Laser Degradation And Mass Spectrometric Analysis Of Organic Polymers, Stanley G. Coloff Mar 1972

Continuous Wave Co2 Laser Degradation And Mass Spectrometric Analysis Of Organic Polymers, Stanley G. Coloff

Chemistry and Chemical Biology ETDs

Previous laser-mass spectrometer and laser-gas-chromatographic instruments for study of organic polymer degradation have used high power pulsed lasers (ruby, Nd-glass) which limit sample characterization due to extensive fragmentation. A new experimental technique has been developed for high temperature pyrolysis and rapid analysis of organic polymers. The experimental technique combines a low power continuous wave CO2 laser for sample pyrolysis and a time-of-flight mass spectrometer to analyze degradation products. The laser built for use in this study was a continuous flow CO2 gas-molecular laser excited by high voltage direct current with maximum power of 10 watts at a wavelength …

On The Mechanism Of Low-Temperature Termolecular Atomic Recombination, Russell T. Pack, Richard L. Snow, Wesley D. Smith Jan 1972

On The Mechanism Of Low-Temperature Termolecular Atomic Recombination, Russell T. Pack, Richard L. Snow, Wesley D. Smith

Faculty Publications

A simple theory of gas-phase termolecular atomic recombination kinetics is presented which treats the bound-complex (BC) mechanism in a fashion which parallels the Roberts, Bernstein, and Curtiss resonance theory of the energy-transfer (ET) mechanism. The ET rate constant has a low temperature maximum, but the BC rate constant does not. The contributions of the two mechanisms to the low-temperature recombination of hydrogen atoms, with He, Ar, and H2 as third bodies, are estimated and compared. The presence (absence) of a low-temperature maximum in the observable total rate constant is determined by absence (presence) of bound vibrational levels of the hydrogen-third …

The Hemoglobin Of Tubifex Tubifex, Lalitchandra Vrajlal Vora Jan 1972

The Hemoglobin Of Tubifex Tubifex, Lalitchandra Vrajlal Vora

Masters Theses

The hemoglobin of Tubifex tubifex is found in solution in the blood of the worm in cells as in vertebrates. The binding of oxygen to the hemoglobin can be followed spectrophotometrically. A peak of absorbtion at 540 nm is fairly constant under different condition, while a peak at 575 nm disappears from the spectrum when oxygen is removed. The hemoglobin contains heme, non-heme iron and protein.

Binding of oxygen to the hemoglobin was found to be dependent on the concentration of the hemoglobin, at low concentrations (4.16 x 10-6 M), oxygen could not be removed from the hemoglobin. Only …

The Conductance Of Some Non-Rigid Bolaform Electrolytes, William Terry Schmidt Jan 1972

The Conductance Of Some Non-Rigid Bolaform Electrolytes, William Terry Schmidt

Masters Theses

Bolaform electrolytes are those which contain two or more charges separated by an inert framework. These are intermediate in structure between simple electrolytes and polyelectrolytes. Since these are similar to both synthetic and biological electrolytes, this investigation has been undertaken to determine some of their properties in solution.

The sodium and magnesium salts of ethane, propane, butane, pentane, hexane and decane α,ω-disulfonates were prepared. Each acid was prepared by refluxing the α,ω-dibromide of the desired salt with sodium sulfite. The sodium and magnesium salts were prepared by ion-exchange techniques.

The conductance of each salt was determined in methanol utilizing the …

Spectrophotometric And Microbiological Determination Of The Stability Of Sodium Ampicillin As Affected By Ph, Enrico Frank Raffanti

University of the Pacific Theses and Dissertations

The specific purpose of this investigation was to determine the effect of pH on the stability of ampicillin sodium for injection. Then, to correlate this information, a pH profile may be established to predict chemical stability with ampicillin sodium alone or when other drugs may be included in the admixture. With a pH profile the pharmacist could predict the length of time after which more than a 10 percent loss of activity would occur. Stability measurements were to be accomplished through a microbiological assay after adding ampicillin sodium to different buffer solutions. At the end of four hours, eight hours, …

The Synthesis Of Some New Condensed Pyridazines, Pyridazine N-Oxides And Pyrimidines As Potential Chemotherapeutic Agents, Donald E. Pichler

Chemistry and Chemical Biology ETDs

The need for new antimicrobial and antifungal agents as well as medicinals possessing antiprotozoal activity against special strains of malaria resistant to most synthetic antimalarials prompted the synthesis of new compounds that may have the desired antimetabolite properties. The structural similarities of the 1,2,5-thiadiazolo[3,4-d] pyridazines and the v-triazolo[4,5-d]pyridazines with the biologically important purines prompted the synthesis of members of these ring systems. The 1,2,5-thiadiazoles synthesized as intermediates ware also screened as potential chemotherapeutic agents. Similarly, the pyridazine N-oxides and the 5-nitro substituted pyrimidines are structurally similar to the pyrazine N-oxides and the pyridine N-oxides, which are known to possess a …
