High Pressure Liquid Chromatographic Methods For The Separations Of Derivatized Alpha-Keto Acids And Thiamine Phosphate Esters, Bruce Clark Hemming

Improved methods have been developed for separation of a-keto acid derivatives and for thiamine and its phosphate esters. Linear gradient reverse phase chromatography using ion-pair formation provides baseline resolution for a seven-component homologous series of a-keto acid dinitrophenylhydrazones having increasing carbon chain lengths. Branched-chain keto acids can also be separated. Aldehyde derivatives were examined as possible interfering compounds. Peak identification in biological material was confirmed for pyruvate by an enzymic peak shift technique. Monitoring near 365 nm permits low nanogram detection. Preliminary studies of keto acids in biological material illustrate the method's applicability. The same system with slight changes separates …

It was desired to help establish whether or not the tertiary structure of a biomolecule is the same in a cry­stal (or an oriented film) as it is in solution. Generally it is accepted that the structures in these diverse environ­ments are similar, if not identical. By comparing the extinction coefficients obtained from oriented film data to those obtained from solution data it was hoped that this question might further be resolved. Upconversion materials have been prepared which may be stimulated by incident 970 nm radiation to emit in the visible region. The couples Yb+3-Ho+3 in a YF3 host and Yb+3-Er+3 in YF3, YPO4, Li3PO4 and Y2O2S hosts have been studied at temperatures from 9K to 273K. For (Y.80Yb.19Er.01)F3 emission bands at 657 nm, 538 nm, 520 nm, 409 nm and 379 nm have been detected and determined to result from 3-, 2-, 2-, 3-, and 3-photon …

Conversion Of Farnesyl Pyrophosphate And Squalene To Sterols By Rat Liver Microsomal Enzymes: Specificity Of Sterol Carrier Protein₁, Kathleen Lobaugh Gavey

The conversion of farnesyl pyrophosphate to squalene by rat liver microsomes was investigated. The enzyme exhibited an apparent maximum velocity at a substrate concentration of 30 μM. No evidence of substrate inhibition was seen at concentrations up to 65 μM. Phosphatases present in the microsomes or in the 105,000 x g supernatant (S105) did not interfere with squalene synthesis under the experimental conditions used.

s105 slightly enhanced the biosynthesis of squalene at the lowest concentrations of microsomal protein studied. The effect was reversed at higher concentrations of microsomes. A 1O-fold increase in the concentration of s105 did not …

The zero‐frequency shear wave dip appearing in the depolarized Rayleigh spectrum in benzonitrile has been studied as a function of concentration and temperature. The solution study was carried out at constant viscosity equal to the viscosity of liquid benzonitrile at each temperature. The result indicates that the presence of shear wave fine structure does not depend on the collective orientational fluctuations. The orientational and vibrational relaxation times of benzonitrile were measured at various concentrations and temperatures. The orientational relaxation times show no concentration dependence at any temperature, suggesting that the pair correlation is negligible at all concentrations. The orientational relaxation …

The rotational and vibrational relaxation times of acetonitrile–carbon tetrachloride solutions were investigated as a function of concentration, viscosity, and temperature using depolarized Rayleigh and Raman scattering. Using a Fabry‐Perot interferometer and single frequency laser source, we have shown that reliable results for the single particle orientational correlation times (τs) for CH3CN can be obtained by carrying out a concentration dependent depolarized Rayleigh scattering study. Raman scattering was shown to yield inconsistent results for τs in CH3CN. At constant viscosity, it was found that the Rayleigh scatteringrelaxation time (τRay) of CH3 …

The photolyses of a series of chromium(III) complexes Cr(en)3 3+, Cr(den)2 3+, Cr(tn)3 3+, and Cr(pn)3 3+, where en= ethylenediamine, den= diethylenetriamine, tn = trimethylenediamine, and pn = propylenediamine, have been investigated. Experiments were carried out on unbuffered and buffered (pH= 2.00) solutions containing 10^-3 – 10^-2 M chromium(III) complexes at room temperature and 10°C. While purging with argon or oxygen, the irradiation was performed with 184.9-253.7 nm u.v. light, corresponding to the charge transfer bands of the complex.

The binuclear tartrate-bridged complexes [Cr2 (tartH)2 (L)2] and Na[Cr2 (tart) (tartH) (L)2]("tart" = C4H2O64-, "tartH" = C4H3O63- and "L"= 1,10-phenanthroline or 2,2'-bipyridyl) have been synthesized where each complex contains either two optically active or two meso bridging ligands. A discussion of the number of isomers and symmetries of these complexes is presented, as well as a complete description of the syntheses of the complexes and their sodium salts. Diethylaminoethyl cellulose was titrated with acid and base in various concentrations of NaCl. The pK values were determined from titration curves using a graphical technique. Two titratable groups were observed from the acid titration curve, a major and a minor group. The minor group, which represented 20% of the acid titration, was barely detectable in the base titration curve and had a salt-independent pK of 6.0. The pK of the major group was strongly salt-dependent and was approximately a linear function of log N NaCl. An equilibrium shift caused by the counter ion may explain the salt dependence. Intermolecular Potential Surfaces From Electron Gas Methods. Ii. Angle And Distance Dependence Of The A’ And A” Ar–No(X^2ii) Interactions, Glen C. Nielson, Gregory A. Parker, Russell T. Pack

Angle dependent intermolecular potential energy surfaces for the two states (2A’ and 2A”) that arise from the interaction of ground (X^2II) state NO with Ar are calculated using the electron gas model to obtain the short range interactions. The average and difference of the two interaction energies are fit to analytic forms convenient for use in scattering calculations and joined smoothly onto the long range van der Waals potential previously determined. The results, which appear to be of useful accuracy, and the applicability of the electron gas model to such open shell–closed shell interactions are discussed.

1. The waters of Las Vegas Bay, a heavily utilized recreational resource, receive discharges from a variety of municipal and industrial waste sources. The U.S. Environmental Protection Agency has determined, on the basis of numerous studies, that the present water quality violates state and federal standards and constitutes a public nuisance. Consultants have advised the Sewage and Wastewater Advisory Committee that rapid abatement of the alleged pollution conditions can be achieved by an advanced wastewater treatment (AWT) plant.

The circulating half-lives of the four isozymes of bovine pancreatic ribonuclease (RNases A, B, C, and D) have been determined in normal and in nephrectomized rats. The isozymes differ only in their glycosyl content. While A contains no sugars, B has a simple oligosaccharide (GlcNAc, Man,+), and C and D each have a complex oligosaccharide (GlcNAc, Man,., Gal, Fuc NeuAc%, and GlcNAc, Mans Gal, Fuc NeuAc,, respectively) attached to Asn-34 of the polypeptide chain. A reaction involving iron as an intermediary electron carrier for the coupling of electron transfer from an endogenous substrate to ferroxidase-I has been proposed.

The enzymic nature of the reacti on has been established and studies with iron chelating agents have demonstrated the involvement of Fe(III). The kinetic parameters of ferroxidase-11 with Fe (lll) were found to be quite similar to those previously established for Fe(Il). The participation by the copper associated with ferroxi dase-11 was ascertained by use of the copper chelating agent, diethyl -di thio-carbamate .

Two free radicals formed when single crystals of potassium hydroxylamine disulfonate are irradiated with gamma rays at 77degrees K were studied by ESR. The ESR spectra result from a predominant center (radical A) exhibiting large 14N and 1H hyperfine splittings and from lesser amounts of Fremy's radical [O-dotN(SO3)2-2]. The g and hyperfine tensors of these radicals have been evaluated and are discussed. Radical A, whose identity remains in doubt, may be regarded as the precursor to Fremy's radical; on warming its lines decay and those of Fremy's radical are formed. Addition reactions of pentafluorosulfur bromide with unsaturated substrates were studied and four new compounds containing the pentafluorosulfur group have been prepared.

The mechanism for these reactions seems to involve a radical addition pathway. In reaction with CH2=CFC1, CF3C≡CH and CH3C≡CH the SF5 radical attacks the carbon atoms carrying the most hydrogens. The reaction with CF2=CHCl is interesting, as by analogy with other fluoroolefins, the SF5 primary attack should occur.at the CHCl group, however, SF5CF2CHC1Br was found as the only product for this reaction. The synthesis of analogs of toxoflavin: 1,3,4,6-tetramethyl-1,5,6, 7-tetrahydropyrimido[4,5-c]-5,7-pyridazinedione; 1,6-dimethyl-3,4-diphenyl-1,5,6,7-tetrahydropyrimido[4,5-c]-5,7-pyridazinedione; 3,5,7-trimethyl-8-phenyl-2,3,4,8-tetrahydropyrido[2,3-d]-2,4-pyrimidinedione are discussed. The synthesis of analogs of fervenulin: 3,4,6-trimethyl-5,6,7,8-tetrahydropyrimido[4,5-c]-5,7-pyridazinedione; 6-methyl-3,4-diphenyl-5,6,7,8-tetrahydropyrimido[4,5-c]-5,7-pyridazinedione; 2-hydroxy-3,5,7-trimethyl-4(3H)pyrido[2,3-d]-2,4-pyrimidinone are discussed. Several intermediates have been prepared: 6 -methylamino-3-methyluracil by a new method; 6-n-propylamino-3-methyluracil: 6-n-butylamino-3-methyluracil and 3-ethyl-2-ethylthio-2-hydroxy-4(3H)pyrimidinone.

When an N-ethoxycarbonylthioamide is treated with dinucleophilic reagents containing a primary or secondary amino group and another nucleophilic group, reaction occurs initially between the amino group of the reagent and the thiocarbonyl of the substrate with elimination of hydrogen sulfide and formation of a carbon, nitrogen double bond. The intermediate thus formed may subsequently cyclize in either of two different manners depending upon the relative positions of the two nucleophilic groups in the reagent. A. Formulas are derived for the van der Waals Cn coefficients for the interaction of a diatomic molecule in a II electronic state with an S-state atom. Two triatomic states arise from the degenerate IIstate. The average of the two energies has the usual Legendre polynomial (PL) angular dependence, but the difference in energies of the two states is shown to have associated Legendre polynomial (PML with M=2) angular dependence. Procedures for including spin orbit coupling are included, and the extension to interactions of Delta- and Phi-state molecules is discussed. Pade approximant methods and experimental frequency dependent polarizabilities are used to calculate reliable van der Waals C6 coefficients for the interaction of CO with He, Ne, Ar, Kr, Xe, H, Li, Na, K, Rb, Cs, H2, N2, O2, CO2, and CO. Error bounds are given for the C6 coefficients and for the CO oscillator strength sums.

An experimental testing program is described which utilizes questions that are partly computer composed, in addition to a section composed by the instructor, and a retesting option to the student. Results from a trial of the program for one term indicate that (1) course grades were improved, (2) the student withdrawal failure rate was unaffected, and (3) the employed students took greater advantage of the retest than did the unemployed students.

A new scheme is presented for the preparation of 3~(4-chlorophenyl)-5,7-dichloro-l-methylnaphthalene, from which, four derivatives, 3~(4-chlorophenyl)-~-di(n-butyl)aminomethyl~5,7-dichloro-l-naphthalene-methanol hydrochloride, 3-(4-chlorophenyl)-5,7-dichloro-~-(2-piperidyl)-1-naphthalenemethanol acetate, l-[3-(4-chlorophenyl)-5,77dichloro-lnaphthyl]-3-di(n-butyl)aminopropanol hydrochloride, and 1-[3-(4-chlorophenyl)-5, 7-dichloro-l-naph,thyl]-2-(2-piperidyl) ethanol hydrochloride, were prepared. These four derivatives were submitted to Walter Reed Army Institute of Research for. the determination of their potential as antimalarial agents.
