A variety of substituted phosphonium salts, phosphorus ylides and phosphonates were studied to assess their utility in ring expansion reactions. The strategy involved formation of fused ring systems whose ring strain could be exploited to accomplish ring expansion to medium sized rings. β-Alkoxy vinylphosphonium salts were found to be unreactive with species such as the dimethylsulfoxonium methylide anion, Simmons-Smith type reagents. and carbenes in attempts to produce cyclopropyl phosphonium salts.
C-Alkylation of β-keto phosphorus ylides using haloalkanes bearing terminal groups that could be converted and reacted with the B-carbonyl, was also examined as a method for accessing strained rings bearing …

Large basis set ab initio calculations have been carried out for a dense grid of points on the He, potential energy surface. Three-body contributions were extracted at every point, and a number of concise functional representations for the three-body potential surface were then examined. Three-body multipolar dispersion terms and other radial and angular terms were used in the representations, and an assessment of relative importance of the different terms is presented. Combined with a two-body He-He potential, the results of this work should offer a high quality interaction potential for simulations of aggregated helium.

A novel PCR technique used to amplify Euplotes crassus macronuclear chromosomes was developed to provide scientific proof that an Euplotes crassus 1.0 kb gene is not genetically related to an Oxytricha nova 1.8 kb β-telomere protein gene. Theories and experimental procedures associated with the development of this PCR technique were the product of research that will have a profound impact on future studies of telomere protein genes and telomeric DNA in a multitude of eukaryotic cells. The hypothesis that a β-telomere protein does not exist in Euplotes crassus was supported by the result of this study.

Semiclassical Quantization Of A Nonintegrable System: Pushing The Fourier Method Into The Chaotic Regime, Karl Sohlberg, Randall B. Shirts

Semiclassical Einstein–Brillouin–Keller (EBK) quantization of the nonintegrable Hénon–Heiles Hamiltonian succeeds using the Fourier transform method of Martens and Ezra. Two innovations are required for this success: (1) the use of tunneling corrected quantizing actions obtained from an approximate, one-dimensional Hamiltonian and (2) exploitation of intermediate-time approximate quasiperiodicity or "vague tori'' wherein the Fourier transform of chaotic motion over 10–100 vibrational periods allows the determination of frequencies and amplitudes which approximate motion during the time interval. Approximate tori, actions, and EBK energy levels are then straightforward. We use an interpolation method to smooth over small resonance zones that are not expected …

The potential dependence in the surface second harmonic response from hydrogen terminated n‐Si(111) and oxidized n‐Si(111) surfaces has been examined in aqueous NH4F and H2SO4 solutions. The relative phase of the nonlinear response as measured by rotational anisotropy experiments is found to be highly sensitive to the presence of the oxide and the field applied across the Si(111)/oxide/electrolyte interface. These observations are attributed to field effects within the space–charge region of the semiconductor which vary with the presence and thickness of the insulating oxide layer on the Si(111) surface.

Planar-supported phopholipid bilayers formed by te adsorpton of vesicles are increasingly used in the investigation of lipK-ependent reactis. We have studied the way in which these bilayers are forned with phopholipid vesicles coaining the btranembrane protein Tssue Factor (TF). TF complexed with te senne protease, factor Vlla, is the primary initiator of bklod coagulation by way of activation of the zymogen factor X. TF has been shown to orient randomly on the inner and outer leaflets of vesicles. This research developed a methodology for assessment of the exposure of a mature corn crop to trichloroethylene from a contaminated aquifer. The methodology was then applied to the case of Hill AFB to determine the ability of the methodology to provide information about a specific exposure. Current procedures sample for food contamination but do not attempt to predict exposure problems. A review of the potential exposure pathways from the aquifer to the crop was conducted. Based on this review, the exposures due to soil gas and irrigation were modeled. Empirical estimated were used to approximate the expected flux of soil …

The deletion of federally mandated fecal coliform limits has led many states to review and modify their wastewater disinfection requirements. One issue in analyzing wastewater disinfection is the discharge of potentially carcinogenic halogenated organics formed during the chlorination process. This research investigates the formation of one class of the halogenated organics, the trihalomethanes. The applicability of using drinking water trihalomethane formation models for use with wastewater effluent is examined. Three models are compared for predictive capability by using measured trihalomethane values from previous research data. Reaction of chromium(VI) with one equivalent of ascorbate was studied by electron paramagnetic resonance spectroscopy in the presence of 0.10 M 5,5-dimethyl-1-pyrroline-1-oxide (DMPO) at room temperature in 0.10 M (N-[2-hydroxyethyl]piperazine-N'-[2-ethanesulfonic acid]) (HEPES) and 0.05 M tris(hydroxymethyl)aminomethane hydrochloride (Tris-HCl) buffers (pH 7.0 room temperature). Chromium(V), ascorbyl radical, and carbon-based DMPO-radical adducts were observed. A higher level of Cr(V) was observed in HEPES buffer and a higher level of the DMPO-radical adducts was observed in Tris-HCl buffer. Chromium-DNA binding studies were carried out in vitro for calf thymus DNA incubated with Cr(VI) and ascorbate in both buffers at 37 degrees C. Higher …

Several common elementary methods of computing molecular properties, prerequisite to molecular dynamics studies, are tested for their validity for CS+2. Judged sufficiently accurate, these methods are then used to aid in investigating the collisionally activated dissociation of CS+2 upon impact with xenon. Rice–Ramsperger–Kassel–Marcus (RRKM) unimolecular decay rate calculations are presented and compared to experimental studies employing collisional activation. RRKM theory is shown to reproduce the experimental results for collision energies near threshold. When corrected for vibrational anharmonicity, the RRKM calculation shows agreement with the experimental results over a slightly wider range of energies. A discussion is given on the applicability …

Antibody 43C9 accelerates the hydrolysis of a p-nitroanilide by a factor of 2.5 x 10(5) over the background rate in addition to catalyzing the hydrolysis of a series of aromatic esters. Since this represents one of the largest rate accelerations achieved with an antibody, we have undertaken a series of studies aimed at uncovering the catalytic mechanism of 43C9. The immunogen, a phosphonamidate, was designed to mimic the geometric and electronic characteristics of the tetrahedral intermediate that forms upon nucleophilic attack by hydroxide on the amide substrate. Further studies, however, revealed that the catalytic mechanism is more complex and involves …

This research expands our limited knowledge on the influence of plants on the fate and effects of trichloroethylene (TCE), providing a screening tool on which to base decisions regarding the need for actual sampling of plants. First, physicochemical properties of TCE--vapor pressure, Henry's Law constant, water solubility, octanol-water partition coefficient, molecular weight, and half-life--were screened against relationships reported in literature to evaluate TCE plant uptake potential. This screening approach indicated TCE may be transferred to plants via retention by root surfaces, root uptake and translocation, and foliar uptake. Currently, amylase determinations are nonspecific for the organ source and are based entirely on the enzymatic properties of amylase to produce a measurable product or byproduct. The determination of pancreatic amylase is important in the diagnosis of acute pancreatitis. Most commercially available tests for amylase employ the measurement of the change in NADH absorbance at 280 nm or of the p-nitrophenol released from a maltotetrose substrate. These are nonspecific measurements of pancreatic amylase and often necessitate other tests to be run such as a serum lipase.

Isothermal titration calorimetry (ITC) is used to study the thermodynamics of assembly of the three DNA oligonucleotides S1 (5'GCCTGCCACCGC), S2 (5'-GCGGTGCGTCCG), and S3AA (5'-CGGACGAAGCAGGC) to form a three-way junction (TWJ) complex consisting of three double-helical arms radiating from a junction region having two unpaired adenosines in one strand (S3AA). The thermodynamics of assembly were measured for three different orders of addition of the component oligonucleotides at four temperatures between 10 and 25 degrees C. The geochemical evolution of Low Molecular Weight Fatty Acids (LMWFA's) in various ground water samples and aquifer solid samples from both contaminated and uncontaminated sites has been studied. An improved analytical method has been established for both qualitative and quantitative determination of LMWFA's. The low molecular weight (LMW) organic acids are metabolic intermediates of the oxidative biodegradation of hydrocarbon contaminants. The hydrocarbons are believed to be the electron donors in these redox reactions. Formic and acetic acids were found to be the major LMWFA's in these ground water samples and aquifer solid samples. Stereolithography, a process by which a computer-guided Ultraviolet (UV) laser is applied onto a liquid monomer resin to form a solid polymer, was used to create solid models of amiloride and two of its analogues. The Cartesian coordinates used as input for the orientation of each atom in a molecule were calculated using ab initio molecular orbital theory. Since the structures of the molecules are calculated in this fashion, they represent a more accurate representation of the molecules than that provided by standard molecular modeling kits.

Dapsone is a drug which has recently been discovered as a potential treatment for the disease of leprosy. Many of the derivatives of dapsone have been sent to the United States by Dr. M. Fibrous carcinogens, such as crocidolite asbestos, are known to catalyze many of the same reactions as iron, namely O2 consumption, generation of reduced oxygen species, and damage to DNA, such as strand breaks, and modifications of bases. Upon inhalation, fibers are also known to become coated with an iron-rich material. The mechanism by which this iron is bound to fibers in the lung is not known, and the effect of this additional iron on the reactivity of the fibers is also not well understood. Inotropic support for the failing myocardium as the therapy for congestive heart failure (CHF) is intended to achieve an increase in cardiac output via positive responses in myocardial contractility and vasodilation. A novel approach to differentiate these two responses is the use of an animal with an implanted total artificial heart (TAH). Three inotropic drugs, dobutamine, enoximone, and pimobendan, were tested in eight animals with their natural heart intact and five animals implanted with a TAH. Baseline values of the TAH and natural heart (NH) were compared to determine their hemodynamic similarities. Each of the three drugs was given randomly …

Conventional metal film sensors can measure flow instabilities of the order of 60 kHz. The turbulence fluctuations in the hypersonic regime are of the order of 200- 400 kHz. Haemophilus ducreyi, a cause of genital ulcer disease in developing countries, appears to facilitate the heterosexual transmission of the human immunodeficiency virus in Africa. Despite an increase in studies of this gram-negative human pathogen, little is known about the pathogenesis of chancroid. Our studies have shown that the lipooligosaccharides (LOS) of H. ducreyi may play an important role in ulcer formation. Monoclonal antibody and mass spectrometric analyses identified a terminal trisaccharide present on H. ducreyi LOS that is immunochemically similar to human paragloboside. This epitope is present on the LOS of Neisseria gonorrhoeae, and it may be the site of …

The resonance Raman spectrum of the 1,3,5-hexatriene radical cation generated by v-irradiation in a Freon glass is reported. The spectrum is excited at 395 nm in resonance with the second absorption band. Identical spectra are obtained from ionized (E)- and (Z)-1,3,5-hexatriene. The presence of more than one rotamer has to be assumed to account for all the observed resonance Raman bands. The bands are assigned to a mixture of the two rotamers, calculated at lowest energy, the ttt an tct rotamers, on the basis of ab initio ROHF/6-31G calculated harmonic frequencies.
