Diepoxybutane, diepoxyoctane, and mechlorethamine are cytotoxic DNA crosslinking agents that vary in their carcinogenic versus chemotherapeutic potentials. Interstrand cross-linking occurs between the N7 positions of deoxyguanosine residues on opposite strands of the DNA duplex. Each synthetic DNA oligomer used in this study contained four 5'-N\GN2CN3 sites (within a 32 base sequence) and we have systematically varied the bases in the N1 and N2 positions to determine the resulting cross-linking efficiencies of each cytotoxic agent. Each duplex was 5' -end labeled and incubated with cross-linking agent. Interstrand cross-links were purified through denaturing polyacrylamide gel electrophoresis and then subjected to piperidine cleavage. …

The impact of phycotoxins and mycotoxins on human and animal health are of important concern for worldwide authorities and the scientific community. Liquid chromatography-mass spectrometry (LC-MS) techniques have been developed, optimised and validated for the analysis of these toxins.

Phycotoxins are potentially toxic substances, which are produced by marine phytoplankton and which may accumulate in shellfish or finfish. Azaspiracid poisoning (AZP) is a recently discovered syndrome that has been implicated in several incidents of human intoxications. This thesis reports the first complete synthesis of monriporynes A and B. isomeric diacetylenes that bear α, β unsaturated ketone functionalities. Recently isolated from the stony coral Montipora sp. these compounds are reported to possess in vitro cytotoxic activity against several human solid tumor cells. We have synthesized both isomers in three simple steps. In the first step, l-iodononyne was prepared by reacting 1-nonyne with N-iodosuccinimide and a catalytic amount of silver nitrate. This iodoalkyne was then coupled with propargyl alcohol using copper iodide as the catalyst to give 2,4dodecadiynyl alcohol. Finally, the diacetylenic alcohol was subjected to the Swern …

In industrial polymer and synthetic rubber production facilities, workers are exposed to 1,3-butadiene. This compound is converted in vivo to 1,2,3,4-diepoxybutane (DEB) and has been linked to increased incidences of cancer in these individuals. Carcinogenesis has been attributed to formation of DEB induced DNA interstrand cross-links. Previous studies have demonstrated that DEB cross-links deoxyguanosine residues within 5'-GNC sequences in synthetic DNA, in restriction fragments, and in defined sequence nucleosomes. The current study utilized the polymerase chain reaction (PCR) to examine DEB damage frequencies within nuclear genes, found within "open" regions of chromatin, as compared to regions of unexpressed sequence that …

In 1952, Dwyer and coworkers began testing a series of metal complexes for potential inhibition of cancer cell proliferation in animals.[l] The complexes tested were unsuitable for such studies due to their high toxicity. Therefore, no further work was done on the project. However, in 1965, Rosenberg and coworkers revisited the possibility of potential metal-based drugs. Serendipitously, they discovered that cis-diamminedichloroplatinum(lI) (cisplatin) inhibits cell division in E. coli.[2] Further studies of this and other platinum compounds revealed inhibition of tumor cell lines sarcoma 180 and leukemia LI2l0 in mice.[l] Cisplatin was approved by the Food and Drug Administration in 1970 …

TiO2-coated conducting glass electrodes can be modified by attachment of redox-active molecules to the TiO2 surface. This thesis describes the synthesis and characterization of a polypyridyl Ru(II) complex that can be covalently bound to a TiO2 surface through the carboxylate groups of 2,2'-bipyridine-4,4' -dicarboxylic acid, H2dcbpy. In addition, the bound complex contains a redox-active alloxazine derivative, pptd, which is capable of 2e-/2H+ transfers. 3-Phenylbenzofuranone, when deprotonated with sodium hydride, forms an extended enolate system which, when treated with excess alkyl chloroformates, affords only oxygen-acylated products, as opposed to the usually desired carbon-functionalized derivatives. It was discovered that such molecules, when treated with a catalytic amount of 4- (N, N-dimethylamino)pyridine (DMAP), rearrange quantitatively to the carbon acylated isomers.

These migrations, which are accompanied by a deep blue color, are proposed to involve a reactive intermediate. This intermediate, which is postulated as an ion pair charge transfer (IPCT) complex, has been closely studied in order to gain a better understanding of the …

A sequential extraction procedure, a three-step protocol proposed by the Standards, Measurements and Testing programme (SM & T--formerly BCR) of the European Union, was applied to soil samples for the determination of metals (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn). This procedure provides measurements of extractable metals from media such as acetic acid (0.11 mol L^{-1}), hydroxylammonium chloride (0.1 mol L^{-1}) and hydrogen peroxide (8.8 mol L^{-1}) plus ammonium acetate (1 mol L^{ - 1}), which are exchangeable, reducible and oxidisable metals, respectively. In this study, 4(1H)-pyridinones with an amide function in Position 2 of the pyridinone ring showed antimicrobial activity. The synthesis and the biological properties of some novel amide derivatives of pyran-4(1H)-one and 4(1H)-pyridinone have been reported. Amide derivatives of pyran-4(1H)-one and 4(1H)-pyridinone were prepared using two different methods. Antimicrobial activities were determined as MIC values using the microdilution broth method against the bacteria Staphylococcus aureus, Enterococcus faecalis, Escherichia coli and Pseudomonas aeruginosa and the fungi Candida albicans, Candida krusei and Candida parapsilosis. Compounds 6 and 9 exhibited the best inhibitor activity against S. aureus.

Effects Of A Yec Apologetics Class On Student Worldview, David A. Dewitt, Steve Deckard, Sharon E. Cargo

Creationism is a significant component of an overall Christian worldview. A Creation Worldview Test (CWT) was administered to students both before and after completion of a required Apologetics course dealing with the creation/evolution controversy. Pre-test analysis revealed that incoming students tend to have a creationist worldview, however their CWT scores reflected a degree of uncertainty or neutrality on scientific creation and the age of the earth. Post-test analysis showed a significant shift toward a young earth creationist (YEC) view. Implications for teaching apologetics and development of a Creationist worldview based on a Six-day Young Earth perspective were explored and identified.

Benzoylecgonine (BE) is the most abundant metabolite of cocaine found in the human body. Analysis of BE in urine by gas chromatography/mass spectrometry is the most common method currently used to detect cocaine abuse. This current method is costly and time consuming, so the investigation of an easier and more cost-effective approach is the goal of this research. Because BE is a zwitterion, it is highly soluble in water and very difficult to extract from urine. Abstract

Background

Combination of biochemical and bioinformatic analyses led to the discovery of oxidative demethylation – a novel DNA repair mechanism catalyzed by the Escherichia coli AlkB protein and its two human homologs, hABH2 and hABH3. This discovery was based on the prediction made by Aravind and Koonin that AlkB is a member of the 2OG-Fe2+ oxygenase superfamily.

Results

In this article, we report identification and sequence analysis of five human members of the (2OG-Fe2+) oxygenase superfamily designated here as hABH4 through hABH8. These experimentally uncharacterized and poorly annotated genes were not associated with the AlkB family …

Structure Property Correlation methods such as regression analysis and pattern recognition are applied to predict the molecular properties of two members of an ampicillin homologous series. The pentyl and hexyl esters of ampicillin are also evaluated for their similarity to other penicillins by use of multiple regression, contingency tables, cluster analysis, correspondence analysis, self organizing tree algorithms, factor analysis, principal component analysis, box plots, and other graphing methods. Other members of the homologous series include methyl, ethyl, propyl, and butyl esters of ampicillin which have been previously synthesized and tested in tissue culture against Escherichia coli. This dissertation is an account of the synthesis, characterization, and application of novel water-soluble chiral calixarenes in spectroscopy and capillary electrophoresis. It is divided into four sections. The first part describes the synthesis and nuclear magnetic resonance (NMR) characterization of four p-t-butylcalix[4]arenes bearing L-amino acid moieties on their lower rims (CX4-AA). The structure and conformation of the derivatives have been determined using one- and two-dimensional NMR techniques. Proton and carbon-13 spectra show that the derivatives are tetra-substituted and adopt a cone conformation. The preparation and characterization of silica-bonded calixarene stationary phases for capillary electrochromatography is also reported in Chapter 2. …

Most of the research in the field of molecular imprinting has been focused on the development of new functional monomers, in order to improve the molecular recognition properties afforded by these materials. The role of the crosslinking monomer has often been overlooked, since it is considered an inert component that only provides a scaffold to support the binding sites. The Aqueous Phase Yield Of Alkyl Nitrates From Roo+No: Implications For Photochemical Production In Seawater, Elizabeth E. Dahl, Eric S. Saltzman, Warren J. De Bruyn

Alkyl nitrates have been observed in remote oceanic regions of the troposphere and in the surface ocean. The mechanism for their production in the oceans is not known. A likely source is the reaction of ROO + NO (where R is an alkyl group). Steady-state laboratory experiments show that alkyl nitrates are produced in the aqueous phase via this reaction, with branching ratios of 0.23 +/- 0.04, 0.67 +/- 0.03, and 0.71 +/- 0.04 for methyl, ethyl, and propyl nitrate respectively. The branching ratios in aqueous solution are significantly higher than in the gas phase. Irradiation of surface seawaters yield …

Proteus mirabilis, a cause of complicated urinary tract infection, produces urease, an essential virulence factor for this species. UreR, a member of the AraC/XylS family of transcriptional regulators, positively activates expression of the ure gene cluster in the presence of urea. To specifically evaluate the contribution of UreR to urease activity and virulence in the urinary tract, a ureR mutation was introduced into P. mirabilis HI4320 by homologous recombination. The isogenic ureR::aphA mutant, deficient in UreR production, lacked measurable urease activity. Expression was not detected in the UreR-deficient strain by Western blotting with monoclonal antibodies raised against UreD. Urease …

Phytoplankton are a major consumer of carbon dioxide. In theory, if phytoplankton are limited in number, more carbon dioxide will be present in the atmosphere and contribute to the greenhouse effect. An increased concentration of iron would, therefore, promote phytoplankton blooms that would consume carbon dioxide. There would then be less carbon dioxide in the atmosphere to prevent heat from radiating away from earth. In 1996, an iron seeding experiment was performed in the open ocean to determine whether phytoplankton density would be increased. Phytoplankton rapidly increased in number.
