To date there are no analytical techniques designed to exclusively measure bioavailable iron in marine environments. The goal of this research is to develop such a technique by isolating the bioavailable iron using the terrestrial siderophore desferrioxamine B (DFB). This project contained many challenging aspects, but the specific goal of this study was to develop a robust analytical technique for quantification of Fe(III)-DFB complexes at nanomolar concentrations. Past work showed that oxalate (Ox) promotes photodissociation of Fe(III)-DFB to Fe(Il), and we are specifically interested in the mechanism of this process. 2-methoxy-2-methylpropylidene was photochemically generated from a non-nitrogenous precursor, 1a,9b-Dihydro-IH-cyclopropa[1]phenanthrene-l-etbane 2-methoxy-2-methyl, available in two steps from phenanthrene. Two competing rearrangement pathways were observed for the p-methoxy carbene, 1,2-methyl shift and C-H insertion, which had varying weights depending on the solvent used. Nevertheless, the C-H insertion was always predominant, contrary to the current understanding of B-substituted carbenes, which apparently needs revision. An attempt was also made to measure by LFP the rate constant of rearrangement and lifetime of the stabilized l-chloro-2-methoxy-2-metbylpropylidene.

The purpose of this project was to develop an upper level undergraduate biochemistry laboratory (CHEM 453) based upon a novel research project, designed to replace an existing instructional laboratory that employs ‘cookbook’ exercises. Methods for the expression, purification, spectral analysis of structural integrity, and electron transfer capability of rabbit cyt b5 were adapted from published work done by Dr. Lucy Waskell of the University of Michigan and Dr. Mansuy of the University of Paris. We report an extensive study on total and partial-ion-yield spectroscopy around both the S 2p and F 1s thresholds in SF6. All positive and negative single-ion channels have been measured. Below the F 1s threshold we detect a large variation in relative intensity of the resonant structures according to the specific channel monitored, indicating selective fragmentation. Above threshold, at variance with previous cases described by us, we detect high-intensity structures related to shape resonances not only in the cation channels but also for the anions. Cisplatin, carboplatin, and oxaliplatin are used clinically for treating some cancers, but are they are not the preferred treatment for breast cancer. The usefulness of these platinum-containing drugs is limited by poor concentration of the active platinum moiety within the tumor before dose-limiting side effects are reached. One way to selectively concentrate a platinum antitumor compound in breast cancer cells is to conjugate the platinum moiety via a malonate linkage to an estrogen which selectively binds to the estrogen receptor (ER) protein. 31P NMR spectroscopy was used to study freeze tolerance of the wood frog, R. sylvatica. Spectra were obtained of living whole body samples and isolated thigh muscle tissue. These results were compared to R. pipiens which is freeze sensitive. R. sylvatica was found to have significantly higher levels of glucose-6-phosphate and several phosphodiesters. Both seasonal and environmental stresses were studied to determine the effect of freezing conditions on the phosphorus compounds present in R. sylvatica. The results show that R. sylvatica is capable of tolerating stresses that are associated with freezing without any drastic changes in the levels of metabolic …

Short Tandem Repeat (STR) analysis has become a valuable tool in identifying the source of biological stains, particularly from the investigation of sexual assault crimes. Difficulties in analysis arise primarily in the interpretation of mixed genotypes when cell separation of the sexual assailant's sperm from the victim's cells is incomplete. The forensic community continues to seek improvements in cell separation methods from mixtures for DNA typing. This report describes the use of laser microdissection (LMD) for the separation of pure populations of spermatozoa from two-donor cell mixtures. The release of the 1000th complete microbial genome will occur in the next two to three years. In anticipation of this milestone, the Fellowship for Interpretation of Genomes (FIG) launched the Project to Annotate 1000 Genomes. The project is built around the principle that the key to improved accuracy in high through put annotation technology is to have experts annotate single subsystems over the complete collection of genomes, rather than having an annotation expert attempt to annotate all of the genes in a single genome. C2C12 mouse myoblast cells, grown in glass vials, were connected to a cryofocusing unit to trap volatile organic compounds (VOCs). The VOCs were eluted from the trap by capacitive discharge into a gas chromatograph with time-of-flight mass spectral capabilities (GC-TOFMS) and were found to include the lipid peroxidation product hexanal. The pro-oxidant cumene hydroperoxide elevated the levels of these lipid peroxidation products, whereas the anti-oxidant butylated hydroxy toluene (BHT) impaired their production. Derivatization of the aldehyde products of lipid peroxidation in the same myoblast cells with pentafluorobenzyl hydroxylamine hydrochloride (PFB) provided evidence for formation of non-volatile products of lipid peroxidation …

Nature has provided a vast number of compounds that have medicinal application. The use of these compounds provided by nature however, is limited by the amount of a specific compound that nature provides. The synthesis of these molecules and their derivatives provides an even greater number of compounds for biological screening. Various pyrroles, the heterocyclic building blocks of porphyrins, can be prepared synthetically over several steps. The implementation of carboranes on pyrrole rings intrinsically provides substrates that can be converted into porphyrins bearing carborane substituents. The impact of phycotoxins on human and animal health is of important concern for worldwide authorities and the scientific community. Domoic acid (DA) is a neurotoxin produced by phytoplankton {Pseudonitzchia spp.). It enters the food chain through the consumption of shellfish that feed on toxic diatoms. DA has been responsible for several outbreaks affecting humans and animals all around the world. Yessotoxin (YTX) is produced by dinoflagellate species {Protoceratiuni reticulatum) which are globally distributed. Its toxicity is low when ingested orally. The cationic ruthenium hydride complex [(PCy3)2(CO)(CH3CN)2RuH]+BF4- was found to be a highly effective catalyst for the C−H bond activation reaction of arylamines and terminal alkynes. The regioselective catalytic synthesis of substituted quinoline and quinoxaline derivatives was achieved from the ortho-C−H bond activation reaction of arylamines and terminal alkynes by using the catalyst Ru3(CO)12/HBF4·OEt2. As technology begins to utilize nanocrystals for many chemical, biological, medical, electrical, and optoelectrical applications, there is a growing need for an understanding of their fundamental properties. The study of melting and solidification of nanocrystals is of interest to fundamental understanding of the effect of reduced size and crystal shape on the solid-liquid phase transition. Melting and solidification of as-deposited and recrystallized indium and bismuth nanocrystals were studied using reflection high-energy electron diffraction (RHEED). Further phytochemical studies on the aerial parts of Phlomis viscosa (Lamiaceae) led to the isolation of 24 compounds: 3 iridoid glycosides, 10 phenylethanoid glycosides, a megastigmane glycoside and a hydroquinone glycoside, as well as 2 lignan glucosides and 7 neolignan glucosides, 1 of which is new (17b). Compound 17b was obtained as a minor component of an inseparable mixture (2:1) of 2 neolignan glucosides (17a/b), and characterized as 3',4-O-dimethylcedrusin 9-O-\beta -glucopyranoside. Full NMR data of the known 8-O-4' neolignan glucoside, erythro-1-(4-O-\beta-glucopyranosyl-3-methoxyphenyl)- 2-{2-methoxyl-4-[1-(E)-propene-3-ol]-phenoxyl}-propane-1,3-diol (18) are also reported. The kinetics of the adsorption of lead ions on active carbon from aqueous solutions was studied at room temperature at different lead ion concentrations (i.e. 0.1-0.5 g/L). Phthalic anhydride was reacted with L-Valine and L-Leucine in a mixture of acetic acid and pyridine at room temperature, and then was refluxed at 90-100 °C and N-phthaloyl-L-valine or N-phthaloyl-L-leucine were obtained in quantitative yields. The imide-acids were converted to N-phthaloyl-L-valine acid chloride and N-phthaloyl-L-leucine acid chloride by reaction with thionyl chloride. Then 2 new derivatives of the chiral isocoumarin with L-valine and L-leucine moieties were synthesised by the condensation reaction of homophthalic acid with respective imide-acid chloride. 3-Phenyl-6-methyl-4(3H)-quinazolinone-2-yl-mercaptoacetic acid hydrazide (3) was reacted with substituted aldehydes in absolute ethanol to obtain a new series, 3-phenyl-6-methyl-4(3H)-quinazolinone- 2-yl-mercaptoacetic acid arylidenehydrazides (4a-q). The structures of 4a-q were elucidated by elemental analyses and from spectrometric (UV, IR, ^1H-NMR, ^{13}C-NMR and EIMS) data. Then 4a-q were evaluated for antibacterial, antifungal and antitubercular activities. No significant activity was observed against the selected microorganisms.

Some new di- and tri-organotin(IV) derivatives of 4 different non-steroidal anti-inflammatory drugs (NSAIDs) with the general formulae R_2SnL_2 and R_3SnL (where R = n-C_{8}H_{17} and C_6H_5CH_2) and L = 2-[(2,3-dimethylphenyl)amino]benzoic acid, 2-(2-fluoro-4-biphenyl)propionic acid, 2-(4-isobutylphenyl) propionic acid and 2-(3-benzoylphenyl)-propionic acid were synthesized. These compounds were structurally characterized by infrared and multinuclear NMR (^1H, ^{13}C, ^{119}Sn) spectroscopies and mass spectrometry. The isotopic effect of tin was studied by comparison of experimental data with the simulated isotopic pattern using the Chemtool software package. Organotin(IV) complexes of 2-(N-maleoylamino)-2-methylpropanoate were prepared by treating the ligand acid with diorganotin dichloride and triorganotin chloride in 1:2 and 1:1 molar ratios, respectively. These reactions proceeded smoothly to completion and the complexes formed were characterized by elemental analysis, infrared, multinuclear NMR (^1H, ^{13}C, ^{119}Sn) and mass spectrometry. The biological activities of the tin complexes were studied by screening the compounds against various animal and plant pathogens. These results show that triorganotin(IV) carboxylates are more active against the animal and plant pathogens tested as compared to diorganotin dicarboxylates.

Research, inspired by nature, provides ample opportunity for discovery, ingenuity, and creative endeavor. For these reasons, the work and accomplishments presented herein focus on the total syntheses and related investigations of a variety of natural product derivatives, specifically (+)-nootkatone and jasmone. While the former consists of several independent projects: (a) an enantioselective synthetic route to (+)-nootkatone and derivatives, (b) examination of the valencane ring system, (c) determination of the absolute configuration of (+)-nootkatone, and (d) rearrangements of various sesquiterpenoid derivatives; the latter focuses on the construction ofƒndelta, epsilon-unsaturated carbonyl compounds, hetero Pauson-Khand substrates, via [3,3]-sigmatropic rearrangements.

Novel symmetric and asymmetric water-soluble tetrabenzoporphyrin are synthesized in relatively good yields. These compounds are synthesized by the condensation of butanopyrrole with several aldehydes that contain masked water solubilizing groups followed by metal insertion, oxidation and demetallation and deprotection of the masked solubilizing groups. The biological evaluation (in vitro and in vivo) studies are described in addition to the photophysical evaluations of the corresponding compounds. Increasing the information content obtainable from a single assay and system miniaturization has continued to be important research areas in analytical chemistry. The research presented in this dissertation involves the development of a two-color, time-resolved fluorescence microscope for the acquisition of both steady-state and time-resolved data during capillary and microchip electrophoresis. The utility of this hybrid fluorescence detector has been demonstrated by applying it to DNA sequencing applications. Coupling color discrimination with time-resolved fluorescence offers increased multiplexing capabilities because the lifetime data adds another layer of information. Spherical Polymeric Particles (SPP) have been prepared from Tanzanian Cashew Nut Shell Liquid (CNSL) by suspension polymerization technique involving either step-growth or chain- growth polymerization mechanisms. The sizes of the SPP, which ranged from 0.1 to 2.0 mm were strongly influenced by the amounts of stabilizers in polymerization recipes used. The particles showed variable surface areas, indicative of the presence of pores in some of the preparations and the highest surface area recorded was about 260 m2g-1. The particles were found to exhibit Langmuir-type adsorption isotherms with saturation capacity of about 9.0 and 44.2 mg/g for Na+ and Ca2+, respectively. …

A study conducted during the period of March/April 2002 aimed at measuring physical and chemical parameters in Tanzanian portion of Lake Victoria. Temperature and conductivity values were homogeneous among and within the study stations. Levels of dissolved oxygen (DO) ranged from 4 - 9 mgl-1 and there was significant difference between stations (P < 0.05). Water transparency was poor (< 2 m secchi depth) and differed from one sampling station to another. Ammonium nitrogen (NH4+-N), with few exceptions, showed the highest concentrations both at the bottom and near-surface followed by total phosphates (TP), nitrate nitrogen (NO2+NO3--N), nitrite nitrogen (NO2--N) and Soluble Reactive Silicon (SRSi). Concentrations of NH4+-N, TP and SRSi were high at bottom waters and the reverse was true for NO2+NO3--N and NO2--N. Differences in nutrients and physical water parameters among sampling stations were statistically significant (P < 0.05). However, temperature and nutrient levels at studied areas were higher than that of 50 years ago and the trend reversed for DO and transparency. This indicates that Lake Victoria is receiving organic materials from the catchment area leading to the decline of water quality. Long-term efforts for rehabilitation, conservation and sustainable management of the lake are suggested.

A 29 cm long core recovered from a water depth of 5 m in a small closed lake located in the Empakai crater northern Tanzania, is used to document the contents of organic carbon and nitrogen, stable isotopes composition of organic carbon and nitrogen, and C/N ratios and to infer climatic changes from these data. The δ 13C and δ15N values for this core, which averages - 16.5±2.1l and 11.0±5.6l respectively, show a general downcore decrease below 5 cm. Spectroscopic studies have been used to describe the mechanism of the decomposition of hydrogen peroxide by solutions of a dimeric Cu(II) complex of a dissymetric Schiff base, [CuSALAD]2.H2O, and imidazole or methyl substituted imidazoles, B, which form monomeric CuSALAD.B2 complexes, in aqueous ethanol solvent. Freezing point depression and vapor pressure lowering studies were carried out to confirm the dimeric nature of the [CuSALAD]2.H2O complex that had been previously reported. The chemistry of Gd3+-based MRI agents has advanced considerably during the past decade toward agents with higher relaxivity and agents that respond to physiology and / or metabolism. This review describes various approaches that have been taken toward the development of responsive contrast agents and discusses the importance of fast water exchange for advancement of targeted Gd3+-based agents with higher sensitivity. The recent discovery of Eu3+ complexes having extraordinarily slow water exchange has opened a new avenue in contrast agent design based upon the chemical exchange saturation transfer (CEST) mechanism.
