A fast and simple differential pulse polarographic (DPP) method for the determination of trace elements in a certain form of cabbage is described. This vegetable is commonly used as food in Turkey, especially in the Black Sea region. Using DPP polarograms of wet digested cabbage samples (leaf) in pH 2 and 4 acetate buffer Mo, Cr, Se, Pb, As and Zn quantities were determined. The best separation and determination condition for Cu and Fe was in EDTA at pH 6.5, and for Ni and Zn it was ammonia buffer at pH 9.8. The trace element quantities in digested leaf samples …

The thiophene analogue of (\pm)-4-ipomeanol (1) was prepared in 2 steps starting from 3-iodothiophene and pent-4-yn-2-ol. The key steps were a Sonogashira reaction, followed by HgO catalysed hydratisation of the triple bond. The resulting thiophene was tested in the MTT assay for its cytotoxic activity against HL 60 cells.

Spectroscopic Characterisation And Biological Applications Of Organotin(Iv) Derivatives Of 3-(N-Naphthylaminocarbonyl)-2-Propenoic Acid, Sajjad Ahmad, Moazzam H. Bhatti, Saqib Ali, Fiaz Ahmed

In an effort to develop new organotin materials for investigation and biocidal evaluation, a series of compounds with the general formula R_{4 - n}SnL_n (where R = CH_3, n-C_4H_9, C_6H_5, C_6H_5CH_2 and L = 3-(N-naphthylaminocarbonyl)-2-propenoic acid) were synthesised, and characterised by elemental analysis, IR, multinuclear (^1H, ^{13}C and ^{119}Sn) NMR, ^{119m}Sn Mössbauer spectroscopy and mass spectrometry. The biological activity of these compounds against various bacteria and fungi was investigated. All of the compounds were active against the fungi tested with a few exceptions. These compounds also showed significant antibacterial activity. LD_{50} data show that the investigated compounds have significant toxicity.

The N-alkyl derivatisation and photochemical dimerisation of 3 o-, m-, and p-nitro substituted 4-azachalcones (1-3) yielded 3 new o-, m-, and p-nitro substituted (E)-N-decyl-4-azachalconium bromides, (2E)-1-(2-nitrophenyl)-3-(N-decyl-4-pyridinium bromide)-2-propen-1-one (4), (2E)-1-(3-nitrophenyl)-3-(N-decyl-4-pyridinium bromide)-2-propen- 1-one (5), and (2E)-1-(4-nitrophenyl)-3-(N-decyl-4-pyridinium bromide)- 2-propen-1-one (6), and 3 new dimers in solution, (1\beta, 2\alpha)-di- (3-nitrobenzoyl)-(3\beta, 4\alpha)-di-(4-pyridinyl) cyclobutane (7), (1\beta, 2\alpha)-di-(4-nitrobenzoyl)-(3\beta,4\alpha)-di-(4-pyridinyl) cyclobutane (8a), and (1\beta, 2\beta)-di-(4-nitrobenzoyl)-(3\beta, 4\alpha)-di-(4-pyridinyl)cyclobutane (8b), stereoselectively. The monomeric compounds showed good antimicrobial activity against test micro-organisms. The most sensitive micro-organisms were Gram-positive bacteria. From the aerial parts of Galium tortumense Ehrend & Schönb.-Tem., 8 iridoid glycosides, a flavonol glycoside, a oleanane-type triterpene acid, an ursan-type triterpene acid and a sterol were isolated. The M(IN)_2Ni(CN)_4 [where M: Cu, Mn, Zn, IN: Isonicotinic acid, abbreviated to M-Ni-IN] tetracyanonickelate and some metal halide complexes with the following stoichiometries: Cu(IN)X_2 (X:Br,I), Cd(IN)_2X_2, (X:Cl,Br), and Zn(IN)_4X_2 (X:Br,I) were synthesized for the first time. Their FT-IR spectra were reported in the 4000-400 cm^{-1} region. Vibrational assignments were given for all the observed bands. The analysis of the vibrational spectra indicates that there are some structure-spectrum correlations. A quantitative structure-property relationship study is suggested for the prediction of the acidity constants of some thiazolidine-4-carboxylic acid derivatives in aqueous solution. Ab initio theory was used to calculate some quantum chemical descriptors, including electrostatic potentials and local charges at each atom, highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) energies, etc. Modeling of the acidity constant of thiazolidine-4-carboxylic acid derivatives as a function of molecular structures was established by means of the partial least squares algorithm. Adsorption isotherms for polyacrylamide (PAA) on polycrystalline gold, from sulfuric acid (potential range 0-2.0 V/she; 293 K), were obtained by means of cyclic voltammetry with simultaneous monitoring of the double-layer capacity. The Frumkin isotherm indicates that relatively strong attractive forces are present (a = 0.8-1.0), and the Gibbs energies range between 43 and 56 kJ mol^{-1}. The coverage (\theta) of the metal surface with polymer molecules depends on the molar concentration of PAA, as well as on its molecular weight, and the size of the statistical polymer coil in the solution (R_G = 7.49 \times 10^{-3} \times M_n^{0.64}/nm). The reactions of 4-benzoyl-5-phenyl-2,3-furandione (1a) and 4-(4-methoxybenzoyl)-5-(4-methoxyphenyl)-2,3-furandione (1b) with acyl hydrazines (2) (namely hydrazides) are reported. From these reactions, novel pyridazinone systems (3a-g) are obtained as well as the cyclization product of 3g at high temperature (4). A series of five-coordinated germanium-substituted tricyclohexylantimony dipropionates have been synthesized and characterized by different instrumental techniques, such as elemental analyses, FT-IR, multinuclear NMR (^1H, ^{13}C) and mass spectrometry. These compounds have also been screened against different microbes and they showed good activity against different bacteria that was comparable to the reference drugs. The crystal structure of the precursor (C_6H_{11})_3SbBr_2 is reported here, which showed that the antimony atom in an asymmetric unit exists in trigonal bipyramidal geometry, having space group C2/c with the monoclinic crystal system.

Template Synthesis And Structural Characterization Of Homo Binuclear Chromium(Iii), Manganese(Iii), Iron(Iii), Cobalt(Iii), And Ruthenium(Iii) Complexes With Octaazamacrocyclic Ligands, Vinod K. Sharma, Shipra Srivastava

The Schiff base octaazamacrocyclic ligands derived from primary diamines and 3,6-dimethyl/diphenyl-4,5-diazaocta-3,5-diene-2,7-dione, and their binuclear complexes [M_2LCl_4]Cl_2 [M = Cr(III), Fe(III), Co(III), or Ru(III)] and [Mn_2L(AcO)_4](AcO)_2 were synthesized by template condensation reactions. Attempts to synthesize the corresponding metal-free macrocyclic ligands did not prove successful. The overall geometry and stereochemistry of these complexes were elucidated by elemental analyses, magnetic susceptibilities, electronic spectra, infrared spectra, molar conductance measurements, ^1H NMR, and thermogravimetric analysis. All the trivalent metal ion complexes appear to be 1:2 electrolytes. An octahedral geometry is proposed for all the complexes.

The goal of the research presented in this dissertation is to develop novel water soluble carboranylporphyrins that can deliver therapeutic dosage of boron specifically to tumors for application in boron neutron capture therapy. In order to accomplish this goal novel water soluble tetra(nido-carboranyl)porphyrin and the octa(nido-carboranyl)porphyrin with high boron contents are synthesized by the condensation of the carborane substituted benzaldehyde with the pyrrole under acidic conditions. Previous work in our laboratory has demonstrated not only a synthetic value of tandem reductive cyclization - carbonylation reactions in the preparation of complex molecules but also the interesting stereoselectivity observed therein. The first part of this work employs the titanium-mediated cyclocarbonylation of tethered dienals for the synthesis of prostaglandin analogues, in particular PGF2α and methyl jasmonate. Based on this hetero Pauson-Khand strategy, we have established an efficient racemic route to both the prostaglandin and the methyl jasmonate core skeletons. In Chapter 2, a simple and reproducible technique for constructing perfectly aligned gaps in fused-silica capillaries has been developed for postcolumn reagent addition with capillary electrophoresis (CE) to take advantage of laser-induced fluorescence (LIF) detection. A major task in proteomics is to identify proteins from a biological sample using two-dimensional (2-D) separation prior to mass spectrometry of peptides generated via proteolytic digestion of the proteins. For 2-D separations, microfluidic devices are superior to bench top and capillary-based systems since they potentially provide higher separation efficiencies due to the minimal dead volumes produced during peak transfer between the two separation dimensions. Sphingosine 1-phosphate is a bioactive sphingolipid that regu- lates cell growth and suppresses programmed cell death. The bio- synthesis of sphingosine 1-phosphate is catalyzed by sphingosine kinase (SK) but the mechanism by which the subcellular localization and activity of SK is regulated in response to various stimuli is not fully understood. To elucidate the origin and structural determi- nant of the specific subcellular localization of SK, we performed biophysical and cell studies of human SK1 (hSK1) and selected mutants. We previously reconstituted a minimal DNA replicase from Pseudomonas aeruginosa consisting of α and ϵ (polymerase and editing nuclease), β (processivity factor), and the essential τ, δ, and δ′ components of the clamp loader complex (Jarvis, T., Beaudry, A., Bullard, J., Janjic, N., and McHenry, C. (2005) J. Biol. Chem. 280, 7890-7900). In Escherichia coli DNA polymerase III holoenzyme, χ and Ψ are tightly associated clamp loader accessory subunits. Mineral aerosol deposition is the dominant source of iron to the open ocean. Soil iron is typically insoluble and understanding the atmospheric processes that convert insoluble iron to the more soluble forms observed over the oceans is crucial. In this paper, we model several proposed processes for the conversion of Fe(III) to Fe(II), and compare with cruise observations. The comparisons show that the model results in similar averaged magnitudes of iron solubility as measured during 8 cruises in 2001–2003. Aromatase, the enzyme responsible for estrogen biosynthesis, is a particularly attractive target in the treatment of hormone-dependent breast cancer. The synthesis and biological evaluation of a series of 2-(4′-pyridylmethyl)thio, 7-alkyl- or aryl-substituted isoflavones as potential aromatase inhibitors are described. The isoflavone derivatives demonstrate IC50 values from 79 to 553 nM and compete with the endogenous substrate, androstenedione. Data supporting the ability of these analogs to suppress aromatase enzyme activity in the SK-BR-3 breast cancer cell line are also presented.

Wilsons disease is an autosomal recessive disorder of copper metabolism which results from mutations in the gene encoding for Wilson disease protein (WDP). The cytosolic N-terminus of WDP comprises six metal binding domains each containing a conserved metal binding motif, GMXCXXC. The contribution of these metal binding domains to copper homeostasis is poorly understood. We have cloned, expressed and purified residues 485 to 633 corresponding to N-terminal metal-binding domains 5 and 6 of WDP (WD5-6). This study examines college student understanding of key concepts that will support future organic chemistry success as determined by university instructors. During four one-hour individual interviews the sixteen subjects attempted to solve general chemistry problems. A think-aloud protocol was used along with a whiteboard where the students could draw and illustrate their ideas. The protocols for the interviews were adapted from the Covalent Structure and Bonding two-tiered multiple choice diagnostic instrument (Peterson, Treagust, & Garnett, 1989) and augmented by the Geometry and Polarity of Moleculessingle-tiered multiple choice instrument (Furió & Calatayud, 1996).
