To date, the use of mitochondrial DNA in forensic analysis has relied on the presence of variations in the control region to differentiate between samples. One problem that this analysis has shown is the occurrence of common Haplogroup H haplotypes or identical sequences. Thus, there is a need to enhance the distinguishing power of this type of analysis. Design And Performance Of A Curved-Crystal X-Ray Emission Spectrometer, A. C. Hudson, Wayne C. Stolte, Dennis W. Lindle, Renaud Guillemin

A curved-crystal x-ray emission spectrometer has been designed and built to measure 2–5 keV x-ray fluorescence resulting from a core-level excitation of gas phase species. The spectrometer can rotate 180°, allowing detection of emitted x rays with variable polarization angles, and is capable of collecting spectra over a wide energy range (20 eV wide with 0.5 eV resolution at the Cl K edge) simultaneously. In addition, the entire experimental chamber can be rotated about the incident-radiation axis by nearly 360° while maintaining vacuum, permitting measurements of angular distributions of emitted x rays.

RNase L is one of the key enzymes involved in the molecular mechanisms of interferon (IFN) actions. Upon binding with its activator, 5′-phosphorylated, 2′-5′ oligoadenylates (2-5A), RNase L plays an important role in the antiviral and anti-proliferative functions of IFN, and exerts proapoptotic activity independent of IFN. In this study, we have found that RNase L retards proliferation in an IFN-dependent and independent fashion. To directly measure the effect of RNase L on tumour growth in the absence of other IFN-induced proteins, human RNase L cDNA was stably expressed in P-57 cells, an aggressive mouse fibrosarcoma cell line. Exploring The Mechanism Of Flexible Biomolecular Recognition With Single Molecule Dynamics, H. Peter Lu, Qiang Lu, Jin Wang

Combining a single-molecule study of protein binding with a coarse grained molecular dynamics model including solvent (water molecules) effects, we find that biomolecular recognition is determined by flexibilities in addition to structures. Our single-molecule study shows that binding of CBD (a fragment of Wiskott-Aldrich syndrome protein) to Cdc42 involves bound and loosely bound states, which can be quantitatively explained in our model as a result of binding with large conformational changes. Protein–protein interactions are very important in the function of a cell. Computational studies of these interactions have been of interest, but often they have utilized classical modelling techniques. In recent years, quantum mechanical (QM) treatment of entire proteins has emerged as a powerful approach to study biomolecular systems. A novel sensory platform based on conjugated polymer was developed, that consists of a monodispersed oligo(phenylene ethynylene) backbone, end-capped with a terminal alkene for further attachment to a solid surface at one terminus, and an anthracene based receptor group at the other terminus. Energy transfer from backbone to end-cap was observed by absorption spectroscopy and fluorescence spectroscopy. Other ongoing projects described herein include the synthesis of longer-chain analogues of the above platform, the synthesis of silica microspheres based conjugated polymer sensory platform and the synthesis of long wavelength (NIR) dye for in vivo amyloid screening studies.

The spectroscopic properties of a chiral boronic acid based resorcinarene macrocycle employed for chiral analysis were investigated. Specifically, the emission and excitation characteristics of tetraarylboronate resorcinarene macrocycle (TBRM) and its quantum yield were evaluated. The chiral selector TBRM was investigated as a chiral reagent for the enantiomeric discrimination of amino acids using steady-state fluorescence spectroscopy. Chiral recognition of amino acids in the presence of the macrocycle was based on diastereomeric complexes. Results demonstrated that TBRM had better chiral discrimination ability for lysine as compared to the other amino acids. Before such MOFs can be attempted, multifunctional ligands of the appropriate geometry for the desired MOFs first must be synthesized. Experimental and theoretical investigations were carried out on 2 sets of chalcones. Set 1 has an OH group on ring A and Set 2 does not, and both have different substituents on ring B at different positions. The experimental investigations comprised cyclic voltammetric (CV) studies to determine the reactivity in terms of redox potentials in DMF at room temperature (23 \pm 1 °C). Phytochemical investigations on the above-ground parts of Lamium garganicum subsp. laevigatum resulted in the isolation of 7 iridoid glucosides: shanzhiside methyl ester (1), barlerin (8-O-acetylshanzhiside methyl ester) (2), 6-O-syringyl-8-O-acetylshanzhiside methyl ester (3), 6\beta-hydroxyipolamiide (4), lamalbide (5), dehydropenstemoside (6), and sesamoside (7). An Electrochemical Dna Biosensor For The Detection Of Dna Damage Caused By Radioactive Iodine And Technetium, Pinar Kara, Kağan Dağdevi̇ren, Mehmet Özsöz

A DNA damage process by radioactive iodine (I-131) and technetium (Tc-99m) is described. The procedure relies on a label-free electrochemical DNA biosensor. Fish sperm double stranded DNA was immobilized onto a carbon paste electrode (CPE) by electro deposition and the changes to the guanine oxidation signal were observed upon exposure to radioactive I-131 and Tc-99m by differential pulse voltammetry (DPV). The changes to the signal of guanine after exposure to therapeutic doses of Tc-99m and I-131 showed conformational changes in DNA molecules.

Di-(1, 3, 8) and triorganotin(IV) complexes (2, 5, 7) with the general formula R_nSnL_{4-n}, where R = Me, Et, n-Bu, n-Oct, Ph, and Bz, and L = the n-C_4H_9OCOC_6H_4COO^- monoanionic ligand have been synthesized by the reaction of the silver salt of the n-butylhydrogen phthalate with di- and triorganotin chloride in dry chloroform. However, di-n-butyl- and di-n-octyltin(IV) derivatives (4 and 6, respectively) were synthesized by the reaction of the corresponding organotin(IV) oxide with n-butylhydrogen phthalate in dry toluene. Dimethyl bis(4-methylpiperidine dithiocarbamato-S,S')-tin(IV) was synthesized and characterized by elemental analysis, IR and mass spectrometry, multinuclear NMR (^1H- and ^{13}C-NMR), and X-ray single crystal analysis. IR data showed that the ligand acts as a bidentate in the solid state. X-ray data showed the unsymmetrical nature of the ligand towards coordination to tin. It crystallized in the monoclinic P2_1/n space group. Its geometry is distorted octahedral. Antimicrobial activity data shows that the complex exhibits significantly more activity than the free ligand.

Achmatowicz rearrangement was carried out with 2-chloro-1-(furan-2-yl) ethanol, which was synthesized starting from 1-(furan-2-yl) ethanone. It was shown that the oxidation of 2-chloro-1-(furan-2-yl) ethanol with m-CPBA produced 2-(chloromethyl)-6-hydroxy-2H-pyran-3 (6H)-one in good yield (70%).

A new series of receptors were designed and synthesized, and their interactions with anions, such as F^-, Cl^-, Br^-, I^-, CH_3COO^-, HSO_4^-, and NO_3^-, in DMSO solvent were investigated using UV-Vis absorption spectroscopy. The results showed that hydrogen-bonding complexes were formed between the receptors and the tested anions, such as CH_3COO^- and F^-. It was also found that the selectivity of the receptors for anions could be efficiently tuned by changing the place of the substituent group at the N-phenyl moiety. Three new diterpenoids, piliferol (2\alpha, 20-dihydroxy-12-oxo- abieta-9(11),13-dien-8,20-ether) (1), salvipiliferol (2\alpha, 10\beta -dihydroxy-12-oxo-norabieta-9(11),13-dien) (2), and piliferalactone (2\alpha ,8-dihydroxy-12-oxo-abieta-9(11),13-dien-20-oic acid-8,20-lactone) (3), together with 7 known compounds, ursolic acid, oleanolic acid, betulinic acid, \alpha -amyrin, lupeol, \beta -sitosterol, and pectolinarigenin, have been isolated from the aerial parts of an endemic Salvia species, S. pilifera. The Schiff base derivative of mixed ligand lanthanocene complexes (1-4) showed better efficiency (Mv = 227 \times 10^3-354 \times 10^3) compared to the methoxyethylindenyl derivative of mixed ligand lanthanocene complexes (5-9) (Mv = 72 \times 10^3, 172 \times 10^3). The effects of the catalyst, temperature, catalyst/MMA molar ratio, and catalyst/Al(i-Bu)_3 molar ratio on the polymerization of MMA at 60 °C suggested that ligand size and radius of rare earth metals play an important role in determining the activity of the catalyst.

The interaction of nicotinamide (NA) with NH_4VO_3 at the physiological pH (pH 7.4) was studied by voltammetry and UV-Vis spectroscopy. The reduction of V(V) ions complexed with NA in phosphate buffer (pH 7.4) was observed as a well-defined reversible reduction peak at --0.35 V. The stoichiometric (metal:ligand) ratio of the complex of V(V) with NA was determined as 1:2 by the mole ratio method.

The series of compounds [M(H2O)2(15-crown-5)](NO3)2, M = Mg, Mn, Co, Cu and Zn, has been extended to include two new phases for M = Fe and two new phases for M = Ni. The system [M(H2O)2(15-crown-5)](NO3)2 is remarkable for having many high-Z’ phases (Z’ > 1) with similar packing and for having solid-solid phase transitions through which there is no significant loss of crystallinity.
