Transition metal catalyzed C–H and C–N functionalization methods have emerged as an innovative tool for designing efficient synthesis of fine chemicals by using readily available starting materials without the formation of wasteful byproducts. The similar nature of the hydrogen atom to its isotope, deuterium, allows for the simple exchange of hydrogen atoms for deuterium atoms in drug molecules to alter the absorption, distribution, metabolism, and excretion properties. Installing the deuterium functionality into a specific site in the molecule is essential. Clostridioides difficile is a Gram-positive anaerobic bacterium that causes infections in humans that costs healthcare systems billions per year. C. difficile infection has high rates of recurrence due to multiple antibiotic resistance. When bacteria are in stressful environments, they produce hyperphosphorylated guanosine ribonucleotide signaling molecules called alarmones. The accumulation of alarmones activates the stringent response (SR), in which bacterial cells induce transcription of stress survival genes to delay growth and replication. The C. difficile SR is regulated by enzymatic activity of a bifunctional synthetase/hydrolase, RelA/SpoT homolog (RSH), and a monofunctional small alarmone synthetase (RelQ). The aim of this study was to enhance students’ science literacy through a discussion-based contextual approach in the learning of the human reproductive system. This quantitative research employed an experimental method with a quasi-experimental posttest-only design. The study involved 60 students, with 30 students each in class XI MIPA 4 serving as the experimental group and class XI MIPA 5 serving as the control group. This research aims to explore kinematic resources in linear kinematics by utilizing different forms of problem representation. The study was conducted by administering 15 reasoned multiple-choice questions to 146 high school students from three districts in Central Sulawesi, Indonesia. The research instrument covers topics in linear kinematics, including distance, displacement, speed, and velocity, presented through diagrammatic representations, graphs, and mathematical equations. The students’ written answers were analyzed both quantitatively and qualitatively. Quantitative analysis was conducted to determine descriptive statistics from the data, while qualitative analysis identified the resources students activated when solving these questions. The burgeoning field of continuous glucose monitoring (CGM) for diabetes management faces significant challenges, particularly in achieving precise and stable biosensor performance under changing environmental conditions such as varying glucose concentrations and O2 levels. To address this, we present a novel biosensor based on the electroless coupling of glucose oxidation catalyzed by flavin-dependent glucose dehydrogenase (FAD-GDH) and O2 reduction catalyzed by bilirubin oxidase (BOD) via a redox polymer, dimethylferrocene-modified linear poly(ethylenimine), FcMe2-LPEI. This report mainly discusses the development of RET Proteolysis targeting chimera PROTAC molecules based on Selpercatinib (LOXO-292) and Sulfono-γ-AApeptide PROTAC. A PROTAC molecule is a bifunctional molecule composed of two active domains and a linker. It works by inducing selective protein intracellular degradation. This technology was first discussed by Kathleen Sakamoto, Ray Deshaies and Craig Crews in 2001. Using various E3 Ligase, such as CRBN, pVHL, beta-TrCP1, Mdm2, DCAF15, DCAF16, RNF114, and c-IAP1, the PROTAC technology has been applied to several novel drug development,. Recent literature reports examples of electrochemical biosensor systems that have been developed based upon on the encapsulation of enzymes and metal nanoparticles into a xerogel. These systems have been utilized for the detection of biologically important compounds, such as glucose. These reports on these systems have found acceptable detection limits and linear ranges. This study aims to encapsulate Cosmos caudatus K. leaf extract in sodium alginate crosslinked with calcium chloride (CaCl2) as coating materials through freeze-drying. Antioxidant assays were performed to evaluate the applicability of the microcapsules as antidiabetic medicines. Their characteristics, antioxidant activity, and encapsulation efficiency (EE) were also examined. Results indicated that the concentration of sodium alginate and pH influenced the manufacturing process of the microcapsules. The highest EE was obtained at pH 6 and alginate concentration of 2% (w/v). The IC50 for the antioxidant activity of the microcapsules was 139.96 ± 1.094 μg/mL. Hydrogen bonding (H-bond) interactions are essential in dictating structures and properties of chemical and biological systems and are pivotal mediators in charge transport reactions and chemical recognition. Accurate description of H-bonds requires thorough characterization of the shared proton’s potential energy surface (PES) and molecular dynamics. A series of quaternary selenides, NaxMGaSe4 (M = Mn, Fe, and mixed Zn/Fe), have been synthesized for the first time employing a high-temperature solid-state synthesis route through stochiometric or polychalcogenide flux reactions. Along with the selenides, a previously reported sulfide analogue, NaxFeGaS4, is also revisited with new findings. These compounds form an interpenetrated structure made up of a supertetrahedral unit. Despite a record number of clinical studies investigating various anti-myeloma treatments, the 5-year survival rate for multiple myeloma (MM) patients in the US is only 55%, and nearly all patients relapse. Poor patient outcomes demonstrate that myeloma cells are "born to survive” which means they can adapt and evolve following treatment. Thus, new therapeutic approaches to combat survival mechanisms and target treatment resistance are required. Importantly, Mcl-1, anti-apoptotic protein, is required for the development of MM and treatment resistance. Enzyme kinetics and inhibition studies are crucial in biochemistry education and research. Conventional methods often require expensive equipment and reagents, potentially limiting their accessibility in limited resource settings. Our approach sought to develop a cost-effective experimental design for studying enzyme kinetics and inhibition. Lactase was chosen as a protein model and its activity was investigated by measuring glucose production from lactose hydrolysis. In the study, commercially available lactase pills were used as an enzyme source, while milk was used as a substrate. Polyvinyl alcohol (PVA)-starch-based bioplastics are widely used in many applications. pH-responsive plastic packaging was produced through the incorporation of senggani (Melastoma malabathricum Linn.) fruit extract into PVA-taro starch-based plastic packaging. The objective of this research was to examine the characteristics of senggani fruit extract under different pH conditions and explore its application as a pH indicator in intelligent packaging. The cofactor of a class A monooxygenase is reduced at an external location of the enzyme and is subsequently pulled back into the active site after the reduction. This observation brings the question; is there any defense mechanism of the active site of a monooxygenase against the formation of the harmful hydrogen peroxide from the reactive C(4a)-(hydro)peroxide intermediate? Injectable hydrogels play an important role in tissue engineering as a filling and repairing material. This study aimed to develop a new injectable hydrogel based on hyaluronic acid (HA) and quince seed gum (QSG) and investigate the effect of QSG on hydrogel performance. The amount of unreacted 1,4-Butanediol diglycidyl ether (BDDE) is maintained at an undetectable level for HA-QSG hydrogels. Amino acid analysis showed that the HA-QSG hydrogel had rich amino acid concentrations of leucine, arginine, and valine. After thermal sterilization, the elastic modulus of HA-QSG gels for dermal and intra-articular filler applications is 63 Pa and 92 Pa respectively. …

Abies is an important genus of the family Pinaceae, with about 50 species found in the highlands of Asia, Europe, North Africa, and North and Central America. The principal aim of the present work was to investigate the chemical content and biological potential of the resin and cone from Abies nordmanniana subsp. bornmulleriana and Abies nordmanniana subsp. equi-trojani, respectively. The flavonoid and phenolic contents of the resin and cones were evaluated using liquid chromatography-high resolution mass spectrometry (LC-HRMS). In this study, we synthesized novel, economically efficient phosphonic acid-functionalized grafted sepiolite nanohybrids for selective elimination of copper ions from water. These nanohybrids were prepared by graft polymerization of glycidyl methacrylate onto sepiolite. We utilized free radical graft polymerization to graft glycidyl methacrylate (GMA) onto silanized sepiolite. The nanohybrids obtained exhibited a grafting percentage of 479% at 0.3 g of KPS initiator, 15% GMA monomer, and after 4 h of reaction. In pursuit of selectively removing metal ions from water, the nanohybrid with the highest grafting (PGE3) was chemically treated with phosphoric acid to introduce phosphonic acid groups on it. …

Rapid Synthesis Of Azoindolizine Derivatives Via Aryldiazonium Salts, Ferruh Lafzi̇

A practical, rapid, and efficient method for the synthesis of azoindolizine derivatives via aryldiazonium salts with excellent yields was reported. Firstly, the corresponding aniline derivatives were synthesized via a simple and rapid method. Then, the optimal reaction conditions were investigated using a variety of protic and aprotic solvents that demonstrating the robustness of the approach. Finally, the applicability of this method to various sources of indolizine and phenyldiazonium tetrafluoroborate salts was expanded.

Chapter 1 introduces computational strategies in computer aided drug design (CADD), focusing on physics-based methodologies to enhance drug discovery. This chapter also discusses the theoretical foundations and practical applications of molecular dynamics (MD) simulations including free energy and stochastic kinetic analysis. The oxygen isotope anomaly (Δ17O = δ17O − 0.52 × δ18O > 0) has proven to be a robust tool for probing photochemical cycling and atmospheric formation pathways of oxidized reactive nitrogen (NOy). Photocatalysis using nanostructured semiconductors for the degradation of organic pollutants has received significant attention as an advanced water treatment method. A photocatalyst absorbs light with energy greater than or equal to its band gap and generates electron-hole pairs. These electrons and holes take part in redox reactions in water to generate reactive oxygen species, such as hydroxyl radicals and superoxide anions, which can break down organic contaminants. However, this method has some challenges, such as electron-hole recombination, which reduces the efficiency, high cost of nanostructure synthesis, and reduced durability of nanostructured photocatalysts in suspension form.
