Controversy exists as to whether the Na-K ATPases of euryhaline fishes are the same enzyme.This study demonstrates that the phosphorylated enzyme from gills of Fundulus heteroclitus adapted to freshwater and salt water migrate to the same position in SDS polyacrylamide gels. Similar behavior of the enzyme from both sources in different ionic media is also shown. Additionally, the possible ability or the enzyme to couple an osmoregulatory influx to an excretory efflux in freshwater fish is demonstrated. The term "ambidirectional" is proposed for this enzyme, and its place in certain membrane models is discussed.

The metabolic secretions from Aspergillus flavus, aflatoxins. are often found as contaminants of agricultural products used for human and animal consumption. Aflatoxin B1 is the most toxic and carcinogenic of the aflatoxins. A reduction in weight gain has been observed when juvenile animals are fed aflatoxin B1 over an extended period of time. This reduction in weight gain may possibly result from an anemic condition induced by an alteration of iron absorption and/or distribution. The effect of chronic sublethal levels of this hepatocarcinogen on weight gain and the uptake and distribution of 59Fe++ were studied in this investigation.

Epidermal melanocytes in mammals are known to occur in a variety of shapes, sizes, and colors. The basic melanocyte morphology is dependent upon genetics, but there are a number of environmental factors that can cause modification.

In the epidermis of PET/\vrnr mice, the population of rnelanocytes reaches a peak and begins to decline during the first post-natal week, and has disappeared within four weeks. Dispersed among the large, highly DOPA-reactive melanocytes are miniature melanocytes. These miniature melanocytes are weakly DOPA-reactive and appear early in the post-natal period. Glycogen content and the activities of phosphorylase (PHRL) and glycogen synthetase (GS) in the hind limb muscle of normal, unilaterally (right) - denervated and in nicotine sulfate (NS) - injected chick embryos were determined to assess the effect of the experimental treatments on glycogen metabolism. Glycogen content increased with increase in age in normal limbs, remained relatively low in denervated limbs, increased with age in left (unoperated) limbs of denervated embryo limbs at a rate lower than normal except between days 13 and 17 of incubation, when glycogen content tripled. Water consumption, and weight loss in response to water deprivation and saline water consumption, were studied in the Eutamias townsendii townsendii from Portland, Multnomah County, Oregon. These chipmunks occupy moist Pacific Northwest forests. The results of these studies suggest that Townsend's chipmunks are dependent on sources of drinking water, at least for normal above ground activity, that their tolerance for water deprivation is comparatively low, and that they are unable to obtain a net gain of water by drinking 0.2 M sodium chloride solutions. Chipmunks cannot drink 0.4 M NaCl solution and survive for more than a few days. Metabolic …

Isolation And Characterization Of Two Bacteriophages Infecting The Myxobacterium Cytophaga Succinicans, Kenneth Wayne Lee

Two bacteriophages which infect the myxobacterium Cytophaga succinicans, strains 14 and 16, were isolated from fish hatchery effluent. Each was submitted to a detailed study. These phages have a limited host range, even within the genus Cytophaga. The turbid plaques formed by these phages are similar in appearance to those produced by coliphage Lambda, and average 0.85 mm in diameter. Both phages have a latent period of 195 minutes and a rise period of an additional 165 minutes. Phage 0CS-16 has a burst size of 137 and phage 0CS-14 has a burst of 45. This thesis was designed to provide a histological and histochemical survey of the small intestine of the mink, Mustela vison. These two objectives were achieved and it was found that the histology and enzyme activity in this animal is in most respects similar to other mammals. Exceptions to this are the absence of a cecum and an ileocecal valve and the absence of lactase activity in the small intestine.

In this study a multivariate statistical analysis utilizes the linear discriminant function that has been used to substantiate the shift of the variety lagocephala (Sharp) Cronq., from Balsamorhiza hirsuta, to B. hookeri (Cronquist, 1955). This method can be used to determine the placement of other questionable taxa.

In addition, this paper deals with the history, morphology and distribution of B. hirsuta, B. hookeri var. hookeri, and B. hookeri var. lagocephala.

In the present study, characteristics of egg masses and egg capsules of the four most abundant California littorinaceans (Lacuna marmorata, L. unifasciata; Littorina planaxis, and L. scutulata) are described. Development within the egg capsule at ambient water temperature is followed to hatching and early weligers are described to facilitate their separation from the plankton.

Holothurin, a biotoxic principle from the Cuvierian glands of the Bahamian sea-cucumber, Actinopyga agassizi, was studied as an anti-tumor agent capable of retarding tumor growth and prolonging the life of tumor-bearing mice. White Swiss mice injected with Sarcoma 180 had a mean survival time of 18.1 days with an average weight gain representing tumor and ascites fluid accumulation amounting to 25.1 grams in 15 days. Of five white Swiss mice with Sarcoma 180 receiving 0.15 mg Holothurin every other day, one was alive at 57 days and the average weight gain of the group was 10.4 grams in 15 …

The present study has attempted to investigate some of the more basic questions concerning the toxicity and accumulation of mercury in the purple shore crab, Hemigrapsus nudus. This rocky intertidal denizen is found in abundance along the entire Pacific coast of the United States from Sitka, Alaska, to the Gulf of California where it dominates the mid-tide pool region. Local specimens were collected just north of the Pacific Marine Stations, Dillon Beach, Marin County, California. The cellular and secretory proteins of the salivary gland of Sciara coprophila during the stages of the larval-pupal transformation were examined by electrophoresis in 0.6 mm sheets of polyacrylamide gel with both SDS-continuous and discontinuous buffer systems. After SDS-electrophoresis, all electrophoretograms of both reduced and nonreduced proteins from single glands stained with Coomassie brilliant blue revealed a pattern containing the same 25 bands during the stages of the larval-pupal transformation. With the staining procedures used in this study, qualitative increases and decreases were detected in existing proteins and enzymes. Recent experiments published in the literature have questioned the importance of the appearance of chromosomal puffs during larval development in Dipterans since Sciara larvae fed the vertebrate hormone cortisone apparently show no puffs without impairment of normal development. We have used Drosophila virilis larvae to test these results because distinct morphological criteria can be used to assess the developmental age of third instar larvae and predict the appearance of a specific puff. The DDT inhibition of (Na+, K+) - activated ATPase in gill preparations of the euryhaline teleost, Fundulus heteroclitus, was measured. The (Na+, K+) -activated ATPase was found to be sensitive to DDT, with 74.6% of the enzyme's specific activity inhibited by 20 ppm DDT. The (Mg++)-activated ATPase was also sensitive to inhibition bv DDT but not to the extent of the (Na+, K+) - activat ed ATPase . Due to previous work which showed that DDT antagonized valinomycin-induced K+ conductance in a synthetic membrane, the effect of K+ on DDT inhibition was examined kinetically. The effects of thyroidectomy, estradiol treatment, and thyroxine treatment on plasma and liver protein concentrations in the water snake, Natrix fasciata fasciata (Linnaeus, 1766), were investigated. There was a significant lowering of liver protein concentrations following thyroidectomy. This deficit could be replaced with thyroxine alone, but could not be replaced by treatment of thyroidectomized snakes with estradiol alone or with a combination of estradiol and thyroxine.

The plasma resolved into nine plasma proteins when electrophoresed in polyacrylamide gels. Although thyroidectomized snakes treated with estradiol only or a combination of estradiol and thyroxine showed enhancement in two protein fractions, the significance …

Urine concentrating ability and kidney structure were investigated in the following insectivorous bats of North America: Myotis lucifugus occultus, Myotis yumanensis, Myotis velifer incautus, Myotis auriculus, Myotis evotis, Myotis thysanodes, Myotis volans, Myotis californicus, Lasionycteris noctivagans, Pipistrellus hesperus, Eptesicus fuscus , Lasiurus cinereus, Euderma maculatum, Plecotus townsendii, Antrozous pallidus and Tadarida brasiliensis. Those species best able to conserve urinary water can survive solute load-water deprivation stresses at higher temperatures and lower water vapor pressures, have relatively longer loops of Henle and tend to live in more arid habitats than those species with relatively poor urine concentrating abilities .

The Role Of The Adrenal Gland In The Survival Of White Rats Subjected To The Parotid Toxin Of The Toad, Bufo Marinus, Mark A. Green

Six groups of white labor atory rats were utilized in this experiment. A sham-operated group and an adrenalectomized group were used as controls. Four other groups were adrenalectomized and given replacement therapy of cortisone, deoxycorticosterone acetate, epinephrine, and norepinephrine. All groups were then given injections of toad toxin from the parotid glands of Bufo marinus. Their heart and breathing rates were recorded for 60 minutes.

All of the untreated adrenalectomized rats showed altered heart and breathing rates and were killed by the toxin. Cortisone and nor ­ epinephrine aided the adrenalectomiz ed rats to survive. but cortisone was the …

The spacing and orientation of sipons of Styela plicata (Lesueur)in swift and slow currents were examined. Within aggre­ crates, the siphons were spaced farther apart in slow currents than in swift currents. The siphons were not horizontally oriented but were vertically oriented in slow currents. The adaptive values of spacing and orientation under different currents are discussed.

A study was undertaken to compare proteins present in the gill membranes of freshwater and salt­ water adapted killifish, Fundulus heteroclitus. For this purpose, a method for the preparation or gill plasma membranes was developed. Polyacrylarnide gel electrophoresis indicated that the two types of gills possessed many membrane proteins in common. However, the freshwater adapted group possessed a single protein not found n the saltwater adapted group and the latter contained a protein not found in the freshwater adapted group.

The initial purpose of this work was to study the rela­tionship between Pantosteus plebeius (Baird and Girard), the Rio Grande mountain-sucker, and Catostomus commersoni (Lacépède ), the common white sucker, in Jemez Creek drainage, Sandoval County, New Mexico. Because insufficient informa­tion on Catostomus commersoni could be obtained, the food and feeding habits of Pantosteus plebeius were studied as an al­ternative.

Mountain-suckers were captured and observed in Jemez Creek drainage. The intestinal contents of 145 specimens were ex­amined in detail. The intestinal contents of other specimens were examined cursorily.

One-hundred and fifty Microtus ochrogaster and one-hundred and fifty-five Peromyscus leucopus were examined for arthropod and helminth parasites during a study from November, 1970 through November, 1971. The mean monthly burdens, per host, of each species of parasite and observations on the seasonal incidence of each species are reported.

Nineteen species of arthropod parasites were found; these include eight species of mites, two of chiggers, one of ticks, two of lice, five of fleas, and one of hots. Infective oncospheres of Oochorlstica osheroffi and Oochoristica scelopori were compared using fifteen histochemical tests. Vital stains were also used.

Carnoy’s-fixed gravid proglottids were vacuum-embedded in a paraffin-piccolyte mixture and cut transversely at 6 microns. Sections were tested for the presence of phosphatases, proteins, sclerotin precursors, nucleic acids, carbohydrates and acid mucopolysaccharides.

Oncospheres were histochemlcally very similar. Heavy concentrations of mucoproteins and glycoproteins were detected in the oncospheral coat of O. scelopori. Acid phosphatase was found in the embryophore of O. osheroffi, but could not be demonstrated in O. scelopori. No evidence of a sclerotized egg capsule was …
