Most multicellular life reproduces sexually, utilizing a single-celled stage that acts as a genetic bottleneck. This bottleneck limits the evolution of selfish cell adaptations by ensuring all cells descend from a single progenitor. We investigated an obligately asexual strain of Schmidtea mediterranea planarians that reproduces by self-bisection and lacks a single-cell bottleneck. Using 2.5 yearlong data on planarian reproductive behavior combined with genotyping, we revealed two previously undescribed genetically distinct substrains within the CIW4 strain. One substrain showed reduced fitness, which correlated with substantial losses of heterozygosity and increased somatic diversity. [Objective ] This study aims to explore the role of Lactobacillus in enhancing host immunocompetence and preventing vaginal infections with Candida albicans.[Method ] A human vaginal epithelial cell (VK2/E6E7 cell) model was employed to screen the most potent strain among ten vaginal Lactobacillus isolates in inducing the antimicrobial peptide LL- 37.A mouse model of vaginal infection with Candida albicans was established.The effects of oral administration of the selected Lactobacillus strain on vaginal mucosal immunity,systemic inflammatory balance,and vaginal microbiota homeostasis were systematically evaluated using hematoxylin-eosin (HE) staining,ELISA,qPCR,and 16S rRNA sequencing.[Result ] Both t he cell lysate and fermentation supernatant of Lacticaseibacillus …

RNA virus infections can transition from an initial acute infection to a prolonged or persistent infection, in which infected cells survive and continue to produce progeny virus. This presents a major clinical problem as persistently infected (PI) cells extend viral shedding, maintain inflammation, and provide potential sources for new viral variants. The RNA virus, Parainfluenza virus 5 (PIV5), with mutations in the P/V gene region (P/V mutant) is a strong inducer of antiviral responses. In 16th century Europe the nearly 2000-year-old discipline of rhetoric, still a pillar of education, underwent significant change. One of the more curious changes occurred in rhetoric’s subdivision known as memory, memoria, that is, memory’s “artificial” cultivation. During the 16th century rhetorical memory was paradoxically both maximized and marginalized as a discipline. And by the early 17th century memoria was all but extinct as a branch of rhetoric. The causal mechanisms of this transformation remain underexamined, particularly by rhetoric and composition scholars, and most explanations remain unclear and uncompelling. This thesis project examines low-cost photogrammetry and 3D walkthrough tours to preserve a collection of artifacts and buildings from Warm Mineral Springs, Florida. The documented objects and structures consist of a small collection of artifacts from Warm Mineral Springs currently held by Florida’s Bureau of Archaeological Research and three buildings designed by noted architect Jack West, a Sarasota School of Architecture member, which suffered extensive damage from a category five hurricane in 2022. Clostridioides difficile is an anaerobic, spore-forming bacterium and one of the leading causes of antibiotic- associated diarrhea. Colonization depends on the metabolism of certain nutrients in the host gut, namely amino acids and carbohydrates. C. difficile ferments amino acids as an integral part of its metabolism via Stickland reactions, utilizing the selenoproteins D-proline reductase (Prd) and glycine reductase (Grd) to generate energy. Specificity of selenocysteine incorporation reflects the intracellular levels of selenophosphate, the activated selenium donor synthesized by selenophosphate synthetase (SelD). By alerting the body’s startle response once a puzzle is completed with a sudden loud noise or visuals, many horror-themed escape rooms utilize the jump scare tactic to heighten their guests’ adrenaline. Neuroinflammation plays a complex and context-dependent role in many neurodegenerative diseases. We identified a key pathogenic function of macrophages in a mouse model of a rare human congenital neuropathy in which SARM1, the central executioner of axon degeneration, is activated by hypomorphic mutations in the axon survival factor NMNAT2. Macrophage depletion blocked and reversed neuropathic phenotypes in this sarmopathy model, revealing SARM1-dependent neuroimmune mechanisms as key drivers of disease pathogenesis. In this study, we investigated the impact of chronic subacute SARM1 activation on the peripheral nerve milieu using single cell/nucleus RNA-sequencing (sc/snRNA-seq).
