As photosynthetic producers, phytoplankton form the foundation of aquaticfood webs. Understanding the relationships among photosynthetic traits inphytoplankton is essential to revealing how diversification of these traitsallows phytoplankton to harvest energy from different light environments. Weinvestigated whether the diversification of 15 species of cryptophytes, a phy-lum of phytoplankton with diverse light-capturing pigments, showed evidenceof trade-offs among photosynthetic performance traits as predicted by the gleaner-opportunist resource exploitation framework. We constructed photo-synthesis versus irradiance (P–E) curves and rapid light curves (RLCs) to estimate parameters characterizing photosynthetic performance and electrontransport rate. We inferred the evolutionary relationships among the 15 specieswith ultraconserved genomic elements and …

Background/aim: Exhausting exercise can damage muscle tissue due to free radical interactions. It is hypothesized that the increase in free radicals following muscle injury, either due to oxidative damage to biomolecules or the activation of inflammatory cytokines, may lead to secondary muscle damage. This study investigated the effects of a novel joint health formula (JHF) containing bisdemethoxycurcumin-enriched curcumin, 3-O-Acetyl-11-keto-beta-boswellic acid-enriched Boswellia (AKBA), and Ashwagandha on exhaustion time, grip strength, antioxidant status, and muscle-signaling proteins in exhaustively exercised rats. Materials and methods: Twenty-eight rats were divided into four groups: Control (C), exercise (E), E + JHF 100 (100 mg/kg), and E …

Characterizing The Causal Role Of Svep1 In Human Disease, Jared S. Elenbaas
Characterizing The Causal Role Of Svep1 In Human Disease, Jared S. Elenbaas
Arts & Sciences Graduate Student Theses and Dissertations
Sushi, von Willebrand factor type A, EGF and pentraxin domain containing 1 (SVEP1) is a poorly understood, putative extracellular matrix protein. Several human traits and diseases are associated with SVEP1, including coronary artery disease, hypertension, type 2 diabetes, glaucoma, dementia, longevity, and platelet reactivity. This dissertation establishes a causal link between SVEP1 and human disease using Mendelian randomization and mouse models. It also provides insights into the disease mechanisms of the protein, including a novel interaction between SVEP1 and the orphan receptor Platelet endothelial aggregation receptor 1. The relevance of this interaction to platelet biology and its potential role in …
Mechanisms Of Electrical Substrate Reprogramming After Cardiac Radiotherapy, David Meng Zhang
Mechanisms Of Electrical Substrate Reprogramming After Cardiac Radiotherapy, David Meng Zhang
Arts & Sciences Graduate Student Theses and Dissertations
Ventricular tachycardia (VT) is frequently associated with morbidity in patients with heart disease and can lead to sudden cardiac death, which accounts for over 350,000 deaths per year and nearly half of all cardiovascular-related deaths in the United States. Despite its status as a major public health concern, arrhythmia management strategies are severely limited in safety and efficacy. Standards-of-care include defibrillators, anti-arrhythmic drugs, and/or ablations. Implantable cardioverter defibrillators improve survival in at-risk populations but severely reduce quality-of-life and increase risks of further-declining ventricular function. Pharmacologic interventions are limited in efficacy and have dozens of off-target effects which are major sources …

This study reports on the first case of the low-cost and environmentally friendly ZnO/SiO2 heterogeneous catalyst from pineapple leaves ash (PLA). Catalyst shows excellent performance in catalyzing the transesterification of waste cooking oil (WCO) with methanol for biodiesel conversion. This study focuses on assessing the influence of Zn content on physicochemical characteristics, using XRD, FTIR, SEM, and N2 adsorption-desorption methods. In addition, three different Zn content levels (20, 25, and 30 %wt) were applied. The results showed that all ZnO/SiO2 samples exhibited characteristics suitable for use as catalyst with an average crystallite size of 31.83-34.15 nm, and a surface area …
Impairment Of The Glial Phagolysosomal System Drives Prion-Like Propagation Of Huntington’S Disease, Graham Davis
Impairment Of The Glial Phagolysosomal System Drives Prion-Like Propagation Of Huntington’S Disease, Graham Davis
Theses and Dissertations
The ability of glia to tightly regulate neuronal health and homeostasis in the CNS is conserved across species. Yet, despite the ability to degrade protein aggregates, glia are vulnerable to the accumulation of neurotoxic amyloid aggregates during neurodegenerative disease progress, and even exacerbate their spread. A developing narrative highlights glia as a double-edged sword in neurodegenerative diseases: initially capable of dynamically responding to amyloid aggregate-ladened dying neurons but also capable of inducing chronic inflammation and creating seeding-competent amyloid oligomers. Thus, uncovering the mechanisms that allow glia to control aggregate deposition while preventing the neurotoxic effects and seed generation is vital …

The implementation of live imaging in reproductive research is crucial for studying the physiological dynamics. Sperm transport is a highly dynamic process regulated by tubular contractions and luminal flows within the male reproductive tract. However, due to the lack of imaging techniques to capture these dynamics in vivo, there is little information on the physiological and biomechanical regulation of sperm transport through the male reproductive tract. Here, we present a functional in vivo imaging approach using optical coherence tomography, enabling live, label-free, depth-resolved, three-dimensional, high-resolution visualization of the mouse testis and epididymis. With this approach, we spatiotemporally captured tubular contractility …

Foraging Rates From Metabarcoding: Predators Have Reduced Functional Responses In Wild, Diverse Prey Communities, Stella F. Uiterwaal, John Delong
School of Biological Sciences: Faculty Publications
Functional responses describe foraging rates across prey densities and underlie many fundamental ecological processes. Most functional response knowledge comes from simplified lab experiments, but we do not know whether these experiments accurately represent foraging in nature. In addition, the difficulty of conducting multispecies functional response experiments means that it is unclear whether interaction strengths are weakened in the presence of multiple prey types. We developed a novel method to estimate wild predators' foraging rates from metabarcoding data and use this method to present functional responses for wild wolf spiders foraging on 27 prey families. These field functional responses were considerably …

Genetic selection has been identified as a promising approach for reducing enteric methane (CH4) emissions; a prerequisite for genetic evaluations; however, these are estimates of the necessary genetic parameters based on a population representative of where the genetic evaluations will be used. The objective of this study was, therefore, to derive genetic parameters for a series of definitions of CH4, carbon dioxide (CO2), and dry matter intake (DMI) as well as genetic correlations between CH4, CO2, and DMI in a bid to address the paucity of studies involving methane emissions …

Stromal Reprogramming By Fak Inhibition Overcomes Radiation Resistance To Allow For Immune Priming And Response To Checkpoint Blockade, Varintra Lander
Arts & Sciences Graduate Student Theses and Dissertations
Pancreatic Ductal Adenocarcinoma (PDAC) is one of the most lethal malignancies. While immune checkpoint blockade (ICB) is effective in many solid cancers, it has not been effective in PDAC. Furthermore, clinical trials combining ICB with standard of care chemo- or radio-therapy (RT), which should be able to prime anti-tumor immunity and unlock ICB, have not been overwhelmingly successful. Thus, understanding the effect of RT on PDAC tumor immunity is critical to plan better combination treatment strategies for this highly recalcitrant cancer. To elucidate if RT can prime antigen specific T cell responses, we first looked at human PDAC resections. In …

Evidence That Aquaporin 11 (Aqp11) In The Spiny Dogfish (Squalus Acanthias) May Represent A Pseudogene, Christopher P. Cutler, Meghan Canicatti, Esosa Omoregie
Biology: Faculty Publications
Various attempts to amplify an AQP11 cDNA from tissues of the spiny dogfish (Squalus acanthias) were made. Two pairs of deoxy-inosine-containing degenerate primers were designed based on conserved amino acid sequences from an AQP11 alignment. These primers yielded some faint bands from gill cDNA that were sequenced. Blast searches with the sequences showed they were not AQP11. An elasmobranch AQP11 nucleotide sequence alignment was produced to identify conserved regions to make further degenerate primers. One primer pair produced a short 148 bp fragment showing particularly strong amplification in gill and intestine. It was sequenced and represented a piece of the …

Pennycress (Thlaspi arvense L.) is an annual cover crop known for its exceptional cold tolerance and high oil and protein yields. Pennycress can be integrated into a corn–soybean rotation in the U.S. However, the utilization of pennycress biomass remains largely unexplored, including assessing compositional changes through its growth and organic matter digestibility. This study harvested pennycress at three growth stages, characterized the biomass for anaerobic digestion (AD), and tested the effects of concurrent alkali pretreatment and ensiling on the biomass methane yield. Results showed that the biomass harvested when the plants were undergoing senescence (“third-harvest”) had higher contents of …

I Spy: Factors Influencing The Observation Of Oxytetracycline In Calcified Structures Of Fishes Viewed Using Standard Light And Fluorescence Microscopy, Meredith B. Pfennig, Derek P. Crane, Nate G. Smith, Dave L. Buckmeier
Biology
The antibiotic oxytetracycline (OTC) is a fluorochrome marker, and fluorescence microscopy is used to view OTC marks in fishes' calcified structures. However, OTC marks have been observed in calcified structures using standard light microscopy for multiple species. Therefore, we conducted an experiment to investigate potential factors (i.e., season, total length of fish, growth rate, and sex) influencing the observation of OTC in calcified structures (otoliths and fin rays or spines) from channel catfish Ictalurus punctatus, gray redhorse Moxostoma congestum, Guadalupe bass Mircopterus treculii, and redbreast sunfish Lepomis auritus viewed using standard light and fluorescence microscopy. OTC stains were not observed …

Honest Signalling Of Age In A Territorial Breeder, Brian A. Hoover, Walter H. Piper
Biology, Chemistry, and Environmental Sciences Faculty Articles and Research
Age often indicates phenotypic quality, ecological experience or social status in animals. Consequently, behavioural signals in territorial animals that accurately convey age to potential mates or rivals might provide useful information for both signaller and receiver about the likelihood of aggression. We tested whether male territorial yodels convey information about age in the common loon, Gavia immer. Using a sample of male loons of known age, we found that the dominant frequency of Fintro3, a dominant syllable in the beginning sequence of a male yodel, increases strongly with age, but we were unable to replicate an earlier finding linking …
Effects Of Hemiparasites In Grassland Restorations Are Not Universal, Anna Scheidel, Victoria Borowicz
Effects Of Hemiparasites In Grassland Restorations Are Not Universal, Anna Scheidel, Victoria Borowicz
Faculty Publications – Biological Sciences
Root hemiparasites infiltrate the vascular tissue of host roots to acquire water and nutrients, which often reduces host growth. Hemiparasites are postulated to be keystone species in grassland communities if they suppress dominant species and increase plant community biodiversity, and ecosystem engineers if they increase nutrient accessibility for surrounding species. We examined keystone effects by evaluating species richness and evenness in 1 m2 plots in a recent prairie restoration where Castilleja sessiliflora was naturally present or absent, and in a longer-established prairie restoration with or without Pedicularis canadensis. We examined ecosystem engineer effects by determining nitrate and phosphate concentrations …

The Effect Of Urbanization And Temperature On Thermal Tolerance, Foraging Performance, And Competition In Cavity-Dwelling Ants, Brooke A. Harris, Dale Stevens, Kaitlyn A. Mathis
Biology
Human disturbance including rapid urbanization and increased temperatures can have profound effects on the ecology of local populations. Eusocial insects, such as ants, have adapted to stressors of increasing temperature and urbanization; however, these evolutionary responses are not consistent among populations across geographic space. Here we asked how urbanization and incubation temperature influence critical thermal maximum (CTmax) and various ecologically relevant behaviors in three ant species in urban and rural locations in Worcester, MA, USA. We did this by incubating colonies of three species of cavity dwelling ant (Aphaenogaster picea, Tapinoma sessile, and Temnothorax longispinosus) from 2 habitat …

Crocodilians are considered to be “ecosystem engineers” because their modification of habitats provides opportunities for feeding, drinking, breeding, and other vital life activities to a wide variety of other animals. One such habitat modification is the construction of nest mounds during the breeding season by most crocodilian species, including American alligators (Alligator mississippiensis). While many reports exist describing wildlife associated with alligator nests, no studies have quantified faunal associates and their corresponding behaviors while visiting nests. To address this data gap, we used automated game cameras to monitor wildlife and their behaviors at alligator nests during the egg incubation period …

The essential role of nicotinamide adenine dinucleotide+ (NAD+) in redox reactions during oxidative respiration is well known, yet the coenzyme and regulator functions of NAD+ in diverse and important processes are still being discovered. Maintaining NAD+ levels through diet is essential for health. In fact, the United States requires supplementation of the NAD+ precursor niacin into the food chain for these reasons. A large body of research also indicates that elevating NAD+ levels is beneficial for numerous conditions, including cancer, cardiovascular health, inflammatory response, and longevity. Consequently, strategies have been created to elevate …

The mechanism of mortality plays a large role in how microorganisms in the open ocean contribute to global energy and nutrient cycling. Salps are ubiquitous pelagic tunicates that are a well-known mortality source for large phototrophic microorganisms in coastal and high-latitude systems, but their impact on the immense populations of smaller prokaryotes in the tropical and subtropical open ocean gyres is not well quantified. We used robustly quantitative techniques to measure salp clearance and enrichment of specific microbial functional groups in the North Pacific Subtropical Gyre, one of the largest ecosystems on Earth. We discovered that salps are a previously …

Testate amoebae (order Arcellinida) are abundant in freshwater ecosystems, including low pH bogs and fens. Within these environments, Arcellinida are considered top predators in microbial food webs and their tests are useful bioindicators of paleoclimatic changes and anthropogenic pollutants. Accurate species identifications and characterizations of diversity are important for studies of paleoclimate, microbial ecology, and environmental change; however, morphological species definitions mask cryptic diversity, which is a common phenomenon among microbial eukaryotes. Lineage-specific primers recently designed to target Arcellinida for amplicon sequencing successfully captured a poorly-described yet diverse fraction of the microbial eukaryotic community. Here, we leveraged the application of …

Gut symbionts influence the physiology and behavior of their host, but the extent to which these effects scale to social behaviors is an emerging area of research. The use of the western honeybee (Apis mellifera) as a model enables researchers to investigate the gut microbiome and behavior at several levels of social organization. Insight into gut microbial effects at the societal level is critical for our understanding of how involved microbial symbionts are in host biology. In this Commentary, we discuss recent findings in honeybee gut microbiome research and synthesize these with knowledge of the physiology and behavior …

Targeting Strategies To Optimize The Therapeutic Potential Of Gold Compounds Against Her2-Positive Breast Cancers, Afruja Ahad
Dissertations, Theses, and Capstone Projects
The overexpression of HER2 accounts for 20-30% of breast cancer tumors and not only serves as a marker for poor predictive clinical outcomes but also as a target for treatment. Antibody-drug conjugates (ADCs) combine the selectivity of monoclonal antibodies (mAbs) with the efficacy of chemotherapeutic drugs to provide targeted treatment without toxicity to normal tissue. Most of the ADCs currently in the clinic for cancer chemotherapy are based on complex organic molecules. In contrast, the conjugation of metallodrugs to mAbs has been overlooked when there is enormous potential in this area with the resurgence of metal-based drugs as prospective cancer …

Modulation of the heart’s immune microenvironment is crucial for recovery after ischemic events such as myocardial infarction (MI). Endothelial cells (ECs) can have immune regulatory functions; however, interactions between ECs and the immune environment in the heart after MI remain poorly understood. We identified an EC-specific IFN responsive and immune regulatory gene signature in adult and pediatric heart failure (HF) tissues. Single-cell transcriptomic analysis of murine hearts subjected to MI uncovered an EC population (IFN-ECs) with immunologic gene signatures similar to those in human HF. IFN-ECs were enriched in regenerative-stage mouse hearts and expressed genes encoding immune responsive transcription factors …

Pesticides are widely used to protect against unwanted insects, but risk assessments of the effects on pollinators have traditionally only been applied on an individual level, not a population level. This has allowed many pesticides to be approved without the knowledge of sub-lethal effects, such as life history characteristics and population structure to name a few. The biopesticide BotaniGard, containing the entomopathogenic fungus Beauveria bassiana, has previously been thought to be safe for non-targeted insects, but recent studies have found it to have significant negative effects on pollinators. Such negative effects can include decrease in longevity and odorant responsiveness with …

In Tanzania, low tomato (Solanum lycopersicum) yield and postharvest marketable fruit qualities remain significant limitations for tomato grower’s profit. Seven endophytic fungal isolates from pyrethrum and Lemongrass leaf and or flowers were evaluated to promote tomato productivity and fruit qualities. The isolates were inoculated by priming the seeds in spore suspension, and the variations in the number of germinated seeds, seedling vigour, yield and fruit qualities were compared across the treatments. It was observed that only three isolates induced more than 50% seed germination. Epf1 and Epl1 isolates showed significant promotion of seed germination (F 0.05 (pdf, 23) = 8.121 …
