Despite a strong literature base relating childhood adversity to pain, the biological mechanisms underlying these associations remain unclear. Theoretical and preliminary empirical evidence supports allostatic load as a potential biological mechanism, though prior studies investigating associations between childhood adversity and elevated allostatic load and/or between elevated allostatic load and poorer pain outcomes have primarily focused on adult populations and individual allostatic load indicators rather than a comprehensive index. Generalized arousal can amplify emotional responses to a range of target stimuli, but it is not yet known whether perceiver characteristics moderate the effects of arousal as target characteristics do. This study investigated the effects of residual arousal from exercise on psychological and physiological reactivity to an active coping task, when individual appraisals of the task differed. Participants who endorsed either a positive or negative attitude toward job interviews underwent either a highly or minimally arousing exercise task, followed by a mock job interview, to which self-reported and cardiovascular responses were assessed. Heart failure (HF) is a chronic, progressive, debilitating illness that disproportionately affects Black individuals, often leading to poor quality of life (QOL). Various sociodemographic, clinical, and psychological characteristics are related to QOL in patients with HF. Because most studies do not include representative Black samples or report findings from racial subgroup analyses, these relationships and any existing racial differences are not well understood. Memory dysfunction is a common symptom of aging, neuropsychiatric disorders, and neurodegenerative disorders, yet truly effective treatments for memory loss do not exist. De novo gene transcription is a molecular requirement for long-term memory formation. The transcription of genes related to synaptic plasticity and learning are regulated in part by histone acetylation, an epigenetic mechanism that regulates chromatin accessibility. Pharmacological compounds that maintain histone acetylation, called histone deacetylase inhibitors (HDACi), enhance memory by preventing deacetylation of core histone proteins, which initiates binding of transcriptional machinery to open chromatin. The purpose of the present study was to summarize and synthesize the research that pertains to the impact of aerobic exercise on adolescents with depression. This review addressed aerobic exercise as a mental health treatment, such as differences between (a) post-intervention and follow-up response and remission rates; (b) exercise in lieu of psychotherapy or exercise as an adjunct to psychotherapy; and (c) response and remission rates amongst minority gender and racial and ethnic groups. Neuropsychological impairments have been observed in both individuals with sleep-disordered breathing and in individuals with sickle cell disease (SCD), but there has been little research on the potential effect of sleep-disordered breathing on neuropsychological function in individuals with SCD. This study aims to examine the effect nocturnal oxygen desaturations have on neuropsychological functioning in individuals with sickle cell disease when compared to those with non-sickle anemia and healthy controls. Sickle cell disease (SCD) is a prevalent genetic disorder involving red blood cells. SCD is a multisystem disease and is connected to various severe medical complications, including debilitating pain. Though pain and inflammation have been connected to adverse childhood experiences (ACEs) in other populations, no prior work has investigated ACEs within a SCD population. The current study examined the prevalence of ACEs as well as the association of ACEs, inflammation, and pain in a sample of youth and young adults with SCD. Episodic memory is a complex process requiring input from several regions of the brain. Coordinated activity in the Dorsal Hippocampus (DH) and medial Prefrontal Cortex (mPFC) is required for episodic memory consolidation. Our laboratory demonstrated that simultaneous subthreshold chemogenetic inactivation of the dorsal hippocampus (DH) and medial prefrontal cortex (mPFC) impairs the consolidation of object placement (OP) and object recognition (OR) memory in female mice (Tuscher et al., 2018), suggesting that these two brain regions work in concert to promote memory consolidation. An activity-based anorexia (ABA) paradigm in adolescent female mice was used to explore whether anorexia affects circuits underlying reward. The ABA paradigm significantly enhanced methamphetamine-induced conditioned place preference (CPP), but not wheel-induced CPP. These results indicate that the ABA paradigm enhances the rewarding properties of methamphetamine, but not wheel running.

While it is well documented that AUD is heritable, the shift from recreational alcohol use to abuse/dependence is poorly understood. In this dissertation, using postmortem brain tissue from individuals with alcohol dependence (AD), we profiled the genome-wide expression of circular RNA (circRNA), microRNA (miRNA), and messenger RNA (mRNA) to better understand the impact of gene expression on the development of AUD. The following study sought to examine the psychological substrates of renewal (e.g.., context dependent extinction processes) for conditioned avoidance behaviors in rats. Using signaled active avoidance conditioning, rats acquired two-way shuttle responding, to two different auditory stimuli. These behaviors were then extinguished through exposure to the auditory stimuli where shuttling behavior was now without consequence. Subjects were then tested for renewal of avoidance in three distinct renewal sequences (e.g., ABA vs ABB, AAB vs AAA, and ABC vs ABB) in three separate groups of rats. Widely effective treatment for alcohol use disorder is not yet available, because the exact biological mechanisms that underlie this disorder are not completely understood. One way to gain a better understanding of these mechanisms is to examine the genetic frameworks that contribute to the risk for developing this disorder. This dissertation examines genetic association data in combination with gene expression networks in the brain to identify functional groups of genes associated with alcohol consumption and dependence.

Introduction. Callous-unemotional (CU) traits predict socially debilitating outcomes including Antisocial Personality Disorder and violent crime in adulthood. Despite significant research, the etiology of CU traits is not well understood. This dissertation incorporates genetic, physiological, neuroanatomical, and self-report measures to investigate the etiology of CU traits. Specifically, this project focuses on measures previously found to associate with impaired fear-processing observed in individuals high on CU. We used a conditioned place preference paradigm to test the rewarding effects of methamphetamine (1mg/kg) in adolescent male and female mice of two strains. We found that methamphetamine induced conditioned place preference in female C57Bl/6 and male 129/SvEv mice, but not female 129/SvEv or male C57Bl/6 mice.

Self-affirmation is the act of focusing on important aspects of the self, such as personal values and characteristic. Benefits of self-affirmation have been documented in past research. However, the immediate impacts of self-affirmation on cardiovascular responses have not been fully explored. Risky alcohol use is a major health concern among college students, with 40.1% reporting binge drinking (5 or more drinks in one occasion) and 14.4% reporting heavy drinking (binge drinking on 5 or more occasions) in the past month. Risky alcohol use is thought to be the result of a complex interplay between genes, biological processes, and other phenotypic characteristics. Understanding this complex relationship is further complicated by known phenotypic heterogeneity in the development of alcohol use. Uncertainty is often associated with subjective distress and a potentiated anxiety response. Occurrence uncertainty (OU), or the inability to predict if a threat will occur, has never been compared experimentally with temporal uncertainty (TU), or the inability to predict when a threat will occur. The current study aimed to 1) directly compare the eye-blink startle responsivity of OU and TU, 2) develop a more effective task for isolating uncertain anticipation, and 3) better understand the relationship between individual differences in the intolerance of uncertainty and uncertain anticipation startle responsivity. Despite widespread misuse of inhalants, there are limited preclinical methods available to study the reinforcement-like properties of inhalants. Pavlovian fear conditioning provides a useful model system for investigating the mechanisms underlying associative learning. In recent years, there has been an increasing interest in "trace" fear conditioning, which requires conscious awareness of the contingency of CS and US therefore considered as a rodent model of explicit fear.
